Molecular Strategies for Vital Pulp Therapy and Endodontic Regeneration

Dear Colleagues,

Recent developments in dental biomaterials demonstrate a clear shift toward molecular strategies that actively influence tissue behaviour in both vital pulp therapy and endodontic regeneration.

Emerging biomaterials increasingly incorporate smart, stimulus-responsive features that interact dynamically with the biological microenvironment. pH-responsive formulations, antimicrobial or immunomodulatory nanocarriers, and drug–device systems can release therapeutic agents precisely in response to local cues. By sensing biochemical or mechanical stimuli, these materials enable the targeted delivery of factors that regulate mineralization, modulate inflammation, or promote antimicrobial activity. Such adaptive mechanisms support minimally invasive, biologically driven approaches in vital pulp therapy.

In regenerative endodontics, molecular strategies are similarly designed to coordinate key processes such as cell migration, angiogenesis, and neurogenesis. Scaffolds with defined degradation profiles and tailored signalling cues can guide the differentiation and spatial organization of cells, promoting hierarchical tissue formation within the root canal. These principles may allow the restoration of a functional pulp–dentin complex.

For this Special Issue, we invite the submission of research featuring the molecular design, characterization, and clinical translation of innovative restorative materials and novel scaffolds for endodontic tissue engineering. By integrating molecular insights into material innovation, these approaches can achieve predictable tissue preservation and regeneration, reshaping the biological foundation of restorative and endodontic therapy.
