This paper reports a statistical analysis of a database archiving information on the strengths of the materials in existing Italian bridges having pre- and post-tensioned concrete beams. Data were collected in anonymous form by analyzing a stock of about 170 bridges built between 1960 and 2000 and located in several Italian regions. To date, the database refers to steel reinforcing bars, concrete, and prestressing steel, whose strengths were gathered from design nominal values, acceptance certificates, and in situ test results, all derived by consulting the available documents for each examined bridge. At first, this paper describes how the available data were collected. Then, the results of a statistical analysis are presented and commented on. Moreover, goodness-of-fit tests are carried out to verify the assumption validity of a normal distribution for steel reinforcing bars and prestressing steel, and a log-normal distribution for concrete. The database represents a valuable resource for researchers and practitioners for the assessment of existing bridges. It may be applied for the use of prior knowledge within a framework where Bayesian methods are included for reducing uncertainties. The database provides essential information on the strengths of the materials to be used for a simulated design and/or for verification in the case of limited knowledge. Goodness-of-fit tests make the collected information very useful, even if probabilistic methods are applied. Full article

Introduction: Biodegradable Temporizing Matrix (BTM) is a new synthetic dermal substitute suitable for wound closure and tissue regeneration. The data in paediatric population remain limited. The study purpose is to review the indications for BTM application in paediatric patients; evaluate the short-term and long-term results, including complications and functional outcomes, as well as to share some unique observations regarding the use of BTM in paediatric population. Patients and Methods: Patients undergoing reconstructive surgery and BTM application during the last three years were included. Data collected included patient demographics, primary diagnosis, previous surgical management, post-operative complications and final outcomes. BTM was used in 32 patients. The indications varied including epidermolysis bullosa (n = 6), burns (n = 4), trauma (n = 7), infection (n = 4), ischemia or necrosis (n = 11). Results: The results were satisfying with acceptable aesthetic and functional outcomes. Complications included haematomaunderneath the BTM leading to BTM removal and re-application (n = 1), BTM infection (n = 1) and split-thickness skin graft failure on top of BTM requiring re-grafting (n = 2). Conclusions: BTM can be a good alternative to large skin grafts, locoregional flaps or even free flaps. The big advantages over other dermal substitutes or skin grafts are that BTM is less prone to infection and offers excellent scarring by preserving the normal skin architecture. Specifically in children, BTM might not require grafting, resulting in spontaneous healing with good scarring. In critically ill patients, BTM reduces the operation time and there is no donor site morbidity. BTM should be considered in the reconstructive ladder when discussing defect coverage options in children and young people. Full article

Posterior capsule opacification (PCO), a frequent complication of cataract surgery, arises from dysregulated wound healing and fibrotic transformation of residual lens epithelial cells. While transcriptomic and machine learning (ML) approaches have elucidated fibrosis-related pathways in other tissues, the molecular divergence between regenerative and fibrotic outcomes in the lens remains unclear. Here, we used an ex vivo chick lens injury model to simulate post-surgical conditions, collecting RNA from lenses undergoing either regenerative wound healing or fibrosis between days 1–3 post-injury. Bulk RNA sequencing data were normalized, log-transformed, and subjected to univariate filtering prior to training LASSO, SVM, and RF ML models to identify discriminatory gene signatures. Each model was independently validated using a held-out test set. Distinct gene sets were identified, including fibrosis-associated genes (VGLL3, CEBPD, MXRA7, LMNA, gga-miR-143, RF00072) and wound-healing-associated genes (HS3ST2, ID1), with several achieving perfect classification. Gene Set Enrichment Analysis revealed divergent pathway activation, including extracellular matrix remodeling, DNA replication, and spliceosome associated with fibrosis. RT-PCR in independent explants confirmed key differential expression levels. These findings demonstrate the utility of supervised ML for discovering lens-specific fibrotic and regenerative gene features and nominate biomarkers for targeted intervention to mitigate PCO. Indoor horticulture requires a substantial quantity of electricity to meet crops extended photoperiodic requirements for optimal photosynthetic rate. Simultaneously, global electricity costs have grown dramatically in recent years, endangering the sustainability and profitability of indoor vertical farms and/or modern greenhouses that use artificial lighting systems to accelerate crop development and growth. This study investigates the growth rate and physiological development of cherry tomato plants cultivated in a pilot indoor vertical farm at the Agricultural University of Athens’ Laboratory of Farm Structures (AUA) under continuous and disruptive lighting. The leaf physiological traits from multiple photoperiodic stress treatments were analyzed and utilized to estimate the plant’s tolerance rate under varied illumination conditions. Four different photoperiodic treatments were examined and compared, firstly plants grew under 14 h of continuous light (C-14L10D/control), secondly plants grew under a normalized photoperiod of 14 h with intermittent light intervals of 10 min of light followed by 50 min of dark (NI-14L10D/stress), the third treatment where plants grew under 14 h of a load-shifted energy demand response intermittent lighting schedule (LSI-14L10D/stress) and finally plants grew under 13 h photoperiod following of a load-shifted energy demand response intermittent lighting schedule (LSI-13L11D/stress). Plants were subjected also under two different light spectra for all the treatments, specifically WHITE and Blue/Red/Far-red light composition. The aim was to develop flexible, energy-efficient lighting protocols that maintain crop productivity while reducing electricity consumption in indoor settings. Results indicated that short periods of disruptive light did not negatively impact physiological responses, and plants exhibited tolerance to abiotic stress induced by intermittent lighting. Post-harvest data indicated that intermittent lighting regimes maintained or enhanced growth compared to continuous lighting, with spectral composition further influencing productivity. Plants under LSI-14L10D and B/R/FR spectra produced up to 93 g fresh fruit per plant and 30.4 g dry mass, while consuming up to 16 kWh less energy than continuous lighting—highlighting the potential of flexible lighting strategies for improved energy-use efficiency. Background/Objectives: There is significant interest in using texture features to extract hidden image-based information. In medical imaging applications using radiomics, AI, or personalized medicine, the quest is to extract patient or disease specific information while being insensitive to other system or processing variables. While we use digital breast tomosynthesis (DBT) to show these effects, our results would be generally applicable to a wider range of other imaging modalities and applications. Methods: We examine factors in texture estimation methods, such as quantization, pixel distance offset, and region of interest (ROI) size, that influence the magnitudes of these readily computable and widely used image texture features (specifically Haralick’s gray level co-occurrence matrix (GLCM) textural features). Results: Our results indicate that quantization is the most influential of these parameters, as it controls the size of the GLCM and range of values. We propose a new multi-resolution normalization (by either fixing ROI size or pixel offset) that can significantly reduce quantization magnitude disparities. We show reduction in mean differences in feature values by orders of magnitude; for example, reducing it to 7.34% between quantizations of 8–128, while preserving trends. Conclusions: When combining images from multiple vendors in a common analysis, large variations in texture magnitudes can arise due to differences in post-processing methods like filters. We show that significant changes in GLCM magnitude variations may arise simply due to the filter type or strength. These trends can also vary based on estimation variables (like offset distance or ROI) that can further complicate analysis and robustness. We show pathways to reduce sensitivity to such variations due to estimation methods while increasing the desired sensitivity to patient-specific information such as breast density. Finally, we show that our results obtained from simulated DBT images are consistent with what we see when applied to clinical DBT images. Full article

Castration remains a common surgical procedure in equids, yet postoperative complications such as hemorrhage, infection, and eventration persist, influencing surgical outcomes. This retrospective study introduced a novel modified semi-closed castration technique aimed at minimizing these complications. Eighty-five sexually intact mature male equids (including horses, ponies, and donkeys) underwent the described inguinal castration procedure under general anesthesia. The technique involved a carefully managed opening and subsequent secure closure of the parietal vaginal tunic, enabling direct inspection of tunic contents while reducing risks associated with fully open or closed approaches. The median surgery duration was 60 min (range, 45–95), with no intraoperative complications. Postoperatively, only two equids (2.35%) exhibited scrotal swelling, which was resolved rapidly with NSAIDs. No cases of hemorrhage, pyrexia, or infection occurred. Follow-up, conducted via owner interviews after a minimum of three months, confirmed complete recovery and primary intention healing in all patients, who returned to normal activity within two days post-surgery. Compared to previously documented complication rates ranging from 2% to 48%, the present approach demonstrates superior results. Additionally, this study proposes refining existing castration technique classifications, focusing explicitly on the vaginal tunic’s status post-procedure (open, semi-closed, closed) for clarity and consistency. This novel semi-closed inguinal technique effectively integrates inspection advantages with closed technique safety, offering significantly reduced complication rates and enhanced postoperative recovery profiles in equids undergoing castration. Doxorubicin (DOX) is widely used for the treatment of several tumors, but considerable dose-dependent side effects on many normal tissues, including bones, have been reported. The aim of the present study was to follow for the first time the kinetics of DOX accumulation/clearance in the non-vascularized intervertebral disc (IVD), as well as to assess the drug’s biological action in the annulus fibrosus (AF) and nucleus pulposus (NP) IVD cells and tissues. DOX was administered intravenously to rabbits before the isolation of IVDs, in which DOX quantification was performed using a highly sensitive LC-HRMS/MS analytical method. The effect of the drug on IVD cells’ physiology was assessed in vitro, while IVD tissue quality post-DOX administration was studied in vivo through histological analysis. DOX delivery was found significantly lower in the IVD compared to the highly vascularized skin, declining from the outer AF to the inner NP. The low DOX concentrations reaching the IVDs had marginal effects on cells’ viability, intracellular redox status, and p38 MAPK activation, while they did not evoke cellular senescence. Most importantly, the drug did not negatively affect ECM integrity, as collagen and proteoglycan content remained stable in vitro and in vivo. Full article

Grapholita molesta is a globally significant fruit pest. Females achieve maximal reproductive output through efficient sperm utilization following a single copulation. Post-mating maturation of eupyrene sperm is a critical step in reproductive success. Here, we report that a male accessory gland-derived serine protease (named GmAGSP1) is essential for this process. GmAGSP1 was only distantly related to other identified sperm-activating SPs, and its transcript was highly expressed in the AG at 48 h after emergence. RNAi-mediated knockdown of GmAGSP1 in males did not affect courtship rate, copulation duration, or mating frequency, whereas male fertility decreased significantly. Mating with GmAGSP1-knockdown males markedly impaired eupyrene sperm maturation in the spermatophores, with phenotypes including failure of eupyrene sperm bundles to dissociate normally and marked reduction in viability of the dissociated eupyrene sperm. Finally, untargeted metabolomic analysis preliminarily demonstrated marked alterations in multiple metabolic pathways within the spermatophore following mating with GmAGSP1-knockdown males. This study advances our understanding of the regulatory mechanism of “sperm activation in the spermatophore’s metabolic microenvironment mediated by male AG-derived SP” while providing critical insights for the development of novel genetic control strategies targeting G. molesta. On 20 April 2024, an extreme rainfall event occurred in Jiangwan Town Shaoguan City, Guangdong Province, China, where a historic 24 h precipitation of 206 mm was recorded. This triggered extensive landslides that destroyed residential buildings, severed roads, and drew significant societal attention. Rapid acquisition of landslide inventories, distribution patterns, and key controlling factors is critical for post-disaster emergency response and reconstruction. Based on high-resolution Planet satellite imagery, landslide areas in Jiangwan Town were automatically extracted using the Normalized Difference Vegetation Index (NDVI) differential method, and a detailed landslide inventory was compiled. Combined with terrain, rainfall, and geological environmental factors, the spatial distribution and causes of landslides were analyzed. Results indicate that the extreme rainfall induced 1426 landslides with a total area of 4.56 km2, predominantly small-to-medium scale. Landslides exhibited pronounced clustering and linear distribution along river valleys in a NE–SW orientation. Spatial analysis revealed concentrations on slopes between 200–300 m elevation with gradients of 20–30°. Four machine learning models—Logistic Regression, Support Vector Machine (SVM), Random Forest (RF), and Extreme Gradient Boosting (XGBoost)—were employed to assess landslide susceptibility mapping (LSM) accuracy. RF and XGBoost demonstrated superior performance, identifying high-susceptibility zones primarily on valley-side slopes in Jiangwan Town. Shapley Additive Explanations (SHAP) value analysis quantified key drivers, highlighting elevation, rainfall intensity, profile curvature, and topographic wetness index as dominant controlling factors. This study provides an effective methodology and data support for rapid rainfall-induced landslide identification and deep learning-based susceptibility assessment. Full article

Background: Acute myeloid leukemia (AML) is a common and aggressive adults hematological malignancies. This study explored megakaryocyte–erythroid progenitors (MEPs) signature genes and constructed a prognostic model. Methods: Uniform manifold approximation and projection (UMAP) identified distinct cell types, with differential analysis between AML-MEP and normal MEP groups. Univariate and the least absolute shrinkage and selection operator (LASSO) Cox regression selected biomarkers to build a risk model and nomogram for 1-, 3-, and 5-year survival prediction. Results: Ten differentially expressed genes (DEGs) related to overall survival (OS), six (AHSP, MYB, VCL, PIM1, CDK6, as well as SNHG3) were retained post-LASSO. The model exhibited excellent efficiency (the area under the curve values: 0.788, 0.77, and 0.847). Pseudotime analysis of UMAP-defined subpopulations revealed that MYB and CDK6 exert stage-specific regulatory effects during MEP differentiation, with MYB involved in early commitment and CDK6 in terminal maturation. Finally, although VCL, PIM1, CDK6, and SNHG3 showed significant associations with AML survival and prognosis, they failed to exhibit pathological differential expression in quantitative real-time polymerase chain reaction (qRT-PCR) experimental validations. In contrast, the downregulation of AHSP and upregulation of MYB in AML samples were consistently validated by both qRT-PCR and Western blotting, showing the consistency between the transcriptional level changes and protein expression of these two genes (p < 0.05). Conclusions: In summary, the integration of single-cell/transcriptome analysis with targeted expression validation using clinical samples reveals that the combined AHSP-MYB signature effectively identifies high-risk MEP-AML patients, who may benefit from early intensive therapy or targeted interventions. Neuroinflammation is a key feature of Alzheimer’s disease (AD), and stem cell therapies have emerged as promising candidates due to their immunomodulatory properties. Neuro-Cells (NC), a combination of unmodified mesenchymal stem cells (MSCs) and hematopoietic stem cells (HSCs), have demonstrated therapeutic potential in models of central nervous system (CNS) injury and neurodegeneration. Here, we studied the effects of NC in APPswe/PS1dE9 mice, an AD mouse model. Twelve-month-old APPswe/PS1dE9 mice or their wild-type littermates were injected with NC or vehicle into the cisterna magna. Five to six weeks post-injection, cognitive, locomotor, and emotional behaviors were assessed. The brain was stained for amyloid plaque density using Congo red, and for astrogliosis using DAPI and GFAP staining. Gene expression of immune activation markers (Il-1β, Il-6, Cd45, Tnf) and plasticity markers (Tubβ3, Bace1, Trem2, Stat3) was examined in the prefrontal cortex. IL-6 secretion was measured in cultured human monocytes following endotoxin challenge and NC treatment. Untreated APPswe/PS1dE9 mice displayed impaired learning in the conditioned taste aversion test, reduced object exploration, and anxiety-like behavior, which were improved in the NC-treated mutants. NC treatment normalized the expression of several immune and plasticity markers and reduced the density of GFAP-positive cells in the hippocampus and thalamus. NC treatment decreased amyloid plaque density in the hippocampus and thalamus, targeting plaques of <100 μm2. Additionally, NC treatment suppressed IL-6 secretion by human monocytes. Thus, NC treatment alleviated behavioral deficits and reduced amyloid plaque formation in APPswe/PS1dE9 mice, likely via anti-inflammatory mechanisms. The reduction in IL-6 production in human monocytes further supports the potential of NC therapy for the treatment of AD. Full article

Background/Objectives: To assess the optical performance of two refractive premium IOLs across pupil sizes and values of corneal spherical aberration (SA). Methods: Two refractive IOLs were evaluated in this study: Tecnis Eyhance and Mini Well. The surface profiles were obtained to calculate the through-object MTF (TO MTF) curves and simulate optotype images. Entrance pupil sizes ranging from 2 to 5.5 and three corneal models were analyzed in the simulation: an average population aberrated cornea, an aberration-free cornea and a post-Lasik myopic cornea. Results: For Model 1 and pupil sizes between 3.0 and 3.5 mm, Mini Well provided acceptable visual quality from far to near distances, whereas Eyhance struggled to maintain visual quality at distances closer than intermediate. For patients with lower-than-normal corneal SA (i.e., more prolate corneas, such as post-hyperopic LASIK) both IOLs exhibited a hyperopic shift in far focus. Conversely, for patients with higher-than-normal corneal SA (i.e., more oblate corneas, such as post-myopic LASIK), the shift occurred in the myopic direction. Despite the implementation of an optimized IOL power to circumvent any shift, the TO MTF nevertheless reflected the interaction between corneal and IOL SA. Furthermore, the Mini Well demonstrated increased tolerance to less negative SA values, while Eyhance exhibited behavior consistent with a monofocal lens for more positive SA values. Conclusions: Surgeons should consider each patient’s corneal asphericity and typical pupil diameter when selecting and calculating the power of the premium IOLs studied, particularly in patients with a history of refractive surgery. Crohn’s disease is a chronic inflammation affecting the gastrointestinal tract. To date, patients are commonly treated with corticosteroids or more aggressive biologics for high-risk subjects. Stem Cell Educator therapy has been successfully utilized to treat patients with type 1 diabetes and other autoimmune conditions. A 78-year-old patient with long-standing Crohn’s disease received one treatment with the Stem Cell Educator therapy, followed by clinical, radiographic, pathological examinations and immune marker testing by flow cytometry. After the treatment with Stem Cell Educator therapy, the patient’s clinical symptoms were quickly improved with normal bowel movements, without abdominal pain or rectal bleeding. Flow cytometry analysis revealed a marked decline in inflammatory markers, such as the percentage of monocyte/macrophage-associated cytokine interleukin-1 beta (IL-1β)+ cells, which reduced from 94.98% at the baseline to 18.21%, and down-regulation of the percentage of chemokine CXCL16+ cells from 91.92% at baseline to 42.58% at 2-month follow-up. Pathologic examination of the biopsy specimens from colonoscopy five weeks and six months post-treatment showed ileal mucosa with no specific abnormality and no significant inflammation or villous atrophy; no granulomas were identified. A follow-up CT scan four and one-half months post-treatment showed no evidence of the previously seen stenosis of the ilio-colonic anastomosis with proximal dilatation. Stem Cell Educator therapy markedly reduced inflammation in the subject with Crohn’s disease, leading to durable clinical, immunological, radiographic, and pathological improvements. Full article

Fluid resuscitation after thermal injury is paramount to avoid burn shock and restore organ perfusion. Both over- and under-resuscitation can lead to unintended consequences affecting patient outcomes. While many studies have examined systemic effects, limited data exist on how fluid resuscitation impacts burn wound progression in the acute period. Furthermore, the mechanisms underlying burn wound progression remain not fully understood. This study used a swine model to investigate how varying resuscitation levels affect peri-burn wound dynamics. Twenty-seven female Yorkshire pigs were anesthetized, subjected to 40% total body surface area burn and 15% hemorrhage, then randomized (n = 9) to receive decision-support-driven (adequate, 2–4 mL/kg/%TBSA), fluid-withholding (under, <1 mL/kg/%TBSA), or high-constant-rate (over, >>4 mL/kg/%TBSA) resuscitation. Pigs were monitored for 24 h in an intensive care setting prior to necropsy. Laser Doppler Imaging (LDI) was conducted pre-burn and at 2, 6, 12, and 24 h post burn to assess perfusion. Biopsies were taken from burn, peri-burn (within 2 cm), and normal skin. RNA was isolated at 24 h for the qRT-PCR analysis of IL-6, CXCL8, and IFN-γ. At hour 2, LDI revealed increased peri-burn perfusion in over-resuscitated animals vs. under-resuscitated animals (p = 0.0499). At hour 24, IL-6 (p = 0.0220) and IFN-γ (p = 0.0253) were elevated in over-resuscitated peri-burn skin. CXCL8 showed no significant change. TUNEL staining revealed increased apoptosis in over- and under-resuscitated peri-burn skin. Differences in perfusion and cytokine expression based on resuscitation strategy suggest that fluid levels may influence burn wound progression. Full article

Mild cognitive impairment (MCI) is the transitional stage between normal cognition and dementia and is associated with arterial stiffness, which may lead to cardiovascular disease. A water-based exercise (W) presents a low-impact activity for the joints and increases resistance compared to exercises performed in the air, which benefits older adults. However, little evidence has been found regarding the effect of W on promoting cognitive and physical performance in older individuals with MCI. Therefore, this study aimed to investigate and compare the post-training effects of W alone and in combination with cognitive training on cognitive function, cardiovascular fitness, and arterial stiffness in older adults with MCI. Forty-six adults with MCI, aged 65 years or older, were enrolled. Participants were divided into two groups: a W group and a water-based exercise combined with cognitive training (W-COG) group. Both groups performed an aerobic exercise program in water for 60 min per/day, 3 day/week, for 12 weeks. Participants in the W-COG group simultaneously performed aerobic exercise and cognitive training in water. Cognitive performance, cardiovascular fitness, and arterial stiffness were examined before and after the intervention. The results revealed improvements in cognitive performance and cardiovascular fitness in both the W and W-COG groups after 12 weeks of intervention. However, there were no significant differences in cognitive and cardiovascular fitness changes between the two groups. Neither the W nor the W-COG groups showed a decrease in brachial pulse wave velocity. Therefore, W interventions have the potential to enhance cognitive function, restore cognition, and improve cardiovascular fitness in older adults with MCI. Full article
