Air pollutants pose a growing public health concern in Puerto Rico (PR), particularly from rapid industrialization, military activities, environmental changes and natural disasters. A total of 193 pollutants, comprising the 187 hazardous air pollutants and the 6 criteria air pollutants—including particulate matter (PM), carbon monoxide (CO), volatile organic compounds (VOC), and heavy metals—coincide with rising respiratory disease rates (e.g., lung cancer) documented in national and regional health registries. This study aimed to review major air pollutants in PR, their molecular carcinogenic mechanisms (mostly focused on respiratory-related cancers), and the geographic areas impacted significantly. We conducted an extensive literature search utilizing peer-reviewed scientific articles (PubMed and Web of Science), governmental reports (EPA, WHO, State of Global Air), public health registries, (Puerto Rico Central Cancer Registry and International Agency for Research on Cancer) and local reports. Data on pollutant type, source, molecular pathways, and carcinogenic properties were extracted and synthesized. Our analysis identified ethylene oxide (EtO), polycyclic aromatic hydrocarbons, and PM from industrial sites as key pollutants. The municipalities of Salinas and Vieques, hubs of industrial activity and military exercises, respectively, emerged as critical hotspots where high concentrations of monitored pollutants (e.g., EtO, formaldehyde, 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and diesel PM) are associated with a significant prevalence of cancer and respiratory diseases. These agents, known to induce genomic instability and chromosomal aberrations, were correlated with elevated local cancer incidence. Our findings underscore the urgent need for targeted public health interventions and support a multi-pronged strategy that includes: (1) enhanced regulatory oversight of EtO and other hazardous air pollutant emissions; (2) community-based biomonitoring of high-risk populations; and (3) investment in public health infrastructure and a transition to cleaner energy sources. Integrating rigorous environmental science with public health advocacy is essential to strengthen PR’s cancer-control continuum and foster resilience in its most vulnerable communities. Full article

The transportation sector mainly relied on fossil fuel and is one of the major causes of climate change and environmental pollution. Advances in smart sensing technology are paving the way for the development of clean and intelligent vehicles that lead to a more sustainable transportation system. In response, the automotive industry is actively engaging in new sensor technologies and innovative control and diagnostic algorithms that improve energy sustainability and reduce vehicle emissions. In particular, recent regulations for diesel vehicles require the integration of smart soot sensors to deal with particulate filter on-board diagnostic (OBD) challenges. Meeting the recent, more stringent OBD requirements will be difficult using traditional diagnostic approaches. This study investigates an advanced diagnostic strategy to assess particulate filter health based on resistive soot sensors and available engine variables. The sensor data are projected to generate a 2D signature that reflects the changes in filtration efficiency. A relevant feature (character) is then extracted from the generated signature that can be transformed into an analytical expression used as an indicator of DPF malfunction. The diagnostic strategy uses an adaptive approach that dynamically adjusts the signature’s characters according to the engine’s operating conditions. A correction factor is calculated using an optimization algorithm based on the integral of engine speed measurements and IMEP set points during each sensor loading period. Different cost functions have been tested and evaluated to improve the diagnostic performance. The proposed adaptive approach is model-free and eliminates the need for subsystem models, iterative algorithms, and extensive calibration procedures. Furthermore, the time-consuming and inaccurate estimation of soot emissions upstream of the DPF is avoided. It was evaluated on a validated numerical platform under NEDC driving conditions with simultaneous dispersions on engine-out soot concentration and soot sensor measurements. The promising results highlight the robustness and superior performance of this approach compared to a diagnostic strategy solely reliant on sensor data. Full article

Background/Objectives: Diesel Particulate Matter (DPM) is a very small particulate matter originating from cities, factories, and the use of fossil fuels in diesel vehicles. When DPM permeates the skin, it causes inflammation, leading to severe atopic dermatitis. Hibiscus cannabinus L. (Kenaf) seeds and leaves possess various beneficial properties, including anti-coagulation, antioxidant, and anti-inflammation effects. In this study, we investigated the anti-inflammatory effects of an ethanol extract of Hibiscus cannabinus L. flower (HCFE) in HaCaT cells stimulated with 100 μg/mL of DPM. Methods: The anthocyanin content of HCFE was analyzed, and its antioxidant capacity was investigated using the DPPH assay. After inducing inflammation with 100 ug/mL of DPM, the cytotoxicity of HCFE 25, 50, and 100 ug/mL was measured, and the inhibitory effect of HCFE on inflammatory mediators was evaluated. Results: Anthocyanin and myricetin-3-O-glucoside were present in HCFE and showed high antioxidant capacity. In addition, HCFE decreased the mRNA expression of inflammatory cytokines and chemokines such as IL-1β, IL-4, IL-6, IL-8, IL-13, and MCP-1, and significantly reduced the gene expression of CXCL10, CCL5, CCL17, and CCL22, which are known to increase in atopic dermatitis lesions. Furthermore, HCFE reduced intracellular reactive oxygen species (ROS) production, and down-regulated the activation of NF-κB, MAPKs. Inhibition of the NLRP-3 inflammasome was observed in DPM-stimulated HaCaT cells. In addition, the restoration of filaggrin and involucrin, skin barrier proteins destroyed by DPM exposure, was confirmed. Conclusions: These data suggest that HCFE could be used to prevent and improve skin inflammation and atopic dermatitis through the regulation of inflammatory mediators and the inhibition of skin water loss. Exposure to air pollution poses a risk to human respiratory health, and a preventive and therapeutic remedy against fine dust-induced respiratory disease is needed. Background/Objectives: The respiratory-protective effects of Lysimachia mauritiana (LM) against airway inflammation were evaluated in a mouse model exposed to a fine dust mixture of diesel exhaust particles and particulate matter with a diameter of less than 10 µm (PM10D). Methods: To induce airway inflammation, PM10D was intranasally injected into BALB/c mice three times a day for 12 days, and LM extracts were given orally once per day. The immune cell subtypes, histopathology, and expression of inflammatory mediators were analyzed from the bronchoalveolar lavage fluid (BALF) and lungs. Results: LM alleviated the accumulation of neutrophils and the number of inflammatory cells in the lungs and the BALF of the PM10D-exposed mice. LM also reduced the release of inflammatory mediators (MIP-2, IL-17, IL-1α, CXCL1, TNF-α, MUC5AC, and TRP receptor channels) in the BALF and lungs. Lung histopathology was used to examine airway inflammation and the accumulation of collagen fibers and inflammatory cells after PM10D exposure and showed that LM administration improved this inflammation. Furthermore, LM extract inhibited the MAPK and NF-κB signaling pathway in the lungs and improved expectoration activity through an increase in phenol red release from the trachea. Conclusions: LM alleviated PM10D-exposed neutrophilic airway inflammation by suppressing MAPK/NF-κB activation. This study indicates that LM extract may be an effective therapeutic agent against inflammatory respiratory diseases. Full article

Air pollution causes various airway diseases. However, many commonly used treatments can have high risks of side effects or are costly. To examine the anti-inflammatory properties of Inula japonica Thunb. and Potentilla chinensis Ser., a mouse model was generated via inhalation of both particulate matter 10 and diesel particulate matter, and 30% ethanol extracts of either I. japonica (IJ) or P. chinensis (PC) and a mixture of both ethanol extracts (IP) were orally administered to BALB/c mice for 12 days. IJ, PC, and IP inhibited immune cell numbers and their regulation in both the bronchoalveolar lavage fluid (BALF) and lungs. These agents suppressed the levels of interleukin (IL)-1α, IL-17, tumor necrosis factor (TNF)-α, C-X-C motif chemokine ligand (CXCL)-1, and CXCL-2 in BALF, and also inhibited F4/80 and IL-1 receptor-associated kinase (IRAK)-1 in lungs. They reduced the gene expression of TNF-α, CXCL-1, inducible NOS, COX-2, Mucin 5AC, and transient receptor potential cation channel subfamily V member 1 in lungs. These extracts also reduced histopathological changes and inflammatory progression, manifested as decreased cell infiltration, collagen deposition, and respiratory epithelial cell thickness. I. japonica and P. chinensis show potential for development as pharmaceuticals that suppress inflammatory progression and alleviate airway inflammation diseases caused by air pollutants. Full article

Exposure to particulate matter (PM) causes considerable breathing-related health risks. Siraitia grosvenorii fruit is a traditional remedial plant used in Korea and China to treat respiratory diseases. Our recently published study showed that S. grosvenorii extract (SGE) ameliorated airway inflammation in lipopolysaccharide- and cigarette-smoke-induced chronic obstructive pulmonary disease in mice. Thus, we aimed to assess the inhibitory effects of SGE on airway inflammation in mice exposed to a fine dust mixture of PM10 (PM diameter < 10 mm) and diesel exhaust particles (DEPs) known as PM10D. The mice (BALB/c) were treated with PM10D via intranasal injection three times over a period of 12 days, and SGE 70% ethanolic extract (50 or 100 mg/kg) was orally administered daily for 12 days. SGE attenuated neutrophil accumulation and the number of immune B and T cells from the lung tissue and bronchoalveolar lavage fluid (BALF) of the PM10D-exposed mice. SGE reduced the secretion of cytokines and chemokines, including interleukin (IL)-1α, tumor necrosis factor (TNF)-α, IL-17, C-X-C motif chemokine ligand (CXCL)1, and macrophage inflammatory protein (MIP)-2 in the BALF. Airway inflammation, infiltration of inflammatory cells, and collagen fibrosis in the lung after PM10D exposure were investigated via histopathological analysis, and SGE treatment ameliorated these symptoms. SGE decreased the mRNA expression of mucin 5AC (MUC5AC), CXCL1, TNF-α, MIP-2, and transient receptor potential ion channels in the lung tissues. Furthermore, SGE ameliorated the activation of mitogen-activated protein kinase (MAPK)/nuclear factor-kappa B (NF-κB) signaling by PM10D in the lungs. We conclude that SGE attenuated PM10D-induced neutrophilic airway inflammation by inhibiting MAPK/NF-κB activation. These results show that SGE may be a candidate for the treatment of inflammatory respiratory diseases. Full article

This study investigated the suitability of stone fruit seed as a source of biodiesel for transport. Stone fruit oil (SFO) was extracted from the seed and converted into biodiesel. The biodiesel yield of 95.75% was produced using the alkaline catalysed transesterification process with a methanol-to-oil molar ratio of 6:1, KOH catalyst concentration of 0.5 wt% (weight %), and a reaction temperature of 55 °C for 60 min. The physicochemical properties of the produced biodiesel were determined and found to be the closest match of standard diesel. The engine performance, emissions and combustion behaviour of a four-cylinder diesel engine fuelled with SFO biodiesel blends of 5%, 10% and 20% with diesel, v/v basis, were tested. The testing was performed at 100% engine load with speed ranging from 200 to 2400 rpm. The average brake specific fuel consumption and brake thermal efficiency of SFO blends were found to be 4.7% to 15.4% higher and 3.9% to 11.4% lower than those of diesel, respectively. The results also revealed that SFO biodiesel blends have marginally lower in-cylinder pressure and a higher heat release rate compared to diesel. The mass fraction burned results of SFO biodiesel blends were found to be slightly faster than those of diesel. The SFO biodiesel 5% blend produced about 1.9% higher NOx emissions and 17.4% lower unburnt HC with 23.4% lower particulate matter (PM) compared to diesel fuel. To summarise, SFO biodiesel blends are recommended as a suitable transport fuel for addressing engine emissions problems and improving combustion performance with a marginal sacrifice of engine efficiency. Air pollutants contribute to the development of diseases such as asthma, chronic obstructive pulmonary disease (COPD), pulmonary cancer, cardiovascular problems, and some skin diseases. We recently found that a major air pollutant, diesel particulate matter (DPM), induces apoptosis in human keratinocytes by increasing a proapoptotic lipid mediator, ceramide. DPM activates nicotinamide adenine dinucleotide phosphate (NADPH) oxidase (NOX), which stimulates sphingomyelinase, leading to an increased conversion of sphingomyelin to ceramide. Interestingly, we characterized that although NOX is a reactive oxygen species (ROS) generator, the activation of sphingomyelinases by NOX is an ROS-independent mechanism. A Korean weed, prostrate spurge Euphorbia supina Rafin (ESR), has been used for centuries as a folk medicine to treat bronchitis, hepatitis, hemorrhage, and skin inflammation. Flavonoids, terpenes and tannins are enriched in ESR, and although ESR has proven antioxidative activity, its biological activities are largely unknown. Here, we investigate whether and how ESR protects keratinocytes against DPM-mediated apoptosis. We found that ESR-extracts (ESR-Ex) protect keratinocytes from DPM-induced apoptosis by inhibiting NOX activation in keratinocytes in response to DPM. We also demonstrated that ESR-Ex suppresses NOX activation via a blockage of the aryl hydrocarbon receptor (AhR) activation-mediated transcription of neutrophil cytosolic factor 1 (NCF1)/p47phox, a subunit of NOX. Our study reveals previously uncharacterized biological activity of ESR-Ex; i.e., its inhibition of Ahr and NOX activation. Thus, because the inhibition of NOX has already been developed to treat NOX-mediated diseases, including various types of cardiovascular diseases and cancers, initiated by air pollutants and because AhR activation contributes to the development of chronic inflammatory diseases, our study provides further advantages for the medical use of ESR. Both intrinsic (i.e., an individual’s body clock) and extrinsic factors (i.e., air pollutants and ultraviolet irradiation) accelerate premature aging. Epidemiological studies have shown a correlation between pollutant levels and aging skin symptoms. Diesel particle matter in particular leads to some diseases, including in the skin. Our recent study demonstrates that diesel particulate extract (DPE) increases apoptosis via increases in an anti-mitogenic/pro-apoptotic lipid mediator, ceramide in epidermal keratinocytes. Here, we investigated whether and how DPE accelerates premature skin aging using cultured normal human dermal fibroblasts (HDF). We first demonstrated that DPE increases cell senescence marker β-galactosidase activity in HDF. We then found increases in mRNA and protein levels, along with activity of matrix metalloprotease (MMP)-1 and MMP-3, which are associated with skin aging following DPE exposure. We confirmed increases in collagen degradation in HDF treated with DPE. Nicotinamide adenine dinucleotide phosphate (NADPH) oxidase (NOX) is activated by DPE and results in increased ceramide production by sphingomyelinase activation in HDF. We identified that ceramide-1-phosphate (C1P) (produced from ceramide by ceramide kinase activation) activates MMP-1 and MMP-3 through activation of arachidonate cascade, followed by STAT 1- and STAT 3-dependent transcriptional activation. Particulate matter (PM) exposure may cause adverse health effects such as respiratory disorders. We evaluated the protective effects of various Opuntia ficus-indica (OFI) extracts on airway inflammation associated with exposure to PM10D with an aerodynamic diameter <10 μm (PM10) and diesel exhaust particles (DEP). BALB/c mice were exposed to PM10D via intranasal tracheal injection three times over a period of 12 days and various OFI extracts (water, 30% ethanolic, or 50% ethanolic extracts) were administered orally for 12 days. All OFI extracts suppressed neutrophil infiltration and the number of immune cells (CD3+/CD4+, CD3+/CD8+, and Gr-1+/CD11b) in bronchoalveolar lavage fluid (BALF) and lungs. OFI extracts decreased the expression of cytokines and chemokines, including chemokine (C-X-C motif) ligand (CXCL)-1, interleukin (IL)-17, macrophage inflammatory protein-2, tumor necrosis factor (TNF)-α, cyclooxygenase-2, IL-1α, IL-1β, IL-5, IL-6, transient receptor potential cation channel subfamily V member 1, and mucin 5AC, and inhibited IRAK-1, TNF-α, and CXCL-1 localization in BALF and lungs of mice with PM10D-induced airway inflammation. Serum asymmetric and symmetric dimethyl arginine levels were also decreased by OFI extracts treatment. Moreover, all OFI extracts restored histopathological damage in the trachea and lungs of mice with PM10D-induced airway inflammation. These results indicate that OFI extracts may be used to prevent and treat airway inflammation and respiratory diseases. Full article

Inhaled particulate air pollution exerts pulmonary inflammation and cardiovascular toxicity through secondary systemic effects due to oxidative stress and inflammation. Catalpol, an iridiod glucoside, extracted from the roots of Rehmannia glutinosa Libosch, has been reported to possess anti-inflammatory and antioxidant properties. Yet, the potential ameliorative effects of catalpol on particulate air pollution—induced cardiovascular toxicity, has not been studied so far. Hence, we evaluated the possible mitigating mechanism of catalpol (5 mg/kg) which was administered to mice by intraperitoneal injection one hour before the intratracheal (i.t.) administration of a relevant type of pollutant particle, viz. diesel exhaust particles (DEPs, 30 µg/mouse). Twenty-four hours after the lung deposition of DEPs, several cardiovascular endpoints were evaluated. DEPs caused a significant shortening of the thrombotic occlusion time in pial microvessels in vivo, induced platelet aggregation in vitro, and reduced the prothrombin time and the activated partial thromboplastin time. All these actions were effectively mitigated by catalpol pretreatment. Likewise, catalpol inhibited the increase of the plasma concentration of C-reactive proteins, fibrinogen, plasminogen activator inhibitor-1 and P- and E-selectins, induced by DEPs. Moreover, in heart tissue, catalpol inhibited the increase of markers of oxidative (lipid peroxidation and superoxide dismutase) and nitrosative (nitric oxide) stress, and inflammation (tumor necrosis factor α, interleukin (IL)-6 and IL-1β) triggered by lung exposure to DEPs. Exposure to DEPs also caused heart DNA damage and increased the levels of cytochrome C and cleaved caspase, and these effects were significantly diminished by the catalpol pretreatment. Moreover, catalpol significantly reduced the DEPs-induced increase of the nuclear factor κB (NFκB) in the heart. In conclusion, catalpol significantly ameliorated DEPs–induced procoagulant events and heart oxidative and nitrosative stress, inflammation, DNA damage and apoptosis, at least partly, through the inhibition of NFκB activation. The current study is focused on platinum recovery from the secondary sources of end-of-life heavy-duty diesel oxidation catalysts (DOCs) and heavy-duty catalyzed diesel particulate filters (c-DPFs) in order to reduce the supply–demand gap within the European Union. The extraction of platinum was based on a hydrometallurgical single-step low acidity leaching system (HCl-H2O2-NaCl) and a calcination step that takes place before the leaching process. The parameters of calcination and leaching process were thoroughly investigated in order to optimize recovery efficiency. The optimized results proved that a calcination step (at 800 °C for 2 h) improves the recovery efficiency by a factor of 40%. In addition, optimal Pt recovery yield was achieved after 3 h of leaching at 70 °C, with a solid-to-liquid (S/L) ratio of 70 g/100 mL (70%) and 3 M HCl-1% vol H2O2-4.5 M NaCl as leaching conditions. The optimum Pt recovery yield was 95% and 75% for DOC and c-DPF, respectively. Since the secondary feedstock investigated is highly concentrated in platinum, the pregnant solution can be used as a precursor for developing new Pt-based catalytic systems. Full article

The impact of the constituent oxides of nitrogen, carbon, sulphur, and other particulate matter which make up the gas emissions from diesel engines has motivated several control techniques for these pollutants. Water-in-diesel emulsions provide a reliable solution, but the wear effects on the fuel injection system (FIS) still pose remarkable concerns. Because pressure signals from the common rail (CR) reflect the dynamics associated with varying emulsion compositions and at varying engine RPMs, an investigative (and diagnostic) study was conducted on a KIA Sorento 2004 four-cylinder line engine at various water-in-diesel emulsion compositions and engine speeds. Alongside visual/microscopic inspections and spectral analyses, the diagnostic framework proposed herein functions on the use of standardized first-order differentials of the CR pressure signals to generate reliable continuous wavelet coefficients (CWCs) which capture discriminative spectral and transient information for accurate diagnosis. The results show that by extracting the CWCs from the first-order CR pressure differentials up to the 512th scale on a Mexican hat wavelet, adequate fault parameters can be extracted for use by a deep neural network (DNN) whose hyperparameters were globally optimized following a grid search. With a test accuracy of 92.3% against other widely-used ML-based diagnostic tools, the proposed DNN-based diagnostics tool was empirically assessed using several performance evaluation metrics. Using biodiesel in diesel engines is beneficial for reducing emissions of carbon monoxide (CO), hydrocarbons (HC) and particulate matters (PM). Biodiesel is usually produced from vegetable oils or animal fats. When produced from plant oil or woody plant sources, biodiesel can reduce a significant amount of carbon dioxide on a life cycle basis. The objective of this study is to produce biodiesel from a non-conventional woody plant source that is, crude tall oil, which is a dark brown viscous liquid extracted and processed in wood pulping plants. It contains a high percentage of fatty acids. From raw crude tall oil, tall oil fatty acids were separated and were successfully used for the production of biodiesel in this study. Although biodiesel produces lower CO, HC and PM than petroleum diesel fuel, it produces higher oxides of nitrogen (NOx) emissions in diesel engines. Water emulsifications of diesel-biodiesel blends are investigated in a direct injection (DI) diesel engine in this work to understand their potential for NOx reduction. When using 10% water in the emulsions, NOx was reduced by nearly 15%. In aldehyde emissions, B100 showed 35% lower aldehydes and B100 with 10% water emulsion produced nearly 90% lower aldehydes than diesel fuel—a substantial reduction. Therefore, this study accomplished the desired goal of producing biodiesel from a non-conventional source, which satisfies ASTM biodiesel standard and results in lower NOx and aldehydes emissions with water emulsifications of diesel-biodiesel blends in a diesel engine compared to that of diesel fuel. This research qualitatively and quantitatively characterises the minor chemical elements in diesel particulate matter (DPM). DPM was extracted from in-use diesel engine passenger vehicles of diverse types and models from major brand car producers in Europe. We analysed particulate matter extracted from the exhaust manifold part from passenger vehicles that are used in daily life environment. To qualitatively and quantitatively characterise the DPM, we employed the high-resolution Laser-Induced Breakdown Spectroscopy analytical technique (LIBS). Qualitative spectrochemical LIBS analyses confirmed the presence of minor chemical elements—silicon, nickel, titan, potassium, strontium, and molybdenum in DPM. For quantification of the LIBS signal, the calibration functions were constructed from internal calibration standards. Different concentrations of detected minor elements Si, Ni, Ti, K, Sr, and Mo were measured with respect to the type of complex particulate matter. Full article
