Special Issue Information

Dear Colleagues,

Translational neurooncology has significantly emerged by the use of novel techniques including high-resolution omics, advanced spatial profiling, and improved preclinical platforms that are capable of investigating how cell–cell interactions shape tumor evolution, therapeutic response, and resistance. However, potent novel treatment options are urgently needed due to the lack of approved systemic therapy options, particularly in progressive tumor situations.

Therefore, recent mechanistic and translational work linking intratumoral heterogeneity, tumor–microenvironment crosstalk (including astrocytes, microglia/macrophages, endothelial cells, and infiltrating lymphocytes and glioma cells), and emergent molecular vulnerabilities are from high translational relevance. Key themes include single-cell and spatial transcriptomic mapping of glioma niches, ligand–receptor signaling networks that sustain stem-like states and immunosuppression, and the evolving landscape of actionable genomic and epigenomic alterations enabling precision approaches.

This Special Issue aims to highlight recent advances spanning from biomarkers of glioma cell–cell interactions, harboring prognostic and predictive value to novel molecular-guided therapies—targeted small molecules, epigenetic modulators, and combination regimens designed to overcome adaptive resistance. We are interested in original research articles and thoughtful reviews mapping the current status of literature.

We consider contributions addressing the most recent advances by integrating mechanistic insights on glioma biomarkers, including cell–cell adhesion molecules with modern molecular stratification systems to accelerate the translation of durable, personalized therapies for primary brain tumors

Topics of interest for publication include, but are not limited to, the following:

  • Assessment of glioma cell–cell interactions and their role in glioma progression.
  • Novel biomarkers harboring predictive and prognostic relevance in CNS tumor malignancies.
  • Novel therapeutic targets including the respective tumor associated microenvironments.
  • Addressing tumor resistance mechanisms in primary and progressive CNS tumors.
  • Molecular-guided therapy regimes for molecular-stratified CNS tumor entities.
