Dear Colleagues,
Nearly all pregnant individuals will take at least one medication, whether prescription or over-the-counter, at some point during pregnancy. Yet, pharmacokinetics and pharmacodynamics in pregnancy often remain a “black box” due to a lack of rigorously performed human studies and limitations in animal and computer modeling studies, as well as incomplete understanding of placental drug transfer and potential fetal effects. In fact, pregnant individuals are often excluded from clinical drug trials, which, on one hand, protects individuals and fetuses from potential unknown harms, but on the other hand, severely limits knowledge of medication safety and efficacy among pregnant populations. The focus of this Special Issue is to highlight recent developments in pharmacokinetics, pharmacodynamics, computer modeling, clinical management, and policy practices regarding drugs used in the periconception, pregnancy, and/or postpartum/breastfeeding time periods. The goals of this Special Issue are to bring to light new research in pharmacotherapy in pregnancy, whether related to novel classes of medications (e.g., GLP1 receptor agonists, immunotherapy, gene therapy), new insights into established medications (e.g., safe alternative drug regimens in pregnancy), and investigations into the physiological changes of pregnancy and metabolism that affect drug properties/effects, as well as reviews and perspectives on balancing maternal and fetal risks and benefits when selecting pharmacotherapy in pregnancy. Original research and reviews are all welcome. Key topic themes include but are not limited to PK studies in human pregnancy, the role of artificial intelligence in pharmacology in pregnancy, emerging medication classes or controversies, drugs intended for fetal benefit/fetal therapy, placental drug transfer and teratology.

Submitted manuscripts should not have been published previously, nor be under consideration for publication elsewhere (except conference proceedings papers). All manuscripts are thoroughly refereed through a single-blind peer-review process.
