Special Issue Information

Dear Colleagues,

Endocrine-Disrupting Chemicals (EDCs) are a paramount concern in environmental health, with pervasive exposure linked to varied adverse effects in both humans and wildlife. This Special Issue, titled "Exposure and Effects of Endocrine Disrupting Chemicals," will compile cutting-edge research that deepens our understanding of the identification, exposure pathways, toxicological mechanisms, and ecological and health impacts of prevalent and emerging EDCs. Our primary focus will be on Bisphenol A (BPA), Per- and Polyfluoroalkyl Substances (PFAS), and their analogues while also welcoming significant contributions on other relevant EDCs.

The scope of this Special Issue is comprehensive and multidisciplinary, inviting original research and review articles that address, but are not limited to, the following key areas: (1) Mixture Toxicity; (2) Next-generation NAMs-based Assessment; (3) In silico/in vitro Mechanistic research; (4) Virtual cells, Virtual life forms; (5) New Pollutant Toxicity Assessment Tools. The ultimate purpose of this Special Issue is to provide a platform for disseminating novel findings that can inform regulatory decisions, advance preventive public health strategies, and stimulate future research directions in the field of endocrine disruption.

This Special Issue is positioned to address several of these frontier areas:

(1) Moving Beyond Single Chemicals;

(2) Elucidating Novel Mechanisms;

(3) Focus on Understudied BPA, PFAS or other Endocrine Disrupting Chemicals;

(4) Big Data Analysis Methods and Tools Based on Artificial Intelligence.

By consolidating research on these cutting-edge themes, this Special Issue will serve as a valuable and timely resource that not only expands upon the existing foundation of EDC knowledge but also charts a course for the next stage of scientific inquiry into the exposure and effects of these critical environmental contaminants.

Endocrine-disrupting chemicals (EDCs) are a heterogeneous group of exogenous compounds capable of interfering with hormonal homeostasis and endocrine-regulated physiological processes. Their widespread occurrence in food, water, air, consumer products and industrial materials has raised increasing concern regarding their contribution to chronic disease burden. This review synthesizes current evidence on the exposure characteristics, molecular mechanisms, health effects, and prevention strategies related to major EDC classes, including bisphenol A and phthalates, dioxins and polychlorinated biphenyls, per- and polyfluoroalkyl substances, pesticides, and brominated flame retardants. Evidence indicates that EDCs may act through receptor-mediated signaling, altered hormone synthesis and metabolism, oxidative stress, mitochondrial dysfunction, immune modulation, and epigenetic mechanisms, with effects that may vary according to dose, timing, sex, age, and developmental susceptibility. Reported health outcomes include metabolic and cardiovascular disorders, reproductive dysfunction, hormone-dependent cancers, thyroid disruption, immune dysregulation, and adverse developmental effects. Although complete avoidance is unrealistic, exposure reduction and risk mitigation can be achieved through coordinated individual, clinical, environmental, and regulatory interventions. A life-course approach is essential to limit the health burden associated with endocrine disruption. Full article
