Some Cretaceous ants belonging to the stem group of Formicidae exhibit bizarre morphology. This wide range of unusual adaptive features is primarily related to the mouthparts and clypeus. The researchers were perplexed by their specific ecology, as modern ant lineages do not exhibit anything similar. Here, we report and describe a new genus based on an extraordinary and mysterious alate ant from Late Eocene Baltic amber. Undoubtedly, the new ant is classified within the subfamily Formicinae (one of the crown groups), yet it displays a highly specialised morphology and an unusual array of features that are not observed in any extant ant lineages. Neither recent nor extinct ants have such a combination of features. While the exact phylogenetic placement of the new ant remains uncertain, we offer a discussion of its potential affinities based on our constrained phylogenetic analyses. We propose that †Eridanomyrma gen. n. should be considered in the new tribe †Eridanomyrmini trib. n. This new taxon highlights the adaptive diversity of a highly specialised, extinct lineage of Eocene crown-group ants. We also present a 3D model based on X-ray computed microtomography (µCT). To explore the potential seed compounds from natural products as anticancer agents against small-cell lung cancer (SCLC), the underground parts of Agapanthus africanus, a plant commonly used for ornamental purposes, were investigated. Three spirostan-type steroidal glycosides (13) were isolated and identified by nuclear magnetic resonance spectral analysis. Compounds 13 exhibited cytotoxicity against SBC-3 human SCLC cells, with IC50 values of 0.56, 1.4, and 7.4 µM, respectively. Compound 1, also known an agapanthussaponin A, demonstrated the most potent cytotoxicity among the isolated compounds and was evaluated for its apoptosis- and ferroptosis-inducing activities. Compound 1 arrested the cell cycle of SBC-3 cells in the G2/M phase and induced apoptosis primarily via the mitochondrial pathway, characterized by caspases-3 and -9 activation, loss of mitochondrial membrane potential, and overproduction of reactive oxygen species. Additionally, 1 triggered ferroptosis via a dual mechanism consisting of enhanced cellular iron uptake through upregulation of transferrin and transferrin receptor 1 expression and impaired glutathione synthesis via downregulation of both xCT and glutathione peroxidase 4 expression. Compound 1 induces cell death via the apoptosis and ferroptosis pathways, suggesting its promise as a seed compound for the development of anticancer therapeutics against SCLC. Full article

Background and Aim: Syphilis, caused by Treponema pallidum, classically presents with genital or anal chancres; rectal involvement is rare and frequently misdiagnosed as inflammatory bowel disease or malignancy. We describe an unusually severe case of syphilitic proctitis in the setting of advanced HIV-related immunosuppression (CD4 39 cells/µL), in which targeted immunophenotyping (ERG and CD38) was a valuable adjunctive tool in the differential diagnosis. Case Presentation: A 46-year-old man with a recent history of erosive gastritis and esophageal candidiasis presented after six months of unintentional 20 kg weight loss, profound fatigue, intermittent fevers, profuse diarrhea, and two episodes of hematemesis. Workup revealed a new diagnosis of HIV infection (CD4: 39 cells/µL; viral load: 87,837 copies/mL). Contrast-enhanced CT demonstrated uniform, concentric rectal wall thickening (“target sign”). Colonoscopic biopsy showed exuberant granulation tissue and dense plasma cell infiltrates. Immunohistochemistry revealed a dense infiltrate of CD38-positive plasma cells and ERG-positive endothelial proliferation. These findings, in the context of positive serology, were highly supportive of a spirochetal etiology and helped differentiate it from potential mimics. Serology was positive for latent late syphilis (VDRL 1:64). The patient received three weekly doses of intramuscular benzathine penicillin; lumbar puncture excluded neurosyphilis. Discussion: This is among the first reported cases of syphilitic proctitis in a patient with CD4 < 50 cells/µL, where advanced immunophenotyping differentiated syphilitic inflammation from neoplastic or inflammatory mimics. Profound immunosuppression accelerates disease progression and yields atypical clinical features. Conclusion: In HIV-infected patients with chronic rectal symptoms, especially those with CD4 < 50 cells/µL, syphilitic proctitis must be considered. Integration of radiologic assessment, histopathology with ERG/CD38 staining, and serologic testing permits prompt diagnosis. Early benzathine penicillin therapy and rigorous clinical and serologic follow-up are essential to prevent complications, including neurosyphilis. Full article

MgO–C refractory materials were developed by incorporating different ratios of alumina/titania nano-additives which were synthesized chemically. Their physical and mechanical properties, oxidation resistance, slag wettability, bulk density, apparent porosity, cold crushing strength, oxidation index, and closed porosity were tested, evaluated, and compared using conventional techniques as well as X-ray micro-computed tomography (µCT). This investigation indicated a slight degradation of physical properties and mechanical strengthening which was stronger for samples with increased alumina content. Oxidation and corrosion extent were tested both with X-ray tomography and conventional methods. The first method allowed for the calculation of the oxidation index, the detection of closed porosity, and an improved analysis of the internal corrosion, avoiding the sectioning of the materials. This result confirms the supremacy of the first technique. On the contrary, although conventional methods such as the Archimedes procedure cannot detect close porosity, they provide more accurate measurements of the physical properties of refractories. This study shows that conventional methods exhibit superiority in investigations of the pore structures of refractories for pore sizes in the range 1–2 μm, while the use of the μCT system is limited for pore sizes equal to or larger than 20 μm. Comparative investigations on homogenous cranial series have demonstrated that metopism is linked to a specific configuration of the cranial vault; however, there are no comparative data concerning the endocranial morphology in this condition. This study aimed to compare the endocranial space in metopic and control crania using morphometric analysis and machine learning algorithms. For this purpose, a series of 230 (184 control and 46 metopic) dry crania of contemporary adult Bulgarian males were scanned using an industrial µCT system. The 3D coordinates of 47 landmarks were collected on the endocranial surface. All possible measurements between the landmarks were calculated as Euclidean distances. The resultant 1081 measurements represented the initial dataset, which was reduced to smaller datasets applying different criteria. The derived datasets were used for learning a set of classification models by machine learning algorithms. The morphometric analysis showed that in the metopic crania some segments of the anterior and middle cranial fossae were significantly longer, and the landmark endobregma was significantly closer to the anterior and middle sections of the cranial base. The most accurate model, with a classification accuracy of 85%, was the Naive Bayes one learned on a dataset of 69 attributes assembled after an attribute selection procedure. Full article

Background/Objectives: Air embolism on contrast-enhanced computed tomography (CECT) scans may have significant consequences, particularly if a right-to-left shunt is present, as seen in hereditary hemorrhagic telangiectasia. We sought to evaluate the frequency of CECT-associated air emboli in a single tertiary care referral center. Methods: Consecutive non-enhanced and contrast-enhanced cardiac CT studies (NECCT and CECCT, respectively) were evaluated prospectively over a 6-month period. Following the University of Alberta’s Health Research Ethics Board approval (code: Pro00042313; date: 1 May 2014), two experts reviewed all studies independently to assess for the presence and location of air emboli. The control group consisted of only NECCTs. All patients, except for the control group in this study, had an IV cannula placed. When present, the number, volume, and location of air emboli were recorded. Results: In this study, 110 subjects underwent intravenous cannula placement and both NECCT and CECCT. Of these, 27 of the NECCT studies (24.5%) and 36 of the CECCT studies (32.7%) demonstrated intravascular air emboli. Of those with air emboli, the average volume of intravascular gas was 19.22 ± 25.35 µL in the NECCT studies, with most of the intravascular air (70.4%) seen in the right atrial appendage (RAA). The average volume of intravascular air was 14.81 ± 26.54 µL in the CECCT studies, with most of the intravascular air also located within the RAA (72.2%). The incidence of intravascular air was higher in the CECCT group (28.6% increase), with lower volumes of intravascular air. None of the subjects in the control group (n = 28), who underwent NECCT without intravenous cannulation, demonstrated air emboli. Conclusions: Air emboli were present in a significant proportion of subjects undergoing intravenous cannulation and subsequent CECT. The use of CECT should be carefully considered in high-risk populations. Full article

Electret filter media are electrostatically charged during the manufacturing process to activate effective electrical separation mechanisms. In order to investigate the influence of humidity on these mechanisms, the electric field, and filtration efficiency, a Direct Numerical Simulation (DNS) study of the aerosol deposition within wetted fibrous nonwoven filter media used in masks was carried out. Initial experimental investigations determined key properties of the filter material, including porosity, fiber diameter, and surface charge density. Using Micro-Computed Tomography (µCT), preferred locations for droplet deposition within the filter were identified. Additional experiments quantified the amount of water absorbed by the filter medium and assessed its impact on the existing electric field. Numerical simulations examined various models with differing porosity and fiber diameter, incorporating different levels of water content to analyze the changes in the electric field, flow velocity, and resulting filtration efficiency. The results provide valuable insights into the significant effects of fiber change on filtration performance, demonstrating the electret filter’s ability to partially compensate for the negative impacts of water. The increasing number of acetabular revision total hip arthroplasties requires the evaluation of alternative materials in addition to established standards using a defined animal experimental defect that replicates the human acetabular revision situation as closely as possible. Defined bone defects in the load-bearing area of the acetabulum were augmented with various materials in an ovine periacetabular defect model (Group 1: NanoBone® (artificial hydroxyapatite-silicate composite; Artoss GmbH, Germany); Group 2: autologous sheep cancellous bone; Group 3: Tutoplast® (processed allogeneic sheep cancellous bone; Tutogen Medical GmbH, Germany)) and bridged with an acetabular reinforcement ring of the Ganz type. Eight months after implantation, a μ-CT examination (n = 8 animals per group) was performed. A μ-CT analysis of the contralateral acetabula (n = 8, randomly selected from all three groups) served as the control group. In a defined volume of interest (VOI), bone volume (BV), mineral volume (MV), and bone substitute volume (BSV), as well as the bone surface (BS) relative to the total volume (TV) and the surface-to-volume ratio (BS/BV), were determined. To assess the bony microarchitecture, trabecular thickness (Tb.Th), trabecular separation (Tb.Sp), and trabecular number (Tb.N), as well as connectivity density (Conn.D), the degree of anisotropy (DA), and the structure model index (SMI), were evaluated. The highest BV was observed for NanoBone® (Group 1), which also showed the highest proportion of residual bone substitute material in the defect. This resulted in a significant increase in BV/TV with a significant decrease in BS/BV. The assessment of the microstructure for Groups 2 and 3 compared to Group 1 showed a clear approximation of Tb.Th, Tb.Sp, Tb.N, and Conn.D to the microstructure of the control group. The SMI showed a significant decrease in Group 1. All materials demonstrated their suitability by supporting biological defect reconstruction. NanoBone® showed the highest rate of new bone formation; however, the microarchitecture indicated more advanced bone remodeling and an approximate restoration of the trabecular structure for both autologous and allogeneic Tutoplast® cancellous bone when using the impaction bone grafting technique. Background: The objective was to evaluate and compare the efficacy of direct fragment impaction, indirect reduction through ligamentotaxis, and the combination of both techniques in burst fractures of the thoracolumbar (TL) spine. Methods: The fractures were categorized using the Arbeitsgemeinschaft für Osteosynthesefragen (AO) classification and assessed via standard computed tomography (CT) scans for spinal canal area (SCA) and mid-sagittal diameter (MSD). The Frankel classification was used to assess neurological deficits. Only single vertebrae AO types A3 and A4 thoracic or lumbar fractures were included. All patients received bisegmental posterior stabilization, one of the reduction techniques, and, if neurological deficits were present, a spinal decompression. Mean preoperative (µSCApre/µMSDpre), postoperative (µSCApost/µMSDpost) and difference (∆SCA/∆MSD) in radiographic values were obtained and analyzed using the Mumford formula. The significance of the reduction from preoperative stenosis was assessed using a t-test, while the effectiveness of the reduction techniques was compared using the Kruskal–Wallis test and Dunn’s post hoc test. The manuscript was focused primarily on radiographic outcomes; therefore, aside from the neurostatus, no other clinical parameters were statistically analyzed. Results: Thirteen patients (38.2%) received stand-alone indirect reduction, 13 patients (38.2%) underwent direct reduction, and a combined reduction was used in eight patients (23.6%). All methods resulted in a statistically significant reduction in spinal canal stenosis (p < 0.05), with a minimal mean ∆SCA of 19%. Patients in the direct reduction group had significantly higher µSCApre values compared to those in the indirect reduction group (p = 0.02). Conclusions: All of the tested reduction techniques provided a significant reduction in spinal canal stenosis. Patients who underwent mere direct reduction had significantly higher preoperative spinal canal stenosis compared to the indirect reduction group. Full article

Insights into the Silver Camphorimine Complexes Interactions with DNA Based on Cyclic Voltammetry and Docking Studies

Cyclic voltammetry (CV) is an accessible, readily available, non-expensive technique that can be used to search for the interaction of compounds with DNA and detect the strongest DNA-binding from a set of compounds, therefore allowing for the optimization of the number of cytotoxicity assays. Focusing on this electrochemical approach, the study of twenty-seven camphorimine silver complexes of six different families was performed aiming at detecting interactions with calf thymus DNA (CT-DNA). All of the complexes display at least two cathodic waves attributed respectively to the Ag(I)→Ag(0) (higher potential) and ligand based (lower potential) reductions. In the presence of CT-DNA, a negative shift in the potential of the Ag(I)→Ag(0) reduction was observed in all cases. Additional changes in the potential of the waves, attributed to the ligand-based reduction, were also observed. The formation of a light grey product adherent to the Pt electrode in the case of {Ag(OH)} and {Ag2(µ-O)} complexes further corroborates the interaction of the complexes with CT-DNA detected by CV. The morphologic analysis of the light grey material was made by scanning electronic microscopy (SEM). The magnitude of the shift in the potential of the Ag(I)→Ag(0) reduction in the presence of CT-DNA differs among the families of the complexes. The complexes based on {Ag(NO3)} exhibit higher potential shifts than those based on {Ag(OH)}, while the characteristics of the ligand (AL-Y, BL-Y, CL-Z) and the imine substituents (Y,Z) fine-tune the potential shifts. The energy values calculated by docking corroborate the tendency in the magnitude of the interaction between the complexes and CT-DNA established by the reaction coefficient ratios (Q[Ag-DNA]/Q[Ag]). The molecular docking study extended the information regarding the type of interaction beyond the usual intercalation, groove binding, or electrostatic modes that are typically reported, allowing a finer understanding of the non-covalent interactions involved. The rationalization of the CV and cytotoxicity data for the Ag(I) camphorimine complexes support a direct relationship between the shifts in the potential and the cytotoxic activities of the complexes, aiding the decision on whether the cytotoxicity of a complex from a family is worthy of evaluation. Background: Vascular imaging is essential for clinical practice, research, and the diagnosis and management of vascular diseases. Super-resolution ultrasound (SRUS) imaging is an emerging high-resolution imaging technique with broad applications in soft tissue vascular imaging. However, the impact of biological and clinical variables on its imaging accuracy is currently unknown. This study investigates these factors in an animal model and compares SRUS with contrast-enhanced µCT. Methods: Kidney scans from 29 Zucker rats (Zucker Diabetic Fatty and Zucker Lean) were retrospectively analysed. The left kidney was imaged in vivo using SRUS during microbubble infusion, then filled with Microfil and excised for ex vivo µCT. SRUS parameters and clinical variables were analysed, and SRUS scans were co-registered with µCT to compare vascular density measurements. Results: Mean arterial blood pressure and anaesthesia time showed significant linear relationships with SRUS microbubble detection and vascular track reconstruction. The anaesthesia time was also strongly correlated with vascular density measurement. Visualisation and velocity estimations of renal arteries were limited with SRUS. Ultrasound signal attenuation had significant impacts, particularly in cortical far-field imaging. Despite differences between kidney regions, the vascular density distribution did not differ considerably between SRUS and µCT datasets for whole-kidney imaging. Conclusions: This study outlines key factors SRUS users must consider for optimal technique use. Careful region selection and control of clinical variables ensure more reliable and comparable images. Further research is necessary to translate these findings from a rat model into clinical application. Full article

Wood–textile composites (WTCs), consisting of polypropylene and woven willow wood, have potential for both interior and exterior applications. However, their basic materials are not inherently resistant to outdoor weathering. This study examines the impact of various climatic conditions on the material behavior of WTCs. The composite and its components were aged under different scenarios, including kiln-drying, frost, standard and tropical climate, and artificial weathering and water storage, and analyzed for dimensional stability, chemical changes (FTIR), mechanical damage (µ-CT), and mechanical performance. While kiln-drying, frost, and tropical climates had only minor effects, water storage caused swelling-related damage, resulting in a 45% decrease in Young’s modulus but increased elongation at break (+88%) and impact strength (+75%). Artificial weathering led to significant degradation: tensile strength declined by 28%, Young’s modulus by 49%, and impact strength by 26%. In the medium term, this degradation compromises the integrity of the composite. The results highlight the need for effective stabilization measures—such as polymer modification or structural protection—to ensure the long-term durability of WTCs in outdoor use. Full article

Development of Multiplex qPCR Method for Accurate Detection of Enzyme-Producing Psychrotrophic Bacteria

Microbial detection in milk is crucial for food safety and quality, as beneficial and harmful microorganisms can affect consumer health and dairy product integrity. Identifying and quantifying these microorganisms helps prevent contamination and spoilage. The study employs advanced molecular techniques to detect and quantify the genomic DNA for the target hydrolytic enzyme coding genes lipA and aprX based on the multi-align sequence conserved region, specific primer pair, and hydrolysis probes designed using the singleplex qPCR and multiplex qPCR. Cultured isolates and artificially contaminated sterilized ultra-high-temperature (UHT) milk were analyzed for their specificity, cross-reactivity, and sensitivity. The finding indicated that strains with lipA and aprX genes were amplified while the other strains were not amplified. This indicated that the designed primer pairs/probes were very specific to the target gene of interest. The specificity of each design primer pair was checked using SYBR Green qPCR using 16 different isolate strains from the milk sample. The quantification specificity of each strain target gene was deemed to be with a mean Ct value for positive pseudomonas strain > 16.98 ± 1.76 (p < 0.0001), non-pseudomonas positive strain ≥ 27.47 ± 1.25 (p < 0.0001), no Ct for the negative control and molecular grade water. The sensitivity limit of detection (LOD) analyzed based on culture broth and milk sample was >105 and >104 in PCR amplification while it was >104 and >103 in real-time qPCR, respectively. At the same time, the correlation regression coefficient of the standard curve based on the pure culture cell DNA as the DNA concentration serially diluted (20 ng/µL to 0.0002 ng/µL) was obtained in multiplex without interference and cross-reactivity, yielding R2 ≥ 0.9908 slope (−3.2591) and intercepting with a value of 37, where the efficiency reached the level of 95–102% sensitivity reached up to 0.0002 ng/µL concentration of DNA, and sensitivity of microbial load was up to 1.2 × 102 CFU/mL. Therefore, multiplex TaqMan qPCR simultaneous amplification was considered the best method developed for the detection of the lipA and aprX genes in a single tube. This will result in developing future simultaneous (three- to four-gene) detection of spoilage psychrotrophic bacteria in raw milk. Full article

Evaluation of RT-LAMP for SARS-CoV-2 Detection in Animal Feces

The wide host range, potential lethality, and zoonotic potential of SARS-CoV-2 infection in animals highlights the need for additional surveillance strategies. We validated a commercial, pH-based, colorimetric RT-LAMP assay for the detection of SARS-CoV-2 RNA in animal feces. The comparator assay was rRT-PCR. The limit of detection of the RT-LAMP assay was 72 genome copies per reaction. RT-LAMP was highly specific for SARS-CoV-2 and did not detect other human or animal coronaviruses. RT-LAMP was robust, with valid results generated for incubation lengths of 30 to 45 min, incubation temperatures of 60 to 70 °C, and reaction volumes of 10 to 25 µL. The diagnostic sensitivity was 100% for clinical fecal samples with high viral loads (Ct ≤ 25), 97.4% for samples with moderate to high viral loads (Ct ≤ 33), and 62% overall (Ct ≤ 40). The diagnostic specificity was 97.9%. Blinded method testing organized by an independent laboratory confirmed the satisfactory reproducibility of the assay. To our knowledge, this study represents the first validation of RT-LAMP for SARS-CoV-2 detection in animals. RT-LAMP testing could detect SARS-CoV-2 infection more rapidly and at the point of care in animals with moderate to high viral loads, allowing for earlier implementation of control measures. Background and Objectives: Padua (1997) and Bland (2000) have already proposed neurophysiological classification scales for patients with carpal tunnel syndrome (CTS), where the absence of orthodromic sensory response is used as a criterion of a severe stage. We hypothesized that antidromic values cannot be used equally for correct staging. Materials and Methods: We performed a consecutive investigation with nerve conduction studies in 60 arms of patients with CTS and prolonged distal motor latency. Results: In 11 out of 60 arms (18.3% of cases), orthodromic sensory nerve action potential (SNAP) was undetectable, while the antidromic SNAP was present. ROC curve analysis with Yoden index calculation were utilized in the study. The cut-off value of antidromic SNAP amplitude as a diagnostic marker of unrecordable orthodromic SNAP was 3.9 µV with high sensitivity and specificity. Conclusions: Our findings conflict with Padua et al.’s assertion that CTS staging can be determined irrespective of the stimulation technique. Antidromic SNAP amplitude is the most reliable parameter for predicting the absent orthodromic SNAP. Our study addresses the bias associated with the application of antidromic stimulation of median nerve sensory fibers for accurately staging moderate to severe CTS. Full article
