In June 2025, a two-year-old female brown bear (Ursus arctos) appeared in the streets of Vilnius, the capital city of Lithuania. This sparked significant public, institutional, and media responses. This paper analyzes the event through ecological, social, and symbolic lenses to explore how large carnivores are perceived and managed at the wildland–urban interface. Through an examination of media reports, policy responses, and theoretical perspectives from environmental sociology and narrative studies, we explore how the bear’s presence became a public safety concern and a culturally significant symbol. Public discourse revealed tensions between institutional authority and local ethical values, as evidenced by hunters’ refusal to carry out a kill permit. This case also illustrates the growing use of technology, such as drones, in urban wildlife management. The bear’s peaceful departure reinforced the effectiveness of nonlethal conflict resolution. This case underscores the importance of integrating ecological realities with social perceptions, media framing, and symbolic interpretations in large carnivore conservation. It emphasizes the need for interdisciplinary approaches that address the emotional and cultural aspects of human–wildlife interactions in rapidly urbanizing areas. Full article

Water temperature significantly affects the physiological balance of aquatic organisms like crustaceans, and heat shock proteins (HSPs) are crucial for stress resistance and pathogen defense. This study conducted a genome-wide analysis to explore the functional characteristics of the Hsp70 gene family in Procambarus clarkii. Fifteen Hsp70 family members were identified, with several genes showing upregulation under non-lethal heat shock (NLHS) and pathogen challenges. RNA-Seq and qPCR analyses confirmed increased expression of certain PcHsp70s during NLHS, indicating NLHS activation of the Hsp70 family to enhance immune regulation. dsRNA-mediated silencing of Hsp70 led to downregulation of TLR pathway genes (e.g., TLR1, TLR6), suggesting Hsp70 regulates the TLR signaling pathway for immune responses. These findings reveal that NLHS-induced Hsp70 upregulation improves pathogen resistance, offering insights for addressing temperature fluctuations and disease outbreaks in aquaculture to optimize management practices. Conflicts between humans and Japanese macaques (Macaca fuscata) have intensified due to urban and agricultural expansion, reducing natural habitats and pushing macaques into human settlements. This review examines strategies for managing human–macaque coexistence in Japan. Since the 1970s, urbanisation and deforestation have led to increased macaque migration into populated areas, resulting in property damage, crop loss, and, in some cases, aggressive encounters. The growing macaque population underscores the urgency of effective management programmes. Strategies include preventive measures, such as weeding to limit food sources, habitat modification, and community-based interventions to reduce attractants. Non-lethal deterrents, including visual and auditory scare tactics, trained guard animals, and electric fences, have been implemented to discourage macaques from entering human spaces. Physical barriers, such as fences and buffer zones, provide long-term mitigation but require maintenance and community cooperation. Fertility control, including sterilisation and hormonal contraception, offers a long-term population management solution but presents logistical challenges. Lethal control and capture-relocation, though controversial, remain options for particularly problematic individuals. Additionally, integrating One Health and One Conservation approaches into macaque management allows for a holistic strategy that considers disease risks, ecological balance, and ethical implications. A balanced management plan that incorporates multiple strategies, community participation, and continuous monitoring is crucial for mitigating conflicts and fostering sustainable coexistence between humans and macaques. The tomato leafminer, Tuta absoluta, is an invasive pest that poses a significant threat to Solanaceae crops, including tomatoes, potatoes, and eggplants. Its ability to adapt to various environmental conditions, especially cold stress, allows it to survive in regions with fluctuating temperatures. In this study, the molecular response of T. absoluta adults to rapid cold hardening (RCH) under gradually decreasing non-lethal temperatures was investigated. Using RNA-seq, 197 upregulated genes and 626 downregulated genes were identified in response to cold hardening. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses revealed that upregulated genes were primarily associated with the cell cycle and cell division, including key genes such as CDC25A, BUB1, ORC2, and PlK1, as well as metal ion transporters like unc-93. Downregulated genes were found to be involved in metabolism and oxidative phosphorylation, including COX4, ATPase_I, and ATP-synt_ab. Weighted gene coexpression network analysis (WGCNA) indicated a positive correlation between cell cycle-related genes and RCH, while metabolic processes, including fatty acid metabolism, were negatively correlated with cold hardening. These findings provide new insights into the transcriptional mechanisms of insect responses to cold stress, and highlight potential targets for environmentally sustainable pest management strategies. The search for effective pain management solutions remains a critical challenge, especially amidst growing concerns over the use of conventional opioids. In the US, opioid-related mortality rates have surged to as many as 80 deaths per 100,000 people in some states, with an estimated economic burden of USD 1.5 trillion annually—exceeding the gross domestic product (GDP) of most US industrial sectors. A remarkable breakthrough lies in the discovery that indole and oxindole alkaloids, produced by several genera within the plant Tribe Naucleeae, act on opioid receptors without activating the beta-arrestin-2 pathway, the primary driver of respiratory depression and overdose deaths. This systematic review explores the pharmacological properties, mechanisms of action, dosing considerations, interactions, and long-term effects of mitragynine and corynoxeine, alkaloids from the Southeast Asian plant Mitragyna speciosa (kratom) and others in the Tribe Naucleeae. Mitragynine, a partial opioid receptor agonist, and corynoxeine, known for its anti-inflammatory and neuroprotective effects, demonstrate significant therapeutic potential for managing diverse pain types—including neuropathic, inflammatory, nociceptive, visceral, and central pain syndromes—with a focus on cancer pain. Unlike traditional opioids, these compounds do not recruit beta-arrestin-2, avoiding key adverse effects such as respiratory depression, severe constipation, and rapid tolerance development. Their distinct pharmacological profiles make them innovative candidates for safer, non-lethal pain relief. However, challenges persist, including the unregulated nature of kratom products, inconsistencies in potency due to crude extract variability, potential for misuse, and adverse drug interactions. Addressing these issues requires establishing standardized quality control protocols, such as Good Manufacturing Practices (GMP), to ensure consistent potency and purity. Clear labeling requirements with dosage guidelines and warnings should be mandated to ensure safe use and prevent misuse. Furthermore, the implementation of regulatory oversight to monitor product quality and enforce compliance is essential. This review emphasizes the urgency of focused research to optimize dosing regimens, characterize the pharmacodynamic profiles of these alkaloids, and evaluate long-term safety. By addressing these gaps, the mitragynine- and corynoxeine-related drug classes can transition from promising plant-derived molecules to validated pharmacotherapeutic agents, potentially revolutionizing the field of pain management. La Graciosa, a Natura 2000 site in the Canary Islands, faces substantial conservation challenges, including a large free-roaming cat population that threatens the island’s native biodiversity. In July 2024, a Trap–Neuter–Return (TNR) campaign achieved an 81.4% sterilization rate within urban areas, highlighting TNR’s short-term effectiveness in reducing reproductive potential and, consequently, mitigating predation pressures primarily through the prevention of new litters and reduced reproductive activity in cats. The campaign’s success relied heavily on the active involvement of the local community, who assisted with identifying, trapping, and monitoring free-roaming cats, thereby facilitating a high sterilization rate. However, administrative restrictions hindered access to peri-urban zones, leaving essential population clusters unsterilized and limiting the campaign’s overall scope. Additionally, strong opposition from conservation groups, amplified by extensive media coverage, halted the project prematurely, reducing the effective sterilization rate to 69.3% within three months. Population Viability Analysis (PVA) suggests that achieving high sterilization rates could lead to population reduction over time; however, the inability to access all population segments and to reach the ideal 93–95% sterilization threshold limits TNR’s potential as a long-term standalone solution. Our findings underscore the need for adaptive, context-specific management frameworks in ecologically sensitive areas that integrate TNR with complementary measures, consider regulatory barriers, and value community involvement. This case study provides crucial insights for policymakers and conservationists seeking to balance biodiversity conservation with humane management practices in protected areas. Full article

Predators are dependent on the capture of prey to meet their energetic and nutritive requirements, which brings the risk of predation to prey. The predation risk is divided into consumptive and non-consumptive effects. Non-consumptive effects may manifest through altered growth and ontogenetic trajectories in prey species, a dynamic modulated by olfactory or other sensory cues from predators. Bactrocera dorsalis Hendel represents a major invasive threat to global horticulture. While earlier research was primarily centered on the consumptive interactions between B. dorsalis and its natural enemies, the potential consequences of non-consumptive interactions on the development of B. dorsalis have been overlooked. In this study, we investigated the impact of predation risk effects, induced by both visual exposure to the predatory mantis Hierodula patellifera Serville and its associated odor, on the life history traits of B. dorsalis. Female B. dorsalis demonstrated a reduced developmental time in the presence of a caged predator (H. patellifera) or predator odors, but showed significantly increased fecundity. Conversely, males displayed no significant change in developmental time. Additionally, neither the female nor male body weight at death was significantly influenced by the predation risk from the caged predator or predator odors. This study investigated the effects of predation risk on the development and reproduction of B. dorsalis, emphasizing the potential importance of odor risk in biological and pest control. The land use change can negatively impact ecosystems, enriching water and soil with heavy metals (HMs). The fragile riparian areas along the Medjerda River of Northern Algeria are particularly affected by this phenomenon, and biological tools able to combine information about both matrices can be preferred in their monitoring. This research aimed to evaluate the suitability of the spiny toad (Bufo spinosus D.) as a biomonitor for assessing the impact of three different land uses (agricultural (AGR), urban (URB), and industrial (IND) managements) on soil and water for Cu, Fe, Pb, and Zn by using the non-lethal skin biopsy. The IND land use, followed by URB, mostly impacted soils for Cu and Pb, whereas management was not able to influence water differently despite worrying Pb levels. The cluster analysis allows to show that toad responds like soil in terms of land use and like water for HMs, as it is possibly related to the metal–chemical affinity. Although the single skin analyses do not display any difference among the managements, the bioaccumulation factor (BF > 1) shows that B. spinosus absorbs more HMs from water; skin accumulates Fe and Zn regardless of the land use, probably for both natural and anthropogenic assimilation and Pb and Cu for URB and IND, as it is related to their ionic forms and human impact. Human-shark conflict has been managed through catch-and-kill policies in most parts of the world. More recently, there has been a greater demand for shark bite mitigation measures to improve protection for water users whilst minimizing harm to non-target and target species, particularly White Sharks (Carcharodon carcharias), given their status as a Threatened, Endangered, or Protected (TEP) species. A new non-lethal shark bite mitigation method, known as the Shark-Management-Alert-in-Real-Time (SMART) drumline, alerts responders when an animal takes the bait and thereby provides an opportunity for rapid response to the catch and potentially to relocate, tag, and release sharks. Thirty-six White Sharks were caught on SMART drumlines in New South Wales, Australia, and tagged with dorsal fin-mounted satellite-linked radio transmitters (SLRTs) and acoustic tags before release. Thirty-one sharks were located within 10 days, 22 of which provided high-quality locations (classes 1 to 3) suitable for analysis. Twenty-seven percent and 59% of these sharks were first detected within 10 and 50 h of release, respectively. For the first three days post-release, sharks moved and mostly remained offshore (>3.5 km from the coast), irrespective of shark sex and length. Thereafter, tagged sharks progressively moved inshore; however, 77% remained more than 1.9 km off the coast and an average of 5 km away from the tagging location, 10 days post-release. Sharks were acoustically detected for an average of 591 days post-release (ranging from 45 to 1075 days). Although five of the 36 sharks were not detected on acoustic receivers, SLRT detections for these five sharks ranged between 43 and 639 days post-release, indicating zero mortality associated with capture. These results highlight the suitability of SMART drumlines as a potential non-lethal shark bite mitigation tool for TEP species such as White Sharks, as they initially move away from the capture site, and thereby this bather protection tool diminishes the immediate risk of shark interactions at that site. Homeless cats are a major problem in Europe, with hundreds of thousands abandoned every year. While many die, others can adapt to a lifestyle of roaming freely and establish community cat populations that tend to cluster together in groups. These groups of cats are typically found in urban areas that offer food and shelter to the cats. Animal welfare organizations often care for these cats, providing them with food, shelter, and medical attention. Despite this, conflicts can arise due to the presence of free-roaming cats, with some individuals advocating for drastic measures such as trapping and killing the cats to reduce their populations. However, it is essential to note that such methods are frequently illegal, inhumane, and ultimately ineffective in most situations. A thorough assessment of the impact of cats on a particular natural area requires a comprehensive cat census, a detailed study of the species being preyed upon, and an investigation into the prevalence of zoonotic or epizootic diseases. Moreover, veterinary experts assert that the public health risks associated with cats are often overstated. This article aims to provide a nuanced perspective on the impact of cats on biodiversity in natural areas, while also discussing their role in transmitting the main zoonotic diseases identified in European countries in recent years, with a particular focus on Spain. Effective cat control programs should focus on non-lethal methods such as trap–neuter–return (TNR) and adoption. TNR has proven to be the most effective and humane method of controlling the free-roaming cat population, but its effectiveness is influenced by several factors, including adoption programs and public education on responsible pet ownership. According to Spanish veterinarians, sustainable and science-based solutions such as TNR programs are the best way to achieve population control of free-roaming cats. The veterinary profession should raise awareness regarding sterilization, vaccination, and identification of cats and the consequences of abandonment. They oppose lethal control and removal of cats from the environment, which are ineffective and unethical methods. To promote animal welfare, veterinary professionals must collaborate with public administrations to implement long-term, sustainable solutions to the problem of cat overpopulation. Greater social awareness regarding the importance of sterilization and identification to prevent abandonment and reduce the number of free-roaming cats is also needed. Despite the challenges presented by homeless cat populations in Spain and the rest of Europe, there are many reasons for optimism. Animal welfare organizations and veterinary professionals are actively collaborating to develop humane and effective solutions to manage community cats, including programs such as TNR and adoption. Furthermore, these initiatives are gaining momentum and support from emerging laws and regulations, such as the recent Spanish animal welfare law. Through these efforts, we can reduce the number of free-roaming cats and improve their quality of life. Full article

Atlantic Goliath Grouper, Epinephelus itajara, the largest grouper in the western North Atlantic, exhibits life history characteristics (e.g., it is slow-growing, long-lived, and late-maturing) that make it particularly susceptible to fishing pressure. Otoliths, the structure typically processed for age determination, are usually collected as part of fishery monitoring, but otoliths are not available from Goliath Grouper due to a harvest moratorium enacted in 1990 for United States waters. Alternative structures—such as dorsal fin rays or spines—can be acquired for ageing via nonlethal sampling and may provide reliable age estimates. Since 2006, the Florida Fish and Wildlife Conservation Commission (FWC) has been collecting incidental Goliath Grouper samples from mortality events including those from cold kills and red tides. Corresponding otolith, dorsal fin ray and dorsal fin spine samples were compared to determine the accuracy and precision of the external ageing structures. Marginal increment analyses indicated that annulus (translucent zone) deposition occurred primarily in March–June on spines and in April–June on rays. While ages determined from both rays and spines exhibited high precision, the accuracy compared to otolith ages was low as a result of systematic underageing of both external structures, particularly at the oldest age classes. A correction factor was applied to rays and spines that remedied the underageing, but the correction factor was more successful for spines. An analysis of ray and spine section accuracy based on nonlethal structure removal from the fish (i.e., comparing ages from sections at the base of the structure within the body to those sectioned from the external surface of the body) determined that spines can be accurately aged when sampled distal of the base of the structure, but that rays cannot. Nonlethal sampling and ageing of Goliath Grouper spines facilitated by public participation could address management goals and help determine the offshore population age structure. Simultaneously, it could contribute critical data needed for a traditional stock assessment, should such a path be warranted by the recovery or management of the species. Full article

Increasing numbers of wild ungulates in human-dominated landscapes in Europe could lead to negative effects, such as damages to forests through browsing. To prevent those effects and, thus, mitigate wildlife-based conflicts while ensuring viable ungulate populations, sustainable management is required. Roe deer, as the most abundant cervid species in Europe, is primarily managed via hunting to decrease population densities through harvesting. Besides direct mortality, non-lethal effects of hunting activities further affect the spatial habitat selection for this species. Accordingly, the spatial distribution of hunting locations might influence game impact on forest vegetation. To examine these relationships in more detail, we linked the spatial distribution of hunting locations for roe deer with forest damage through browsing in 20 regions in Upper Austria. Consistent with our hypothesis, an avoidance of forests by hunters was found in regions with <20% forest cover and intolerable browsing impact. When hunters in certain regions, however, used forests according to their availability, game impact on forest vegetation was tolerable. Although forest damage by ungulates depends on numerous factors, we conclude that careful consideration of hunting locations might be an additional approach to reduce browsing intensity by roe deer, at least in regions with low forest cover. Emergent infectious diseases have an increasing impact on both farmed animals and wildlife. The ability to screen for pathogens is critical for understanding host–pathogen dynamics and informing better management. Ranavirus is a pathogen of concern, associated with disease outbreaks worldwide, affecting a broad range of fish, amphibian, and reptile hosts, but research has been limited. The traditional screening of internal tissues, such as the liver, has been regarded as the most effective for detecting and quantifying Ranavirus. However, such methodology imposes several limitations from ethical and conservation standpoints. Non-lethal sampling methods of viral detection were explored by comparing the efficacy of both buccal swabbing and fin clipping. The study was conducted on two Iberian, threatened freshwater fish (Iberochondrostoma lusitanicum and Cobitis paludica), and all samples were screened using qPCR. While for C. paludica both methods were reliable in detecting Ranavirus, on I. lusitanicum, there was a significantly higher detection rate in buccal swabs than in fin tissue. This study, therefore, reports that fin clipping may yield false Ranavirus negatives when in small-bodied freshwater fish. Overall, buccal swabbing is found to be good as an alternative to more invasive procedures, which is of extreme relevance, particularly when dealing with a threatened species. Full article

Sperm cryopreservation and biobanking are emerging as tools for supporting genetic management of small and threatened populations in amphibian conservation programs. However, there is little to no evidence demonstrating reproductive maturity and viability of offspring generated with cryopreserved sperm, potentially limiting widespread integration of these technologies. The purpose of this report is to demonstrate that amphibian sperm can be cryopreserved and thawed to successfully produce individuals of an F1 generation that can reach adulthood and reproductive maturity, to generating viable gametes and an F2 generation. Species-specific exogenous hormones were administered to both F0 and F1 adults to stimulate spermiation and oviposition in the eastern tiger salamander (Ambystoma tigrinum), dusky gopher frog (Lithobates sevosa), and Puerto Rican crested toad (Peltophryne lemur). Sperm cells collected non-lethally from F0 adults were cryopreserved, thawed, and used for in vitro fertilization (IVF) to produce F1 offspring. Individuals of the F1 generation are shown to reach adulthood, express viable gametes, and produce offspring through facilitated breeding, or IVF. The production of amphibian F2 generations shown here demonstrates that amphibian sperm collected non-lethally can be banked and used to generate reproductively viable animals of subsequent generations, thus maintaining valuable genetic linages and diversity in threatened amphibian species. The incredible value that cryopreservation of sperm has for long-term genetic management aids in the sustainability of both in situ and ex situ conservation efforts for this taxon. Full article

Mayaro virus (MAYV) is a neglected arthropod-borne virus found in the Americas. MAYV infection results in Mayaro fever, a non-lethal debilitating disease characterized by a strong inflammatory response affecting the joints and muscles. MAYV was once considered endemic to forested areas in Brazil but has managed to adapt and spread to urban regions using new vectors, such as Aedes aegypti, and has the potential to cause serious epidemics in the future. Currently, there are no vaccines or specific treatments against MAYV. In this study, the antiviral activity of a series of synthetic cyclic ketones were evaluated for the first time against MAYV. Twenty-four compounds were screened in a cell viability assay, and eight were selected for further evaluation. Effective concentration (EC50) and selectivity index (SI) were calculated and compound 9-(5-(4-chlorophenyl]furan-2-yl)-3,6-dimethyl-3,4,5,6,7,9-hexahydro-1H-xanthene-1,8(2))-dione (9) (EC50 = 21.5 µmol·L−1, SI = 15.8) was selected for mechanism of action assays. The substance was able to reduce viral activity by approximately 70% in both pre-treatment and post-treatment assays. Full article
