Harnessing Epigenetic Modifiers Reveals MAPK-Mediated Regulation Mechanisms in Hadal Fungi of Alternaria alternata Under High Hydrostatic Pressure

High hydrostatic pressure (HHP) significantly modulates microbial metabolism, while chemical epigenetic modifiers are known to reactivate silent biosynthetic gene clusters and induce novel natural products. However, the mechanisms by which these epigenetic modifiers regulate fungal responses under differential pressure conditions, and how such regulation affects natural product biosynthesis, remain completely unexplored. Here, we investigated the hadal fungus Alternaria alternata CIEL23 isolated from 7332 m sediments in the Mariana Trench under epigenetic modifier treatment with contrasting pressures (0.1 MPa vs. 40 MPa). Our results revealed that epigenetic perturbations and high pressure significantly altered fungal phenotypes, gene expression, and secondary metabolite composition. Transcriptome-level analysis of epigenetic regulatory mechanisms under epigenetic modifiers in both pressure conditions (0.1 MPa and 40 MPa) demonstrated that the addition of epigenetic modifiers regulated MAPK pathway-related gene expression in response to the environment stimuli. Under dual stress conditions, the IG, CWI, and HOG branches of the MAPK pathway showed significantly altered activity patterns. These changes were associated with differential the regulation of genes related to hyphal growth, cell wall remodeling, cell cycle progression, and osmolyte synthesis, suggesting the coordinated modulation of multiple cellular processes. These findings provide the mechanistic link between epigenetic modification induced HHP-response changes and regulation in hadal fungi. Our study not only advances understanding of hadal fungal response to dual stressors but also unlocks new possibilities for harnessing their stress-driven metabolic versatility for biotechnological applications. This work proposes a mid-fidelity load-mapping method for the structural analysis of semisubmersible floating offshore wind turbine substructures. Building on a hybrid linear potential flow and strip-theory dynamic analysis, the method maps hydrodynamic, current, hydrostatic, gravitational, inertial, mooring, and turbine loads onto a shell-based finite element (FE) model. The functionality of the proposed method is demonstrated through two case studies involving ultimate limit state analysis of a structurally reinforced OC4 DeepCwind semisubmersible platform. The analyses were conducted for two design load cases (DLCs) formulated to represent the metocean conditions at the Utsira Nord site, located off the coast of Norway. The accuracy of the mapped hydrostatic and potential flow loads is validated against dynamic simulation data, while a mesh convergence study is used to ensure reliable FE model performance. Results show that the highest von Mises stresses occur at unsupported heave-plate regions, internal stiffeners, and welded joints, with peak stresses safely below the steel’s yield strength. The more severe conditions of DLC 6.1 lead to a broader distribution of high-stress locations compared to DLC 1.6 but only a modest increase in peak stress. Full article

To address concrete cracking in submerged box-shaped hollow thin-walled piers under static ice and hydrostatic pressure, this study proposes a composite strengthening method employing externally bonded steel plates coupled with concrete infill blocks. Based on mechanical theoretical derivation, the strengthened structure is simplified as a cooperative system comprising compression–truss and suspended-cable mechanisms. Key design parameters—including steel plate span, thickness, infill block height, and plate corner configuration—are optimized using a genetic algorithm. The optimization objective minimizes strengthening cost, subject to constraints of concrete crack resistance, steel plate strength, and deformation control, ultimately determining the numerically optimal composite strengthening solution. Validation through planar finite element models demonstrates that: (1) the proposed system effectively suppresses cracking in the original structure; (2) peak stresses in the steel plates remain below the yield strength of Q345 steel; and (3) the theoretical design is reasonable and effective, which can solve the cracking problem of the wading-tank hollow thin-walled pier under the action of surrounding load. Bentonite materials are extensively used in cutoff walls at landfill sites. This study calculates the stress and permeability characteristics of bentonite materials using the piezocone penetration test (CPTU) and ABAQUS simulations. The lateral effective stress of bentonite materials is evaluated using arching models, lateral squeezing models, and a modified lateral squeezing model. Pore pressure dissipation types are categorized into standard and non-standard, with the coefficient of consolidation obtained using the half dissipation time of excess pore pressure (t50) method. In the standard dissipation type, the excess pore pressure gradually dissipates over time after the cone stops penetrating. In contrast, the non-standard dissipation type is characterized by an initial increase in pore pressure until it reaches a maximum value, followed by a decrease to hydrostatic pressure. Additionally, the pore pressure dissipation process in bentonite cutoff walls is recorded and analyzed over various time intervals. Finally, the relationship between hydraulic conductivity and t50 at landfill sites is established based on standard and non-standard dissipation types using CPTU and ABAQUS methods. The t50 method is used for the standard dissipation type, while a modified t50m method is used for the non-standard dissipation type from CPTU and a t50m method is used in the non-standard dissipation type from CPTU. The t50m is the modified value derived from t50. Cutoff walls made from bentonite materials offer the advantage of enhancing the isolation effects and meeting the design requirement of permeability (1.0 × 10−7 cm/s). Full article

To study and quantify the impact of water storage on lake slope stability after the closure of an open-pit mine, we targeted slope control measures by large-scale parallel computing methods and strength reduction theory. This was based on a three-dimensional refined numerical model to simulate the evolution of slope stability under different water storage levels and backfilling management conditions, and to quantitatively assess the risk of slope instability through the spatial distribution of stability coefficients. This study shows that during the impoundment process, the slope stability has a nonlinear decreasing trend due to the decrease in effective stress caused by the increase in pore water pressure. When the water storage was at 0 m, the instability range is the largest, and the surface range is nearly 200 m from the edge of the pit; when the water level continued to rise to 50 m, the hydrostatic pressure of the pit lake water on the slope support effect began to appear, and the stability was improved, but there is still a wide range of unstable areas at the bottom. In view of the unstable area of the steep slope with soft rock in the north slope during the process of water storage, the management scheme of backfilling the whole bottom to −150 m was proposed, and the slope protection and pressure footing were formed by discharging the soil to −40 m in steps to improve the anti-slip ability of the slope. With the rise of carbon capture and storage, liquefied carbon dioxide (LCO2) has emerged as a promising medium for large-scale marine transport. This study evaluates the structural integrity of an IMO Type C cargo tank for a medium-range LCO2 carrier under four conditions: ultimate limit state, accidental limit state, hydrostatic pressure test, and fatigue limit state, based on IGC Code and classification rules. Seventeen load cases were analyzed using finite element methods with multi-step loading to ensure stability. The highest stress occurred at the pump dome–shell junction due to geometric discontinuities, but all stress and buckling criteria were satisfied. The fatigue damage from wave-induced loads was negligible, with low-cycle fatigue from loading/unloading operations governing the fatigue life, which exceeded 31,000 years. The findings confirm the tank’s structural robustness and its suitability for safe, efficient medium-pressure LCO2 transport. Full article

The formation of hot tearing during direct chill casting of aluminum alloys, specifically AA6111, is a significant challenge in the production of ingots for industrial applications. This study investigates the role of hydrostatic pressure and tensile stress in the formation of hot tearing during direct chill casting of rectangular ingots. Combining experimental results and finite element modeling with ABAQUS/CAE 2022, the mechanical behavior of the semi-solid AA6111 alloy was analyzed under different cooling conditions. “Hot” (low water flow) and “Cold” (high water flow) conditions were the two types of cooling conditions that produced cracked and sound ingots, respectively. The outcomes indicate that high tensile stress and localized negative hydrostatic pressure in the hot condition are the main factors promoting the initiation and propagation of cracks in the mushy zone, whereas the improvement of the cooling conditions reduces these defects. To address the persistent challenges of substantial land occupation, intricate construction sequencing, and extended project timelines inherent to conventional substation accident oil sumps, this research introduces a novel integrally prefabricated circular cross-section oil containment structure. The study establishes a finite element representation of this prefabricated system to systematically examine structural deformation mechanisms and failure patterns under combined hydrostatic and geostatic loading scenarios. Through parametric analysis of the oil tank structure, the influences of longitudinal reinforcement diameter, thickness–diameter ratio, height–diameter ratio, and concrete-strength grade on the mechanical characteristics of the structure are explored. Utilizing the response surface methodology for the parametric optimization in finite element analysis, a comprehensive optimization of critical geometric design variables is conducted. These results indicate that longitudinal reinforcement diameter and concrete-strength grade exert negligible influence on concrete stress except for stress increase under internal pressure, with higher concrete grades. The thickness-to-diameter ratio dominantly regulates structural responses: response surface optimization achieved 12% stress reduction and 14% displacement mitigation at 220 mm wall thickness under internal pressure, despite a 4% stress increase under external loading. Height-dependent effects require specific optimization, with 18% stress reduction beyond 3000 mm under external pressure but 20% stress increase at 3400 mm under top loads. Geometric refinements enable 34–50% displacement reduction in critical zones, providing validated references for prefabricated oil tanks. Full article

This study investigates the shell hydroforming of 1.2 mm-thick AA5052 aluminum alloy sheets to produce hemispherical domes which possess inherent spatial symmetry about their central axis. Shell hydroforming is widely used in fabricating lightweight, high-strength components for aerospace, automotive, and energy applications. The forming process was driven by a spatially symmetrical internal pressure distribution applied uniformly across the blank to maintain balanced deformation and minimize geometrical distortion. Experimental trials aimed at achieving a dome depth of 50 mm revealed wrinkle formation at the blank periphery caused by circumferential compressive stresses symmetrical in nature with respect to the dome’s central axis. To better understand the forming behavior, a validated 3D finite element (FE) model was developed, capturing key phenomena such as material flow, strain rate evolution, hydrostatic stress distribution, and wrinkle development under symmetric boundary conditions. The effects of the internal pressure (IP), blank holding force (BHF), coefficient of friction (CoF), and flange radius (FR) were systematically studied. A strain rate of 0.1 s−1 in the final stage improved material flow, while a symmetric tensile hydrostatic stress of 160 MPa facilitated dome expansion. Although tensile stresses can induce void growth, the elevated strain rate helped suppress it. An optimized parameter set of IP = 5.43 MPa, BHF = 140 kN, CoF = 0.04, and FR = 5.42 mm led to successful formation of the 50 mm dome with 19.38% thinning at the apex. Internal pressure was identified as the most critical factor influencing symmetric formability. A process window was established to predict symmetric failure modes such as wrinkling and bursting. When deep-buried tunnels are excavated using the drill-and-blast method, the surrounding rock is subjected to combined cyclic blasting loads and excavation-induced stress unloading. Understanding the distribution characteristics of rock damage zones under these conditions is crucial for the design and safety of building-integrated underground structures. This study investigates the relationship between surrounding rock damage and in situ stress conditions through numerical simulation methods. A constitutive model suitable for simulating rock mass damage was developed and implemented in the LS-DYNA (version R12) code via a user-defined material model, with parameters determined using the Hoek–Brown failure criterion. A finite element model was established to analyze surrounding rock damage under cyclic blasting loads, and the model was validated using field data. Simulations were then carried out to explore the evolution of the damage zone under various stress conditions. The results show that with increasing hydrostatic pressure, the extent of the damage zone first decreases and then increases, with blasting-induced damage dominating under lower pressure and unloading-induced shear failure prevailing at higher pressure. When the hydrostatic pressure is less than 20 MPa, the surrounding rock stabilizes at a distance greater than 12.6 m from the tunnel face, whereas at hydrostatic pressures of 30 MPa and 40 MPa, this distance increases to 29.4 m. When the lateral pressure coefficient is low, tensile failure occurs mainly at the vault and floor, while shear failure dominates at the arch waist. As the lateral pressure coefficient increases, the failure mode at the vault shifts from tensile to shear. Additionally, when the horizontal stress perpendicular to the tunnel axis (σH) is less than the vertical stress (σv), variations in the axial horizontal stress (σh) have a significant effect on shear failure. The pressure dependence of structural behavior in the orthorhombic (Pccn, PI) and monoclinic (P21/c, PII) polymorphs of the compound [Fe(PM-BiA)2(NCS)2], where PM–BiA = (N–(2′–pyridylmethylene)–4-amino–bi–pheynyl), is studied with synchrotron single-crystal X-ray diffraction and vibrational spectroscopy. Both polymorphs are stable up to ∼1.5 GPa, with a spin state transition occurring only in polymorph PII under hydrostatic conditions as documented by single-crystal synchrotron diffraction. The diffraction data also provide evidence of the formation of superstructures for both PI, with a doubled c axis, and PII, with a doubled b axis, on applying pressures above 2 GPa. The LS and HS states seem to coexist at high-pressures for both polymorphs studied with synchrotron infrared spectroscopy at quasi-hydrostatic conditions. Such results indicate that the occurrence of spin-crossover transformations in [Fe(PM-BiA)2(NCS)2] might strongly depend on the stress in the sample. Full article

This study presents, for the first time, a detailed linear stability analysis (LSA) of bedform evolution under low-flow conditions using a one-dimensional vertically averaged and moment (1D-VAM) approach. The analysis focuses exclusively on bedload transport. The classical Saint-Venant shallow water equations are extended to incorporate non-hydrostatic pressure terms and a modified moment-based Chézy resistance formulation is adopted that links bed shear stress to both the depth-averaged velocity and its first moment (near-bed velocity). Applying a small-amplitude perturbation analysis to an initially flat bed, while neglecting suspended load and bed slope effects, reveals two distinct modes of morphological instability under low-Froude-number conditions. The first mode, associated with ripple formation, features short wavelengths independent of flow depth, following the relation F2 = 1/(kh), and varies systematically with both the Froude and Shields numbers. The second mode corresponds to dune formation, emerging within a dimensionless wavenumber range of 0.17 to 0.9 as roughness increases and the dimensionless Chézy coefficient C decreases from 20 to 10. The resulting predictions of the dominant wavenumbers agree well with recent experimental observations. Critically, the model naturally produces a phase lag between sediment transport and bedform geometry without empirical lag terms. The 1D-VAM framework with Exner equation offers a physically consistent and computationally efficient tool for predicting bedform instabilities in erodible channels. This study advances the capability of conventional depth-averaged models to simulate complex bedform evolution processes. Full article

An analysis of the impacts of shock-wave propagation media, explosive distance, charge equivalence, hydrostatic pressure, and shell thickness on the dynamic response of these bottom-sitting shell structures is conducted. The findings reveal that the deformation of semi-spherical steel shells subjected to underwater shock waves is significantly greater than that of shells subjected to air shock waves, with effective stress reaching up to 831.4 MPa underwater. The mechanical deformation of curved steel shells exhibits a gradual increase with increasing explosive equivalents. The center displacement of the hemispherical shell at 800 kg equivalent is 6 times that at 50 kg equivalent. Within the range of 0 to 2.0092 MPa, hydrostatic pressure leads to an approximate 26.34% increase in the center vertical displacement of the semi-cylindrical shell compared with 0 MPa, while restricting horizontal convex deformation. Increasing thickness from 0.025 m to 0.05 m results in a reduction of approximately 60% in the center vertical displacement of the semi-cylindrical shell. These quantitative correlations provide critical benchmarks for enhancing the blast resilience of underwater foundation systems. Full article

This study proposes a novel unified constitutive model that systematically integrates the microstructure evolution and macroscopic stress–strain response during the hot deformation of a Ni-based superalloy. The proposed model incorporates a suite of microstructural variables, including damage fraction, recrystallization fraction, δ phase content, average grain size, and dislocation density. Furthermore, the model explicitly considers critical macroscopic stress state parameters, specifically the magnitude and orientation of maximum principal stress, hydrostatic stress component, and Mises equivalent stress. A comparative analysis of rheological curves derived from uniaxial tension and compression experiments reveals that the prediction errors of the proposed model are less than 3%. The model is subsequently implemented to investigate the evolution characteristics of the damage accumulation fraction and δ phase content under varying stress directions and initial δ phase contents. An advanced computational framework integrating the finite element method with the proposed constitutive model is established through customized subroutines. The framework exhibits exceptional predictive accuracy across critical regions of disk forging, as evidenced by a close agreement between computational and experimental results. Specifically, the relative errors for predicting recrystallization fraction and average grain size remain consistently below 8% under varying stress–strain conditions. Testing results from four representative regions demonstrate a good alignment of high-temperature tensile properties with the macroscopic stress–strain distributions and microstructure characteristics, thereby confirming the model’s reliability in simulating and optimizing the forging process. Composite pressure vessels offer significant advantages over traditional metal-lined designs due to their high strength-to-weight ratio, corrosion resistance, and design flexibility. This study investigates the structural design, winding process, finite element analysis, and experimental validation of a glass fiber-reinforced composite low-pressure vessel. A high-density polyethylene (HDPE) liner was designed with a nominal thickness of 1.5 mm and manufactured via blow molding. The optimal blow-up ratio was determined as 2:1, yielding a wall thickness distribution between 1.39 mm and 2.00 mm under a forming pressure of 6 bar. The filament winding process was simulated using CADWIND software (version 10.2), resulting in a three-layer winding scheme consisting of two helical layers (19.38° winding angle) and one hoop layer (89.14°). The calculated thickness of the composite winding layer was 0.375 mm, and the coverage rate reached 107%. Finite element analysis, conducted using Abaqus, revealed that stress concentrations occurred at the knuckle region connecting the dome and the cylindrical body. The vessel was predicted to fail at an internal pressure of 5.00 MPa, primarily due to fiber breakage initiated at the polar transition. Experimental hydrostatic burst tests validated the simulation, with the vessel exhibiting failure at an average pressure of 5.06 MPa, resulting in an error margin of only 1.2%. Comparative tests on vessels without adhesive sealing at the head showed early failure at 2.46 MPa, highlighting the importance of head sealing on vessel integrity. Scanning electron microscopy (SEM) analysis confirmed strong fiber–matrix adhesion and ductile fracture characteristics. The close agreement between the simulation and experimental results demonstrates the reliability of the proposed design methodology and validates the use of CADWIND and FEA in predicting the structural performance of composite pressure vessels.
