This article examines the impact of neoliberal political–economic orientations on urban planning policies through a case study of Ireland’s Strategic Housing Development (SHD) fast-track planning scheme. By evaluating SHD applications from 2017 to 2021, this study assesses the scheme’s effectiveness in housing development in County Dublin and addresses how neoliberal policies affect planning objectives. The findings reveal that, although the SHD scheme expedited planning approvals, it faced significant challenges, including high rates of judicial review and low commencement rates for approved developments. These issues hindered the scheme’s capacity to address housing shortages effectively. The research highlights that market-driven policies often shape urban development patterns in ways that misalign with policy goals, potentially compromising social equity and long-term sustainability. By exploring the tensions between planning efficiency, industry interests, and broader policy objectives, this study contributes to ongoing debates about the role of neoliberalism in urban planning. It concludes by advocating for a more integrated approach to urban governance—one that balances economic imperatives with social and environmental priorities to promote sustainable and equitable development. This paper provides a study of free-flight air traffic behaviour in increasingly constrained airspace environments. Traffic assumes three different free-flight operational constructs with airspace constraints considered as restricted (no-fly) regions. Simulations combine path planning and Monte Carlo techniques to qualitatively analyse emergent traffic behaviour and quantitatively assess spatial–temporal airspace conflict as the airspace constraints vary. Findings indicate that airspace constraints have a much stronger influence on aircraft behaviour than the free-flight operational construct, with any benefits of free flight rapidly diminishing as the airspace becomes more constrained. We conclude that structured traffic route (or network) designs and associated risk modelling approaches should be considered for safe and efficient traffic management of highly constrained and congested (or dense) airspace. This work therefore provides evidence to inform new airspace design and management initiatives, including low-altitude uncrewed traffic. Full article

Chronic lymphocytic leukemia (CLL) is a malignancy of mature B cells, and it is the most frequent form of leukemia diagnosed in Western countries. It is characterized by the proliferation and accumulation of neoplastic B lymphocytes in the blood, lymph nodes, bone marrow and spleen. We report the synthesis and antiproliferative effects of a series of novel ethanoanthracene compounds in CLL cell lines. Structural modifications were achieved via the Diels–Alder reaction of 9-(2-nitrovinyl)anthracene and 3-(anthracen-9-yl)-1-arylprop-2-en-1-ones (anthracene chalcones) with dienophiles, including maleic anhydride and N-substituted maleimides, to afford a series of 9-(E)-(2-nitrovinyl)-9,10-dihydro-9,10-[3,4]epipyrroloanthracene-12,14-diones, 9-(E)-3-oxo-3-phenylprop-1-en-1-yl)-9,10-dihydro-9,10-[3,4]epipyrroloanthracene-12,14-diones and related compounds. Single-crystal X-ray analysis confirmed the structures of the novel ethanoanthracenes 23f, 23h, 24a, 24g, 25f and 27. The products were evaluated in HG-3 and PGA-1 CLL cell lines (representative of poor and good patient prognosis, respectively). The most potent compounds were identified as 20a, 20f, 23a and 25n with IC50 values in the ranges of 0.17–2.69 µM (HG-3) and 0.35–1.97 µM (PGA-1). The pro-apoptotic effects of the potent compounds 20a, 20f, 23a and 25n were demonstrated in CLL cell lines HG-3 (82–95%) and PGA-1 (87–97%) at 10 µM, with low toxicity (12–16%) observed in healthy-donor peripheral blood mononuclear cells (PBMCs) at concentrations representative of the compounds IC50 values for both the HG-3 and PGA-1 CLL cell lines. The antiproliferative effect of the selected compounds, 20a, 20f, 23a and 25n, was mediated through ROS flux with a marked increase in cell viability upon pretreatment with the antioxidant NAC. 25n also demonstrated sub-micromolar activity in the NCI 60 cancer cell line panel, with a mean GI50 value of 0.245 µM. This ethanoanthracene series of compounds offers potential for the further development of lead structures as novel chemotherapeutics to target CLL. Full article

High-density urban development is promoted by both global and local policies in response to socio-economic and environmental challenges since it increases mobility of different land uses, decreases the need for traveling, encourages the use of more energy-efficient buildings and modes of transportation, and permits the sharing of scarce urban amenities. It is therefore argued that increased density and mixed-use development are expected to deliver positive outcomes in terms of contributing to three pillars (social, economic, and environmental domains) of sustainability in the subject themes. Territorial quality of life (TQL)—initially proposed by the ESPON Programme—is a composite indicator of the socio-economic and environmental well-being and life satisfaction of individuals living in an area. Understanding the role of urban density in TQL can provide an important input for urban planning debates addressing whether compact development can be promoted by referring to potential efficiencies in high-density, mixed land use and sustainable transport provisions. Alternatively, low-density suburban development is preferable due to its benefits of high per capita land use consumption (larger houses) for individual households given lower land prices. There is little empirical evidence on how TQL is shaped by high-density versus low-density urban forms. This paper investigates this topic through providing an approach to spatially map and examine the relationship between TQL, residential expansion, and densification processes in the so-called NUTS2 (nomenclature of terrestrial units for statistics) regions of European Union (EU) member countries. The relative importance of each TQL indicator was determined through the entropy weight method, where these indicators were aggregated through using the subject weights to obtain the overall TQL indicator. The spatial dynamics of TQL were examined and its relationship with residential expansion and densification processes was analysed to uncover whether the former or the latter process is positively associated with the TQL indicator within our study area. From our regression models, the residential expansion index is negatively related to the TQL indicator, implying that high levels of residential expansion can result in a reduction in overall quality of life in the regions if they are not supported by associated infrastructure and facility investments. Fisheries-independent data on the diversity, relative abundance, and demographic structure of poorly studied, threatened oceanic sharks are absent from much of the western North Atlantic Ocean, where multiple oceanic shark species have experienced significant population declines. Resource-limited management approaches require the identification of critical habitats or aggregation sites worthy of protection and enforcement. Data were collected on oceanic sharks using pelagic longline surveys, targeted baiting, and opportunistic encounters in oligotrophic open-ocean habitats of north-eastern Exuma Sound (NES), The Bahamas. The oceanic epipelagic shark community was also characterized using targeted baiting off Columbus Point, Cat Island (CI), a seamount north of San Salvador (SSSM), and the northwestern tip of Mayaguana. Pelagic longline surveys suggested that the relative abundance of sharks at NES was low (shark catch-per-unit-effort: 0.0007 sharks hook−1 h−1; 2.3 sharks per 1000 hooks). Silky sharks Carcharhinus falciformis, particularly juveniles (134 ± 39 cm stretched total length; mean ± SD STL), were the most common. Targeted baiting suggested oceanic whitetip sharks C. longimanus were abundant at CI, where large adults (245 ± 23 cm STL), most of which were females (83.8%, n = 98 of 117) that were gravid (65.7%; n = 46 of 70 assessed for pregnancy), dominated the aggregation. Many (20.5–26.5%, n = 24–31 of 117 depending on assumptions regarding tag loss) were recaptured or resighted at CI for up to five years. Silky sharks dominated catches at SSSM. Oceanic sharks, particularly adults, were sometimes caught or observed alongside short-finned pilot whales Globicephala macrorhynchus or tunas. Although The Bahamas offers threatened oceanic sharks refuge from fishing across its entire jurisdiction, these data suggest that some fixed features, including sites such as CI and potentially SSSM, are important aggregation sites with high regional conservation value and should be prioritized by fisheries managers and enforcement officials. Chronic lymphocytic leukaemia (CLL) is a malignancy of the immune B lymphocyte cells and is the most common leukaemia diagnosed in developed countries. In this paper, we report the synthesis and antiproliferative effects of a series of (E)-9-(2-nitrovinyl)anthracenes and related nitrostyrene compounds in CLL cell lines and also in Burkitt’s lymphoma (BL) cell lines, a rare form of non-Hodgkin’s immune B-cell lymphoma. The nitrostyrene scaffold was identified as a lead structure for the development of effective compounds targeting BL and CLL. The series of structurally diverse nitrostyrenes was synthesised via Henry–Knoevenagel condensation reactions. Single-crystal X-ray analysis confirmed the structure of (E)-9-chloro-10-(2-nitrobut-1-en-1-yl)anthracene (19f) and the related 4-(anthracen-9-yl)-1H-1,2,3-triazole (30a). The (E)-9-(2-nitrovinyl)anthracenes 19a, 19g and 19i–19m were found to elicit potent antiproliferative effects in both BL cell lines EBVMUTU-1 (chemosensitive) and EBV+ DG-75 (chemoresistant) with >90% inhibition at 10 μM. Selected (E)-9-(2-nitrovinyl)anthracenes demonstrated potent antiproliferative activity in CLL cell lines, with IC50 values of 0.17 μM (HG-3) and 1.3 μM (PGA-1) for compound 19g. The pro-apoptotic effects of the most potent compounds 19a, 19g, 19i, 19l and 19m were demonstrated in both CLL cell lines HG-3 and PGA-1. The (E)-nitrostyrene and (E)-9-(2-nitrovinyl)anthracene series of compounds offer potential for further development as novel chemotherapeutics for CLL. Full article

While astrocyte-to-neuron (AtN) reprogramming holds great promise in regenerative medicine, the molecular mechanisms that govern this unique biological process remain elusive. To understand the function of miRNAs during the AtN reprogramming process, we performed RNA-seq of both mRNAs and miRNAs on human astrocyte (HA) cultures upon NeuroD1 overexpression. Bioinformatics analyses showed that NeuroD1 not only activated essential neuronal genes to initiate the reprogramming process but also induced miRNA changes in HA. Among the upregulated miRNAs, we identified miR-375 and its targets, neuronal ELAVL genes (nELAVLs), which encode a family of RNA-binding proteins and were also upregulated by NeuroD1. We further showed that manipulating the miR-375 level regulated nELAVLs’ expression during NeuroD1-mediated reprogramming. Interestingly, miR-375/nELAVLs were also induced by the reprogramming factors Neurog2 and ASCL1 in HA, suggesting a conserved function to neuronal reprogramming, and by NeuroD1 in the mouse astrocyte culture and spinal cord. Functionally, we showed that miR-375 overexpression improved NeuroD1-mediated reprogramming efficiency by promoting cell survival at early stages in HA and did not appear to compromise the maturation of the reprogrammed neurons. Lastly, overexpression of miR-375-refractory ELAVL4 induced apoptosis and reversed the cell survival-promoting effect of miR-375 during AtN reprogramming. Together, we demonstrated a neuroprotective role of miR-375 during NeuroD1-mediated AtN reprogramming. Full article

Varves accumulating below the chemocline of meromictic Crawford Lake, Milton, Ontario, Canada, consist of dark-colored organic matter laminations that are primarily deposited during the fall plankton die-off, alternating with light-colored laminations comprising calcite crystals that are precipitated during a relatively narrow water temperature and pH-controlled depositional window in the summer. A novel high-resolution imaging protocol was used to photograph the varve record in the 87 cm-long freeze core CRA19-2FT-B2, collected from the deepest part (~23 m) of the lake in February 2019. High-resolution images were used to: (1) characterize varve couplets deposited between AD 1870 and 2000 (chronology verified through 137Cs/210Pb analysis of freeze core CRA22-1FRA-3, and consistent with the historic record of nuclear fallout and other proxies of the Great Acceleration); (2) document distinctive varves that permit a correlation between cores throughout the deep basin of Crawford Lake; (3) measure the thickness of individual dark and light- colored laminations, which were found to vary between 0.04 mm and 3.76 mm; and (4) carry out wavelet and spectral time series analyses based on varve thickness data that can be correlated to climatic trends and cycles. Time series analyses identified cycles with statistically significant periodicities that were attributed to the Quasi-biennial Oscillation (2.3 years), El Niño Southern Oscillation (2–7 years), the 11-year Schwabe Sunspot cycle and a possible Pacific Decadal Oscillation (50–70 years). This research not only provides baseline chronostratigraphic data that allow the correlation between freeze cores subsampled for various proxies, but also documents the dynamics of the climate drivers that influence the deposition of both organic matter and inorganically precipitated calcite. Crawford Lake is currently a candidate site under consideration for the Global boundary Stratotype Section and Point (GSSP) to define the Anthropocene series/epoch. Full article

The uptake of COVID-19 vaccination in Wales is high at a population level but many inequalities exist. Household composition may be an important factor in COVID-19 vaccination uptake due to the practical, social, and psychological implications associated with different living arrangements. In this study, the role of household composition in the uptake of COVID-19 vaccination in Wales was examined with the aim of identifying areas for intervention to address inequalities. Records within the Wales Immunisation System (WIS) COVID-19 vaccination register were linked to the Welsh Demographic Service Dataset (WDSD; a population register for Wales) held within the Secure Anonymised Information Linkage (SAIL) databank. Eight household types were defined based on household size, the presence or absence of children, and the presence of single or multiple generations. Uptake of the second dose of any COVID-19 vaccine was analysed using logistic regression. Gender, age group, health board, rural/urban residential classification, ethnic group, and deprivation quintile were included as covariates for multivariable regression. Compared to two-adult households, all other household types were associated with lower uptake. The most significantly reduced uptake was observed for large, multigenerational, adult group households (aOR 0.45, 95%CI 0.43–0.46). Comparing multivariable regression with and without incorporation of household composition as a variable produced significant differences in odds of vaccination for health board, age group, and ethnic group categories. These results indicate that household composition is an important factor for the uptake of COVID-19 vaccination and consideration of differences in household composition is necessary to mitigate vaccination inequalities. Spatial planning systems and institutions have a significant role in managing non-agricultural land growth in Europe and the assessment of how their implementation impacts on agricultural land consumption is of great significance for policy and institutional improvement. Reducing the area of agricultural land taken for urban development, or eliminating such conversion, is an international policy priority aiming to maintain the amount and quality of land resources currently available for food production and sustainable development. This study aimed to evaluate the impact of land use planning systems and institutional settings on urban conversion of agricultural land in the 265 NUTS2 level EU27 and UK regions. Taking these regions as the unit of our analysis, the research developed and used global and local econometrics models to estimate the effect based on socio-economic, institutional and land use data for the 2000–2018 period. There is limited research focusing on the impacts of institutional settings and planning types of the European countries on the conversion of agricultural land. Furthermore, existing research has not considered the spatial relationships with the determinants of agricultural land conversion and the response variable, therefore, our research aimed to contribute to the literature on the subject. The results showed that the types of spatial planning systems and institution variables significantly impact the conversion of agricultural land to urban uses. Socio-economic indicators and areas of agricultural and urban land have significant impact on agricultural land conversion for any type of spatial planning system. A further result was that decentralization and political fragmentation were positively associated with agricultural land conversion while quality of regional government and governance was negatively associated. A local regression model was assessed to explore the different spatial patterns of the relationships driving agricultural land conversion. The main empirical finding from this model was that there was spatial variation of driving factors of agricultural land conversion in Europe. Full article

Lymphomas (cancers of the lymphatic system) account for 12% of malignant diseases worldwide. Burkitt’s lymphoma (BL) is a rare form of non-Hodgkin’s lymphoma in which the cancer starts in the immune B-cells. We report the synthesis and preliminary studies on the antiproliferative activity of a library of 9,10-dihydro-9,10-ethanoanthracene based compounds structurally related to the antidepressant drug maprotiline against BL cell lines MUTU-1 and DG-75. Structural modifications were achieved by Diels-Alder reaction of the core 9-(2-nitrovinyl)anthracene with number of dienophiles including maleic anhydride, maleimides, acrylonitrile and benzyne. The antiproliferative activity of these compounds was evaluated in BL cell lines EBV MUTU-1 and EBV+ DG-75 (chemoresistant). The most potent compounds 13j, 15, 16a, 16b, 16c, 16d and 19a displayed IC50 values in the range 0.17–0.38 μM against the BL cell line EBV MUTU-1 and IC50 values in the range 0.45–0.78 μM against the chemoresistant BL cell line EBV+ DG-75. Compounds 15, 16b and 16c demonstrated potent ROS dependent apoptotic effects on the BL cell lines which were superior to the control drug taxol and showed minimal cytotoxicity to peripheral blood mononuclear cells (PBMCs). The results suggest that this class of compounds merits further investigation as antiproliferative agents for BL. Full article

Despite rapid changes in vehicle technology and the expansion of IT-based mobility solutions, travel habits must be changed to address the environmental and health implications of increasing car dependency. A significant amount of research focuses on commuting, which comprises the largest share of annual vehicle miles travelled. However, non-work trips are also significant, especially when considering trip frequency. Using empirical data (N = 1298) from an urban-rural region and bivariate statistical analysis, the relationship between the land use–transport configuration (6 types) and travel behaviour patterns is examined for 14 non-work destinations. The land use characterisation used in this research includes an updated means of representing a land use mix. By defining the typologies of land use and transport for use in the analysis, the findings can be directed towards contrasting area types in the region. A strong statistically significant association between the land use–transport configuration and mode-share for 14 non-work journey purposes is found. Using regression modelling, income and car ownership are identified as key influences on travel behaviour patterns. The results of both analyses show that, for non-work trips, the transport–land use relationship is as important as key socio-demographic indicators. However, the results for reductions in car travel are relatively small for the area typologies outside the inner-city core. This indicates that efforts to provide alternatives to car travel in order to mitigate car dependency should be prioritised in these outer urban areas. Appropriate management of spatial structure for non-work activity types such that active mode use is possible is essential. This will resolve some of the important environmental and health impacts of car dependency. Full article

In this study, environmental sustainability implications of planned rail infrastructure investments on the urban form and development in the Greater Dublin Region (GDR) have been analysed incorporating the scenario analysis approach. Various scenarios are developed using the MOLAND Model applications including: A baseline scenario incorporating a continuation of the present dispersed pattern of urban development and an alternative scenario with rail-oriented corridor development, under varying conditions of economic growth. An alternative scenario was also developed for the recessionary development case considering the prolonged recession in the GDR. Further explorations incorporating a Cost-Benefit Analysis (CBA) approach are developed to evaluate the sustainability implications of different land development scenarios in the Dublin Region. This is assisted by focussing on the impacts of rail investments on urban form and development as raised in the international comparative literature. The findings from the CBA assessment positively indicate that containment policies-as represented by the public transport oriented development indicate benefits over the dispersed development case by reducing the negative consequences of sprawl type of developments. In contrast, dispersed development in the baseline scenario indicates costs of continuation of such development patterns exceed the benefits in the long term. This study will contribute to policy support evaluation measures relating to the integration of scenario analysis tool with the CBA approach in assisting the evaluation of new transport infrastructure proposals. In the context of sustainable urban development, the application of selected indicators integrated with scenario simulation and analysis can contribute to evidence-based decision making. This paper discusses the application of land use modelling and opportunity mapping approaches to evaluate regional development scenarios for the Greater Dublin Region in the period to 2026 evolving from research initially developed with the Dublin Regional Authority. This involved the simulation of four different future regional development scenarios using an adapted version of the MOLAND model with opportunity maps based on combined spatial indicators corresponding to these scenarios. The results produce valuable information for policy makers and planners assisting the evaluation of the consequences of their decisions in both a spatial and temporal context. This paper aims to show how current and future planning and economic policy can make targeted and evidence-based policy interventions and achieve resource efficiencies through the use of scenario analysis.
