Skin functionalities are instrumental in four main domains: protection, regulation, sensation, and support. However, excessive exposure to ultraviolet (UV) radiation can compromise skin integrity and, in turn, affect its functions, by generating reactive oxygen species (ROS). Aiming to protect skin from UV radiation, sunscreens incorporate UV filters and antioxidants that absorb/reflect UV rays and neutralise free radicals, respectively. Nevertheless, undesired side and ecological effects of conventional UV filters have spurred the search for safer alternatives. Among synthetic antioxidants, thioethers have attracted attention for their redox power and potential medicinal properties. In this context, a library of aminopyrimidine–arylthioether conjugates was synthesised and evaluated for their antioxidant, enzyme-inhibitory and antibacterial activities, as well as for their cytotoxicity in HaCaT cells and potential photoprotective properties. Among the aminopyrimidine-thioethers studied, compound C5 stood out for its antioxidant potential, exhibiting a value of 566.39 mM FeSO4 equivalents per mM of the compound, while compound C2 showed the highest anti-enzymatic potential, inhibiting elastase (45.58%) and tyrosinase activities (34.66%). Regarding photoprotective activity, compound C13 reduced by 33.74% the ROS production induced by UV radiation exposure, at 100 μM, a non-cytotoxic concentration. Finally, compound C7 inhibited the growth of Staphylococcus epidermidis, Staphylococcus hominis and Cutibacterium acnes, at 30 μM. These preliminary results demonstrate that aminopyrimidine–arylthioethers constitute a new class of compounds warranting further investigation for skin protection. Compound C5 showed antioxidant activity in the FRAP assay, comparable to that of the positive control, BHT. Full article

Flowers of melliferous plants, such as Tilia L. and Calluna vulgaris L., are rich in highly active polyphenols and flavonoids. The aim of this study was to evaluate the biological properties of extracts and kombucha ferments from Tilia L. and Calluna vulgaris L. flowers. The phytochemical profile was analyzed by LC-MS. Antioxidant potential was assessed using ABTS and DPPH assays, as well as by measuring intracellular ROS levels in skin cells. Cytotoxicity was determined using Alamar Blue assay in in vitro models. Anti-inflammatory activity was assessed based on IL-6, TNF-α, and COX-2 levels, and antibacterial properties were assessed against Gram-positive and Gram-negative bacteria. ABTS and DPPH assays showed the greatest antioxidant potential for Calluna vulgaris L. 7-day ferments, achieving 90.41 and 63.24% reductions in these radicals, respectively. Resazurin tests showed that in most cases, sample concentrations not exceeding 250 µg/mL did not cause a cytotoxic effect. In anti-inflammatory tests, the compound most strongly inhibited IL-6, reducing the level of this cytokine from 3.55-fold (for the positive control with LPS) to only 2.53-fold. The results demonstrated antimicrobial activity, with a zone of inhibition for S. aureus of 16 mm. Overall, the results indicate that Tilia L. and Calluna vulgaris L. flower extracts and kombucha ferments represent promising natural sources of bioactive compounds with antioxidant, antibacterial, and anti-inflammatory properties. Coleus Lour. and Plectranthus L’Hér. (Lamiaceae) species are recognized as promising sources of bioactive compounds for skin health, supported by their traditional use in skin-related conditions; however, scientific validation of these traditional uses remains limited for several species. In this work, the biological activities of eight Coleus and Plectranthus species (C. barbatus, C. cylindraceus, C. grandidentatus, C. hadiensis, C. madagascariensis, P. ambiguus, P. ecklonii, and P. fruticosus) were evaluated to support their ethnomedicinal relevance. Extracts were assessed for general toxicity (Artemia salina model), antimicrobial activity (well diffusion and microdilution methods) and antioxidant capacity (DPPH, TBARS and cellular assays). In addition, anti-inflammatory activity (ability to suppress nitric oxide), cytotoxicity in skin cell lines (HaCaT and HFF-1) and inhibition of skin-related enzymes (elastase, tyrosinase and collagenase) were evaluated. Antibacterial activity was predominantly observed against Gram-positive bacteria, with no general toxicity observed. P. ambiguous and P. ecklonii showed moderate-to-high antioxidant activity, while anti-inflammatory effects were observed for C. hadiensis and C. madagascariensis and P. ambiguous. None of the extracts showed cytotoxicity. Enzyme inhibition assays highlighted C. cylindraceus and P. ecklonii. HPLC-DAD analysis confirmed the presence of rosmarinic acid. Overall, the findings support the traditional use of Coleus and Plectranthus species for skin-related conditions and highlight their potential as sources of bioactive compounds for dermatological and cosmeceutical applications. Macrococcus is a genus of Gram-positive cocci in the Staphylococcaceae family and a close phylogenetic relative of Staphylococcus. It is not a significant human pathogen but is known to widely colonize different environments, including animal skin and food products. Phylogenetically, Macrococcus is distinct from yet closely related to Staphylococcus, particularly the sciuri group. The species is effectively identified through such molecular markers as hsp60 and 16S rDNA. A key biochemical feature is an identified FAD-dependent oleate hydratase in Macrococcus equipercicus (M. equipercicus). Critically, Macrococcus carries various mobile antibiotic-resistance genes, especially against β-lactams (e.g., mecB, mecD) and macrolides (e.g., mef(F), msr(G)); these genes are located on plasmids, SCCmec-like elements, or resistance islands (e.g., McRImecD), which facilitates their horizontal transfer. Surveillance confirms the widespread presence of methicillin-resistant Macrococcus, often with a multidrug-resistant phenotype, in food animals and their products. Although its own pathogenicity is low, Macrococcus acts as a reservoir and transmission platform for resistance genes: through horizontal gene transfer, it can potentially confer resistance to pathogenic Staphylococcus, thereby posing a threat to animal and public health. This review summarizes the basic biological characteristics and drug resistance-related research progress of the genus Macrococcus; it aims to provide a reference for subsequent studies as well as to establish technical support and a theoretical basis for the epidemiological investigation, drug-resistant strain identification, and clinical drug-resistance risk prevention and control of Macrococcus. Radiotherapy is a highly effective, safe cancer treatment, and about half of all cancer treatments involve lifesaving radiotherapy. Despite huge advances in technology that have made it safer and more effective, it is still not without side effects. They differ from patient to patient and can include fatigue, nausea, skin reactions, and hair loss, but dysbiosis is the most common complication associated with radiotherapy. Probiotics aimed at restoring the microbiome have found widespread use, but the problem of their rapid inactivation in the gastrointestinal tract has not yet been solved. Our study aims to confirm the effectiveness of a novel biomineral complex, based on a powdered clinoptilolite containing a rock loaded with lactobacilli for restoring the intestinal microbiome of mice exposed to radiation. Based on the 16S rRNA gene analysis, alpha-diversity and dynamics of changes in the fecal metagenome, as well as the functional potential of mice exposed to radiation, were studied, and the prospects of administering the biomineral complex to achieve positive effects were assessed. NMR analysis of the mineral carrier was carried out, and its safety was confirmed. Moreover, per os administration of the complex following irradiation led to a reduction in the level of chromosomal aberrations induced by irradiation. Thus, the biomineral complex has a microbiome-restoring effect and reduces radiation-induced clastogenesis. Full article

Background/Objectives: Chronic inflammatory skin diseases are frequently associated with pruritus, in which the neurokinin-1 receptor (NK1R) and its ligand substance P (SP) play a central role. The development of compounds combining anti-inflammatory and antipruritic effects represents a promising therapeutic strategy. This study aims to identify fiscalin B derivatives as anti-inflammatory agents with high affinity to NK1R using an integrated in silico and in vitro approach. Methods: A library of fiscalin B derivatives was screened through molecular docking against NK1R to identify high-affinity ligands. Selected compounds were further evaluated using in silico ADMET and toxicity predictions. In vitro assays were conducted in HaCaT keratinocytes, RAW264.7 macrophages, and NIH/3T3 fibroblasts to assess cytotoxicity, nitric oxide production, inflammatory proteins expression, and cell migration. Results: Docking studies identified several derivatives with predicted binding affinities comparable to or exceeding those of aprepitant, a well-established NK1R antagonist. Several compounds, particularly 2, 3, 4, 6, and 7, reduced lipopolysaccharide-induced nitric oxide production to 41–51% without relevant cytotoxicity. This effect was associated with reduced iNOS protein levels, suggesting modulation of inflammatory pathways rather than direct nitric oxide scavenging. Most compounds showed positive safety profiles, although in silico analysis indicated limited biodegradability and potential aquatic toxicity. Conclusions: The fiscalin B derivatives, 2, 3, and 4, demonstrate potential as anti-inflammatory agents, in vitro, and as NK1R high affinity ligands, in silico. These findings support their potential as lead compounds for topical therapies for inflammatory skin disorders associated with pruritus, although further optimization and validation are required. Gram-positive infections are associated with significant morbidity and healthcare burden, often requiring prolonged intravenous therapy. Dalbavancin, a long-acting lipoglycopeptide, has emerged as a promising option beyond its approved indication for acute bacterial skin and skin structure infections (ABSSSI). A systematic review was conducted according to the PRISMA guidelines (PROSPERO: CRD420261296328). MEDLINE, Embase, CENTRAL, and Web of Science were searched from inception. Randomized controlled trials (RCTs) and observational studies evaluating dalbavancin in adult patients with Gram-positive infections were included. Outcomes of interest were clinical effectiveness, safety, and healthcare resource utilization. Risk of Bias was assessed using RoB 2 and the Newcastle–Ottawa Scale. Twenty-one studies were included. Randomized trials confirmed non-inferior efficacy of dalbavancin compared with standard therapy in ABSSSI. Observational studies demonstrated high clinical success rates across a range of infections, including osteo-articular infections, bloodstream infections, and infective endocarditis (IE), particularly in acute settings. Lower effectiveness was observed in biofilm-related infections without adequate source control. Dalbavancin was frequently used as sequential or consolidation therapy in complex patients. Its use was consistently associated with reduced length of hospital stay, facilitation of outpatient management, and potential cost savings. The safety profile was favorable, including in prolonged or multi-dose regimens. In conclusion, dalbavancin represents an effective and well-tolerated option for Gram-positive infections, with expanding evidence supporting its use in complex and off-label settings. Its pharmacokinetic profile enables simplified treatment strategies and improved healthcare resource utilization, although appropriate patient selection and source control remain essential. The skin serves as a primary defensive barrier to protect the body from environmental contaminants, infections and trauma. Unfortunately, skin barrier’s structural and functional integrity can be compromised, disrupted or impaired due to a combination of internal and external factors, making it vulnerable and often leading to a wide range of skin conditions characterized by dryness, heightened sensitivity, and increased susceptibility to damage and infections. In addition, the integrity of the skin barrier tends to deteriorate progressively with age. As people age, their skin naturally changes and can also be compromised by a plethora of factors that reduce its strength and resilience. The aging skin becomes thinner and more sensitive, coinciding with a variety of structural–functional alterations, decreased levels of natural moisturizing factor (NMF), lipid content and hydration, increased transepidermal water loss (TEWL), altered skin surface pH (pHss) and microbiome diversity. All these age-related skin integrity alterations make the skin drier, flakier, itchy, and fragile, and more susceptible to damage and breakdown, thus diminishing its ability to effectively protect, repair and heal efficiently. Identifying skin integrity issues before they progress will foster positive outcomes through effective preventive measures. Hence, it is important to understand the impact of skincare formulations on skin integrity in compromised aging skin. A well-considered, evidence-based approach to skincare can provide cleansing, moisturizing and protective benefits, while aiding the reduction in skin integrity issues like dry and itchy skin, sensitive skin, bruising, skin tears, pressure injuries (PIs), lower leg ulcers and moisture-associated skin damage (MASD). Managing skin integrity in compromised aging skin begins with gentle skin cleansing, adequate moisturization and protective barrier care to ensure the skin’s function is maximized. Full article

Diabetic wounds are a common and severe complication of diabetes mellitus, characterized by delayed healing due to persistent inflammation, impaired angiogenesis, and cellular dysfunction. Conventional therapeutic approaches remain limited in efficacy. In recent years, exosomes have attracted considerable attention in wound healing and regenerative medicine because of their crucial role in intercellular communication and tissue repair. However, rapid clearance of exosomes in vivo greatly limits their therapeutic efficacy. To address this critical limitation, we engineered a decellularized extracellular matrix (dECM)-based hydrogel system functionalized with exosomes derived from skin-derived precursor cells (SKPs). This biomimetic scaffold was designed to serve as a local exosome-delivery platform at the wound site, with the aim of improving exosome utilization and augmenting their regenerative effects. Comprehensive in vitro characterization demonstrated that the exosome-loaded composite hydrogels exhibited robust pro-angiogenic activity, as evidenced by enhanced endothelial cell proliferation, migration, and tube formation. Moreover, the hydrogels displayed significant antibacterial effects against wound-relevant pathogens and potent reactive oxygen species (ROS)-scavenging capacity, thereby mitigating oxidative damage. Notably, the composite hydrogels also promoted the phenotypic polarization of macrophages toward the pro-regenerative M2 phenotype. In parallel, in vivo studies using a streptozotocin-induced diabetic rat wound model confirmed that treatment with the composite hydrogels significantly accelerated wound closure rates compared to control groups. Histological and immunohistochemical analyses revealed enhanced angiogenesis, as evidenced by increased CD31-positive microvessel density, as well as improved collagen deposition, re-epithelialization, and an attenuated local inflammatory microenvironment characterized by reduced pro-inflammatory cytokine expression and elevated M2 macrophage infiltration. Collectively, the SKPs exosome-loaded dECM based composite hydrogels developed in this study represent a potential therapeutic strategy for the treatment of diabetic wounds. Full article

Topical retinoids are the cornerstone of the treatment of multiple dermatological conditions. Long established in acne therapy, they exert effects on keratinization, inflammation, fibroblast activity, and collagen remodeling, suggesting a potential role in both the prevention and treatment of acne scars. This narrative review summarizes current evidence on the use of topical retinoids in acne vulgaris and acne scarring, focusing on different retinoid molecules, formulation technologies, and combination strategies. A review of published clinical and experimental studies evaluating tretinoin, adapalene, tazarotene, and trifarotene was performed, including their use as monotherapy and in combination with other topical agents or procedural interventions. The available data indicate that topical retinoids have a well-established position in acne treatment, can improve the appearance of atrophic acne scars, reduce the progression of scarring, and support skin remodeling. Advances in formulation technologies have improved tolerability, while combination approaches with agents such as benzoyl peroxide, antibiotics or procedural techniques have shown additive or synergistic effects, particularly in more severe cases. Nevertheless, much of the evidence regarding novel formulations is derived from small or heterogeneous study populations. In conclusion, topical retinoids represent a relevant therapeutic option in acne vulgaris and acne scarring, from monotherapy in mild cases to components of multimodal treatment protocols in more severe disease. Further large-scale, comparative studies are needed to better define the optimal clinical use of advanced drug delivery systems for topical retinoids. Full article

Although many studies have demonstrated the positive effects of natural visual and auditory stimuli on human physiological and psychological states, there is limited empirical evidence on the effects on subjective comfort under different thermal environments. This study used a climatic chamber experiment to evaluate the impact of three types of natural stimuli (visual, auditory, and combined audio-visual) on physiological and psychological responses under three operative temperature conditions (26 °C, 28 °C, and 32 °C). In total, 24 participants were recruited. Physiological indicators, including heart rate variability, skin conductance level (SCL), skin temperature (ST), and blood pressure, as well as psychological indicators including thermal sensation (TSV), thermal comfort (TCV), visual comfort (VCV), and acoustic comfort (ACV), were collected. The results show that TCV was significantly and positively correlated with both VCV and ACV. The visual stimuli produced the most significant decrease in TSV and the greatest increase in TCV, while combined audio-visual stimuli had the most significant impact on physiological responses. At 26 °C, the combined audio-visual stimuli group reduced heart rate by 6.08%. However, at 32 °C, most physiological and psychological restoration indicators showed no significant changes. These findings provide theoretical references for health-oriented multisensory environmental design in urban areas. Full article

Objectives: To describe the efficacy and safety of tocilizumab (TCZ) and rituximab (RTX) in real-world patients with systemic sclerosis (SSc), across different clinical phenotypes and lines of therapy, evaluating both global clinical outcomes and lung function. Methods: SSc patients treated with TCZ (n = 27) or RTX (n = 23) were retrospectively followed for 12–24 months. Clinical measures, including modified Rodnan Skin Score (mRSS), C-reactive protein (CRP), and revised EUSTAR activity index 2017 (RAI), as well as spirometric parameters, were recorded at baseline and 6, 12, and 24 months. Statistical methods for repeated measures were applied to investigate outcome trends. Given the baseline differences, between-group comparisons were considered exploratory. Results: RTX was used earlier in disease course, while TCZ was mainly used as a rescue therapy. In both groups, mRSS, CRP levels and RAI significantly decreased over time. RTX-treated patients showed a greater absolute mRSS improvement, in line with higher baseline skin scores. No treatment discontinuations due to adverse events occurred in either group; one death and one discontinuation due to inefficacy were observed in the TCZ group. Among SSc-ILD patients, FVC% showed a modest decline in both groups, while DLCO% remained overall stable, and only a few patients met the OMERACT criteria for functional progression. Conclusions: In this real-world, single-center cohort of SSc patients, both agents were associated with a positive impact on disease activity, with a low rate of lung progression and with favorable safety profiles. Owing to substantial baseline imbalances and confounding by indication, between-group comparisons do not allow firm conclusions on comparative effectiveness. Thermal spring waters (TSWs) have long been used in dermatology for chronic inflammatory dermatoses and sensitive skin and are increasingly positioned as cosmeceutical active ingredients. This review summarizes studies on the use of TSW and their hydrobiome derivatives in dermocosmetics and cosmeceuticals for skin health. TSW exhibits anti-inflammatory, antioxidant, soothing, hydrating and barrier-restoring effects in vitro, ex vivo and in clinical studies, improving conditions such as atopic dermatitis, psoriasis, acne, sensitive skin, radiation dermatitis and post-procedure erythema. In parallel, the hydrobiome of TSW has enabled the development of postbiotic and paraprobiotic ingredients, which modulate skin immunity, microbiota, barrier function and clinical signs of inflammatory and sensitive skin. Despite robust preclinical and growing clinical evidence, cosmeceuticals remain regulated as cosmetics in most regions, highlighting the need for specific regulatory frameworks and standardized approaches to demonstrate the safety and efficacy of TSW-based cosmeceuticals, as well as defining acceptable claim categories and minimum evidence thresholds. High-quality, well-balanced feeding strategies are essential for optimizing poultry growth performance and for ensuring the safety of poultry products. Here, we evaluated the effects of chelated vs. inorganic trace minerals on the growth, survival and carcass characteristics of Cobb 500 broiler chickens. A large-scale feeding trial involved four dietary treatments at 21–35 days of age, with Group 1 receiving a control diet with mineral supplements of zinc (Zn), copper (Cu) and manganese (Mn) in sulfate form, based on a standard formulation. A further three experimental diets contained chelated forms of Zn (Group 2), Cu (Group 3) and a mineral premix MINTREX® that provides Zn, Cu and Mn (Group 4). Broilers were raised to 42 days of age under commercial production conditions. Feeding chelated trace minerals resulted in significantly higher body weights during the rearing period, including a 10% increase in pre-slaughter weight compared to the controls (p < 0.01). Moreover, survival rates improved from 87% in the controls to 95–96% in the treatment groups (p < 0.001), and carcass evaluation revealed a 15–17% increase in eviscerated carcass weight in birds fed chelated supplements (p < 0.05). Greater yields of muscle, liver and skin with subcutaneous fat were observed, as was improved production of total edible carcass components. Among the treatments, MINTREX® provided the greatest enhancement effects in performance and slaughter traits. These findings demonstrate that dietary chelated minerals, whether single or combined, may have positive effects on the broiler carcass yield and support their inclusion in poultry production systems. Background: Bone and joint infections (BJIs), including osteomyelitis, septic arthritis, and periprosthetic joint infections, typically require prolonged antimicrobial therapy and often involve complex outpatient management. Oritavancin, a long-acting lipoglycopeptide approved for the treatment of acute bacterial skin and skin structure infections caused by Gram-positive bacteria, has emerged as a potential off-label option for BJIs owing to its favourable pharmacokinetic and pharmacodynamic properties. Objectives: To provide a comprehensive overview of the pharmacological rationale, microbiological activity, and available clinical evidence supporting the use of oritavancin in BJIs. Methods: A comprehensive narrative review of the literature was performed using MEDLINE and the Cochrane Central Register of Controlled Trials (CENTRAL), focusing on publications from 2011 to 2025. Observational studies, case series, and case reports describing the off-label use of oritavancin in BJIs were considered. Results: The available literature primarily consists of observational studies and real-world experiences. Eighteen studies met the inclusion criteria. Oritavancin was most frequently evaluated for osteomyelitis (n = 14 studies), prosthetic joint infections (n = 10) and septic arthritis (n = 5). Multi-dose regimens, typically including a 1200 mg loading dose followed by weekly doses of 800–1200 mg, were the most commonly described strategies. Reported clinical success rates generally ranged from approximately 70% to over 90%. Oritavancin was overall well tolerated, with adverse events being mostly mild and self-limiting. Conclusions: Current evidence suggests that oritavancin may represent an effective and well-tolerated off-label option for selected patients with Gram-positive BJIs. Its use may offer practical advantages, including reduced hospitalization and avoidance of prolonged intravenous antimicrobial therapy, particularly in patients for whom standard treatment approaches are challenging.
