Zingerone as a Neuroprotective Agent Against Cognitive Disorders: A Systematic Review of Preclinical Studies

Cognitive problems are associated with impaired learning ability and memory dysfunction. Neuroinflammation has been identified as an important factor in the progression of anxiety and depressive disorders. Zingerone is a phenolic alkanone derived from ginger (Zingiber officinale Roscoe), which is known for its antioxidant and anti-inflammatory properties. A number of studies have investigated the effect of zingerone on neuroinflammation and cognitive impairment. However, this evidence has not been systematically reviewed. This study sought to systematically review the effect of zingerone on neuroinflammation and neurobehavioural changes associated with memory and learning impairment and anxiety-like and depressive-like behaviours. A systematic review was conducted using pre-defined search criteria on Google Scholar, Scopus and Web of Science. The records obtained were screened based on inclusion criteria, and data was extracted from the included studies. Out of the 482 studies that were identified, only 9 studies met the inclusion criteria. Neuroinflammatory markers such as interleukin 1β (IL-1β), interleukin 6 (IL-6), tumour necrosis factor-alpha (TNF-α) and ionized calcium binding adaptor molecule (IBA-1), as well as behavioural parameters including Morris water maze, Y-Maze, recognition test, passive avoidance test, elevated plus maze, sucrose preference test and forced swimming test were measured. Zingerone exhibited anti-neuroinflammatory effects by improving IL-1β, IL-6 and TNF-α levels. However, zingerone did not show any significant changes on activated microglia. The anti-neuroinflammatory mechanisms of zingerone were linked to the inhibition of nuclear factor kappa B (NF-kB) activation and the NOD-like receptor family, pyrin domain-containing 3 (NLRP3) inflammasome, as well as the reduction in neuronal nitric oxide synthase (nNOS). The anxiolytic and anti-depressive effects of zingerone were also associated with an improvement in cortical cholinergic transmission, the mitigation of oxidative stress and the upregulation of neurotransmitters such as serotonin and dopamine. This review provides scientific evidence on the cognitive enhancing and neuroprotective mechanisms of zingerone, which may be beneficial for future experimental investigations. Full article

Background/Objectives: The present study was aimed at documenting S. maltophilia occurrence in dogs with skin ailments, investigating its virulence, biofilm-forming ability, antimicrobial susceptibility, and zoonotic potential to inform preventive and therapeutic strategies against multidrug resistant S. maltophilia infections. Methods: Skin swabs (n = 300) were collected from dogs with dermatological ailments. Isolation was performed using selective media and confirmed with molecular methods, validated by MALDI Biotyper. Antimicrobial susceptibility testing and efflux activity assessment were conducted. Resistance genes related to sulfonamides, quinolones, and β-lactams were screened. Virulence was assessed by biofilm formation, motility, and virulence gene profiling. Results: In total, 15 S. maltophilia (5%) isolates were identified. All 15 isolates were susceptible to trimethoprim-sulfamethoxazole, enrofloxacin, gatifloxacin, levofloxacin, minocycline, and tigecycline, but resistant to cefpodoxime and aztreonam. The following resistance genes qnr (93.3%), blaOXA-48 (46.7%), blaKPC (33.3%), blaNDM (33.3%), blaCTX-M (20%), blaSHV (20%), and blaTEM (6.7%) were detected. All 15 isolates displayed high efflux activity. Overall, 9 isolates (60%) were strong biofilm producers, and 6 (40%) were moderate. Virulence genes such as virB, motA, rmlA, and fliC were present in all 15 isolates, with others varying in frequency. All isolates exhibited swimming motility. Heat map clustering showed diverse profiles, with no identical isolate patterns. Correlation analysis indicated positive associations between several antimicrobial resistance and virulence genes. Conclusions: This study underscores the zoonotic potential of S. maltophilia from dogs, advocating for a One Health approach to mitigate infection risks and limit the spread of virulent multidrug resistant pathogens. Sperm cryopreservation and assisted reproduction are powerful tools for the conservation of endangered species. The domestic cat has been a useful model for studying wild felid reproductive biology due to the limited availability of endangered individuals for experimental research. Here, we investigate the effect of cryodiluents (TEST vs. Biladyl) and the timing of glycerol addition (before vs. after refrigeration, in one vs. three steps, respectively) on post-thaw sperm quality (motility, acrosome integrity) and their subsequent in vitro fertilization (IVF) ability with homologous oocytes. The results showed no statistically significant differences in sperm traits when samples were cryopreserved in TEST or Biladyl, or when glycerol was added in one or three steps. Motile sperm and intact acrosomes were significantly correlated before and after cryopreservation, indicating consistent relationships in fresh and thawed samples. The use of Biladyl significantly reduced IVF rates after cryopreservation compared to fresh sperm. Cryopreservation in TEST led to IVF rates that were not significantly different from those of fresh sperm. Using swim-up after thawing, or adding 1 mM pentoxifylline, did not enhance IVF results. Overall, a TEST cryodiluent with 4% glycerol added in one step is a reliable option for preserving epididymal cat spermatozoa. Full article

Background: To cope with their horizontal swimming phases, water polo players use different swimming techniques, such as specific variants of the crawl swimming style. Thus, this study aimed to investigate the swimming skills of young water polo players. Methods: An all-out 25-m sprint swimming test in crawl style was completed by 273 international youth water polo players (age = 14.0 ± 0.8 yrs) in two modalities: basic crawl with the head in the water (25CHeadIN), and a crawl performed while dribbling the ball (25CBall). Results: We registered an average time of 14.79 and 15.64 s for 25CHeadIN and 25CBall, respectively, in which the ball dribbling increased to 5% of the swimming time. A swimming skill index (25CSIC) was calculated to account for differences in ball dribbling speeds, which, considering our international sample and in the absence of previous data, we could speculate as the first international standard value for 14-year-old male water polo players competing at international level. The averaged values for 25CSI and 25CSIC were 0.94 ± 0.04 (a.u.) and 1.52 ± 0.15 (a.u.), respectively. Factor analysis indicated that swimming with and without the ball are structurally distinct technical skills, highlighting the specificity of these water polo players’ abilities. Moreover, the study shows significant differences (p < 0.05) between players from different countries and despite some limitations, its results provide valuable insights for the assessment and development of sprint swimming skills in youth water polo players. Conclusions: In summary, the findings of this research provide practical implications for training, player selection, player development and the optimization of youth water polo player performance. Objectives: Water polo players ought to possess various physical capacities and well-developed cognitive functions that reflect the requirements of their specific playing position. Therefore, the objective of this study was to compare the cognitive performance, anthropometric characteristics and specific swimming capacities of youth water polo players in different playing positions. Methods: The present cross-sectional study involved 106 youth water polo players. The subjects were recruited as part of a project for talent identification and selection for the Croatian National Water Polo Team. Testing included anthropometric measurements, specific swimming capacities and cognitive performance (Stroop test). Results: Among the 106 youth water polo players, there were 15 goalkeepers (14.2%), 21 center-defenders (19.8%), 17 center-forwards (16.0%), 34 drivers (32.1%) and 19 wings (17.9%), with the mean age of 14.14 ± 0.38 years. The wings performed faster than center-forwards in both StroopOff time (wings: 57.14 ± 10.04 s vs. center-forwards: 67.03 ± 9.72 s, p = 0.016) and StroopOn time (wings: 66.18 ± 15.86 s vs. center-forwards: 80.24 ± 15.64 s, p = 0.019). Conclusions: In conclusion, this study demonstrated significant differences between different playing positions in youth water polo players, specifically between center-forwards and wings. They performed faster than center-forwards in all tested variables of the Stroop test, measures of psychomotor ability, response inhibition and motor speed, as well as in specific swimming capacities measured in the 50 m crawl and the 400 m crawl. The results of this study provide a valuable foundation for establishing developmental recommendations for different playing positions, aimed at improving player’s performance. These recommendations should take into account anthropometric characteristics, specific functional swimming capacities and cognitive functions that influence players’ game intelligence, which can be enhanced through properly designed training programs. Background: Flowering members of the globally diffused Rosaceae family include popular plants, such as apple, almond, and cherry, which play a fundamental role as honeybee nectariferous and polleniferous agents. Through the production of honey, these plants can also play an indirect role in the prevention and treatment of many diseases, including infections, fighting the occurrence of resistant microorganisms, and concurrently stimulating the growth of beneficial bacteria. Objectives: This study focused on the effect of some Rosaceae plants’ honey, including hawthorn, cherry, raspberry, almond, and apple, against the pathogens Acinetobacter baumannii, Escherichia coli, Klebsiella pneumoniae, Listeria monocytogenes, Pseudomonas aeruginosa, and Staphylococcus aureus. Results: Results demonstrated the honey’s ability to impair swimming motility. A crystal violet test indicated that honey could inhibit the formation and stabilization of biofilms, with inhibition rates up to 59.43% for immature biofilms (showed by apple honey against A. baumannii) and 39.95% for sessile bacterial cells in mature biofilms (when we used cherry honey against S. aureus). In the test with 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, cherry and apple honey were the most effective in inhibiting sessile cell metabolism honey in both immature (56.47% cherry honey vs. K. pneumoniae) and mature biofilms (54.36% apple honey vs. A. baumannii). Honey stimulated the growth of Lactobacillus bulgaricus, Lacticaseibacillus casei Shirota, Lactobacillus gasseri, Lacticaseibacillus plantarum, and Lacticaseibacillus rhamnosus; hawthorn, raspberry, and almond honey significantly increased the in vitro adhesion capacity of L. bulgaricus and L. casei Shirota. Tests with probiotic supernatants demonstrated honey’s ability to inhibit the biofilm formation and metabolism of the pathogens. Conclusions: Our results encourage further studies to assess the potential application of Rosaceae honey for food preservation and in the health field, as it could fight the antimicrobial resistance of food and clinical pathogens, and potentially enhance the host’s gut wellness. The use of honey for nanotechnological and biotechnological approaches could be suggested too. Full article

The Agrobacterium fabrum C58 is a phytopathogen able to infect numerous species of cultivated and ornamental plants. During infection, bacteria genetically transform plant cells and induce the formation of tumours at the site of invasion. Bacterial cell wall components play a crucial role in the infection process. Lipopolysaccharide is the main component of Gram-negative bacteria’s outer leaflet of outer membrane. Its lipophilic part, called lipid A, is built of di-glucosamine backbone substituted with a specific set of 3-hydroxyl fatty acids. A. fabrum incorporates a very long chain hydroxylated fatty acid (VLCFA), namely 27-hydroxyoctacosanoic acid (28:0-(27OH)), into its lipid A. A. fabrum C58 mutants deprived of this component due to mutation in the VLCFA’s genomic region, have been characterised. High-resolution mass spectrometry was used to establish acylation patterns in the mutant’s lipid A preparations. The physiological properties of mutants, as well as their motility, ability to biofilm formation and plant infectivity, were tested. The results obtained showed that the investigated mutants were more sensitive to environmental stress conditions, formed a weakened biofilm, exhibited impaired swimming motility and were less effective in infecting tomato seedlings compared to the wild strain. Background: Parkinson’s disease (PD) is a neurodegenerative disorder marked by motor deficits and non-motor symptoms, such as depression, which are associated with dopaminergic loss and α-synuclein aggregation in the brain. Objectives: This study investigated the neuroprotective effects of a hydroalcoholic extract of the purple fruit of Eugenia uniflora (PFEU) on motor ability and depressive-like behaviors in a PD model induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) in female Wistar rats. Methods: Rats received intranasal administration of MPTP or vehicle, followed by 14 days of oral administration of PFEU (300 or 2000 mg/kg, administered once daily) or vehicle. Depressive-like behavior was assessed using the splash and forced swimming tests, while motor ability was evaluated using the rotarod and open field tests. On day 15, hippocampal tissue was collected for immunoreactivity analysis. Results: MPTP treatment induced depressive-like behavior, which was significantly reversed by PFEU, as evidenced by increased grooming and decreased immobility. No motor coordination or locomotion deficits were observed. Furthermore, PFEU treatment prevented the MPTP-induced increase in hippocampal α-synuclein, p-p53, and Bax while restoring Bcl-2 levels, suggesting neuroprotective effects through the modulation of apoptotic pathways and α-synuclein. Conclusions: These findings support PFEU’s potential as a neuroprotective agent for MPTP-induced depressive-like behavior in female rats, highlighting its molecular mechanisms. Full article

Mature, vital, and motile spermatozoa are essential for reaching the oocyte and binding to hyaluronic acid (HA) in the cumulus oophorus matrix. This study aims to determine the relationship between sperm-migration ability and HA-binding potential, as well as the relationship between sperm concentration and motility. Semen samples were collected from 702 men aged 20–56 years (median 34.8). We evaluated the sperm concentration and motility from basic semen analysis, the swim-up test (expressed as millions per mL and the migration efficiency percentage), and the hyaluronan-binding assay (HBA). A moderate positive correlation was found between the migration test results and HBA (R = 0.48). The highest correlation was observed between the concentration of motile spermatozoa and the migration test results (R = 0.85) and HBA (R = 0.4). The sperm migration efficiency strongly correlated with progressive motility (R = 0.6). Although significantly higher sperm migration was observed in patients with normal HBA results, the results of the functional tests were found to differ in some cases. For infertility treatment, the current diagnostic algorithm should be enhanced with more comprehensive seminological methods that assess the sperm-migration ability and HA-binding potential. We also recommend incorporating the swim-up method into the diagnostic protocol before planning assisted reproductive technology (ART) treatment. Full article

Nicotine dependence is an important cause of excessive exposure to tobacco combustion compounds in most smokers. Nicotine replacement therapy is the main method to treat nicotine dependence, but it still has its shortcomings, such as the inability to mitigate withdrawal effects and limited applicability. It has been hypothesized that a combination of low-dose nicotine and caffeine could achieve the same psychological stimulation effect as a high dose of nicotine without causing nicotine withdrawal effects. To establish a model of nicotine dependence, male C57BL/6J mice were subcutaneously injected four times a day with nicotine (2 mg/kg) for 15 days and fed with water containing nicotine at the same time. They were randomly divided into four groups. After 24 h of withdrawal, different groups were injected with saline, nicotine (0.25 mg/kg or 0.1 mg/kg), or nicotine (0.1 mg/kg) and caffeine (20 mg/kg). Behavioral and physiological changes were evaluated by an assessment of physical signs, open field tests, elevated plus maze experiments, forced swimming tests, hot plate tests, and new-object-recognition tests. The changes in dopamine release in the prefrontal cortex (PFC) and ventral tegmental area (VTA) in the midbrain were analyzed using ELISA. The results showed that a combination of caffeine and nicotine could effectively relieve nicotine withdrawal syndrome, increase movement ability and pain thresholds, reduce anxiety and depression, enhance memory and cognitive ability, and increase the level of dopamine release in the PFC and VTA. Thus, caffeine combined with nicotine has potential as a stable and effective treatment option to help humans with smoking cessation. Full article

The purpose of this study was to evaluate the motor development of infants aged 3–12 months participating in water activities. Participants in the study included 43 infants aged 3–12 months who attended water activities classes at Warsaw City swimming pools (n = 21) and infants not attending any classes (n = 22). Two methods of assessing motor development were used in the study: the Alberta Infant Motor Scale (AIMS) and the Early Motor Development Questionnaire (EMQ). The raw scores of the AIMS test and the EMQ questionnaire were statistically significantly different (p < 0.05) in both groups between the first and second examinations. In contrast, there was a statistically significant improvement in motor development measured by AIMS and expressed in percentiles (p = 0.002) and in percentiles for Polish children (p = 0.030) in the water group. The age-independent total score of the EMQ before and after the intervention did not change significantly (p = 0.149). The water environment has the potential to support the motor development of infants aged 3–12 months. Full article

In this research, several analyses were carried out on concentrated fractions of Mexican oregano essential oil (Poliomintha longiflora Gray) in order to determine its ability to inhibit the growth and the motility of Escherichia coli (swimming), Pseudomonas aeruginosa (swimming), and Proteus vulgaris (swarming); these Gram-negative bacteria associated with urinary tract infections are motile due to the presence of flagella, which is considered an important virulence factor that favors their motility when trying to reach the target organ and cause an infection. Also, the resistance pattern to antibiotics of each strain was determined. The results showed resistance pattern (8 out of 12 antibiotics tested) for P. aureginosa, while E. coli and P. vulgaris were resistant to 4 antibiotics out of the 12 tested. On the other hand, fractionated oregano caused an inhibition of growth and a reduction in motility, varying between fractions and among bacteria. Fraction 4 showed major growth reduction, with MBC values ranging from 0.002 to 23.7 mg/mL. Treatment with fractionated oregano (F1, F2, F3, F4) reduced the motility by 92–81% for P. vulgaris, 90–83% for E. coli, and 100–8.9% for P. aeruginosa. These results demonstrated a higher performance with a lower application dose due to its high content of Carvacrol and Thymol; unlike other concentrated fractions, this synergy of oxygenated monoterpenes may cause greater antimicrobial activity. The present study aimed to explore whether water flow velocity could affect the swimming ability and overall energy metabolism of wild Amur grayling (Thymallus grubii). Swimming performance was assessed by measuring critical swimming speed (Ucrit), burst speed (Uburst), and oxygen consumption rate (MO2) based on the stepped velocity test method. Our results showed that the absolute values of Ucrit and Uburst tended to increase with body length. In contrast, the relative values of Ucrit and Uburst tended to decrease and increase, respectively. MO2 in Amur grayling was elevated with increasing velocity, suggesting relatively high swimming efficiency. We also measured the biochemical indices related to energy metabolism. Lactate dehydrogenase, hexokinase, and pyruvate kinase activities significantly increased (p < 0.05). Hepatic glycogen, glucose, and muscle glycogen contents decreased with the increasing trend of velocity (p < 0.05), the lactic acid contents of the blood and muscles increased significantly with the increase in velocities (p < 0.05), and changes in creatine phosphate content showed no significant difference (p > 0.05). The results not only denote the relationship between body size and swimming speed but also show the effects of water flow velocity on energy metabolism in Amur grayling. The results provide basic data for the construction of fish passage. Full article

Methamphetamine use disorder (MuD) is a global health problem, with no FDA-approved medications. Our prior work demonstrated that repeated methamphetamine exposure alters the gut microbiota in male rats and results in depressive-like behaviors. In this study, we extend our findings to females and determine whether probiotics block these effects. Male and female rats were administered methamphetamine (2 mg/kg; SC) or saline twice daily with either a combination of two probiotics (Lactobacillus helveticus R0052 and Bifidobacterium longum R0175) or placebo solution for 14 days. Fecal samples were collected at baseline and other days after treatment cessation. Tests of anxiety- and depressive-like behaviors were conducted using open-field and forced-swim assays. Methamphetamine induced anxiety-like behavior in females and anxiety-like and depressive-like behaviors in males. Probiotics blocked the depressive-like effect in males but did not alter anxiety-like effects in either sex. Methamphetamine exposure decreased levels of alpha diversity in both sexes, but sex differences were seen in the ability of probiotics or methamphetamine to alter levels of various bacteria. These findings support the role of the gut–brain microbiome in the depressive effects of repeated methamphetamine exposure in males, suggesting that probiotics may be a viable treatment option for MuD. The Dynamics of the Development of Apneic Breathing Capacity Specific to Synchronized Swimming in Girls Aged 7–14 Years

The purpose of the study was to identify the durations of maintaining apnea, in different static positions, with and without the use of a nose clip, in girls aged between 7 and 14 years,. The study included a total number of 92 girls, grouped by age into four groups of 2-year spans (7–8, 9–10, 11–12, 13–14 years), and depending on the experience of practicing synchronized swimming (6–42 months). In the study we applied five physical tests where apnea maintenance times were recorded in different static positions: Apnea Test of Facial floatation with and without nose clip, Apnea Test of Front tuck with and without nose clip and Apnea Test of Front layout with support to scull. The statistical analysis was performed with SPSS-24. During the study, a program of specific exercises to learn/consolidate the apneic breathing specific to artistic swimming was implemented, for a time interval of 3 months. The results were recorded at the beginning of the study (TI) and at the end of the study (TF). Analyzing the results of the study, we found positive and statistically significant improvements, related to age and experience. The most significant progress, taking into account the averages between the final and initial tests, was recorded in relation to Facial Flotation for 1.301 s for the 7–8-year-old group and 1.110 s for the 9–10-year-old group; the 11–12-year-old group recorded the most positive effect in the Facial Flotation test with a nose clip, with a result of 0.853 s, and in the 13–14-year-old group in the front tuck with nose clip test, a result of 0.807 s was reached. In all tests of the study, the Cohen’s values in all groups fell between 0.184 and 0.478, the size of the effect being small and medium. The ANOVA analysis of variance showed that the differences were statistically significant for p < 0.05 between the arithmetic means of the four groups according to age and sport experiences. For all groups, the value of Wilks’ Lambda was 0.009 (p < 0.01) for age and 0 (p < 0.01) for sports experience, highlighting large differences between groups. We conclude that the development of the ability to maintain apnea specific to synchronized swimming shows an upward trajectory, being conditioned by the training methodology, the age of the subjects and the sports experience. The small and medium values of the effect size highlight the fact that the improvement in apnea maintenance time is dependent on the duration and frequency of the apnea exercises performed in technical conditions specific to synchronized swimming. The training methodology must be adapted to the particularities of age, sports experience and the characteristics of synchronized swimming. Full article
