Increased market demand for plant herbs has prompted growers to ensure a continuous and assured supply of superior nutritional quality over the years. Apart from the nutritional value, culinary herbs contain phytochemical benefits that can improve human health. However, a significant amount of research has focused on enhancing yield, frequently overlooking the impact of production practices on the antioxidant and phytonutritional content of the produce. Thus, the study aimed to evaluate the yield, phytonutrients, and essential mineral profiling in selected aromatic herbs and their intricate role in nutritional quality when grown under different production systems. Five selected aromatic herbs (coriander, rocket, fennel, basil, and moss-curled parsley) were evaluated at harvest when grown under three production systems: in a gravel-film technique (GFT) hydroponic system and in soil, both under the 40% white shade-net structure, as well as in a soilless medium using sawdust under a non-temperature-controlled plastic tunnel (NTC). The phytonutritional quality properties (total phenolic, flavonoids, β-carotene-linoleic acid, and condensed tannins contents) as well as 1,1-diphenyl-2-picrylhydrazyl (DPPH) were assessed using spectrophotometry, while vitamin C and β-carotene were analyzed using HPLC-PDA, and leaf mineral content was evaluated using ICP-OES (Inductively Coupled Plasma Optical Emission Spectrometry). The results show that the health benefits vary greatly owing to the particular culinary herb. The fresh leaf mass (yield) of coriander, parsley, and rocket was not significantly affected by the production system, whereas basil was high in soil cultivation, followed by GFT. Fennel had a high yield in the GFT system compared to in-soil and in-soilless cultivation. The highest levels of vitamin C were found in basil leaves grown in GFT and in soil compared to the soilless medium. The amount of total phenolic and flavonoid compounds, β-carotene, β-carotene-linoleic acid, and DPPH, were considerably high in soil cultivation, except on condensed tannins compared to the GFT and soilless medium, which could be a result of Photosynthetic Active Radiation (PAR) values (683 μmol/m2/s) and not favoring the accumulation of tannins. Overall, the mineral content was greatly influenced by the production system. Leaf calcium and magnesium contents were highly accumulated in rockets grown in the soilless medium and the GFT hydroponic system. The results have highlighted that growing environmental conditions significantly impact the accumulation of health-promoting phytonutrients in aromatic herbs. Some have positive ramifications, while others have negative ramifications. As a result, growers should prioritize in-soil production systems over GFT (under the shade-net) and soilless cultivation (under NTC) to produce aromatic herbs to improve the functional benefits and customer health. Plant-parasitic nematodes are highly damaging pests responsible for heavy losses in a considerable number of plant crops. Common pest management strategies rely on the use of synthetic chemical nematicides, which have led to serious concerns regarding their impact on human health and the environment. The essential oils (EOs) obtained from aromatic plant species can provide a good source of agents for the sustainable control of nematodes, due to higher biodegradability, generally low toxicity for mammals, fish, and birds, and lower bioaccumulation in the environment. This study aimed to evaluate the nematicidal and nematostatic properties of EOs extracted from plant species relevant to Macaronesia flora or with widespread use as culinary herbs in Mediterranean cuisine. Eighteen EOs were chemically characterized and evaluated by direct contact and hatching bioassays on the root-knot nematode Meloidogyne javanica. The EOs that showed a significant effect on M. javanica second-stage juveniles’ (J2) mortality (≥40%) were also used in chemotaxis assays. From the eighteen EOs, seven showed strong nematicidal activity (>80%) and hatching inhibition. The chemotaxis assays revealed that only Mentha pulegium exhibited repellent behavior for M. javanica J2, and the rest of EOs had attractive behavior. Furthermore, EOs were assessed against the root-lesion nematode Pratylenchus penetrans and the pinewood nematode Bursaphelenchus xylophilus. Cinnamomum burmanni was the EO with the highest nematicidal activity for the three nematode species. Among the terpene-rich EOs, high mortality values and hatching inhibition for M. javanica were observed for the carvacrol chemotype Origanum vulgare, albeit with low activity for P. penetrans and B. xylophilus. Mentha pulegium, mainly composed of monoterpene ketones and monoterpenoids, demonstrated moderate-to-high mortality activity (from 30% for P. penetrans to 99% for M. javanica) for the three nematode species. Foeniculum vulgare Mill, commonly known as fennel, is an aromatic herb traditionally used for culinary and medicinal purposes, with potential therapeutic effects on neurological disorders. However, limited research has focused on its neurotrophic impact, particularly on neuronal maturation and synaptic development. This study investigates the neurotrophic effects of F. vulgare ethanol extracts (FVSE) on the maturation of rat primary hippocampal neurons. Results show that FVSE and its prominent component, anethole, significantly promote neurite outgrowth in a dose-dependent manner. Optimal axonal and dendritic growth occurred at concentrations of 40 µg/mL FVSE and 20 µM anethole, respectively, without causing cytotoxicity, underscoring the safety of FVSE for neuronal health. Additionally, FVSE enhances the formation of synapses, essential for neuronal communication. Network pharmacology analysis revealed that FVSE components influence critical neurotrophic pathways, including PI3K-AKT and Alzheimer’s disease pathways. Specifically, FVSE modulates key proteins, including tropomyosin receptor kinase (Trk), glycogen synthase kinase 3 (GSK3βser9), phosphatidylinositol 3-kinase (PI3K), and extracellular signal-regulated protein kinase (Erk1/2). Anethole was found to play a key role in regulating these pathways, which was confirmed by immunocytochemistry experiments demonstrating its effect on promoting neuronal growth and synaptic development. In conclusion, this study highlights the neurotrophic properties of FVSE, with anethole emerging as a critical bioactive compound. These findings provide valuable insights into the therapeutic potential of fennel in treating neurological disorders, offering a basis for future research into interventions promoting neuronal growth and survival. Background: Active compounds from plants and herbs are increasingly incorporated into modern medical systems to address cardiovascular diseases (CVDs). Foeniculum vulgare Mill., commonly known as fennel, is an aromatic medicinal plant and culinary herb that is popular worldwide. Methods: Protective effects against cellular damage were assessed in the H9C2 cardiomyocyte hypoxia/reoxygenation (H/R) experimental model. The identities of phytochemicals in FVSE were determined by GC-MS analysis. The phytochemical’s potential for nutrients and pharmacokinetic properties was assessed by ADMET analysis. Results: GC-MS analysis of the ethanol extracts of F. vulgare identified 41 bioactive compounds, with four prominent ones: anethole, 1-(4-methoxyphenyl)-2-propanone, ethoxydimethylphenylsilane, and para-anisaldehyde diethyl acetal. Among these, anethole stands out due to its potential for nutrients and pharmacokinetic properties assessed by ADMET analysis, such as bioavailability, lipophilicity, flexibility, and compliance with Lipinski’s Rule of Five. In the H/R injury model of H9C2 heart myoblast cells, FVSE and anethole suppressed H/R-induced reactive oxygen species (ROS) generation, DNA double-strand break damage, nuclear condensation, and the dissipation of mitochondrial membrane potential (ΔΨm). Conclusions: These findings highlight the therapeutic potential of FVSE and its prominent component, anethole, in the treatment of CVDs, particularly those associated with hypoxia-induced damage. Culinary aromatic herbs (CAHs), used worldwide for culinary and industrial purposes, are recognized for their wide range of beneficial health effects including antimicrobial, antioxidant, anti-hyperlipidemic, anti-inflammatory, anti-type 2 diabetes mellitus, antitumorigenic and anticarcinogenic, and anti-hypertensive properties, in addition to glucose- and cholesterol-lowering activities as well as properties that affect mental health and cognition via their phytochemical constituents, such as polyphenols (flavonoids and non-flavonoids), sulfur- and nitrogen-containing compounds, alkaloids, minerals, and vitamins. Moreover, the volatile organic metabolites (VOMs) found in CAHs offer unique analytical biosignatures linked to their sensory qualities and organoleptic characteristics. This study aimed to establish the volatilomic pattern of CAHs commonly used in Europe and in the Mediterranean region, oregano (Origanum vulgare L.) and two savory species: savory (Satureja hortensis L.) and lemon savory (Satureja montana L. var. citriodora). The volatilomic pattern of CAHs was established using headspace solid-phase microextraction (HS-SPME) followed by gas chromatography–mass spectrometry (GC-MS) determination. This is a powerful strategy to unravel the potential health benefits related to the most important VOMs identified in each aromatic herb. This comprehensive understanding will aid in establishing the authenticity of these herbs, while also safeguarding against possible fraudulent activities and adulterations. A total of 112 VOMs from different chemical families were identified. Terpenoids amounted to the major chemical family in the investigated aromatic herbs accounting for 96.0, 95.1, and 79.7% of the total volatile composition for savory, lemon savory, and oregano, respectively. Apart from contributing to flavor profiles, certain identified VOMs also possess bioactive properties, opening interesting avenues for potential application in the food, pharmaceutical, and cosmetic sectors. The volatilomic pattern combined with unsupervised principal component analysis facilitated the differentiation of the aromatic herbs under investigation, revealing the most related VOMs in each sample, which can be used as markers for the authentication of these valuable aromatic herbs, such as caryophyllene oxide (103), camphene (6), p-cymene (23), and borneol (74), among others. In addition, some VOMs have a high influence on the aromatic herb’s bioactive potential, helping to prevent certain diseases including cancer, inflammatory-related diseases, diabetes, and cardiovascular diseases. Petroselinum crispum (Mill.) Fuss (Apiaceae), popularly known as parsley, is a ubiquitous aromatic herb used for culinary and medicinal purposes worldwide. In addition to the richness in nutrients, several bioactive secondary metabolites, especially the flavone apigenin and its glycosides, have been described in this species. Parsley has already been shown to have several health-promoting activities, such as antithrombotic, antihypertensive, and hypolipidemic properties. The multiple studies conducted in animal models so far suggest this species is a potential source of cardioprotective agents. This review provides up-to-date information and perspectives on the potential of parsley and its bioactive compounds for the development of nutraceutical products and drugs for promoting cardiovascular health. It includes not only a discussion of parsley’s reported bioactivities but also the knowledge of supplements and food additives developed as innovative parsley-based products. Full article

Basil (Ocimum species) represents an extraordinary group of aromatic plants that have gained considerable economic importance, primarily due to their essential oils, which have applications in medicine, culinary, and perfumery. The Ocimum genus encompasses more than 60 species of herbs and shrubs originally native to tropical regions. This genus stands out for its remarkable diversity, displaying a wide spectrum of variations in phenotype, chemical composition, and genetic makeup. In addition to genetic factors, the growth, development, and essential oil production of basil are also influenced by environmental conditions, ontogeny, and various other factors. Consequently, the primary objective of this study was to explore the diversity in both the morphological characteristics and essential oil composition among basil genotypes preserved within the gene bank of the Hungarian University of Agriculture and Life Sciences’ Department of Medicinal and Aromatic Plants. The investigation involved the assessment of fifteen basil genotypes, representing four distinct species: Ocimum basilicum (including ‘Anise’, ‘Clove’, ‘Fino Verde’, ‘Licorice’, ‘Mammoth’, ‘Mrs. Burns’, ‘Thai tömzsi’, ‘Thai hosszú’, and ‘Vietnamese basil’), Ocimum sanctum (green holy basils), Ocimum citrodora (Lemon basil), and Ocimum gratissimum (African and Vana holy basil). The genotypes exhibited significant variations in their morphological growth, essential oil content (EOC), and composition. African basil produced more biomass (408.3 g/plant) and showed robust growth. The sweet basil cultivars clove, licorice, Thai tömzsi, and Thai hosszú also exhibited similar robust growth trends. Vietnamese basil, on the other hand, displayed the lowest fresh biomass of 82.0 g per plant. Both holy basils showed EOC levels below 0.5%, while Mrihani basil stood out with the highest EOC of 1.7%. The predominant constituents of the essential oil among these genotypes comprised estragole, thymol, methyl cinnamate, linalool, and eugenol. In conclusion, this study showed that the genotypes of basil stored in the department’s gene bank exhibit a wide range of variability, both within and between species. Full article

Garlic (Allium sativum L.) is an aromatic herb known for its culinary and medicinal uses for centuries. Both unprocessed (white) and processed (black) garlic are known to protect against the pathobiology of neurological disorders such as Alzheimer’s disease (AD) and Parkinson’s disease (PD), which has been attributed to their anti-inflammatory and antioxidant properties. The information on the effects of processed and unprocessed garlic on neuronal process outgrowth, maturation, and synaptic development is limited. This study aimed at investigating and comparing the effects of the ethanol extracts of unprocessed (white garlic extract, WGE) and processed (black garlic extract, BGE) garlic on the maturation of primary hippocampal neurons. Neurite outgrowth was stimulated in a dose-dependent manner by both WGE and BGE and the most effective doses were 15 μg/mL and 60 μg/mL, respectively, without showing cytotoxicity. At this optimal concentration, both extracts promoted axonal and dendritic growth and maturation. Furthermore, both extracts substantially increased the formation of functional synapses. However, the effect of WGE was more robust at every developmental stage of neurons. In addition, the gas chromatography and mass spectrometry (GC-MS) analysis revealed a chemical profile of various bioactives in both BGE and WGE. Linalool, a compound that was found in both extracts, has shown neurite outgrowth-promoting activity in neuronal cultures, suggesting that the neurotrophic activity of garlic extracts is attributed, at least in part, to this compound. By using network pharmacology, linalool’s role in neuronal development can also be observed through its modulatory effect on the signaling molecules of neurotrophic signaling pathways such as glycogen synthase kinase 3 (GSK3β), extracellular signal-regulated protein kinase (Erk1/2), which was further verified by immunocytochemistry. Overall, these findings provide information on the molecular mechanism of processed and unprocessed garlic for neuronal growth, survival, and memory function which may have the potential for the prevention of several neurological disorders. Anethum graveolens L. has been known as an aromatic, medicinal, and culinary herb since ancient times. The main purpose of this study was to determine the chemical composition, antibacterial, antibiofilm, and anti-quorum sensing activities of the essential oil (EO) obtained by hydro-distillation of the aerial parts. Twelve components were identified, representing 92.55% of the analyzed essential oil. Limonene (48.05%), carvone (37.94%), cis-dihydrocarvone (3.5%), and trans-carvone (1.07%) were the main identified constituents. Results showed that the obtained EO was effective against eight bacterial strains at different degrees. Concerning the antibiofilm activity, limonene was more effective against biofilm formation than the essential oil when tested using sub-inhibitory concentrations. The results of anti-swarming activity tested against P. aeruginosa PAO1 revealed that A. graveolens induced more potent inhibitory effects in the swarming behavior of the PAO1 strain when compared to limonene, with a percentage reaching 33.33% at a concentration of 100 µg/mL. The ADME profiling of the identified phytocompounds confirms their important pharmacokinetic and drug-like properties. The in-silico study using molecular docking approaches reveals a high binding score between the identified compounds and known target enzymes involved in antibacterial and anti-quorum sensing (QS) activities. Overall, the obtained results highlight the possible use of A. graveolens EO to prevent food contamination with foodborne pathogenic bacteria. Full article

Climate change is causing a decline in the availability of crucial resources such as water and fertile soil, leading to a detrimental impact on crop yields. Basil (Ocimum basilicum L.), an annual aromatic plant used worldwide for culinary, cosmetic, and medicinal purposes, is especially at risk due to its high water demand and specific growing requirements typical of the Mediterranean climate. In Italy, basil is an essential part of any herb garden and is also commonly found in households as a potted plant. Nowadays, the conventional practice of growing basil in pots with peat as the primary medium is not environmentally sustainable, which underscores the need for alternative and sustainable cultivation techniques to ensure the continued growth of this majestic king of herbs. A greenhouse factorial experiment was conducted to study the impact of peat replacement and reduced water supply on the growth and biochemical traits of basil plants. The study included two cultivars (‘Genovese’ and ‘Valentino’), three substrate mixes with phytoremediated dredged sediment (TS) at varying volume percentages (0%, 12.5%, and 25%), and three levels of water irrigation (normal, reduced by 25%, and reduced by 37.8%). Increasing the TS percentage resulted in higher germination rates and greater biomass production in both cultivars compared with the control. ‘Valentino’ potted plants produced a higher yield than those of ‘Genovese’, while the latter had higher pigment contents due to its genetic characteristics. All combinations of substrates mixed with the highest water amounts of irrigation positively impacted seed germination and biomass-based outputs, while control peat with reduced water availability led to lower plant yields and germination capacity. Substrates with TS had suitable physicochemical characteristics for plant development. Our findings suggest that using peat–TS mixtures can produce quality results comparable to or even better than conventional soilless culture using only peat. Thus, replacing peat with moderate percentages of TS can be an effective and sustainable practice for recycling phytoremediated sediments. Full article

Rosmarinus officinalis L. (rosemary) is an aromatic culinary herb. Native to the Mediterranean region, it is currently cultivated worldwide. In addition to its use as a condiment in food preparation and in teas, rosemary has been widely employed in folk medicine and cosmetics. Several beneficial effects have been described for rosemary, including antimicrobial and antioxidant activities. Here, we investigated the mechanisms accounting for the antioxidant activity of the glycolic extract of R. officinalis (Ro) in isolated rat liver mitochondria (RLM) under oxidative stress conditions. We also investigated its protective effect against acetaminophen-induced hepatotoxicity in vivo. A crude extract was obtained by fractionated percolation, using propylene glycol as a solvent due to its polarity and cosmeceutical compatibility. The quantification of substances with recognized antioxidant action revealed the presence of phenols and flavonoids. Dereplication studies carried out through LC-MS/MS and GC-MS, supported by The Global Natural Product Social Molecular Networking (GNPS) platform, annotated several phenolic compounds, confirming the previous observation. In accordance, Ro decreased the production of reactive oxygen species (ROS) elicited by Fe2+ or t-BOOH and inhibited the lipid peroxidation of mitochondrial membranes in a concentration-dependent manner in RLM. Such an effect was also observed in liposomes as membrane models. Ro also prevented the oxidation of mitochondrial protein thiol groups and reduced glutathione (GSH). In model systems, Ro exhibited a potent scavenger activity toward 2,2′-diphenyl-1-picrylhydrazyl (DPPH) radicals and superoxide anions. It also demonstrated an Fe2+ chelating activity. Moreover, Ro did not exhibit cytotoxicity or dissipate the mitochondrial membrane potential (∆Ψ) in rat liver fibroblasts (BRL3A cells). To evaluate whether such antioxidant protective activity observed in vitro could also be achieved in vivo, a well-established model of hepatotoxicity induced by acute exposure to acetaminophen (AAP) was used. This model depletes GSH and promotes oxidative-stress-mediated tissue damage. The treatment of rats with 0.05% Ro, administered intraperitoneally for four days, resulted in inhibition of AAP-induced lipid peroxidation of the liver and the prevention of hepatotoxicity, maintaining alanine and aspartate aminotransferase (ALT/AST) levels equal to those of the normal, non-treated rats. Together, these findings highlight the potent antioxidant activity of rosemary, which is able to protect mitochondria from oxidative damage in vitro, and effects such as the antioxidant and hepatoprotective effects observed in vivo. Culinary herbs are commercially cultivated for their wide range of volatile compounds that give characteristic aromas and tastes. Rosemary (Salvia rosmarinus Spenn.) is an excellent model for assessment of methods improvement of volatile production as cultivars offer a wide variety of aromatic profiles due to the large family of terpene synthase genes. Arbuscular mycorrhizal fungi (AMF) associations have been shown to improve essential oil production in aromatic plants and offer one approach to enhance aroma in commercial herb production. Changes in the expression of seven different terpene synthases were compared in six rosemary cultivars in response to addition of AMF to a peat substrate. Addition of AMF profoundly influenced terpene synthase expression in all cultivars and did so without impacting the optimised plant size and uniformity achieved in these conditions. In addition, two methods for AMF application, developed with the horticultural industry in mind, were tested in this study. Uniform incorporation of AMF mixed into the growing substrate prior to planting of a root plug produced the most consistent root colonisation. Overall, our findings demonstrate the potential for the use of AMF in the improvement of aroma in culinary herbs within a commercial setting but show that outcomes are likely to greatly vary depending on variety. Flavoured oils arouse great interest among consumers in many countries due to their variety of flavours and versatility, especially in the culinary field. The aromatization of oils seeks to improve their sensory and nutritional properties, and extend their useful life due to the added substances can be beneficial as antioxidant and antimicrobial agent. In this research, olive oils of the ‘Madural’ variety from Trasos Montes region of Portugal have been obtained and flavoured with different aromatics herbs and condiments (flower of salt and bay leaf, garlic, rosemary and dehydrated lemon peel). The objective is to study the influence of the aromatic herbs and condiments on the physicochemical parameters of the oils: quality, purity, oxidative stability and microbiological analysis. It can be noted that the flavourings do not significantly alter the quality of the monovarietal oil, although, for some parameters, the excessive contact times can affect the category of the oil. On the other hand, the high content of antioxidants provided by flavouring agents can favour its stability and prolong its expiration. In this sense, the flavouring agent that contributes to stop the oxidation of the oil over time is salt + bay leaves, as higher oxidative stability values were detected than those obtained in monovarietal oil. However, oils flavoured with rosemary or lemon show a decrease over time for this parameter, which could indicate that this flavouring accelerates oxidation. In the case of oxidative stability referred to those flavoured with garlic, they present similar values to the monovarietal. The effect exerted by flavourings on the different parameters of the oils is complex, since it is influenced by the method followed and the operating variables established for flavouring. Basil (Ocimum basilicum L.) is one of the most common aromatic herbs, a rich source of bioactive compounds, and is used extensively to add aroma and flavor to food. The leaves, both in fresh and dried form, are used as a culinary ingredient in different cultures. O. basilicum is also famous for its therapeutic potential and preservation effects. The present study investigated the cytotoxicity of basil at three different growth stages (GS), i.e., GS-1 (58 days of growth), GS-2 (69 days of growth), and GS-3 (93 days of growth) using the brine shrimp lethality assay. The results revealed that cytotoxicity was influenced by GS and the concentration of extracts. Aqueous extracts of basil at a concentration of 10 to 1000 µg/mL did not show notable toxicity. The lowest mortality rate, i.e., 8.9%, was recorded for GS-2 at the highest tested dose of basil extracts. The mortality rate at GS-1, GS-2, and GS-3 was found to be 26.7 ± 3.34%, 8.91 ± 0.10%, and 16.7 ± 0.34%, respectively, at 1000 µg/mL. GS-2 basil powder with the lowest toxicological risk was extracted with different solvents, viz., n-hexane, dichloromethane, ethanol, and water. The highest concentration of plant secondary metabolites including total phenolic acid, flavonoids, and tannin content was observed in ethanol extracts. Ethanol extracts also exhibited the highest antioxidant activity in DPPH, FRAP and H2O2 assays. LC-ESI-MS/MS analysis presented ethanol extracts of basil as a promising source of known health-promoting and therapeutic compounds such as rosmarinic acid, ellagic acid, catechin, liquiritigenin, and umbelliferone. The results suggest basil, a culinary ingredient, as a potential source of bioactive compounds which may offer an array of health promoting and therapeutic properties. Full article

In this work, Hexagonal Mesoporous Silica (HMS) and Santa Barbara Amorphous-15 (SBA-15) mesostructured silicas were synthesized and functionalized with sulfonic acid groups. The materials (HMS-SO3 and SBA-15-SO3) were evaluated as strong cation exchange sorbents for sample extract clean-up, by solid phase extraction (SPE) and dispersive solid phase extraction, to determine atropine (At) and scopolamine (Sc) in commercial culinary aromatic herbs. Under optimized conditions, 0.25 g of sample was subject to solid–liquid extraction with acidified water (pH 1.0), and good recovery percentages were achieved for At and Sc using 75 mg of HMS-SO3 in SPE as the clean-up stage, prior to their determination by HPLC-MS/MS. The proposed method was validated in a thyme sample showing recoveries in the range of 70–92%, good linearity (R2 > 0.999), adequate precision (RSD ≤ 14%) and low limits (MDL 0.8–2.2 µg/kg and MQL 2.6–7.2 µg/kg for both analytes). Sixteen aromatic herbs samples (dried thyme, basil and coriander leaves) were analysed and At was found in fourteen samples over an interval of <5–42 μg/kg, whereas Sc was found in three of the sixteen samples studied (between <5–34 μg/kg). The amount of At and Sc found in some analysed samples confirms the importance of setting maximum levels of At and Sc in culinary aromatic herbs. Full article
