Myocardial Strain Measurements Obtained with Fast-Strain-Encoded Cardiac Magnetic Resonance for the Risk Prediction and Early Detection of Chemotherapy-Related Cardiotoxicity Compared to Left Ventricular Ejection Fraction

Background: Breast and hematological cancer treatments, especially with anthracyclines, have been shown to be associated with an increased risk of cardiotoxicity (CTX). An accurate prediction of cardiotoxicity risk and early detection of myocardial injury may allow for effective cardioprotection to be instituted and tailored to reverse cardiac dysfunction and prevent the discontinuation of essential cancer treatments. Objectives: The PRoactive Evaluation of Function to Evade Cardio Toxicity (PREFECT) study sought to evaluate the ability of fast-strain-encoded (F-SENC) cardiac magnetic resonance imaging (CMR) and 2D echocardiography (2D Echo) to stratify patients at risk of CTX prior to initiating cancer treatment, detect early signs of cardiac dysfunction, including subclinical CTX (sub-CTX) and CTX, and monitor for recovery (REC) during cardioprotective therapy. Methods: Fifty-nine patients with breast cancer or lymphoma were prospectively monitored for CTX with F-SENC CMR and 2D Echo over at least 1 year for evidence of cardiac dysfunction during anthracycline based chemotherapy. F-SENC CMR also monitored myocardial deformation in 37 left ventricular (LV) segments to obtain a MyoHealth risk score based on both longitudinal and circumferential strain. Sub-CTX and CTX were classified based on pre-specified cardiotoxicity definitions. Results: CTX was observed in 9/59 (15%) and sub-CTX in 24/59 (41%) patients undergoing chemotherapy. F-SENC CMR parameters at baseline predicted CTX with a lower LVEF (57 ± 5% vs. 61 ± 5% for all, p = 0.05), as well as a lower MyoHealth (70 ± 9 vs. 79 ± 11 for all, p = 0.004) and a worse global circumferential strain (GCS) (−18 ± 1 vs. −20 ± 1 for all, p < 0.001). Pre-chemotherapy MyoHealth had a higher accuracy in predicting the development of CTX compared to CMR LVEF and 2D Echo LVEF (AUC = 0.85, 0.69, and 0.57, respectively). The 2D Echo parameters on baseline imaging did not stratify CTX risk. F-SENC CMR obtained good or excellent images in 320/322 (99.4%) scans. During cancer treatment, MyoHealth had a high accuracy of detecting sub-CTX or CTX (AUC = 0.950), and the highest log likelihood ratio (indicating a higher probability of detecting CTX) followed by F-SENC GLS and F-SENC GCS. CMR LVEF and CMR LV stroke volume index (LVSVI) also significantly worsened in patients developing CTX during cancer treatment. Conclusions: F-SENC CMR provided a reliable and accurate assessment of myocardial function during anthracycline-based chemotherapy, and demonstrated accurate early detection of CTX. In addition, MyoHealth allows for the robust identification of patients at risk for CTX prior to treatment with higher accuracy than LVEF. Full article

IgA nephropathy is the most common primary glomerulonephritis, with pathohistological changes described by the Oxford classification, while the Banff classification is used in transplant pathology. This study included 253 patients with IgA nephropathy in native kidneys, divided into the pediatric (n = 105) and adult (n = 148) groups. It aimed to examine clinical, and Oxford and Banff morphological parameters in relation to age, correlations of clinical data with pathohistological parameters, and predictors of the disease outcome. Pediatric patients more frequently presented with macroscopic hematuria, while adults showed higher urea and creatinine levels, and lower eGFR. Examining Oxford classification parameters, chronic glomerular and tubulointerstitial lesions were more common in adults. Banff parameters revealed more frequent chronically active glomerular, inflammatory, chronic tubulointerstitial, and vascular lesions in adults. All inflammatory, chronic tubulointerstitial, and vascular parameters correlated with serum urea levels, eGFR and CKD stage in adults, while less frequent in pediatric patients. Soil sealing and land take are increasingly recognised as critical environmental and land use planning challenges across Europe. Although these issues have received limited attention in Baltic policymaking and the academic literature to date, available data indicate ongoing land consumption despite population decline. This study aims to analyse soil sealing patterns in Estonia, Latvia, and Lithuania between 2018 and 2021 using CLC+ Backbone data, linking them to demographic shifts and local planning frameworks. Results reveal that soil sealing increased in nearly all municipalities across the Baltic states, regardless of population trends. The analysis highlights that shrinking municipalities, constrained by limited resources and declining populations, are structurally disadvantaged in terms of land use efficiency, particularly when measured by sealed area per capita. Moreover, this study discusses emerging policy tensions, including the narrowing conceptual gap between land take and soil sealing in the proposed EU Soil Monitoring and Resilience Directive, as well as the risk of overlooking broader land artificialisation. The findings underscore the need for context-sensitive, multi-scalar approaches to land use monitoring and governance, particularly in sparsely populated and demographically imbalanced regions, such as the Baltic states. Kennaugh Elements Allow Early Detection of Bark Beetle Infestation in Temperate Forests Using Sentinel-1 Data

Climate change is generally having a negative impact on forest health by inducing drought stress and favouring the spread of pest species, such as bark beetles. The terrestrial monitoring of bark beetle infestation is very time-consuming, especially in the early stages, and therefore not feasible for extensive areas, emphasising the need for a comprehensive approach based on remote sensing. Although numerous studies have researched the use of optical data for this task, radar data remains comparatively underexplored. Therefore, this study uses the weekly and cloud-free acquisitions of Sentinel-1 in the Bavarian Forest National Park. Time series analysis within a Multi-SAR framework using Random Forest enables the monitoring of moisture content loss and, consequently, the assessment of tree vitality, which is crucial for the detection of stress conditions conducive to bark beetle outbreaks. High accuracies are achieved in predicting future bark beetle infestation (R2 of 0.83–0.89). These results demonstrate that forest vitality trends ranging from healthy to bark beetle-affected states can be mapped, supporting early intervention strategies. The standard deviation of 0.44 to 0.76 years indicates that the model deviates on average by half a year, mainly due to the uncertainty in the reference data. This temporal uncertainty is acceptable, as half a year provides a sufficient window to identify stressed forest areas and implement targeted management actions before bark beetle damage occurs. The successful application of this technique to extensive test sites in the state of North Rhine-Westphalia proves its transferability. For the first time, the results clearly demonstrate the expected relationship between radar backscatter expressed in the Kennaugh elements K0 and K1 and bark beetle infestation, thereby providing an opportunity for the continuous and cost-effective monitoring of forest health from space. Turbidity currents pose significant threats to offshore seabed infrastructures, including subsea hydrocarbon production facilities and submarine communication cables. These powerful underwater flows can damage pipelines, potentially causing hydrocarbon spills that endanger local communities, the environment, and negatively impact energy production infrastructures. Therefore, a comprehensive understanding of the spatio-temporal development and destructive force of turbidity currents is essential. While numerical computation of 3D flow, sediment transport, and substrate exchange is possible, field-scale simulations are computationally intensive. In this study, we develop a simplified morphodynamic approach to model the flow properties of channelized turbidity currents and the associated trends of sediment accretion and erosion. This model is applied to the Baco–Malaylay submarine system to investigate the dynamics of a significant turbidity current event that impacted a submarine pipeline offshore the Philippines. The modeling results align with available seabed assessments and observed erosion trends of the protective rock berm. Our simplified modeling approach shows good agreement with simulations from a fully 3D numerical model, demonstrating its effectiveness in providing valuable insights while reducing computational demands. (1) Background: Physical inactivity is a major public health concern in modern society. Group fitness programs are widely used to promote physical activity, combining choreographed movements with various dance steps and music. This study aimed to examine the effects of Zumba and MoFit group fitness programs on body composition parameters in women. (2) Methods: The study included 98 female participants (Mean age = 27.8 ± 2.9 years), divided into three groups: E1 (n = 33), which followed the experimental Zumba program; E2 (n = 31), which followed the experimental MoFit program; and a control group (n = 34), which continued with their usual daily activities for 10 weeks. Body composition was assessed using 14 variables measured with the InBody 270 analyser. Statistical analyses included paired t-tests, MANCOVA, and ANCOVA. (3) Results: The findings confirmed the positive effects of both group fitness programs on most body composition parameters in women (p < 0.001). However, Bonferroni post hoc test results indicated that the Zumba program led to significantly greater improvements in most body composition variables compared to the MoFit program. (4) Conclusions: Both Zumba and MoFit programs were effective in reducing body fat, increasing muscle mass, total body water, and mineral content, whereas the control group did not achieve positive changes. Evidence of Graft Incompatibility and Rootstock Scion Interactions in Cacao

This study sought to quantify and characterize diverse rootstock scion interactions in cacao around graft compatibility, disease resistance, nutrient use efficiency, vigor traits, and translocation of nonstructural carbohydrates. In total, 106 grafts were performed with three scion cultivars (Matina 1/6, Criollo 22, Pound 7) and nine diverse open-pollinated seedling populations (BYNC, EQX 3348, GNV 360, IMC 14, PA 107, SCA 6, T 294, T 384, T 484). We found evidence for both local and translocated graft incompatibility. Cross sections and Micro-XCT imaging revealed anatomical anomalies, including necrosis and cavitation at the junction and accumulation of starch in the rootstock directly below the graft junction. Scion genetics were a significant factor in explaining differences in graft take, and graft take varied from 47% (Criollo 22) to 72% (Pound 7). Rootstock and scion identity both accounted for differences in survival over the course of the 30-month greenhouse study, with a low of 28.5% survival of Criollo 22 scions and a high of 72% for Pound 7 scions. Survival by rootstocks varied from 14.3% on GNV 360 to 100% survival on T 294 rootstock. A positive correlation of 0.34 (p = 0.098) was found between the graft success of different rootstock–scion combinations and their kinship coefficient, suggesting that relatedness of stock and scion could be a driver of incompatibility. Significant rootstock–scion effects were also observed for nutrient use efficiency, plant vigor, and resistance to Phytophthora palmivora. These findings, while preliminary in nature, highlight the potential of rootstock breeding to improve plant nutrition, resilience, and disease resistance in cacao. Full article

Waiting Times for Surgery and Radiotherapy Among Breast Cancer Patients in Switzerland: A Cancer Registry-Based Cross-Sectional and Longitudinal Analysis

Background: Delays in breast cancer treatment negatively affect prognosis and have increased over time. Data on waiting times in Switzerland are limited. Patients and Methods: This study analyzed cancer registry data from 2003 to 2005 (2628 patients) and 2015 to 2017 (421 patients) to evaluate waiting times for diagnosis, surgery, and radiotherapy; temporal trends; and survival in women with stage I–III invasive breast cancer treated with surgery without chemotherapy. Associations with demographic/clinical factors and overall survival (OS) were assessed using ANOVA, uni-/multivariable regression, Kaplan–Meier, and Cox regression. Results: From 2003 to 2005, mean intervals were biopsy-to-diagnosis 4.3 days, diagnosis-to-surgery 18.8 days, biopsy-to-surgery 26.8 days, and surgery-to-radiotherapy 56.7 days. Longer diagnosis-to-surgery times were associated with metropolitan areas, public hospitals, basic insurance, mastectomy, and older age (all p < 0.001). Radiotherapy delays were also longer in metropolitan areas and after mastectomy (p < 0.001). Between 2003–2005 and 2015–2017, diagnosis-to-surgery times rose in Eastern Switzerland (from 21.3 to 31.2 days), while radiotherapy timing remained stable. Five-year overall survival improved (from 76.7% to 88.4%), but was not significantly impacted by diagnosis-to-surgery intervals. Conclusions: Despite timely surgery in Switzerland (2003–2005), disparities existed, and time to surgery increased by 2015–2017. Reducing waiting times remains important despite no significant short-term OS impact. Full article

The expression of recombinant proteins in heterologous hosts is a common strategy to obtain larger quantities of the “protein of interest” (POI) for scientific, therapeutic or commercial purposes. However, the experimental success of such an approach critically depends on the choice of an appropriate host system to obtain biologically active forms of the POI. The correct folding of the molecule, mediated by disulfide bond formation, is one of the most critical steps in that process. Here we describe the recombinant expression of hirudin, a leech-derived anticoagulant and thrombin inhibitor, in the yeast Komagataella phaffii (formerly known and mentioned throughout this publication as Pichia pastoris) and in two different strains of Escherichia coli, one of them being especially designed for improved disulfide bond formation through expression of a protein disulfide isomerase. Cultivation of the heterologous hosts and expression of hirudin were performed at different temperatures, ranging from 22 to 42 °C for the bacterial strains and from 20 to 30 °C for the yeast strain, respectively. The thrombin-inhibitory potencies of all hirudin preparations were determined using the thrombin time coagulation assay. To our surprise, the hirudin preparations of P. pastoris were considerably less potent as thrombin inhibitors than the respective preparations of both E. coli strains, indicating that a eukaryotic background is not per se a better choice for the expression of a biologically active eukaryotic protein. The hirudin preparations of both E. coli strains exhibited comparable high thrombin-inhibitory potencies when the strains were cultivated at their respective optimal temperatures, whereas lower or higher cultivation temperatures reduced the inhibitory potencies. Background/Objectives: Cerebrovascular reactivity (CVR) is a key marker of cerebrovascular function, facilitating the early detection of neurovascular dysfunction. Breath-hold functional MRI (bh-fMRI) is a non-invasive method for assessing CVR. This study evaluates the reproducibility of bh-fMRI using short breath-hold periods, which are practical for clinical use. Methods: In a prospective study, 50 healthy subjects underwent three self-paced, end-expiration bh-fMRI sessions with 9 s breath-hold periods at 3T. A 30 min break between the second and third sessions was included. The reproducibility of the percentage signal change (PSC) in predefined volumes of interest for a ±0 s, ±3 s and ±6 s interval around the cerebellar peak (IAP)) was evaluated. The intraclass correlation coefficient (ICC) and the intra-personal coefficient of variation (CVintra) were calculated between the individual sessions. Results: This study demonstrated excellent reproducibility, with an ICC (2, k) for a ±0 s IAP across all sessions at 0.887 (95% CI: 0.882–0.892). The ICC values remained within an excellent range even when the participants left the scanner between sessions. The CVintra for the ±0 s IAP (14.54% ± 8.54%) remained below the 33% fiducial limit. A larger IAP revealed higher ICC values but higher CVintra values and lower PSC values. Conclusions: Bh-fMRI with 9 s breath-hold periods yields highly reproducible CVR assessments, supporting its feasibility for clinical implementation. Full article

The southern conifer Agathis australis (D.Don) Lindl. is a large and long-lived species endemic to Aotearoa New Zealand. It is threatened due to past logging activities, pathogen attack and potentially climate change, with increasing severity and frequency of drought and heatwaves across its distribution. Like many large tree species, little is known about the carbon dynamics of this ecologically and culturally significant species. We explored seasonal variations in non-structural carbohydrates (NSCs) and growth in trees ranging from 20 to 175 cm diameter at breast height (DBH). NSCs were seasonally stable with no measurable pattern across seasons. However, we found growth rates standardised to basal area and sapwood area (growth efficiency) declined with tree age and stem NSC concentrations (including total NSCs, sugars and starch) all increased as trees aged. Total NSC concentrations were 0.3%–0.6% dry mass for small trees and 0.8%–1.8% dry mass for larger trees, with strong relationships between DBH and total NSC, sugar and starch in stems but not roots. Cumulative growth efficiency across the two-year study period declined as tree size increased. Furthermore, there was an inverse relationship between growth efficiency across the two-year study period and NSC concentrations of stems. This relationship was driven by differences in carbon dynamics in trees of different sizes, with trees progressing to a more conservative carbon strategy as they aged. Simultaneously declining growth efficiency and increasing NSC concentrations as trees age could be evidence for active NSC accumulation to buffer against carbon starvation in larger trees. Our study provides new insights into changing carbon dynamics as trees age and may be evidence for active carbon accumulation in older trees. This may provide the key for understanding the role of carbon processes in tree longevity. Full article

One of the advantages of the EPR spectroscopy method in assessing structural defects caused by irradiation is the fact that using this method it is possible to determine not only the concentration dependences of the defect structure but to also establish their type, which is not possible with methods such as X-ray diffraction or scanning electron microscopy. Based on the data obtained, the role of variation in the ratio of components in Li4SiO4–Li2TiO3 ceramics on the processes of softening under high-dose irradiation with protons simulating the accumulation of hydrogen in the damaged layer, as well as the concentration of structural defects in the form of oxygen vacancies and radiolysis products on the processes of high-temperature degradation of ceramics, was determined. It was found that the main changes in the defect structure during the prolonged thermal exposure of irradiated samples are associated with the accumulation of oxygen vacancies, the density of which was estimated by the change in the intensity of singlet lithium, characterizing the presence of E-centers. At the same time, it was found that the formation of interphase boundaries in the structure of Li4SiO4–Li2TiO3 ceramics leads to the inhibition of high-temperature degradation processes in the case of post-radiation thermal exposure for a long time. Also, during the conducted studies, the role of thermal effects on the structural damage accumulation rate in Li4SiO4–Li2TiO3 ceramics was determined in the case when irradiation is carried out at different temperatures. During the experiments, it was determined that the main contribution of thermal action in the process of proton irradiation at a fluence of 5 × 1017 proton/cm2 is an increase in the concentration of radiolysis products, described by changes in the intensities of spectral maxima, characterized by the presence of defects such as ≡Si–O, SiO43− and Ti3+ defects. Immunorecovered but Exhausted: Persistent PD-1/PD-L1 Expression Despite Virologic Suppression and CD4 Recovery in PLWH

Background/Objectives: While ART effectively suppresses HIV viremia, many PLWH exhibit persistent immune dysfunction. This study aimed to assess immune recovery and immune exhaustion (PD-1/PD-L1 expression) in newly diagnosed versus long-term ART-treated individuals. Methods: We analyzed 79 PLWH: 52 newly diagnosed individuals (12-month follow-up) and 27 long-term-treated patients (Ukrainian refugees). Flow cytometry was used to evaluate CD4+ and CD8+ counts, the CD4+/CD8+ ratio, and PD-1/PD-L1 expression on CD3+, CD4+, and CD19+ lymphocytes. ART regimen and HIV subtype were included as covariates in linear regression models. Results: At 12 months, CD4+ counts were similar between groups (median 596.5 vs. 621 cells/μL, p = 0.22), but newly diagnosed patients had higher CD8+ counts (872 vs. 620 cells/μL, p = 0.028) and a lower CD4+/CD8+ ratio (0.57 vs. 1.05, p = 0.0027). Immune exhaustion markers were significantly elevated in newly diagnosed individuals: CD4+ PD-1+ T cells (24.4% vs. 3.85%, p = 0.0002) and CD3+ PD-1+ T cells (27.3% vs. 12.35%, p < 0.0001). Linear regression confirmed group membership independently predicted higher CD3+ (β = +21.92, p < 0.001), CD4+ (β = +28.87, p < 0.0001), and CD19+ (β = +8.73, p = 0.002) percentages. Lipid parameters and SCORE2 did not differ significantly. Conclusions: Despite virologic suppression and CD4+ recovery, immune exhaustion markers remain elevated in newly diagnosed PLWH, suggesting incomplete immune normalization. Traditional parameters (CD4+ count and CD4+/CD8+ ratio) may not fully capture immune status, warranting broader immunologic profiling in HIV care. Full article

Advanced Oxidation of Dexamethasone by Activated Peroxo Compounds in Water Matrices: A Comparative Study

The continuous occurrence of steroidal pharmaceutical dexamethasone (DXM) in aqueous environments indicates the need for an efficient removal technology. The frequent detection of DXM in surface water could be substantially reduced by the application of photo-induced advanced oxidation technology. In the present study, Fe2+ and UVA-light activated peroxo compounds were applied for the degradation and mineralization of a glucocorticoid, 25.5 µM DXM, in ultrapure water (UPW). The treatment efficacies were validated in real spring water (SW). A 120 min target pollutant degradation followed pseudo first-order reaction kinetics when an oxidant/Fe2+ dose 10/1 or/and UVA irradiation were applied. Acidic conditions (a pH of 3) were found to be more favorable for DXM oxidation (≥99%) regardless of the activated peroxo compound. Full conversion of DXM was not achieved, as the maximum TOC removal reached 70% in UPW by the UVA/H2O2/Fe2+ system (molar ratio of 10/1) at a pH of 3. The higher efficacy of peroxymonosulfate-based oxidation in SW could be induced by chlorine, bicarbonate, and carbonate ions; however, it is not applicable for peroxydisulfate and hydrogen peroxide. Overall, consistently higher efficacies for HO-dominated oxidation systems were observed. The findings from the current paper could complement the knowledge of oxidative removal of low-level DXM in real water matrices. Full article

The coastal developments in the Middle East put low priority on tsunami risk assessment due to the rare occurrence and absence of genuine tsunami track records on the coastline in the past. Tsunami-vulnerable coasts, including the east coast of the UAE, need to prepare for, and pay attention to, the impact of future tsunamis due to increased earthquake activity in the region. This study investigated the tsunami characteristics of the nearshore from hypothetical tsunami conditions by applications of numerical modeling and Artificial Neural Network (ANN) methods. The modeling results showed that the maximum tsunami depth at the shore was highest in Khor Fakkan and Mirbih for the given tsunami boundary conditions, while the tsunami withdrawal was greater on the southern bathymetry compared to that on the northern bathymetry when the tsunami period increased. ANN results confirmed that the still sea depth and seabed slope were more important than the tsunami period when predicting the maximum tsunami depth at the shore. Full article
