Special Issue Information

Dear Colleagues,

This Special Issue provides a comprehensive framework for understanding brain metastasis, encompassing its sequential steps, underlying molecular mechanisms, and emerging therapeutic strategies. Recent advances in oncology have significantly enhanced the understanding and management of brain metastases, a common and serious complication of advanced malignancies. Progress in molecular profiling has identified tumor-specific driver proteins and critical signaling pathways that facilitate cancer cell traversal across the blood–brain barrier, enabling improved risk stratification and the development of targeted therapies. Targeted agents and next-generation tyrosine kinase inhibitors with enhanced central nervous system penetration have demonstrated promising intracranial efficacy across multiple cancer types. In parallel, immunotherapies, particularly immune checkpoint inhibitors, have shown durable responses in selected patients with brain metastases. Advances in liquid biopsy technologies are further improving early detection and monitoring of treatment. From a therapeutic standpoint, stereotactic radiosurgery and other precision radiotherapy techniques have reduced neurotoxicity while maintaining effective local control. Collectively, these developments are transforming brain metastasis management toward personalized, multidisciplinary care, thereby improving survival and quality of life.

The purpose of this Special Issue is to present the new insights into the development of brain metastases, focusing on protein-based biomarkers, underlying molecular mechanisms, and advances in therapeutic strategies.

This Special Issue welcomes reviews as well as original research articles, which should be submitted by 5 December 2026.
