In addressing the challenges of low path tracking accuracy and poor robustness during tractor autonomous operation, this paper proposes a path tracking control method for tractors that integrates prescribed performance with sliding mode control (SMC). A key feature of this control method is its inherent immunity to system parameter perturbations and external disturbances, while ensuring path tracking errors are constrained within a predefined range. First, the tractor is simplified into a two-wheeled vehicle model, and a path tracking error model is established based on the reference operation trajectory. By defining a prescribed performance function, the constrained tracking control problem is transformed into an unconstrained stability control problem, guaranteeing the boundedness of tracking errors. Then, by incorporating SMC theory, a prescribed performance sliding mode path tracking controller is designed to achieve robust path tracking and error constraint for the tractor. Finally, both simulation and field experiments are conducted to validate the method. The results demonstrate that compared with the traditional SMC method, the proposed method effectively mitigates the impact of complex farmland conditions, reducing path tracking errors while enforcing strict error constraints. Field experiment data shows the proposed method achieves an average absolute error of 0.02435 m and a standard deviation of 0.02795 m, confirming its effectiveness and superiority. This article presents a comparative study of the storage of energy produced by photovoltaic panels by means of two types of batteries: Lead–Acid and Lithium-Ion batteries. The work involved the construction of a model in MATLAB-Simulink for controlling the loading/unloading of storage batteries with energy produced by photovoltaic panels through a buck-type DC-DC convertor, controlled by means of the MPPT algorithm implemented through the method of incremental conductance based on a MATLAB function. The program for the MATLAB function was developed by the author in the C++ programming environment. The MPPT algorithm provides maximum energy transfer from the photovoltaic panels to the battery. The electric power taken over at a certain moment by Lithium-Ion batteries in photovoltaic panels is higher than the electric power taken over by Lead–Acid batteries. Two types of batteries were successively used in this model: Lead–Acid and Lithium-Ion batteries. Based on the results being obtained and presented in this work it may be affirmed that the storage battery Lithium-Ion is more performant than the Lead-Acid storage battery. At the Laboratory of Electrical Machinery and Drives of the Engineering Faculty of Bacau, an experimental stand was built for a storing system for electric energy produced by photovoltaic panels. For controlling DC-DC buck-type convertors, a program was developed in the programming environment Arduino IDE for implementing the MPPT algorithm for incremental conductance. The simulation part of this program is similar to that of the program developed in C++. Through conducting experiments, it was observed that, during battery charging, along with an increase in the charging voltage, an increase in the filling factor of the PWM signal controlling the buck DC-DC convertor also occurred. The findings of this study may be applicable to the storage of battery-generated electrical energy used for supplying electrical motors in electric cars. Full article

Background: During infection with the cytomegalovirus (CMV), the membrane system of the infected cell is remodelled into a megastructure called the assembly compartment (AC). These extensive changes may involve the manipulation of the host cell proteome by targeting a pleiotropic function of the cell such as ubiquitination (Ub). In this study, we investigate whether the Ub system is required for the establishment and maintenance of the AC in murine CMV (MCMV)-infected cells Methods: NIH3T3 cells were infected with wild-type and recombinant MCMVs and the Ub system was inhibited with PYR-41. The expression of viral and host cell proteins was analyzed by Western blot. AC formation was monitored by immunofluorescence with confocal imaging and long-term live imaging as the dislocation of the Golgi and expansion of Rab10-positive tubular membranes (Rab10 TMs). A cell line with inducible expression of hemagglutinin (HA)-Ub was constructed to monitor ubiquitination. siRNA was used to deplete host cell factors. Infectious virion production was monitored using the plaque assay. Results: The Ub system is required for the establishment of the infection, progression of the replication cycle, viral gene expression and production of infectious virions. The Ub system also regulates the establishment and maintenance of the AC, including the expansion of Rab10 TMs. Increased ubiquitination of WASHC1, which is recruited to the machinery that drives the growth of Rab10 TMs, is consistent with Ub-dependent rheostatic control of membrane tubulation and the continued expansion of Rab10 TMs. Conclusions: The Ub system is intensively utilized at all stages of the MCMV replication cycle, including the reorganization of the membrane system into the AC. Disruption of rheostatic control of the membrane tubulation by ubiquitination and expansion of Rab10 TREs within the AC may contribute to the development of a sufficient amount of tubular membranes for virion envelopment. The lateral vibration of propulsion shafting is a critical factor affecting the acoustic stealth performance of underwater vehicles. As the main vibration isolation component in transmitting vibrational energy, the damping efficiency of the propulsion shafting support system (PSSS) holds particular significance. This study investigates the dynamic characteristics of the PSSS with the integral squeeze film damper (ISFD). A dynamic model of ISFD–PSSS is developed to systematically analyze the effects of shaft speed and external load on its dynamic behavior. Three test bearings (conventional, 1S, and 3S structure) are designed and manufactured to study the influence of damping structure layout scheme, damping fluid viscosity, unbalanced load, and shaft speed on the vibration reduction ability of ISFD–PSSS through axis orbit and vibration velocity. The results show that the damping effects of ISFD–PSSS are observed across all test conditions, presenting distinct nonlinear patterns. Suppression effectiveness is more pronounced in the vertical direction compared to the horizontal direction. The 3S structure bearing has better vibration reduction and structural stability than other schemes. The research results provide a reference for the vibration control method of rotating machinery. Articulated Tugs and Barges (ATBs) are increasingly recognised for their effectiveness in transporting chemicals, petroleum, bulk goods, and containers, primarily due to their exceptional flexibility and fuel efficiency. Recent projections indicate that the ATB market is on track for significant growth, which is expected to lead to an increase in the annual growth rate from 2025 to 2032. This study aims to analyse the current advancements in ATB technology and provide insights into the ATB fleet and the systems that connect tugboats and barges. Furthermore, it highlights the advantages of this transportation system, especially regarding its role in enhancing energy efficiency within the maritime transport sector. Currently, there is limited information available in the public domain about ATBs compared to other commercial vessels. The analysis reveals that much of the required information for modern ATB design is not accessible outside specialised design companies. The study also focuses on conceptual design aspects, which include the main dimensions, articulated connections, propulsion systems, and machinery, concluding with an evaluation of economic viability. Special emphasis is placed on defining the main dimensions, which is a critical part of the complex design process. In this context, the ratios of length to beam (L/B), beam to draft (B/D), beam to depth (B/T), draft to depth (T/D), and power to the number of tugs cubed (Pw/N3) are established as design control parameters in the conceptual design phase. This aspect underscores the novelty of the present study. Additionally, the economic viability is analysed in terms of both CAPEX (capital expenditures) and OPEX (operational expenditures). While CAPEX does not significantly differ between the methods used in different types of commercial ships, OPEX should account for the unique characteristics of ATB vessels. Full article

Chinese hamster ovary (CHO) cells are the most common protein production platform for glycosylated biopharmaceuticals due to their relatively efficient secretion systems, post-translational modification (PTM) machinery, and quality control mechanisms. However, high productivity and titer demands can overburden these processes. In particular, the endoplasmic reticulum (ER) can become overwhelmed with misfolded proteins, triggering the unfolded protein response (UPR) as evidence of ER stress. The UPR increases the expression of multiple genes/proteins, which are beneficial to protein folding and secretion. However, if the stressed ER cannot return to a state of homeostasis, a prolonged UPR results in apoptosis. Because ER stress poses a substantial bottleneck for secreting protein therapeutics, CHO cells are both selected for and engineered to improve high-quality protein production through optimized UPR and ER stress management. This is vital for optimizing industrial CHO cell fermentation. This review begins with an overview of common ER-stress related markers. Next, the optimal UPR profile of high-producing CHO cells is discussed followed by the context-dependency of a UPR profile for any given recombinant CHO cell line. Recent efforts to control and engineer ER stress-related responses in CHO cell lines through the use of various bioprocess operations and activation/inhibition strategies are elucidated. Finally, this review concludes with a discussion on future directions for engineering the CHO cell UPR. Full article

Rail transit as a high-energy consumption field urgently requires the adoption of clean energy innovations to reduce energy consumption and accelerate the transition to new energy applications. As an energy-saving fluid machinery, the ejector exhibits significant application potential and academic value within this field. This paper reviewed the recent advances, technical challenges, research hotspots, and future development directions of ejector applications in rail transit, aiming to address gaps in existing reviews. (1) In waste heat recovery, exhaust heat is utilized for propulsion in vehicle ejector refrigeration air conditioning systems, resulting in energy consumption being reduced by 12~17%. (2) In vehicle pneumatic pressure reduction systems, the throttle valve is replaced with an ejector, leading to an output power increase of more than 13% and providing support for zero-emission new energy vehicle applications. (3) In hydrogen supply systems, hydrogen recirculation efficiency exceeding 68.5% is achieved in fuel cells using multi-nozzle ejector technology. (4) Ejector-based active flow control enables precise ± 20 N dynamic pantograph lift adjustment at 300 km/h. However, current research still faces challenges including the tendency toward subcritical mode in fixed geometry ejectors under variable operating conditions, scarcity of application data for global warming potential refrigerants, insufficient stability of hydrogen recycling under wide power output ranges, and thermodynamic irreversibility causing turbulence loss. To address these issues, future efforts should focus on developing dynamic intelligent control technology based on machine learning, designing adjustable nozzles and other structural innovations, optimizing multi-system efficiency through hybrid architectures, and investigating global warming potential refrigerants. These strategies will facilitate the evolution of ejector technology toward greater intelligence and efficiency, thereby supporting the green transformation and energy conservation objectives of rail transit. Chronic Morphine Treatment Leads to a Global DNA Hypomethylation via Active and Passive Demethylation Mechanisms in mESCs

Epigenetic regulation, particularly DNA methylation, plays a crucial role in embryonic development by controlling gene expression patterns. The disruption of this regulation by environmental factors can have long-lasting consequences. Opioid drugs, such as morphine, are known to cross the placental barrier and affect the developing central nervous system, yet their precise epigenetic effects during early development remain unclear. This study aimed to elucidate the impact of chronic morphine exposure on the DNA methylation landscape and gene expression in mouse embryonic stem cells (mESCs). mESCs were chronically exposed to morphine (10 μM for 24 h). Genome-wide bisulfite sequencing was performed to identify DNA methylation changes, while RNA sequencing (RNA-Seq) assessed corresponding gene expression alterations. Global levels of 5-methylcytosine (5mC) and 5-hydroxymethylcytosine (5hmC) were quantified using mass spectrometry. Morphine exposure induced global DNA hypomethylation and identified 16,808 differentially methylated genes (DMGs) related to development, cell signalling, metabolism, and transcriptional regulation. Integrative transcriptomic analysis with RNA-Seq data revealed 651 overlapping genes, including alterations in key epigenetic regulators involved on DNA methylation machinery. Specifically, Tet1 was upregulated with promoter hypomethylation, while Dnmt1 was downregulated, without changes in promoter methylation after morphine exposiure. Mass spectrometry results confirmed a global decrease in 5mC levels alongside increased 5hmC, indicating the involvement of both passive and active demethylation pathways. These findings demonstrate for the first time that morphine disrupts the epigenetic homeostasis of mESCs by promoting global and gene-specific DNA demethylation, which might be key to the phenotypic changes that occur in adulthood. This work provides novel mechanistic insights into how opioid exposure during early development may lead to persistent epigenetic alterations, with potential long-term implications for neurodevelopment and disease susceptibility. Industrial maintenance has shifted from reactive repairs and calendar-based servicing toward data-driven predictive strategies. This paper presents a non-intrusive, low-cost IoT hardware platform for sustainable predictive maintenance of rotating machinery. The system integrates an ESP32-S3 sensor node that captures vibration (100 kHz) and temperature data, performs local logging, and communicates wirelessly. An automated spectral band segmentation framework is introduced, comparing equal-energy, linear-width, nonlinear, clustering, and peak–valley partitioning methods, followed by a weighted feature scheme that emphasizes high-value bands. Three unsupervised one-class classifiers—transformer autoencoders, GANomaly, and Isolation Forest—are evaluated on these weighted spectral features. Experiments conducted on a custom pump test bench with controlled anomaly severities demonstrate strong anomaly classification performance across multiple configurations, supported by detailed threshold-characterization metrics. Among 150 model–segmentation configurations, 25 achieved perfect classification (100% precision, recall, and F1 score) with ROC-AUC = 1.0, 43 configurations achieved ≥90% accuracy, and the lowest-performing setup maintained 81.8% accuracy. The proposed end-to-end solution reduces the downtime, lowers maintenance costs, and extends the asset life, offering a scalable, predictive maintenance approach for diverse industrial settings. Autonomous earthwork machinery is gaining traction as a means to boost productivity and safety on space-constrained urban sites, yet the fast-growing literature has not been fully integrated. To clarify current knowledge, we systematically searched Scopus and screened 597 records, retaining 157 peer-reviewed papers (2015–March 2025) that address autonomy, integrated control, or risk mitigation for excavators, bulldozers, and loaders. Descriptive statistics, VOSviewer mapping, and qualitative synthesis show the output rising rapidly and peaking at 30 papers in 2024, led by China, Korea, and the USA. Four tightly linked themes dominate: perception-driven machine autonomy, IoT-enabled integrated control systems, multi-sensor safety strategies, and the first demonstrations of fleet-level collaboration (e.g., coordinated excavator clusters and unmanned aerial vehicle and unmanned ground vehicle (UAV–UGV) site preparation). Advances include centimeter-scale path tracking, real-time vision-light detection and ranging (LiDAR) fusion and geofenced safety envelopes, but formal validation protocols and robust inter-machine communication remain open challenges. The review distils five research priorities, including adaptive perception and artificial intelligence (AI), digital-twin integration with building information modeling (BIM), cooperative multi-robot planning, rigorous safety assurance, and human–automation partnership that must be addressed to transform isolated prototypes into connected, self-optimizing fleets capable of delivering safer, faster, and more sustainable urban construction. Soil compaction is a critical challenge in perennial fruit production, limiting root growth, water infiltration, and nutrient uptake—factors essential for climate-resilient and sustainable orchard systems. In subtropical Asian pear (Pyrus pyrifolia Nakai) orchards under the annual top-working system, intensive machinery traffic exacerbates subsurface hardpan formation and tree performance. This study evaluated the effectiveness of pneumatic subsoiling, a minimally invasive method using high-pressure air injection, in alleviating soil compaction without disturbing orchard surface integrity. Four treatments varying in radial distance from the trunk and pneumatic application were tested in a mature orchard in central Taiwan. Pneumatic subsoiling 120 cm away from the trunk significantly reduced soil penetration resistance by 15.4% at 34 days after treatment (2,302,888 Pa) compared to the control (2,724,423 Pa). However, this reduction was not sustained at later assessment dates, and no significant improvements in vegetative growth, fruit yield, and fruit quality were observed within the first season post-treatment. These results suggest that while pneumatic subsoiling can modify subsurface soil physical conditions with minimal surface disturbance, its agronomic benefits may require longer-term evaluation under varying moisture and management regimes. Overall, this study highlights pneumatic subsoiling may be a potential low-disturbance strategy to contribute to longer-term soil physical resilience. Full article

To analyze the wear performance induced by rotor–stator rubbing in an aero-engine sealing structure under authentic operating conditions, a transonic rotor system with double bearing is constructed. This system incorporates the disk, shaft, blades, joint bolts, and auxiliary support structure. The system was evaluated in terms of its critical speed, vibration characteristics, component strength under operational conditions, and response characteristics in abnormal extreme scenarios. A ball screw-type feeding system is employed to achieve precise rotor–stator rubbing during rotation by controlling the coating feed. Additionally, a quartz lamp heating system is used to apply thermal loads to coating specimens, and the appropriate heat insulation and cooling measures are implemented. Furthermore, a high-frequency rubbing force test platform is developed to capture the key characteristics caused by rubbing. The test rig can conduct response tests of the system with rotor–stator rubbing and abrasion tests with tip speeds reaching 425 m/s, feed rates ranging from 2 to 2000 μm/s, and heating temperatures up to 1200 °C. Test debugging has confirmed these specifications and successfully executed rubbing tests, which demonstrate stability throughout the process and provide reliable rubbing force test results. Advances in Industry 4.0 and the emergence of Industry 5.0 are driving the development of intelligent, sustainable manufacturing systems, where embedded sensing and real-time health diagnostics play a critical role. However, implementing robust predictive maintenance in production environments remains challenging due to the variability in machine operations and the lack of access to internal control data. This paper introduces a lightweight, embedded-compatible framework for health status signature extraction based on empirical mode decomposition (EMD), leveraging only data from a single triaxial accelerometer. The core of the proposed method is a cycle-synchronized segmentation strategy that uses accelerometer-derived velocity profiles and cross-correlation to align signals with machining cycles, eliminating the need for controller or encoder access. This ensures process-aware decomposition that preserves the operational context across diverse and dynamic machining conditions to address the inadequate segmentation of unstable process data that often fails to capture the full scope of the process, resulting in misinterpretation. The performance is evaluated on a challenging real-world manufacturing benchmark where the extracted intrinsic mode functions (IMFs) are analyzed in the frequency domain, including quantitative evaluation. As results show, the proposed method shows its effectiveness in detecting subtle degradations, following a low computational footprint, and its suitability for deployment in embedded predictive maintenance systems on brownfield or controller-limited machinery. Identifying the key factors influencing agricultural carbon emissions and accurately predicting future trends are essential for achieving carbon peak and carbon neutrality goals. This study aims to assess carbon emissions in agriculture from 1997 to 2022, construct an accurate model to identify the key influencing factors, and predict carbon emissions in agriculture from 2023 to 2030 with an intelligent prediction system to discuss risk management. Additionally, the Dagum method was employed to explore regional differences in agricultural carbon emissions across China. The results reveal that China’s agricultural carbon emissions exhibited a fluctuating trend from 1997 to 2022, peaking in 2015, followed by a period of decline and a moderate rebound in recent years. Elastic Net Regression identified eleven key variables, including Agricultural Machinery Level (MA), Numbers of Agricultural Tools (AT), and Agricultural Industrial Structure Upgrading (AICE), as major determinants of agricultural carbon emissions. Furthermore, the RF-PSO method demonstrated the highest predictive accuracy, forecasting a minor peak in agricultural carbon emissions in 2027, followed by stabilization. Regionally, imbalances in emissions were observed, with the intensity of transvariation accounting for 37.078% of the disparity. Therefore, the Chinese government is advised to implement region-specific strategies for controlling agricultural carbon emissions, cultivate new high-quality agricultural productivity, and promote advanced technologies. Hydraulic excavators operate in harsh environments where direct measurement of actuator chamber pressures and boom displacement is often unreliable or infeasible. This study presents a novel control strategy that estimates actuator chamber pressures from work port pressures using differential equations, eliminating the need for direct pressure or position sensors. A fuzzy logic-based proportional–derivative (PD) controller is developed to mitigate boom oscillations, particularly under high-inertia load conditions and variable operator inputs. The controller dynamically adjusts gains through fuzzy logic-based gain scheduling, enhancing adaptability across a wide range of operating conditions. The proposed method addresses the limitations of classical PID controllers, which struggle with the nonlinearities, parameter uncertainties, and instability introduced by counterbalance valves and pressure-compensated proportional valves. Experimental data is used to design fuzzy rules and membership functions, ensuring robust performance. Simulation and full-scale experimental validation demonstrate that the fuzzy PD controller significantly reduces pressure overshoot (by 23% during extension and 32% during retraction) and decreases settling time (by 31.23% and 28%, respectively) compared to conventional systems. Frequency-domain stability analysis confirms exponential stability and improved damping characteristics. The proposed control scheme enhances system reliability and safety, making it ideal for excavators operating in remote or rugged terrains where conventional sensor-based systems may fail. This approach is generalizable and does not require modifications to the existing hydraulic circuit, offering a practical and scalable solution for modern hydraulic machinery. Full article
