Fresh fruit are highly perishable commodities, facing significant postharvest losses primarily due to physiological deterioration and microbial spoilage. Conventional preservation methods often face limitations regarding safety, residue, and environmental impact. Because of its rapid decomposition and low-residue-impact characteristics, ozone has proven superior as an efficient and eco-friendly solution for preserving fruit quality after harvest. The maturation and aging processes of kiwifruit are closely linked to the involvement of reactive oxygen species (ROS) metabolism. This study aimed to investigate the effects of intermittent ozone treatment (21.4 mg/m3, applied for 0, 1, 3, or 5 h weekly) on ROS metabolism, the antioxidant defense system, and storage quality of kiwifruit during cold storage (0.0 ± 0.5 °C). The results showed ozone treatment slowed the decline in titratable acid (TA) content and fruit firmness, inhibited increases in total soluble solids (TSSs) and weight loss, and maintained the storage quality. Additionally, ozone treatment enhanced the activities of antioxidant-related enzymes. This includes superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and ascorbate peroxidase (APX). Furthermore, it delayed the reduction in ascorbate (ASA), glutathione (GSH), total phenolic compounds, and flavonoid content, while also preventing the accumulation of ROS and the rise in malondialdehyde (MDA) levels. In summary, the results indicate that ozone treatment enhances the antioxidant capacity of kiwifruit by increasing the structural integrity of cell membranes, preserving the structural integrity of cell membranes, and effectively maintaining the storage quality of the fruit. In an increasingly challenging agricultural environment, the identification of novel tools for protecting crops from stress agents while securing marketable production is a key objective. Here we investigated the effects of three previously characterized Prosystemin-derived functional peptide fragments as protective agents against salt stress and as biostimulants modulating tomato yield and quality traits. The treatments of tomato plants with femtomolar amounts of the peptides alleviated salt stress symptoms, likely due to an increase in root biomass up to 18% and the upregulation of key antioxidant genes such as APX2 and HSP90. In addition, the peptides exhibited biostimulant activity, significantly improving root area (up to 10%) and shoot growth (up to 9%). We validated such activities through two-year field trials carried out on industrial tomato crops. Peptide treatments confirmed their biostimulant effects, leading to a nearly 50% increase in marketable production compared to a commonly used commercial product and consistently enhancing fruit °Brix values. This study aimed to investigate the efficacy of a composite coating composed of 150 mg/L ε-Poly-L-lysine (ε-PL) and 5 g/L chitosan (CTS) in extending the shelf life and maintaining the postharvest quality of fresh Tremella fuciformis. Freshly harvested T. fuciformis were treated by surface spraying, with distilled water serving as the control. The effects of the coating on storage quality, physicochemical properties, reactive oxygen species (ROS) metabolism, and membrane lipid metabolism were evaluated during storage at (25 ± 1) °C. The results showed that the ε-PL/CTS composite coating significantly retarded quality deterioration, as evidenced by reduced weight loss, maintained whiteness and color, and higher retention of soluble sugars, soluble solids, and soluble proteins. The coating also effectively limited water migration and loss. Mechanistically, the coated T. fuciformis exhibited enhanced antioxidant capacity, characterized by increased superoxide anion (O2) resistance capacity, higher activities of antioxidant enzymes (SOD, CAT, APX), and elevated levels of non-enzymatic antioxidants (AsA, GSH). This led to a significant reduction in malondialdehyde (MDA) accumulation, alongside improved DPPH radical scavenging activity and reducing power. Furthermore, the ε-PL/CTS coating preserved cell membrane integrity by inhibiting the activities of lipid-degrading enzymes (lipase, LOX, PLD), maintaining higher levels of key phospholipids (phosphatidylinositol and phosphatidylcholine), delaying phosphatidic acid accumulation, and consequently reducing cell membrane permeability. In conclusion, the ε-PL/CTS composite coating effectively extends the shelf life and maintains the quality of postharvest T. fuciformis by modulating ROS metabolism and preserving membrane lipid homeostasis. This study provides a theoretical basis and a practical approach for the quality control of fresh T. fuciformis. Full article

Soil salinization severely limits plant growth and productivity. Mulberry (Morus alba L.), an economically and ecologically important tree, is widely cultivated, yet its salt-tolerance mechanisms at the seedling stage remain insufficiently understood. This study investigated the physiological and biochemical responses of two-year-old mulberry (‘Tailai Sang’) seedlings subjected to six NaCl treatments (0, 50, 100, 150, 200, and 300 mmol L−1) for 28 days. Results showed that growth parameters and photosynthetic gas exchange exhibited dose-dependent declines. The reduction in net photosynthetic rate (Pn) was attributed to both stomatal limitations (decreased stomatal conductance) and non-stomatal limitations, as evidenced by a significant decrease in the maximum quantum efficiency of photosystem II (Fv/Fm) under high salinity. To cope with osmotic stress, seedlings accumulated compatible solutes, including soluble sugars, proteins, and proline. Critically, mulberry seedlings demonstrated effective ion homeostasis by sequestering Na+ in the roots to maintain a high K+/Na+ ratio in leaves, a mechanism that was compromised above 150 mmol L−1. Concurrently, indicators of oxidative stress—malondialdehyde (MDA) and H2O2—rose significantly with salinity, inducing the activities of antioxidant enzymes (SOD, CAT, APX, and GR), which peaked at 150 mmol L−1 before declining under extreme stress. A biomass-based LC50 of 179 mmol L−1 NaCl was determined. These findings elucidate that mulberry salt tolerance is a coordinated process involving three key mechanisms: osmotic adjustment, selective ion distribution, and a robust antioxidant defense system. This study establishes an indicative tolerance threshold under controlled conditions and provides a physiological basis for further field-based evaluations of ‘Tailai Sang’ mulberry for cultivation on saline soils. The transition from conchocelis to conchosporangia in Pyropia haitanensis represents a pivotal stage in its life cycle. As a commercially vital red alga, P. haitanensis plays a dominant role in global nori production. The transition governing its sporulation efficiency is pivotal for aquaculture success, yet the underlying regulatory mechanisms, especially their integration with metabolic cues such as polyamines, remain poorly understood. This study uncovered a critical role for the polyamine spermine (SPM) in promoting conchosporangial formation, mediated through the signaling activity of superoxide anions (O2·). Treatment with SPM markedly elevated O2· levels, an effect that was effectively inhibited by the NADPH oxidase inhibitor diphenyliodonium chloride (DPI), underscoring the role of O2· as a key signaling molecule. Transcriptomic analysis revealed that SPM enhanced photosynthesis, carbon assimilation, and respiratory metabolism, while simultaneously activating antioxidant enzymes, such as superoxide dismutase (SOD), ascorbate peroxidase (APX), and catalase (CAT), to regulate hydrogen peroxide (H2O2) levels and maintain redox homeostasis. Furthermore, SPM upregulated genes associated with photosynthetic carbon fixation and the C2 oxidative photorespiration pathway, supplying the energy and metabolic resources necessary for this developmental transition. These findings suggested that SPM orchestrated O2· signaling, photosynthetic activity, and antioxidant defenses to facilitate the transition from conchocelis to conchosporangia in P. haitanensis. The functional role of MAPKK genes in potato (Solanum tuberosum L.) under high-temperature stress remains unexplored, despite their critical importance in stress signaling and yield protection. We characterized StMAPKK1, a novel group D MAPKK localized to plasma membrane/cytoplasm. Quantitative real-time polymerase chain reaction (qRT-PCR) revealed cultivar-specific upregulation in potato (‘Atlantic’ and ‘Desiree’) leaves under heat stress (25 °C, 30 °C, and 35 °C). Transgenic lines overexpressing (OE) StMAPKK1 exhibited elevated antioxidant enzyme activity, including ascorbate peroxidase (APX), catalase (CAT), superoxide dismutase (SOD), and peroxidase (POD), mitigating oxidative damage. Increased proline and chlorophyll accumulation and reduced oxidative stress markers, hydrogen peroxide (H2O2) and malondialdehyde (MDA), indicate improved cellular redox homeostasis. The upregulation of key antioxidant and heat stress-responsive genes (StAPX, StCAT1/2, StPOD12/47, StFeSOD2/3, StMnSOD, StCuZnSOD1/2, StHSFA3 and StHSP20/70/90) strengthened the enzymatic defense system, enhanced thermotolerance, and improved photosynthetic efficiency, with significant improvements in net photosynthetic rate (Pn), transpiration rate (E), and stomatal conductance (Gs) under heat stress (35 °C) in StMAPKK1-OE plants. Superior growth and biomass (plant height, plant and its root fresh and dry weights, and tuber yield) accumulation, confirming the positive role of StMAPKK1 in thermotolerance. Conversely, RNA interference (RNAi)-mediated suppression of StMAPKK1 led to a reduction in enzymatic activity, proline content, and chlorophyll levels, exacerbating oxidative stress. Downregulation of antioxidant-related genes impaired ROS scavenging capacity and declines in photosynthetic efficiency, growth, and biomass, accompanied by elevated H2O2 and MDA accumulation, highlighting the essential role of StMAPKK1 in heat stress adaptation. These findings highlight StMAPKK1’s potential as a key genetic target for breeding heat-tolerant potato varieties, offering a foundation for improving crop resilience in warming climates. Air pollution acts as a pervasive oxidative stressor, disrupting global crop production and ecosystem health through the overproduction of reactive oxygen species (ROS). Hazardous pollutants impair critical physiological processes—photosynthesis, respiration, and nutrient uptake—triggering oxidative damage and yield losses. This review synthesizes current knowledge on plant defense mechanisms, emphasizing the integration of enzymatic (SOD, POD, CAT, APX, GPX, GR) and non-enzymatic (polyphenols, glutathione, ascorbate, phytochelatins) antioxidant systems to scavenge ROS and maintain redox homeostasis. We highlight the pivotal roles of transcription factors (MYB, WRKY, NAC) in orchestrating stress-responsive gene networks, alongside MAPK and phytohormone signaling (salicylic acid, jasmonic acid, ethylene), in mitigating oxidative stress. Secondary metabolites (flavonoids, lignin, terpenoids) are examined as biochemical shields against ROS and pollutant toxicity, with evidence from transcriptomic and metabolomic studies revealing their biosynthetic regulation. Furthermore, we explore biotechnological strategies to enhance antioxidant capacity, including overexpression of ROS-scavenging genes (e.g., TaCAT3) and engineering of phenolic pathways. By addressing gaps in understanding combined stress responses, this review provides a roadmap for developing resilient crops through antioxidant-focused interventions, ensuring sustainability in polluted environments. The enhancement of flavonoid content and antioxidant capacity in plants remains a significant area of focus in the investigation of plant-derived functional foods. This study systematically investigated the impact of exogenous zinc sulfate (5 mM ZnSO4) stress on flavonoid content and antioxidant capacity in finger millet (Eleusine coracana L.) sprouts, along with its underlying molecular mechanisms. The results demonstrated that treatment with 5 mM ZnSO4 significantly increased the flavonoid content in sprouts, reaching a maximum value of 5.59 μg/sprout on the 6th day of germination. ZnSO4 stress significantly enhanced the activities of PAL, 4CL, and C4H, while also considerably upregulating the expression levels of flavonoid-biosynthesis-related genes. Physiological indicators revealed that ZnSO4 stress increased the contents of malondialdehyde, hydrogen peroxide, and superoxide anion in the sprouts, while inhibiting sprout growth. As a stress response, ZnSO4 stress enhances the antioxidant system by increasing antioxidant capacity (ABTS, DPPH, and FRAP), antioxidant enzyme activity (POD and SOD), and related gene expression (POD, CAT, and APX) in sprouts. This study provides experimental evidence for ZnSO4 stress to improve flavonoid accumulation and antioxidant capacity in finger millet sprouts and provides important theoretical and practical guidance for the development of high-quality functional foods. Drought is a serious environmental problem that limits the yield of wheat around the world. Using biochemical and microscopy methods, it was shown that methyl jasmonate (MeJA) has the ability to induce the oxidative stress tolerance in roots of wheat plants due to the regulation of antioxidant enzymes activity, proline (Pro), and wheat germ agglutinin (WGA) accumulation. During the first hours of 12% polyethylene glycol (PEG) exposure, stress increased the superoxide radical (O2•−) and the hydrogen peroxide (H2O2) accumulation, the activity of superoxide dismutase (SOD), total peroxidase (POD), ascorbate peroxidase (APX), catalase (CAT), the percent of dead cells (PDC), malondialdehyde accumulation (MDA), and electrolyte leakage (EL) of wheat roots as compared to the control. Stress enhanced proline (Pro) and wheat germ agglutinin (WGA) contents in roots and the plant’s nutrient medium, as well as decreased the mitotic index (MI) of cells of root tips in comparison to the control. During PEG exposure, 10−7 M MeJA pretreatment increased the parameter of MI, declined O2•− and H2O2 generation, PDC, MDA, and EL parameters as compared to MeJA-untreated stressed seedlings. During 1 day of drought, MeJA pretreatment additionally increased the activity of SOD, total POD, APX, CAT, Pro, and WGA accumulation in wheat roots in comparison to MeJA-untreated stressed plants. During stress, MeJA pretreatment caused a decrease in Pro exudation into the growth medium, while WGA content in the medium was at the control level. Full article

Idesia polycarpa is a valuable woody oil plant with potential for horticultural and industrial applications. However, limited information is available regarding its drought tolerance during the seedling stage. In this study, one-year-old seedlings were subjected to five treatments based on soil relative water content (RWC): moderate drought (T1, 40 ± 5%), severe drought (T2, 20 ± 5%), control (CK, 70 ± 5%), and rewatering following moderate (T3) and severe drought stress (T4), with RWC restored to 70 ± 5%. Under drought stress, seedlings exhibited adaptive responses including reduced growth, enhanced antioxidant enzyme activity, osmotic regulation, and changes in endogenous hormone levels. Seedlings showed good tolerance and recovery under moderate drought, but severe drought caused substantial damage and limited post-rewatering recovery. Pearson correlation and principal component analyses revealed that betaine, APX, SA, IAA, ABA, chlorophyll (a + b) content, and crown growth were strongly associated with drought response and could serve as key indicators for drought resistance assessment in I. polycarpa. These findings provide insights into the physiological mechanisms of drought adaptation and support the development of a reliable evaluation system for drought tolerance in this promising species. Climate change has intensified drought stress, threatening global food security by affecting sensitive crops like maize (Zea mays). This study evaluated the potential of Antarctic fungal endophytes (Penicillium chrysogenum and P. brevicompactum) to enhance maize drought tolerance under field conditions with different irrigation regimes. Drought stress reduced soil moisture to 59% of field capacity. UAV-based multispectral imagery monitored plant physiological status using vegetation indices (NDVI, NDRE, SIPI, GNDVI). Inoculated plants showed up to two-fold higher index values under drought, indicating improved stress resilience. Physiological analysis revealed increased photochemical efficiency (0.775), higher chlorophyll and carotenoid contents (45.54 mg/mL), and nearly 80% lower lipid peroxidation in inoculated plants. Lower proline accumulation suggested better water status and reduced osmotic stress. Secondary metabolites such as phenolics, flavonoids, and anthocyanins were elevated, particularly under well-watered conditions. Antioxidant enzyme activity shifted: SOD, CAT, and APX were suppressed, while POD activity increased, indicating reprogrammed oxidative stress responses. Yield components, including cob weight and length, improved significantly with inoculation under drought. These findings demonstrate the potential of Antarctic endophytes to enhance drought resilience in maize and underscore the value of integrating microbial biotechnology with UAV-based remote sensing for sustainable crop management under climate-induced water scarcity. Fusarium wilt, caused by Fusarium solani, is a devastating disease that leads to significant losses in ginger (Zingiber officinale) crops worldwide. To explore the molecular mechanisms underlying F. solani infection and disease progression, we performed a comparative transcriptome analysis of ginger rhizomes during storage, comparing inoculated and non-inoculated samples. A total of 647 and 6398 DEGs were identified in the 1.5- and 2-day infection groups, respectively. KEGG analysis revealed that most DEGs were enriched in the plant–pathogen interaction pathway, with both PTI and ETI being activated. Six DEGs in this pathway were validated by qRT-PCR at two time points, showing a strong correlation with FPKM values from the transcriptome data. Furthermore, transient expression analysis in Nicotiana benthamiana leaves demonstrated that overexpressing ZoCEBiP1 helped scavenge excess ROS, thereby reducing disease severity. Transcriptional profiling of DEGs in the plant–pathogen interaction pathway revealed significant changes in genes involved in ROS and NO metabolism. In F. solani-infected ginger rhizomes, levels of H2O2 and O2 were elevated, along with increased activities of antioxidant enzymes (POD, CAT, SOD, and APX) and higher NO content and NOS activity. These findings elucidated the early defense response of ginger rhizomes to F. solani infection and provided insights for developing effective strategies to manage fungal diseases. Full article

Harnessing a Microbial Consortium and Compost to Control Grapevine Pathogens: A Sustainable Viticulture Strategy for Disease Suppression and Quality Enhancement

Beneficial microorganisms are emerging as promising alternatives to conventional pesticides for the biological control of plant diseases. This study evaluated the efficacy of a consortium composed of Pseudomonas yamanorum and Trichoderma longibrachiatum and compost against three grapevine pathogens, Botrytis cinerea, Erysiphe necator, and Plasmopara viticola, in three cultivars: Victoria, Superior Seedless, and Early Sweet. The microbial consortium (P. yamanorum + T. longibrachiatum) combined with compost (treatment T4) significantly outperformed the individual treatments, reducing disease severity indices (DSIs) to 7.72, 5.35, and 3.37% in Victoria; 5.70, 6.95, and 3.32% in Superior Seedless; and 4.98, 2.35, and 2.84% in Early Sweet. The treatment also enhanced physiological traits, such as the chlorophyll content, and defense responses, including ascorbate peroxidase (APX), peroxidase (POX), and catalase (CAT) enzyme activities. Biochemical markers, including the total protein content, phenolic content, and reduced malondialdehyde (MDA) levels, indicated an improved oxidative stress tolerance. The soil analysis confirmed an increased pH, organic matter, nitrogen content, and microbial biomass. T4 further reduced the fruit disease incidence and improved quality attributes, including the sugar content and size, while lowering nitrate accumulation. These findings highlight the synergistic benefits of combining a microbial consortium with compost as a sustainable strategy to promote grapevine health, productivity, and soil resilience. Investigating the Mechanisms Underlying Citral-Induced Oxidative Stress and Its Contribution to Antifungal Efficacy on Magnaporthe oryzae Through a Multi-Omics Approach

Citral, an organic compound found in lemongrass (Cymbopogon citratus) oil and Litsea cubeba essential oil, has been reported to exhibit notable antifungal activity against Magnaporthe oryzae (M. oryzae), the pathogen of rice blast, which causes significant economic losses in rice production. However, the role of citral in inducing oxidative stress related to antifungal ability and its underlying regulatory networks in M. oryzae remain unclear. In this study, we investigated the oxidative effects of citral on M. oryzae and conducted transcriptomic and widely targeted metabolomic (WTM) analyses on the mycelia. The results showed that citral induced superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) activities but reduced glutathione S-transferase (GST) activity with 25% maximal effective concentration (EC25) and 75% maximal effective concentration (EC75). Importantly, citral at EC75 reduced the activities of mitochondrial respiratory chain complex I, complex III and ATP content, while increasing the activity of mitochondrial respiratory chain complex II. In addition, citral triggered a burst of reactive oxygen species (ROS) and a loss of mitochondrial membrane potential (MMP) through the observation of fluorescence. Furthermore, RNA-seq analysis and metabolomics analysis identified a total of 466 differentially expression genes (DEGs) and 32 differential metabolites (DAMs) after the mycelia were treated with citral. The following multi-omics analysis revealed that the metabolic pathways centered on AsA, GSH and melatonin were obviously suppressed by citral, indicating a disrupted redox equilibrium in the cell. These findings provide further evidences supporting the antifungal activity of citral and offer new insights into the response of M. oryzae under oxidative stress induced by citral. Full article

Asparagus (Asparagus officinalis L.) is a highly nutritious vegetable rich in various bioactive compounds. Ensuring both yield improvement and quality preservation is a shared goal for producers and researchers. As novel green yield-enhancing technologies in facility agriculture, electrostatic fields and elevated CO2 application hold significant potential. This study investigated the effects of the interaction between electrostatic fields and elevated CO2 on the growth and physiological characteristics of asparagus. The results demonstrated that the combined treatment of electrostatic fields and elevated CO2 significantly increased total yield, tender stem number, and single tender stem weight of asparagus, while also shortening the harvesting period and promoting rapid shoot growth. Additionally, the treatment markedly enhanced the total chlorophyll content in asparagus leaves, improving photosynthetic capacity. By boosting antioxidant enzyme activities (e.g., SOD, APX) and reducing malondialdehyde (MDA) levels, the treatment maintained the redox homeostasis of asparagus shoots, effectively mitigating oxidative damage. In terms of nutrient accumulation, the interaction between electrostatic fields and elevated CO2 significantly promoted the synthesis and accumulation of key nutrients, including soluble sugars, reducing sugars, soluble proteins, total phenolics, total flavonoids, and ascorbic acid, thereby substantially improving the nutritional quality of asparagus. Comprehensive analysis using fuzzy membership functions revealed that the combined treatment of electrostatic fields and elevated CO2 outperformed individual treatments in enhancing asparagus growth and physiological characteristics. This study provides important theoretical insights and technical support for the efficient and sustainable cultivation of asparagus in facility agriculture. Full article
