This work examines the utilization of the natural product shilajit resin, sourced from the Himalayas, for the extraction of diverse components suitable to the synthesis of gold nanoparticles (AuNPs) and selenium nanoparticles (SeNPs). The study evaluated the extraction efficiency of Uro-A, a polar phenolic component, from shilajit samples utilizing various solvent systems, followed by the eco-friendly synthesis and characterization of AuNPs and SeNPs. The LC-MS/MS analysis demonstrated that the methanol/ethyl acetate (MeOH/EA) extract was particularly effective and contained the highest concentration of Uro-A. A robust positive association was discovered between Uro-A content in shilajit and the efficiency of nanoparticle production. Sample 2, with 0.002 ppm Uro-A, produced suboptimal yields (15% for AuNPs; 34% for SeNPs), while sample 1, with 2.504 ppm Uro-A, noticeably enhanced the yields to 90% (AuNPs) and 96% (SeNPs). The synthesized AuNPs and SeNPs were characterized using SEM, EDS, PSD, ZP, XRD, FTIR, and XPS techniques, demonstrating spherical particles with an average diameter of around 100 nm. XPS spectra confirmed the elemental form of the obtained AuNPs and SeNPs. Antimicrobial studies revealed that SeNPs exhibit moderate efficacy (MIC = 0.125–1 mg/mL) against bacteria (e.g., Bacillus subtilis, Staphylococcus aureus) and fungi (e.g., Penicillium italicum, Mucor mucedo), but higher in comparison to AuNPs (MIC 0.25–5 mg/mL). Flexible sensors have emerged as critical interfaces for information exchange between soft biological tissues and machines. Here, we present a dual-mode stretchable sensor system capable of synchronous strain and electromyography (EMG) signal detection, integrated with wireless WIFI transmission for real-time joint movement monitoring. The system consists of two key components: (1) A multi-channel gel electrode array for high-fidelity EMG signal acquisition from target muscle groups, and (2) a novel capacitive strain sensor made of stretchable micro-cracked gold film based on Styrene Ethylene Butylene Styrene (SEBS) that exhibits exceptional performance, including >80% stretchability, >4000-cycle durability, and fast response time (<100 ms). The strain sensor demonstrates position-independent measurement accuracy, enabling robust joint angle detection regardless of placement variations. Through synchronized mechanical deformation and electrophysiological monitoring, this platform provides comprehensive movement quantification, with data visualization interfaces compatible with mobile and desktop applications. The proposed technology establishes a generalizable framework for multimodal biosensing in human motion analysis, robotics, and human–machine interaction systems. Full article

Gold nanoparticles (AuNPs) are widely employed in biosensors; however, conventional synthesis methods require additional surface modification to confer colloidal stability and bioconjugation capability. Here, we report a facile strategy to synthesize carboxymethyl dextran (CMD)-coated AuNPs (AuNP@CMD) that simultaneously serve as a plasmonic label, a stabilizing agent, and a functional scaffold. The CMD was prepared directly via partial carboxymethylation of dextran in a one-pot reduction of HAuCl4, enabling the synthesis of AuNP@CMD with tunable particle sizes and excellent colloidal stability for at least one month at 4 °C. The CMD coating on AuNPs can prevent nanoparticle aggregation, suppress nonspecific adsorption, and introduce surface carboxyl groups for conjugation of bioprobes. Such characteristics are important to develop plasmonic nanoparticle-linked sorbent assays as an alternative to the conventional colorimetric enzyme-linked immunosorbent assay. When applied to a fiber-optic nanogold-linked sorbent assay, AuNP@CMD enabled ultrasensitive detection of a single-stranded DNA, achieving a detection limit at the femtomolar (fM) concentration level without nucleic acid amplification. Different ginseng species, such as Panax ginseng and Panax quinquefolius, have various medicinal and economic values. Industrially and commercially, it is important to differentiate them. We have adopted the DNA hybridization method based on a single-nucleotide polymorphism (SNP) found in the ginseng DNA. The ginseng DNA samples were placed on a nitrocellulose membrane, and hybridization of the target sample with the probes immobilized on the membrane occurred, resulting in red spots for unaided eye visualization. We managed to demonstrate a spot test and then a lateral flow assay. Genomic DNAs were extracted from ginseng root samples and DNA amplification was used to generate the PCR products that flank the SNP site. We conclude that ginseng DNA can be differentiated based on a DNA lateral flow assay, detecting ~3 ng (in 1 μL) of PCR amplicons. Full article

The ongoing COVID-19 pandemic has underscored the urgent need for rapid, sensitive, and versatile diagnostic tools. In this study, we developed a nanogold-based lateral flow immunoassay (LFIA) capable of detecting both SARS-CoV-2 nucleocapsid (N) protein antigens and anti-N IgG antibodies at the point of care. Following optimization of colloidal gold nanoparticle size, pH, and protein conjugation parameters, LFIA strips were assembled in two formats: a competitive assay for antigen detection and a sandwich assay for antibody detection. In the competitive format, gold nanoparticles (AuNPs)-conjugated N protein were used to detect varying concentrations of free N protein. The test line signal inversely correlated with antigen concentration, confirming the assay’s specificity and effectiveness. For antibody detection, the sandwich LFIA format employed immobilized anti-human IgG to capture anti-N antibodies in serum samples from COVID-19 patients. Strong test line signals were observed in samples collected ≥11 days post-symptom onset, indicating a time-dependent increase in IgG detectability. These results demonstrate that the AuNP-based LFIA platform provides a flexible, rapid, and low-cost diagnostic solution, suitable for both early antigen detection and serological monitoring during SARS-CoV-2 infection and recovery. This study introduces a novel nucleic acid testing (NAT) protocol that integrates rapid deoxyribonucleic acid (DNA) extraction, isothermal amplification, and visual detection to enable efficient analysis of opportunistic pathogens. Polyethylenimine-functionalized iron oxide (PEI-Fe3O4) nanoparticles were prepared by combining PEI, acting as a stabilizing agent, with iron salt, which was utilized as the metal ion precursor by the ultrasonication-assisted co-precipitation method, and characterized for structural, optical, and magnetic properties. PEI-Fe3O4 exhibited cationic and anionic behavior in response to pH variations, enhancing adaptability for DNA binding and release. PEI-Fe3O4 enabled efficient extraction of E. faecium DNA within 10 min at 40 °C, yielding 17.4 ng/µL and achieving an extraction efficiency of ~59% compared to a commercial kit (29.5 ng/µL). The extracted DNA was efficiently amplified by loop-mediated isothermal amplification (LAMP) at 65 °C for 45 min. Pyrogallol-rich poly(tannic acid)-stabilized gold nanoparticles (PTA-AuNPs) served as colorimetric probes for direct visual detection of the DNA amplified using LAMP. The magnetic-nanogold (PEI-Fe3O4/PTA-AuNPs) hybrid system achieved a limit of quantification of 1 fg/µL. To facilitate field deployment, smartphone-based RGB analysis enabled quantitative and equipment-free readouts. Overall, the PEI-Fe3O4/PTA-AuNPs hybrid system used in NAT offers a rapid, cost-effective, and portable solution for DNA detection, making the system suitable for microbial monitoring. Full article

Background/Objectives: In this study, AuDNs/EPLE composite electrodes with hierarchical dendritic nanogold structures were fabricated using the in situ electrodeposition of gold nanoparticles through the i-t method. Methods: A conductive polymer composite membrane, PEDOT, was synthesized via the electropolymerization of EDOT and the negatively charged PSS. The negatively charged SO3 groups on the surface of the PEDOT membrane were electrostatically adsorbed with the glucose oxidase (GOD) enzyme and a positively charged chitosan co-solution (GOD/chit+). Using a layer-by-layer self-assembly approach, GOD was incorporated into the multilayers of the composite electrode to create the composite GOD/chit+/PEDOT/AuDNs/EPLE. Results: Electrochemical analysis revealed a GOD surface coverage of 8.5 × 10−10 mol cm−2 and an electron transfer rate of 1.394 ± 0.02 s−1. The composite electrode exhibited a linear response to glucose in the concentration range of 6.923 × 10−2 mM to 1.54 mM, with an apparent Michaelis constant of 0.352 ± 0.02 mM. Furthermore, the GOD/chit+/PEDOT/AuDNs/EPLE also showed good accuracy of glucose determination in human serum samples. Conclusions: These findings highlight the potential of the GOD/chit+/PEDOT/AuDNs/EPLE composite electrode in the development of efficient enzymatic biofuel cells for glucose sensing and energy harvesting applications. This study presents an easy and rapid two-step electrodeposition method for the synthesis of a novel hierarchical dendritic AuPt bimetallic nanocomposite electrode. Ascorbic acid served as both a reducing and directing agent, while a roughened carbon substrate facilitated the formation of the unique dendritic nanostructure. The structural and compositional properties of the synthesized material were comprehensively characterized using X-ray photoelectron spectroscopy (XPS), X-ray diffraction (XRD), selected area electron diffraction (SAED), and transmission electron microscopy (TEM). The resulting nanocomposite exhibited a significantly enhanced specific surface area of 6.97 m2 g−1, compared to commercial Pt/C. Electrochemical investigations demonstrated superior electrocatalytic activity and durability for methanol oxidation in the prepared AuPt nanocomposite electrode, suggesting its promising potential for fuel cell applications. Traditional detection methods such as atomic absorption spectroscopy offer high sensitivity and accuracy for heavy metal ion detection; however, they are often limited to laboratory environments due to bulky equipment and complex procedures. To meet the demand for rapid on-site detection, this study employs electrochemical analysis and utilizes Micro-Electro-Mechanical Systems (MEMS) technology to fabricate a microelectrode sensor chip for the electrochemical detection of heavy metal ions, Hg(II) and As(III). Nano-gold particles were electrodeposited on the sensing area of the working electrode of this chip using a constant-potential deposition method. Uniform distribution of the nanoparticles was obtained, which enhanced the effective specific surface area and electrochemical activity of the working electrode. Therefore, wide detection concentration ranges for Hg(II) of 5 to 1000 µg/L and for As(III) of 5 to 5000 µg/L were displayed, with detection limits of 1.4 µg/L and 2.4 µg/L, respectively. Moreover, the sensor exhibited satisfactory reproducibility, stability and anti-interference capability. These characteristics enable the developed microelectrode sensor chip to be utilized in the monitoring of a diverse range of pollution sources. Full article

Silica aerogels are extensively used as catalyst supports due to their mesoporous structure and chemical inertness. In this study, SiO2–AuNP aerogels containing gold nanoparticles (AuNPs) were synthesized using the sol-gel method followed by supercritical CO2 drying. The inclusion of polyvinyl pyrrolidone (PVP) as a stabilizing agent preserved the gold particle sizes during the gelation process. In contrast, aerogels synthesized without PVP contained enlarged AuNP aggregates, resulting in a shift in the plasmon resonance color from red to bluish or blue–grey. Thermal treatment of these bluish-colored aerogels at high temperatures restored their red coloration, visually indicating the breakdown of large gold clusters into individual nanoparticles. Both types of aerogels were characterized using SEM, TEM, 3D optical microscopy, UV–vis and ATR-IR spectroscopy, and N2 porosimetry, with their properties analyzed as a function of annealing temperature. Their catalytic activity was evaluated through the reduction of 4-nitrophenol with sodium borohydride, and both aerogel types demonstrated catalytic activity. This thermal conversion of large clusters into individual nanoparticles within an aerogel matrix introduces a new and promising approach for creating catalytically active nanogold-containing aerogel catalysts. Insulated gate bipolar transistors (IGBTs), as an important power semiconductor device, are susceptible to thermal stress, thermal fatigue, and mechanical stresses under high-voltage, high-current, and high-power conditions. Elevated heat dissipation within the module leads to fluctuating rises in temperature that accelerate its own degradation and failure, ultimately causing damage to the module as a whole and posing a threat to operator safety. Through ANSYS Workbench simulation analysis, it is possible to accurately predict the temperature distribution, equivalent stress, and equivalent strain of solder materials under actual working conditions, thus revealing the changing laws of the heat–mechanical interaction in solder materials. Simulation analysis results show that, under steady-state operating conditions, the highest point of the IGBT module’s overall junction temperature occurs in the center of the chip. Nanogold exhibited the best performance in terms of temperature and equivalent stress-strain among the five solders studied in this paper; defects near the edges caused greater harm to the module compared to those closer to the solder layer’s center. In terms of stress, defects located near the edge corners produced larger strains. Crazing damage in joints allows for a faster transfer of heat sources away from the center; in terms of stress, crazing has fewer detrimental effects on the integrity of the module as compared to through cracks. Simulation analysis can model the interaction of heat and equipment under realistic work conditions, comparing and evaluating different types of solder materials to select the most suitable solder material for product design and material selection. This aids in enhancing design precision and reliability. Full article

Gold nanoparticles (Au NPs) are a promising target for research due to their small size and the resulting plasmonic properties, which depend, among other things, on the chosen reducer. This is important because removing excess substrate from the reaction mixture is problematic. However, Au NPs are an excellent component of various materials, enriching them with their unique features. One example is hydrogels, which provide a good, easily modifiable base for multiple applications such as cosmetics. For this purpose, various compounds, including hyaluronic acid (HA) and its derivatives, are distinguished by their high water-binding capacity and many characteristics resulting from their natural origin in organisms, including biocompatibility, biodegradability, and tissue regeneration. In this work Au NPs were synthesized using a green chemistry method, either by using onion extract as a reductant or chemically reducing them with sodium citrate. A complete characterization of the nanoparticles was carried out using the following methods: Fourier-Transform Infrared Spectroscopy (FT-IR), Electrophoretic (ELS), and Dynamic Light Scattering (DLS) as well as Transmission Electron Microscope (TEM) and Scanning Electron Microscope (SEM). Their antioxidant activity was also tested using the 2,2-diphenyl-1-picrylhydrazyl radical (DPPH). The results showed that the synthesized nanoparticles enrich the hydrogels with antioxidant properties and new surface properties (depending on the reducing agent, they can be more hydrophilic or hydrophobic). Preliminary observations indicated low cytotoxicity of the nanomaterials in both liquid form and as a hydrogel component, as well as their lack of penetration through pig skin. The cosmetic properties of hydrogel masks were also confirmed, such as increasing skin hydration. Full article

Prostate-specific antigen (PSA) is a well-established tumour marker for prostatic carcinoma. In this study, we present a novel, real-time, and ultrasensitive Love-mode surface acoustic wave (L-SAW) immunosensor for PSA detection enhanced by MoS2@Cu2O-Au nanocomposite conjugation. The MoS2@Cu2O-Au nanocomposites were analyzed by SEM, XRD, and EDS. The experiments show a significant improvement in sensitivity and detection limit compared with the previous detection methods utilizing nanogold alone to detect PSA biomolecules. The experimental results show a good linear relationship when the range of PSA concentrations between 200 pg/mL and 5 ng/mL was tested. The experimental results also show good specificity against alpha 1 fetoprotein and L-tryptophan disruptors. Full article

For years, gold nanoparticles (AuNPs) have been widely used in medicine and industry. Although various experimental procedures have been reported for their preparation and manipulation, none of them is optimal for all purposes. In this work, we engineered the N-terminus of the pIII minor coat protein of bacteriophage (phage) M13 to expose a novel HLYLNTASTHLG peptide that effectively and specifically binds gold. In addition to binding gold, this engineered phage could synthesize spherical AuNPs of 20 nm and other sizes depending on the reaction conditions, aggregate them, and precipitate gold from a colloid, as revealed by transmission electron microscopy (TEM), atomic force microscopy (AFM), and scanning electron microscopy (SEM), as well as ultraviolet–visible (UV–vis) and Fourier-transform infrared (FTIR) spectroscopic methods. We demonstrated that the engineered phage exposing a foreign peptide selected from a phage-displayed library may serve as a sustainable molecular factory for both the synthesis of the peptide and the subsequent overnight preparation of AuNPs from gold ions at room temperature and neutral pH in the absence of strong reducing agents, such as commonly used NaBH4. Taken together, the results suggest the potential applicability of the engineered phage and the new, in vitro-identified gold-binding peptide in diverse biomimetic manipulations. Full article

DNA is fundamental for storing and transmitting genetic information. Analyzing DNA or RNA base sequences enables the identification of genetic disorders, monitoring gene expression, and detecting pathogens. Traditional detection techniques like polymerase chain reaction (PCR) and next-generation sequencing (NGS) have limitations, including complexity, high cost, and the need for advanced computational skills. Therefore, there is a significant demand for enzyme-free and amplification-free strategies for rapid, low-cost, and sensitive DNA detection. DNA biosensors, especially those utilizing plasmonic nanomaterials, offer a promising solution. This study introduces a novel DNA-functionalized waveguide-enhanced nanoplasmonic optofluidic biosensor using a nanogold-linked sorbent assay for enzyme-free and amplification-free DNA detection. Integrating plasmonic gold nanoparticles (AuNPs) with a glass planar waveguide (WG) and a microfluidic channel, fabricated through cost-effective, vacuum-free methods, the biosensor achieves specific detection of complementary target DNA sequences. Utilizing a sandwich architecture, AuNPs labeled with detection DNA probes enhance sensitivity by altering evanescent wave distribution and inducing plasmon resonance modes. The biosensor demonstrated exceptional performance in DNA detection, achieving a limit of detection (LOD) of 33.1 fg/mL (4.36 fM) with a rapid response time of approximately 8 min. This ultrasensitive, rapid, and cost-effective biosensor exhibits minimal background nonspecific adsorption, making it highly suitable for clinical applications and early disease diagnosis. The innovative design and fabrication processes offer significant advantages for mass production, presenting a viable tool for precise disease diagnostics and improved clinical outcomes. Full article
