Enhancing forage protein is key to sustainable ruminant nutrition. The nutritive value of Hedysarum coronarium L. was investigated by studying different preservation systems (fresh vs. dehydrated pellet vs. hay) (Exp. 1) and morphological fractions (flowers vs. leaves vs. stems) (Exp. 2). For the fresh and pelleted systems, two cuts were used. Proximately, total polyphenols and condensed tannins were detected. In vitro fermentation characteristics were studied by incubating samples with buffered sheep rumen fluid, estimating methane production by volatile fatty acids. Fresh and pelleted sulla were more nutritionally advantageous than hay, in terms of metabolizable energy and protein-to-fiber ratio. Pelleting at the beginning of flowering proved to be a suitable forage for sheep feeding due to protein (15.1% DM), metabolizable energy (9.64 MJ/kg DM), structural carbohydrates (39.5% DM), and total polyphenols (13.5 GAE g/kg DM) content. This cut showed an in vitro fermentation rate (9.86 mL/h), organic matter degradability (55.7%), and volatile fatty acids (87.3 mmoL/g) that were higher (p < 0.05) than hay. Flowers and leaves showed higher (p < 0.05) levels of secondary metabolites than stems as well as lower methane production. These results suggest the potential influence of these compounds in reducing rumen emissions. Dehydration and pelleting resulted in an effective preservation method for maintaining nutrients in sulla forage. Full article

Seeds of Cucurbitaceae crops represent a promising yet underexplored source of bioactive compounds with potential applications beyond nutrition, particularly in the cosmetics industry. This review examines the seeds of Citrullus lanatus (watermelon), Cucumis melo (melon), and Cucurbita pepo (pumpkin), focusing on their biochemical composition and evaluating their functional value in natural cosmetic development. Although these fruits are widely consumed, industrial processing generates substantial seed by-products that are often discarded. These seeds are rich in polyunsaturated fatty acids, proteins, carbohydrates, and phytochemicals, positioning them as sustainable raw materials for value-added applications. The incorporation of seed-derived extracts into cosmetic formulations offers multiple skin and hair benefits, including antioxidant activity, hydration, and support in managing conditions such as hyperpigmentation, acne, and psoriasis. They also contribute to hair care by improving oil balance, reducing frizz, and enhancing strand nourishment. However, challenges such as environmental instability and low dermal permeability of seed oils have prompted interest in nanoencapsulation technologies to improve delivery, stability, and efficacy. This review summarizes current scientific findings and highlights the potential of Cucurbitaceae seeds as innovative and sustainable ingredients for cosmetic and personal care applications. The market for plant-based alternatives to animal foods has increased rapidly in the past decade, mainly due to consumer demand. Little evidence is available regarding nutritional impacts, drivers, and barriers to using these products as substitutes for animal foods in real-life conditions. This pilot study followed 16 Danish adults (30 ± 11 years old; 11 females) for 4 weeks with substituting milk, cheese, and yogurt with plant-based analogues to dairy (PBADs) and assessed their drivers and barriers to applying the intervention with a mixed-method approach. PBADs are constantly compared to their animal counterparts, both regarding product characteristics, such as price and sensory properties, as well as cultural roles and subjective memories. The mixed methods showed dairy attachment, price, and taste were the main barriers to consuming PBAD, while changes in life and social circles were drivers (qualitative data). As for the liking of PBADs, plant-based yoghurt was the preferred intervention product (73.5/100, p < 0.05), followed by plant-based drinks (65.9/100), while plant-based cheese was the lowest rated (47.9/100, p < 0.05). As for dietary changes, a lower average intake of sugars, saturated fatty acids, cholesterol, calcium, phosphorus, and zinc was observed after the intervention. Additionally, this study describes the attachment of the study population to milk and dairy products. It shows that choosing dairy is beyond nourishment but is connected to tradition, culture, pleasure, memories, and a sense of belonging. In contrast, there is no history or attachment to PBADs. This study examines the impact of hydrothermal pretreatment operation time (10, 20, and 30 min) on the following four lignocellulosic feedstocks with different lignin content: sugar beet pulp (SBP), brewers spent grain (BSG), orange peel (OP), and rice husk (RH). The objective of pretreatment is twofold, as follows: (1) to enhance the organic matter solubilization and the release of value-added bioproducts, such as total reducing sugars (TRS), total proteins (PR), and volatile fatty acids (VFAs); and (2) to improve VFA and hydrogen production during a subsequent stage of acidogenic anaerobic digestion (Dark Fermentation, DF). In this context, OP reported the highest overall yields across all pretreatment durations. Specifically, at 30 min, it achieved a maximum solubilization of 57.3 gO2/L in terms of soluble chemical oxygen demand (sCOD), 19.1 gTRS/L and 20.6 gPR/L. Regarding VFA and hydrogen production via dark fermentation, the best results were obtained with SBP pretreated for 20 and 30 min, yielding 15.1 g H-Ac/L and 97.5 mL H2 (n.c.)/g (d.m.), respectively. BSG displayed an intermediate performance, whereas RH consistently showed the lowest yields across all evaluated parameters, primarily due to its high lignin content. These findings highlight the pivotal role of pretreatment duration in the valorization of lignocellulosic biomasses, primarily aimed at the recovery of high-value-added biochemicals and biofuels, such as hydrogen, thereby supporting the development of integrated biorefinery systems. The retina is highly sensitive to oxygen and blood supply, and hypoxia plays a key role in retinal diseases such as diabetic retinopathy (DR) and age-related macular degeneration (AMD). Müller glial cells, which are essential for retinal homeostasis, respond to injury and hypoxia with reactive gliosis, characterized by the upregulation of the glial fibrillary acidic protein (GFAP) and vimentin, cellular hypertrophy, and extracellular matrix changes, which can impair retinal function and repair. The retinal pigment epithelium (RPE) supports photoreceptors, forms part of the blood–retinal barrier, and protects against oxidative stress; its dysfunction contributes to retinal degenerative diseases such as AMD, retinitis pigmentosa (RP), and Stargardt disease (SD). Extracellular vesicles (EVs) play a crucial role in intercellular communication, protein homeostasis, and immune modulation, and have emerged as promising diagnostic and therapeutic tools. Understanding the role of extracellular vesicles’ (EVs’) signaling machinery of glial cells and the retinal pigment epithelium (RPE) is critical for developing effective treatments for retinal degeneration. In this study, we investigated the bidirectional EV-mediated crosstalk between RPE and Müller cells under hypoxic conditions and its impact on cellular metabolism and retinal cell integrity. Our findings demonstrate that RPE-derived extracellular vesicles (RPE EVs) induce time-dependent metabolic reprogramming in Müller cells. Short-term exposure (24 h) promotes pathways supporting neurotransmitter cycling, calcium and mineral absorption, and glutamate metabolism, while prolonged exposure (72 h) shifts Müller cell metabolism toward enhanced mitochondrial function and ATP production. Conversely, Müller cell-derived EVs under hypoxia influenced RPE metabolic pathways, enhancing fatty acid metabolism, intracellular vesicular trafficking, and the biosynthesis of mitochondrial co-factors such as ubiquinone. Proteomic analysis revealed significant modulation of key regulatory proteins. In Müller cells, hypoxic RPE-EV exposure led to reduced expression of Dyskerin Pseudouridine Synthase 1 (DKc1), Eukaryotic Translation Termination Factor 1 (ETF1), and Protein Ser/Thr phosphatases (PPP2R1B), suggesting alterations in RNA processing, translational fidelity, and signaling. RPE cells exposed to hypoxic Müller cell EVs exhibited elevated Ribosome-binding protein 1 (RRBP1), RAC1/2, and Guanine Nucleotide-Binding Protein G(i) Subunit Alpha-1 (GNAI1), supporting enhanced endoplasmic reticulum (ER) function and cytoskeletal remodeling. Functional assays also revealed the compromised barrier integrity of the outer blood–retinal barrier (oBRB) under hypoxic co-culture conditions. These results underscore the adaptive but time-sensitive nature of retinal cell communication via EVs in response to hypoxia. Targeting this crosstalk may offer novel therapeutic strategies to preserve retinal structure and function in ischemic retinopathies. Full article

Development and Validation of a Custom-Built System for Real-Time Monitoring of In Vitro Rumen Gas Fermentation

While the Ankom RF system facilitates efficient high-throughput in vitro fermentation studies, its high cost and limited flexibility constrain its broader applicability. To address these limitations, we developed and validated a low-cost, modular gas monitoring system (FerME), assembled from commercially available components. To evaluate its performance and reproducibility relative to the Ankom RF system (Ankom Technology, Macedon, NY, USA), in vitro rumen fermentation experiments were conducted under strictly controlled and identical conditions. Whole rumen contents were collected approximately 2 h post-feeding from individual mid- or late-lactation dairy cows and immediately transported to the laboratory. Each fermenter received 50 mL of processed rumen fluid, 100 mL of anaerobically prepared artificial saliva buffer, and 1.2 g of the donor cow’s diet. Bottles were sealed with the respective system’s pressure sensors, flushed with CO2, and incubated in a 50 L water bath maintained at 39 °C. FerME (New Taipei City, Taiwan) and Ankom RF fermenters were placed side-by-side to ensure uniform thermal conditions. To assess the effect of filter bag use, an additional trial employed Ankom F57 filter bags (Ankom Technology, Macedon, NY, USA; 25 μm pore size). Trial 1 revealed no significant differences in cumulative gas production, volatile fatty acids (VFAs), NH3-N, or pH between systems (p > 0.05). However, the use of filter bags reduced gas output and increased propionate concentrations (p < 0.05). Trial 2, which employed filter bags in both systems, confirmed comparable results, with the FerME system demonstrating improved precision (CV: 4.8% vs. 13.2%). Gas composition (CH4 + CO2: 76–82%) and fermentation parameters remained consistent across systems (p > 0.05). Importantly, with 12 pressure sensors, the total cost of FerME was about half that of the Ankom RF system. Collectively, these findings demonstrate that FerME is a reliable, low-cost alternative for real-time rumen fermentation monitoring and could be suitable for studies in animal nutrition, methane mitigation, and related applications. Hydrotreated ester and fatty acids to jet (HEFA-tJ) is presently the most developed and economically attractive pathway to produce sustainable aviation fuel (SAF). An ongoing systematic study of the critical variables of different pathways to SAF has revealed significantly lower greenhouse gas (GHG) reduction potential for the HEFA-tJ pathway compared to competing markets using the same resources for road diesel production. Moderate yield variations between air and road pathways lead to several hundred thousand tons less GHG reduction per project, which is generally not evaluated thoroughly in standard environmental assessments. This work demonstrates that, although the HEFA-tJ market seems to have more attractive features than biodiesel/renewable diesel, considerable viability risks might manifest as HEFA-tJ fuel market integration rises. The need for more transparent data and effort in this regard, before envisaging making decisions regarding the volume of HEFA-tJ production, is emphasized. Overall, reducing the carbon intensity of road diesel appears to be less capital-intensive, less risky, and several times more efficient in reducing GHG emissions. Full article

Although the strawberry tree (A. unedo L.) has been long considered a neglected species of the Mediterranean maquis, the valorization of its fruit production may enhance its socioeconomic value, especially in rural areas. In this study, strawberry trees from different Sicilian sites were investigated in terms of macronutrients, fatty acid (FA) composition, tocopherols, total phenols, carotenoids, and minerals. Sicilian berries were a good source of carbohydrates (mainly fructose, glucose and sucrose) and dietary fiber. They were low in fat; however, the FA composition revealed the abundance of unsaturated FAs over saturated FAs and an advantageous n-6/n-3 ratio. Additionally, Sicilian berries showed an inversed linoleic/α-linolenic acid ratio with respect to berries from other Mediterranean regions, that had previously investigated in literature. This evidence suggests that this ratio may have a chemotaxonomic relevance. Considering antioxidants, the fruits had levels of tocopherols, particularly α-tocopherol, total phenols and carotenoids similar to those of certain commercial fruits. Precious amounts of minerals, such as Ca, K, Zn and Fe were also determined. Interestingly, berries harvested near a Sicilian volcanic area had higher levels of minerals, as well as tocopherols, phenols and carotenoids, than fruits from other Sicilian sites, thereby advancing the hypothesis that fruits from volcanic areas may have a superior nutritional value. Overall, data from this study elaborated by a proper statistical analysis revealed that the geographical origin was a relevant variable to consider in the reliable study of this fruit species. Effects of Ultrafine Bubble Water on Gut Microbiota Composition and Health Markers in Rats

Ultrafine bubbles (UFBs) represent an emerging technology with unique physicochemical properties. This study investigated the effects of air-filled UFBs infused in drinking water on gut microbiota composition and the associated health markers in Sprague Dawley rats over a 12-week period. Using a two-phase design, UFB concentration was increased from 1.7 × 106 to 6.5 × 109 UFBs/mL at week 7 to assess dose-dependent effects. Administration of UFBs in drinking water induced significant shifts in gut microbiome populations, characterized by increased Bacteroidetes (+122% weeks 8–12) and decreased Firmicutes (−43% weeks 8–12) compared to controls. These microbial shifts coincided with enhanced short-chain fatty acid production (butyrate +56.0%, p ≤ 0.001; valerate +63.1%, p ≤ 0.01) and reduced inflammatory markers (TNF-α −84.0%, p ≤ 0.05; IL-1β −41.0%, p ≤ 0.05; IL-10 −69.8%, p ≤ 0.05). UFB effects demonstrated systematic concentration-dependent threshold responses, with 85.7% of parameters exhibiting directional reversals between low (1.7 × 106 UFBs/mL) and high (6.5 × 109 UFBs/mL) concentration phases rather than linear dose–response relationships. The systematic nature of these threshold effects, with 71.4% of parameters achieving statistical significance (p ≤ 0.05), indicates concentration-dependent biological mechanisms rather than random effects on gut biology. Despite current metagenomic techniques identifying only 25% of the total gut microbiome, the observed changes in characterized species and metabolites demonstrate UFB technology’s therapeutic potential for conditions requiring microbiome modulation, providing new insights into UFB influence on complex biological systems. Understanding the components of the diet, food groups, and nutritional strategies that help prevent MASLD (Metabolic Dysfunction-Associated Steatotic Liver Disease) is essential for identifying dietary behaviors that can stop the progression of this condition, which currently affects over one-quarter of the global population. This review highlights the importance of including antioxidant nutrients in the diet, such as vitamins C and E, CoQ10, and polyphenolic compounds. It also emphasizes substances that support lipid metabolism, including choline, alpha-lipoic acid, and berberine. Among food groups, it is crucial to choose those that help prevent metabolic disturbances. Among carbohydrate-rich foods, vegetables, fruits, and high-fiber products are recommended. For protein sources, eggs, fish, and white meat are preferred. Among fat sources, plant oils and fatty fish are advised due to their content of omega-3 and omega-6 fatty acids. Various dietary strategies aimed at preventing MASLD should include elements of the Mediterranean diet or be personalized to provide anti-inflammatory compounds and substances that inhibit fat accumulation in liver cells. Other recommended dietary models include the DASH diet, the flexitarian diet, intermittent fasting, and diets that limit fructose and simple sugars. Additionally, supplementing the diet with spirulina or chlorella, berberine, probiotics, or omega-3 fatty acids, as well as drinking several cups of coffee per day, may be beneficial. Full article

Low-commercial-value natural pistachios (broken, closed, or immature) can be revalorised through oil extraction, obtaining a high-quality oil and partially defatted flour as by-product. This study evaluated the techno-functional and nutritional properties of the flours obtained by hydraulic press (HP) and single-screw press (SSP) systems, combined with pretreatment at 25 °C and 60 °C. The extraction method significantly influenced flour’s characteristics, underscoring the need to tailor processing conditions to the specific technological requirements of each food application. HP-derived flours presented lighter colour, greater tocopherol content, and higher water absorption capacity (up to 2.75 g/g), suggesting preservation of hydrophilic proteins. SSP-derived flours showed higher concentration of protein (44 g/100 g), fibre (12 g/100 g), and minerals, and improved emulsifying properties, enhancing their suitability for emulsified products. Pretreatment at 25 °C enhanced functional properties such as swelling power (~7.0 g/g) and water absorption index (~5.7 g/g). The SSP system achieved the highest oil extraction yield, with no significant effect of pretreatment temperature. The oils extracted showed high levels of unsaturated fatty acids, particularly oleic acid (~48% of ω-9), highlighting their nutritional and industrial value. The findings support the valorisation of pistachio oil extraction by-products as functional food ingredients, offering a promising strategy for reducing food waste and promoting circular economy approaches in the agri-food sector. Adipose tissue inflammation contributes to obesity-induced insulin resistance. However, increasing evidence shows that high BMI (obesity) is not an accurate predictor of poor metabolic health in individuals. The molecular mechanisms regulating the metabolically activated M1 macrophage phenotype in the adipose tissues leading to insulin resistance remain largely unknown. Although the Janus Kinase (Jak)/signal transducer and activator of transcription 3 (Stat3) signaling in myeloid cells are known to promote the M2 phenotype in tumors, we demonstrate here that the Jak2/Stat3 pathway amplifies M1-mediated adipose tissue inflammation and insulin resistance under metabolic challenges. Ablating Jak2 in the myeloid compartment reduces insulin resistance in obese mice, which is associated with a decrease in infiltration of adipose tissue macrophages (ATMs). We show that the adoptive transfer of Jak2-deficient myeloid cells improves insulin sensitivity in obese mice. Furthermore, the protection of obese mice with myeloid-specific Stat3 deficiency against insulin resistance is also associated with reduced tissue infiltration by macrophages. Jak2/Stat3 in the macrophage is required for the production of pro-inflammatory cytokines that promote M1 macrophage polarization in the adipose tissues of obese mice. Moreover, free fatty acids (FFAs) activate Stat3 in macrophages, leading to the induction of M1 cytokines. Silencing the myeloid cell Stat3 with an in vivo siRNA targeted delivery approach reduces metabolically activated pro-inflammatory ATMs, thereby alleviating obesity-induced insulin resistance. These results demonstrate Jak2/Stat3 in myeloid cells is required for obesity-induced insulin resistance and inflammation. Moreover, targeting Stat3 in myeloid cells may be a novel approach to ameliorate obesity-induced insulin resistance. Full article

A comprehensive metabolic profiling of Catharanthus roseus (L.) G. Don was performed using tandem mass spectrometry, along with an evaluation of the biological activities of its various solvent extracts. Among these, the methanolic leaf extract exhibited mild radical scavenging activity, low to moderate antimicrobial activity, and limited cytotoxicity in both the brine shrimp lethality assay and MTT assay against HeLa and A549 cell lines. High-performance liquid chromatography–electrospray ionization–high-resolution tandem mass spectrometry (HPLC-ESI-HRMS/MS) analysis led to the annotation of 34 metabolites, primarily alkaloids. These included 23 indole alkaloids, two fatty acids, two pentacyclic triterpenoids, one amino acid, four porphyrin derivatives, one glyceride, and one chlorin derivative. Notably, two metabolites—2,3-dihydroxypropyl 9,12,15-octadecatrienoate and (10S)-hydroxypheophorbide A—were identified for the first time in C. roseus. Furthermore, Global Natural Products Social Molecular Networking (GNPS) analysis revealed 18 additional metabolites, including epoxypheophorbide A, 11,12-dehydroursolic acid lactone, and 20-isocatharanthine. These findings highlight the diverse secondary metabolite profile of C. roseus and support its potential as a source of bioactive compounds for therapeutic development. Full article

This study evaluates the production of base oils for biolubricants using fatty acid methyl esters (FAMEs) derived from sunflower oil as the raw material. The production process involved the synthesis of oleochemical esters through a single-step alkaline transesterification reaction with a high-molecular-weight polyol, such as trimethylolpropane (TMP). To assess the effectiveness of the developed catalytic system in conducting the transesterification reactions and its impact on the properties of the final product, two types of alkaline catalysts were used. Specifically, the reactions were carried out using either Ca/TEA alkoxide or sodium methoxide as catalysts in various configurations and concentrations to determine the optimal catalyst concentration and reaction conditions. Sodium methoxide served as the commercial benchmark catalyst, while the Ca/TEA alkoxide was prepared in the laboratory. The optimal concentration of Ca/TEA was determined to be 3.0% wt. in the presence of iso-octane and 3.5% wt. under vacuum, while the corresponding concentrations of CH3ONa for both cases were determined to be 2.0% wt. The synthesized biolubricant esters exhibit remarkable performance characteristics, such as high kinematic viscosities and low pour points—ranging from 33–48 cSt at 40 °C, 7.68–10.03 cSt at 100 °C, to −14 to −7 °C, respectively—which are comparable to or improved over those of mineral oils such as SN-150 or SN-500, with the Ca/TEA alkoxide-catalyzed systems showing superior oxidation stability and reduced byproduct formation.
