Dear Colleagues,
Cancer treatment has advanced significantly in the past few decades, yet therapy resistance continues to limit long-term clinical benefit. Many patients initially respond to endocrine therapy, chemotherapy, targeted therapy, or immunotherapy, but then either relapse or progress occurs. One reason for this lack of durability is the ability of cancer cells to adapt to treatment pressure. Metabolic reprogramming is now recognized as a critical part of this adaptation, enabling tumor cells to sustain energy production, survive stress, and acquire more aggressive characteristics.
These metabolic alterations are often widespread and context-dependent. Resistant cancer cells can modify glucose metabolism, lipid synthesis and oxidation, amino acid metabolism, mitochondrial activity, and redox balance. The tumor microenvironment can further modulate these processes. For instance, interactions with stromal cells, immune cells, extracellular metabolites, and nutrient-deprived conditions may influence how cancer cells respond to treatment. Therefore, a better understanding of metabolic adaptation could help identify new therapeutic vulnerabilities in resistant tumors.
This Special Issue focuses on the role of cancer metabolism in therapy resistance. We invite original research articles, review papers, and perspectives on metabolic plasticity, tumor–microenvironment interactions, metabolic biomarkers, and therapeutic strategies targeting cancer metabolism. Studies using metabolomics, multi-omics analysis, systems biology, experimental cancer models, and translational or clinical approaches are particularly encouraged.
We are especially interested in studies related to endocrine-resistant breast cancer, treatment-induced metabolic adaptation, and precision oncology approaches based on metabolic signatures. We also welcome manuscripts investigating natural products and bioactive compounds that regulate cancer metabolism or improve the response to anticancer therapy.
Through this Special Issue, we will bring together basic, translational, and clinical studies that provide new insight into metabolic mechanisms of therapy resistance. We hope that this collection will help identify actionable metabolic targets and support the development of more effective strategies for patients with treatment-resistant cancers.
