Hydraulic actions may compromise dam foundation stability. Helical anchors have been used in dam foundation reinforcement projects because of the advantages of large uplift and compression bearing capacity, fast installation, and convenient recovery. However, the research on the anchor plate, which plays a key role in the bearing performance of helical anchors, is insufficient at present. Based on the finite element model of helical anchor, this study reveals the failure mode and influencing factors of the anchor plate and establishes the theoretical model of deformation calculation. The results showed that the helical anchor plate had obvious bending deformation when the dam foundation reinforced with a helical anchor reached large deformation. The helical anchor plate can be simplified to a flat circular disk. The stress distribution of the closed flat disk and the open flat disk was consistent with that of the helical disk. The maximum deformation of the closed flat disk was slightly smaller than that of the helical disk (less than 6%), and the deformation of the open flat disk was consistent with that of the helical disk. The results fill the blank of the design basis of helical anchor plate and provide a reference basis for the engineering design. Full article

Due to the effect of room acoustics on musical interpretation, a musician’s rehearsal may be greatly enhanced by leveraging virtual and augmented reality technology. This paper presents a preliminary study on a rehearsal tool designed for musicians, enabling practice in a virtual acoustic environment with audience-positioned playback. Fourteen participants, both professional and non-professional musicians, were recruited to practice with the rehearsal tool prior to performing in an unfamiliar venue. Throughout the rehearsal, the subjects either played in a virtual environment that matched the acoustics of the performance venue or one that was acoustically different. A control group rehearsed in an acoustically dry room with no virtual acoustic environment. The tool’s effectiveness was evaluated with two 16-item questionnaires that assessed quality, usefulness, satisfaction with the rehearsal, and aspects of the performance. Findings indicate that rehearsing in a virtual acoustic environment that matches the performance venue improves acoustic awareness during the performance and enhances ease and comfort on stage compared to practising in a different environment. These results support the integration of virtual acoustics in rehearsal tools to help musicians better adapt their performance to concert settings. Full article

Urban parks play an essential role in mitigating the urban heat island (UHI) effect driven by urbanization. A rigorous understanding of the cooling effects of urban parks can support urban planning efforts aimed at mitigating the UHI effect and enhancing urban sustainability. However, previous research has primarily focused on the maximum cooling impact, often overlooking the accumulative effects arising from spatial continuity. The present study fills this gap by investigating 74 urban parks located in the central area of Jinan and constructing a comprehensive cooling evaluation framework through two dimensions: maximum impact (Park Cooling Area, PCA; Park Cooling Efficiency, PCE) and cumulative impact (Park Cooling Intensity, PCI; Park Cooling Gradient, PCG). We further systematically examined the influence of park attributes and the surrounding urban structures on these metrics. The findings indicate that urban parks, as a whole, significantly contribute to lowering the ambient temperatures in their vicinity: 62.3% are located in surface temperature cold spots, reducing ambient temperatures by up to 7.77 °C. However, cooling intensity, range, and efficiency vary significantly across parks, with an average PCI of 0.0280, PCG of 0.99 °C, PCA of 46.00 ha, and PCE of 5.34. For maximum impact, PCA is jointly determined by park area, boundary length, and shape complexity, while smaller parks generally exhibit higher PCE—reflecting diminished cooling efficiency at excessive scales. For cumulative impact, building density and spatial enclosure degree surrounding parks critically regulate PCI and PCG by influencing cool-air aggregation and diffusion. Based on these findings, this study classified urban parks according to their cooling characteristics, clarified the functional differences among different park types, and proposed targeted recommendations. In response to the escalating climate crisis, reducing greenhouse gas emissions (GHG) has become a top priority for both the public and private sectors. The pavement industry plays a key role in this transition, offering innovative technologies that minimize environmental impacts without compromising performance. Among these, the incorporation of recycled tire rubber and warm-mix asphalt (WMA) additives represents a promising strategy to reduce energy consumption and resource depletion in road construction. This study conducts a comparative life cycle assessment (LCA) to evaluate the environmental performance of an asphalt pavement incorporating recycled rubber and a WMA additive—referred to as R-W asphalt—against a conventional hot-mix asphalt (HMA) pavement. The analysis follows the ISO 14040/44 standards, covering material production, transport, construction, and maintenance. Two service-life scenarios are considered: one assuming equivalent durability and another with a five-year extension for the R-W pavement. The results demonstrate environmental impact reductions of up to 57%, with average savings ranging from 32% to 52% across key impact categories such as climate change, land use, and resource use. These benefits are primarily attributed to lower production temperatures and extended maintenance intervals. The findings underscore the potential of R-W asphalt as a cleaner engineering solution aligned with circular economy principles and climate mitigation goals. Full article

The TP53 gene, frequently mutated across multiple cancer types, plays a pivotal role in regulating the cell cycle and apoptosis through its protein, p53. Missense variants of uncertain significance (VUSs) in TP53 present challenges in understanding their impact on protein function and complicate clinical interpretation. This study aims to analyze the effects of missense VUSs in p53, as reported in the gnomAD database, with a specific focus on their impact on protein stability and phosphorylation. In this study, 33 missense VUSs in TP53 reported in the gnomAD database were analyzed using in silico tools, including PhosphositePlus v6.7.4, the Kinase Library v0.0.11, and Dynamut2. Of these analyzed variants, five disrupted known phosphorylation sites, while another five created new consensus sequences for phosphorylation. Moreover, 20 variants exhibited a moderate destabilizing effect on the protein structure. At least three missense VUSs were identified as potentially affecting p53 function, which may contribute to cancer development. These findings highlight the importance of integrating in silico structural and functional analysis to assess the pathogenic potential of missense VUSs. Full article

The placement of hydraulic elements in existing pool-type fishways to make them more suitable for Cyprinid fish is an issue of increasing interest in fishway research. Hydrodynamic characteristics and fish behavior at the representative pool of the fishway with bottom orifices and notches were assessed at the Dagdelen hydropower plant in the Ceyhan River Basin, Türkiye. Three-dimensional velocity measurements were taken in the pool of the fishway using an Acoustic Doppler velocimeter. The measurements were taken with and without a brush block at two different vertical distances from the bottom, which were below and above the level of bristles tips. A computational fluid dynamics (CFD) analysis was conducted for the studied fishway. The numerical model utilized Large Eddy Simulation (LES) combined with the Darcy–Forchheimer law, wherein brush blocks were represented as homogenous porous media. Our results revealed that the relative submergence of bristles in the brush block plays a very important role in velocity and Reynolds shear stress (RSS) distributions. After the placement of the submerged brush block, flow velocity and the lateral RSS component were reduced, and a resting area was created behind the brush block below the bristles’ tips. Fish movements in the pool were recorded by underwater cameras under real-time operation conditions. The heatmap analysis, which is a 2-dimensional fish spatial presence visualization technique for a specific time period, showed that Capoeta damascina avoided the areas with high turbulent fluctuations during the tests, and 61.5% of the fish presence intensity was found to be in the low Reynolds shear regions in the pool. This provides a clear case for the real-world ecological benefits of retrofitting existing pool-weir fishways with such flexible hydraulic elements. Full article

The scarab species Protaetia brevitarsis, an edible insect, has been used in traditional medicine, as animal feed, and for converting agricultural organic wastes into biofertilizer. The intestinal tract, which contains a diverse array of microbiota, including viruses, plays a critical role in animal health and homeostasis. We previously conducted a comparative analysis of the gut microbiota of third-instar larvae of P. brevitarsis obtained from five different farms and found significant differences in the composition of the gut bacterial microbiota between farms. To better understand the gut microbiota, the composition of DNA viruses in the hindgut contents of P. brevitarsis larvae obtained from five farms was investigated using metagenomic sequencing in this study. The β-diversity was significantly different between metagenomic data obtained from the five farms (PERMANOVA, pseudo-F = 46.95, p = 0.002). Family-based taxonomic analysis indicated that the relative abundance of viruses in the gut overall metagenome varied significantly between farms, with viral reads comprising approximately 41.2%, 15.0%, 4.3%, 4.0%, and 1.6% of metagenomic sequences from the farms Tohamsan gumbengi farm (TO), Secomnalagum gumbengi (IS), Gumbengi brothers (BR), Kyungpook farm (KB), and Jhbio (JH), respectively. More than 98% of the DNA viruses in the hindgut were bacteriophages, mainly belonging to the Siphoviridae family. At the species level, Phage Min1, infecting the genus Microbacterium, was detected in all farms, and it was the most abundant bacteriophage in intestinal microbiota, with a prevalence of 0.9% to 29.09%. The detected eukaryotic DNA viruses accounted for 0.01% to 0.06% of the intestinal microbiota and showed little or no relationship with insect viruses. Therefore, they most likely originated from contaminated feed or soil. These results suggest that the condition of substrates used as feed is more important than genetic factors in shaping the intestinal viral microbiota of P. brevitarsis larvae. These results can be used as reference data for understanding the hindgut microbiota of P. brevitarsis larvae and, more generally, the gut virome of insects. Full article

Background/Objectives: This study aimed to investigate the influence of shade and thickness on the polymerization of SDR® flow+, a low-viscosity bulk-fill composite, by assessing its degree of conversion (DC). Methods: An in vitro study was conducted using SDR® flow+ composite resin. Specimens were prepared at two thicknesses (2 mm and 4 mm) and four shades (Universal, A1, A2, A3). Polymerization was performed using a high-intensity LED curing unit. The DC was assessed using Fourier-transform infrared spectroscopy (ATR-FTIR). Results: Both shade and thickness significantly influenced DC. Thicker specimens (4 mm) exhibited reduced polymerization compared to thinner specimens (2 mm). Darker shades, particularly A3, demonstrated the lowest DC values due to their higher chroma, which limits light penetration. In contrast, the Universal shade achieved higher DC values, even at increased depths, likely due to its greater translucency. Conclusions: Shade and thickness play a critical role in the polymerization of bulk-fill composites. Ensuring adequate polymerization is essential for the longevity of pediatric restorations. Further in vivo research is needed to confirm these findings and assess their clinical implications. Full article

It is believed that caspases may play a significant role in the development of cancer, and the expression levels of genes encoding these proteins may influence the prognosis and clinical course of cancer. Taking into account the information presented, we examined the expression profiles of 11 genes from the caspase family in patients diagnosed with triple-negative breast cancer (TNBC). We qualified 29 patients with TNBC. A fragment of the tumor and a fragment of normal tissue surrounding the tumor were collected from each patient. Then, RNA was isolated, and the reverse transcription process was performed. The expression levels of caspase family genes were determined using the real-time PCR method. The obtained data were correlated with clinical data and compared with data from the Cancer Genome Atlas database using the Breast Cancer Gene Expression Miner v4.8 and Ualcan. Based on the results of the conducted research, it can be assumed that the levels of expression of caspase family genes may be correlated with the clinical course of cancer in patients with TNBC, and further research may indicate that profiling the expression levels of these genes may be used in selecting personalized treatment methods. Sport volunteering plays an important role in achieving the goals of sustainable development by supporting the social dimension of sustainability, fostering social integration, and promoting a healthy lifestyle. However, there is a lack of systematic research in Poland on the motivations of sport volunteers, particularly in the context of mass endurance events. This study employed a quantitative, cross-sectional design involving 148 sport volunteers engaged in mass endurance events in Poznan, Poland. To measure motivation, the Polish adaptation of the VMS-ISE scale was used. Data analysis was conducted using one-way analysis of variance (ANOVA). The results showed that volunteer motivations were relatively homogeneous regardless of gender and education level, with the exception of passion for sport, which was significantly stronger among men (p = 0.037). Significant differences were found based on place of residence: residents of medium-sized cities demonstrated the highest motivation for personal development (p < 0.001), whereas individuals from rural areas exhibited stronger patriotism, a greater need for interpersonal interaction, and a higher valuation of external rewards (p < 0.05). The motivations of sport volunteers in Poland are complex and sensitive to environmental factors. Understanding these differences allows for better alignment of recruitment and volunteer management strategies, which can enhance both the effectiveness and sustainability of volunteer engagement. It is recommended to develop volunteer programs that take into account the demographic and socio-cultural characteristics of participants. Full article

The increasing demand for rapid, sensitive, and eco-friendly methods for the detection of trace heavy metals in environmental samples, attributed to their serious threats to health and the environment, has spurred considerable interest in the development of sustainable sensor materials. Toxic metal ions, namely, lead (Pb2+), cadmium (Cd2+), mercury (Hg2+), arsenic (As3+), and chromium, are potential hazards due to their non-biodegradable nature with high toxicity, even at trace levels. Acute health complications, including neurological, renal, and developmental disorders, arise upon exposure to such metal ions. To monitor and mitigate these toxic exposures, sensitive detection techniques are essential. Pre-existing conventional detection methods, such as atomic absorption spectroscopy (AAS) and inductively coupled plasma-mass spectrometry (ICP-MS), involve expensive instrumentation, skilled operators, and complex sample preparation. Electrochemical sensing, which is simple, portable, and eco-friendly, is foreseen as a potential alternative to the above conventional methods. Carbon-based nanomaterials play a crucial role in electrochemical sensors due to their high conductivity, stability, and the presence of surface functional groups. Biochar (BC), a carbon-rich product, has emerged as a promising electrode material for electrochemical sensing due to its high surface area, sustainability, tunable porosity, surface rich in functional groups, eco-friendliness, and negligible environmental footprint. Nevertheless, broad-spectrum studies on the use of biochar in electrochemical sensors remain narrow. This review focuses on the recent advancements in the development of biochar-based electrochemical sensors for the detection of toxic heavy metals such as Pb2+, Cd2+, and Hg2+ and the simultaneous detection of multiple ions, with special emphasis on BC synthesis routes, surface modification methodologies, electrode fabrication techniques, and electroanalytical performance. Finally, current challenges and future perspectives for integrating BC into next-generation sensor platforms are outlined. Full article

Cholesterol accumulation plays a significant role in the pathogenesis of metabolic-dysfunction-associated steatotic liver disease (MASLD), yet changes in cholesterol homeostasis in MASLD remain insufficiently investigated. This study aimed to examine alterations in cholesterol synthesis and absorption by measuring plasma levels of endogenous cholesterol precursors (as markers of synthesis) and phytosterols (as indicators of absorption). A total of 124 MASLD patients and 43 healthy individuals were included. Our results showed higher plasma concentrations of lathosterol in the MASLD group (p = 0.006), in parallel with comparable concentrations of desmosterol (p = 0.472) and all analyzed phytosterols in both groups. Correlation analysis showed that both lathosterol and desmosterol were positively associated with non-invasive hepatic steatosis indices: FLI, HSI, and TyG index (p < 0.01, p < 0.01, and p < 0.05, respectively). Multivariate linear regression further confirmed that these synthesis markers remained significant predictors of FLI (p = 0.010), HSI (p = 0.013), and TyG index (p = 0.002), even after adjusting for other relevant variables. These findings indicate that MASLD is associated with a shift in cholesterol homeostasis towards enhanced endogenous cholesterol synthesis. Full article

The oral–gut microbiota axis is a relatively new field of research. Although most studies have focused separately on the oral and gut microbiota, emerging evidence has highlighted that the two microbiota are interconnected and may influence each other through various mechanisms shaping systemic health. The aim of this review is therefore to provide an overview of the interactions between oral and gut microbiota, and the influence of diet and related metabolites on this axis. Pathogenic oral bacteria, such as Porphyromonas gingivalis and Fusobacterium nucleatum, can migrate to the gut through the enteral route, particularly in individuals with weakened gastrointestinal defenses or conditions like gastroesophageal reflux disease, contributing to disorders like inflammatory bowel disease and colorectal cancer. Bile acids, altered by gut microbes, also play a significant role in modulating these microbiota interactions and inflammatory responses. Oral bacteria can also spread via the bloodstream, promoting systemic inflammation and worsening some conditions like cardiovascular disease. Translocation of microorganisms can also take place from the gut to the oral cavity through fecal–oral transmission, especially within poor sanitary conditions. Some metabolites including short-chain fatty acids, trimethylamine N-oxide, indole and its derivatives, bile acids, and lipopolysaccharides produced by both oral and gut microbes seem to play central roles in mediating oral–gut interactions. The complex interplay between oral and gut microbiota underscores their crucial role in maintaining systemic health and highlights the potential consequences of dysbiosis at both the oral and gastrointestinal level. Some dietary patterns and nutritional compounds including probiotics and prebiotics seem to exert beneficial effects both on oral and gut microbiota eubiosis. A better understanding of these microbial interactions could therefore pave the way for the prevention and management of systemic conditions, improving overall health outcomes. Full article

DNA connects the domains of genetic regulation and environmental interactions and plays a crucial role in neural development and cognitive function. The complex roles of genetic and epigenetic processes in brain development, synaptic plasticity, and higher-order cognitive abilities were reviewed in this study. Neural progenitors are formed and differentiated according to genetic instructions, whereas epigenetic changes, such as DNA methylation, dynamically control gene expression in response to external stimuli. These processes shape behavior and cognitive resilience by influencing neural identity, synaptic efficiency, and adaptation. This review also examines how DNA damage and repair mechanisms affect the integrity of neurons, which are essential for memory and learning. It also emphasizes how genetic predispositions and environmental factors interact to determine a person’s susceptibility to neurodegenerative disorders, such as Parkinson’s and Alzheimer’s diseases. Developments in gene-editing technologies, such as CRISPR, and non-viral delivery techniques provide encouraging treatment avenues for neurodegenerative disorders. This review highlights the fundamental role of DNA in coordinating the intricate interactions between molecular and environmental factors that underlie brain function and diseases. Full article

The tolerance of the fall armyworm (Spodoptera frugiperda) to plant-derived secondary compounds gradually increases with instars. Therefore, even if plant-based additives are applied at early stages, such as the second or third instar, they may have a differential impact on the ecofriendly control of S. frugiperda. In this study, S. frugiperda larvae were exposed to vanillic acid or sinapic acid at the second and third instar, and physiological and growth parameters were measured. The results showed that the effects of vanillic acid treatment on S. frugiperda were similar at the different instars. They can significantly affect the larval carboxylesterase, glutathione S-transferase, and mixed-function oxidase activities. By reducing larval food intake, food conversion, and utilization efficiency while increasing the food consumption rate, it inhibits weight accumulation. This leads to a significant extension of the development of both the larval and pupal stages, and the adult longevity was reduced. Treatment with sinapic acid at the second instar extended the negative effects on the pupal duration of S. frugiperda when compared to treatment at the third instar, but did not affect adult longevity. Therefore, vanillic acid treatment at the second or third instar stage, can play an important role in the ecofriendly control process of S. frugiperda. The results of this study are of great significance for integrated pest management. Full article
