This article provides a critical review of the current design and research challenges in contemporary timber architecture. Conducted from the perspective of a designer-researcher, the review focuses on the role of wood as a material at the intersection of environmental performance, cultural meaning, and spatial practice. The study adopts a conceptual, problem-oriented approach, eschewing the conventional systematic aggregation of existing data. The objective of this study is to identify, interpret and categorise the key issues that are shaping the evolving discourse on timber architecture. The analysis is based on peer-reviewed literature published between 2020 and 2025, sourced from the Scopus and Web of Science Core Collection databases. Fifteen thematic challenges have been identified and classified according to their recognition level in academic and design contexts. The subjects under discussion include well-established topics, such as life cycle assessment and carbon storage, as well as less commonly explored areas, such as symbolic durability, social acceptance, traceability, and the upcycling of low-grade wood. The review under consideration places significant emphasis on the importance of integrating technical, cultural, and perceptual dimensions when evaluating timber architecture. The article proposes an interpretive framework combining design thinking and transdisciplinary insights. This framework aims to bridge disciplinary gaps and provide a coherent structure for understanding the complexity of timber-related challenges. The framework under discussion here encourages a broader understanding of wood as not only a sustainable building material but also a vehicle for systemic transformation in architectural culture and practice. The study’s insights may support designers, educators, and policymakers in identifying strategic priorities for the development of future-proof timber-based design practices. Full article

With the advancement of low-carbon and sustainable development initiatives, electric scooters, recognized as essential transportation tools and leisure products, have gained significant popularity, particularly among young people. However, the current electric scooter market is plagued by severe product similarity. Once the initial novelty fades for users, the usage frequency declines, resulting in considerable resource wastage. This research collected user needs via surveys and employed the KJ method (affinity diagram) to synthesize fragmented insights into cohesive thematic clusters. Subsequently, a hierarchical needs model for electric scooters was constructed using analytical hierarchy process (AHP) principles, enabling systematic prioritization of user requirements through multi-criteria evaluation. By establishing a house of quality (HoQ), user needs were transformed into technical characteristics of electric scooter products, and the corresponding weights were calculated. After analyzing the positive and negative correlation degrees of the technical characteristic indicators, it was found that there are technical contradictions between functional zoning and compact size, lightweight design and material structure, and smart interaction and usability. Then, based on the theory of inventive problem solving (TRIZ), the contradictions were classified, and corresponding problem-solving principles were identified to achieve a multi-functional innovative design for electric scooters. This research, leveraging a systematic industrial design analysis framework, identified critical pain points among electric scooter users, established hierarchical user needs through priority ranking, and improved product lifecycle sustainability. It offers novel methodologies and perspectives for advancing theoretical research and design practices in the electric scooter domain. Full article

Objective: To analyze the composition of the oral microbiome in periodontal pocket lesions and on the tongue dorsum of patients with Helicobacter pylori-associated gastric disease. Materials and Methods: Patients diagnosed with gastric disease and H. pylori (HP+) were evaluated in comparison to a control group of H. pylori-negative patients without gastric disease (HP−). Periodontal and oral health clinical parameters (PPD, BoP, PSE, plaque score and modified DMFT) were assessed for each patient. Microbiological samples were collected from the deepest periodontal pockets and tongue dorsum, followed by DNA extraction, 16S rRNA PCR amplification, and Next-Generation-Sequencing (NGS) analyses. Results: Sixty-seven patients (27F; 40M, aged 35–85 years) were enrolled. Of these, 52 were HP+ and 15 were HP−. HP+ patients exhibited a significantly higher presence of decayed teeth (p < 0.05) and slightly fewer missing teeth (p > 0.05). The plaque score was significantly higher in HP+ patients (p < 0.05), while PPD and BoP showed no significant differences (p > 0.05). NGS analysis revealed no presence of H. pylori in any samples of both periodontal and tongue sites. HP+ patients showed a distinct microbial composition, including higher prevalence of Capnocytophaga, Fusobacterium, and Peptostreptococcus genera in both locations (pockets and tongue dorsum). Conclusions: The study demonstrated that HP+ patients exhibit distinct oral microbial profiles compared to HP− patients, especially in areas with deeper periodontal pockets. H. pylori was not detected in the oral microbiomes of either group. Full article

The artichoke (Cynara cardunculus L. subsp. scolymus) is an endemic perennial plant of the Mediterranean area commonly consumed as food. It is known since ancient times for its beneficial properties for human health, among which its antioxidant activity due to polyphenolics stands out. In the frame of our ongoing studies aiming to highlight the biodiversity and the chemodiversity of natural resources, we investigated the phenolic and saponin content of the cultivar “Carciofo di Procida” collected at Procida, an island of the Gulf of Naples (Italy). Along with the edible part of the immature flower, we included in our analyses the stem and the external bracts, generally discarded for food consuming or industrial preparations. The LCMS quali-quantitative profiling of polyphenols (including anthocyanins) and cynarasaponins of this cultivar is reported for the first time. In addition to antioxidant properties, we observed a significant cytotoxic activity due to extracts from external bracts against human neuroblastoma SH-SY5Y cell lines with 43% of cell viability, after 24 h from the treatment (50 μg/mL), and less potent but appreciable effects also against human colorectal adenocarcinoma CaCo-2 cells. This suggests that the different metabolite composition may be responsible for the bioactivity of extracts obtained from specific parts of artichoke and foresees a possible exploitation of the discarded material as a source of beneficial compounds. The Fernald Feed Materials Production Center (FMPC), located in Fernald, Ohio, USA, released radon (Rn) as a byproduct of the processing of uranium materials during the years from 1951 to 1989. Rn is a colorless, odorless gas that emits charged alpha radiation that interacts with cells in the lung and trachea-bronchial tree, leading to DNA damage, mutations, and tumor initiation. The purpose of this project was to use evidence collected by the Fernald Dosimetry Reconstruction Project and other sources to estimate the outdoor Rn exposure to individuals in the community immediately surrounding the FMPC during the years of plant operation. Using previously tabulated source terms, diffusion and meteorological data, and self-reported detailed residential histories, we estimated radon exposure for approximately 9300 persons who lived at more than 14,000 addresses. The results indicated that a portion of the population cohort experiences mean annual Rn exposure exceeding the U.S. Environmental Protection Agency (EPA) action limit of 4 pCiL−1. These exposure estimates support the analysis of the incidence of lung cancer in the Fernald Community Cohort (FCC). Full article

An important task for the European Union is to transpose agreements and international standards in regulation and directives that are binding on member states. The resultant European action plans and directives identify priority areas in the building and energy sectors where circular economy principles can be applied. Italy records a general circular materials rate of 20.8%, surpassing the mean European value. But low recycling rates are still registered in the construction sector. This paper aims to assess the position of Italy with respect to the European regulatory framework on circularity in the energy transition of buildings. Firstly, the government’s initiatives and technical standards are introduced and commented upon. Secondly, the study illustrates the current Italian platforms, networks, and public and private initiatives highlighting opportunities and obstacles that the energy sector has to overcome in the area of circularity. It emerges that Italian policies still use voluntary tools that are not sufficiently in line with an effective circular economy model. Moreover, data collection plays a crucial role in accelerating the implementation of future actions. Italy should consider the foundation of a National Observatory for the Circular Economy to elaborate European directives, harmonize regional policies, and promote the implementation of effective practices. This paper aims to explore the potentialities and systemic relationships between the ‘regenerative’ process and ‘circular economy’ concept within the conservation and reuse of a built cultural heritage framework through contextualizing the concept of ‘process programming’ of the Preventive and Planned Conservation methodology. As a case study, it depicts a decommissioned industrial agricultural silo in Corbetta—a small historic city with its hinterland located in the protected Southern Milan Regional Agricultural Park. The context includes the industrial agricultural lands of the 20th century, together with historical water infrastructure, farmhouses, and the typical flora of the Lombardy region, all evidences of Corbetta’s rural archaeological values and the sophisticated material culture of its past collective production/economy system—the locus in which the silo was once one of the main productive symbols of Corbetta’s agricultural identity. Within such a complex and challenging context, this paper argues in favor of the constructive role of such a methodology in upholding circular economy principles within the process of conservation and reuse of the silo, highlighting its broader application of the ‘coevolution’ concept from a multidisciplinary long-term perspective. Background: this research presents an integrated approach to enhancing the sustainability of plastic bag supply chains in Indonesia by addressing critical issues related to ineffective post-consumer waste management and low recycling rates. The objective of this study is to develop a combined framework of material flow analysis (MFA) and sustainable supply chain planning to improve demand forecasting and inflow management across the plastic bag lifecycle. Method: the research adopts a quantitative method using the XGBoost algorithm for forecasting and is supported by a polymer-based MFA framework that maps material flows from production to end-of-life stages. Result: the findings indicate that while production processes achieve high efficiency with a yield of 89%, more than 60% of plastic bag waste remains unmanaged after use. Moreover, scenario analysis demonstrates that single interventions are insufficient to achieve circularity targets, whereas integrated strategies (e.g., reducing export volumes, enhancing waste collection, and improving recycling performance) are more effective in increasing recycling rates beyond 35%. Additionally, the study reveals that increasing domestic recycling capacity and minimizing dependency on exports can significantly reduce environmental leakage and strengthen local waste management systems. Conclusions: the study’s novelty lies in demonstrating how machine learning and material flow data can be synergized to inform circular supply chain decisions and regulatory planning. Full article

Prothrombotic Genetic Mutations Are Associated with Sub-Clinical Placental Vascular Lesions: A Histopathological and Morphometric Study

Background: Inherited thrombophilia is increasingly recognized as a contributing factor to placental vascular pathology and adverse pregnancy outcomes. While the clinical implications are well-established, fewer studies have systematically explored the histopathological changes associated with specific genetic mutations in thrombophilic pregnancies. Materials and Methods: This retrospective observational study included two cohorts of placental samples collected between September 2020 and September 2024 at a tertiary maternity hospital. Group 1 included women diagnosed with hereditary thrombophilia, and Group 2 served as controls without known maternal pathology. Placentas were examined macroscopically and histologically, with pathologists blinded to group allocation. Histological lesions were classified according to the Amsterdam Consensus and quantified using a composite score (0–5) based on five key vascular features. Results: Placental lesions associated with maternal vascular malperfusion—including infarctions, intervillous thrombosis, stromal fibrosis, villous stasis, and acute atherosis—were significantly more frequent in the thrombophilia group (p < 0.05 for most lesions). A combination of well-established thrombophilic mutations (Factor V Leiden, Prothrombin G20210A) and other genetic polymorphisms with uncertain clinical relevance (MTHFR C677T, PAI-1 4G/4G) showed moderate-to-strong correlations with histopathological markers of placental vascular injury. A composite histological score ≥3 was significantly associated with thrombophilia (p < 0.001). Umbilical cord abnormalities, particularly altered coiling and hypertwisting, were also more prevalent in thrombophilic cases. Conclusions: Thrombophilia is associated with distinct and quantifiable placental vascular lesions, even in pregnancies without overt clinical complications. The use of a histological scoring system may aid in the retrospective identification of thrombophilia-related placental pathology and support the integration of genetic and histologic data in perinatal risk assessment. In vitro propagation of avocado faces several limitations. To optimize the establishment phase, we evaluated three plant material types: etiolated shoots, 30-day covered field shoots, and uncovered field shoots, collected at two time points. Biochemical and anatomical analyses were conducted to understand material performance during establishment. Across both collection times, etiolated shoots exhibited minimal oxidation, enhanced bud sprouting, reduced malondialdehyde (MDA) and reactive oxygen species (ROS) levels, increased peroxidase (POD) activity, and improved xylem development, consistently outperforming field-derived materials. Using etiolated shoots, we optimized disinfection and in vitro multiplication protocols. Pre-disinfection with 3 mL L−1 Phyton 27® and 2% sodium hypochlorite yielded the highest survival rates. In multiplication experiments, varying concentrations of 6-benzylaminopurine (BAP) and meta-topolin (MT), supplemented with gibberellic acid (GA3), did not significantly affect growth variation. However, 8.88 µM BAP with 0.29 µM GA3 resulted in the greatest number of sprouted buds. Full article

Forests are considered one of the most valuable natural areas in metropolitan region landscapes. Considering the sensitivity and ecosystem services provided by trees, the definition of urban forest ecosystems is nowadays based on a comprehensive understanding of the entire urban ecosystem. The effective capturing of particulate matter is one of the ecosystem services provided by urban forests. These ecosystems function as efficient biological filters. Plants accumulate pollutants passively via their leaves. Therefore, another ecosystem service provided by city forests could be the use of tree organs as bioindicators of pollution. This paper aims to estimate differences in trace metal pollution between the wooded urban areas of Vienna and Krakow using leaves of evergreen and deciduous trees as biomonitors. An additional objective of the research was to assess the ability of the applied tree species to act as biomonitors. Plant samples of five species—Norway spruce, Scots pine, European larch, common white birch, and common beech—were collected within both areas, in seven locations: four in the “Wienerwald” Vienna forest (Austria) and three in the “Las Wolski” forest in Krakow (Poland). Concentrations of Cr, Cu, Cd, Pb, and Zn in plant material were determined. Biomonitoring studies with deciduous and coniferous tree leaves showed statistically higher heavy metal contamination in the “Las Wolski” forest compared to the “Wienerwald” forest. Based on the conducted analyses and the literature study, it can be concluded that among the analyzed tree species, only two: European beech and common white birch can be considered potential indicators in environmental studies. These species appear to be suitable bioindicators, as both are widespread in urban woodlands of Central Europe and have shown the highest accumulation levels of trace metals. Full article

Materials and Methods: A prospective observational study was conducted on 76 patients undergoing lower limb reconstruction with fascio-cutaneous perforator flaps between 2022 and 2024. Perforator mapping was performed concurrently with IRT and Doppler ultrasonography (D-US), with intraoperative confirmation. Flap design variables and systemic parameters were recorded. Postoperative monitoring employed thermal imaging on days 1 and 7. Outcomes were correlated with thermal, anatomical, and systemic factors using statistical analyses, including t-tests and Pearson correlation. Results: IRT showed high sensitivity (97.4%) and positive predictive value (96.8%) for perforator detection. A total of nine minor complications occurred, predominantly in patients with diabetes mellitus and/or elevated glycemia (p = 0.05). Larger flap-to-defect ratios (A/C and B/C) correlated with increased complications in propeller flaps, while smaller ratios posed risks for V-Y and Keystone flaps. Thermal analysis indicated significantly lower flap temperatures and greater temperature gradients in flaps with complications by postoperative day 7 (p < 0.05). CRP levels correlated with glycemia and white blood cell counts, highlighting systemic inflammation’s impact on outcomes. Conclusions: IRT proves to be a reliable, non-invasive method for perforator localization and flap monitoring, enhancing surgical planning and early complication detection. Combined with D-US, it improves perforator selection and perfusion assessment. Thermographic parameters, systemic factors, and flap design metrics collectively predict flap viability. Integration of IRT into surgical workflows offers a cost-effective tool for optimizing reconstructive outcomes in lower limb surgery. Full article

Reverse-transcription real-time polymerase chain reaction (RRT-PCR) is the fastest, and most sensitive assay for detecting CSF virus (CSFV) genomic material. Previously, we demonstrated that spleen swabs outperformed spleen homogenates for the detection of ASFV genomic material by RRT-PCR. In this study, we compared CSFV genome detection in paired spleen homogenates and spleen swabs generated using 49 frozen and 33 fresh spleen samples collected from experimentally inoculated pigs with acute infection. The results show that the CSFV genome detection in spleen swabs is comparable to that in spleen homogenates. The study also demonstrated that the CSFV genomic material can be detected in spleen swabs during early CSFV infections, and the viruses can be successfully isolated from the swabs. The use of spleen swabs instead of spleen tissue homogenates for CSF detection will reduce labor, decrease costs associated with reporting, and increase the diagnostic throughput. Full article

Background and Objectives: Multiple sclerosis (MS) is a chronic immune-mediated neurological disorder that primarily affects young adults and is frequently accompanied by psychiatric comorbidities such as depression and anxiety, both of which significantly diminish patients’ quality of life (QoL). This study investigated the effect of two oral disease-modifying therapies (DMTs), fingolimod and cladribine, on mental health and QoL in patients with relapsing-remitting MS (RRMS). The aim of the study was to compare levels of depression, anxiety, and health-related quality of life (HRQoL) in RRMS patients treated with fingolimod or cladribine, and to evaluate their associations with clinical and radiological parameters. Materials and Methods: Eighty RRMS patients aged 18 to 50 years with Expanded Disability Status Scale (EDSS) scores of 3.0 or less, no recent disease relapse, and no history of antidepressant use were enrolled. Forty patients were treated with fingolimod and forty with cladribine. Depression and anxiety were assessed using the Hamilton Depression Rating Scale (HDRS) and the Hamilton Anxiety Rating Scale (HARS). QoL was evaluated using the Multiple Sclerosis QoL-54 (MSQOL-54) instrument. Additional clinical data, including MRI-based lesion burden, EDSS scores, age, disease duration, and occupational status, were collected. Results: No statistically significant differences were observed between the two groups regarding HDRS and HARS scores (p > 0.05). However, patients treated with fingolimod had significantly higher scores in the Energy/Fatigue subdomain (7.55 ± 2.02 vs. 6.56 ± 2.57, p = 0.046) and Composite Mental Health (CMH) score (64.73 ± 15.01 vs. 56.00 ± 18.93, p = 0.029) compared to those treated with cladribine. No significant differences were found in the independent items of the MSQOL-54. A negative correlation was identified between total lesion load and QoL scores. Conclusions: Although fingolimod and cladribine exert comparable effects on depression and anxiety levels, fingolimod may be associated with better mental health outcomes and reduced fatigue in RRMS patients. Furthermore, lesion burden and clinical parameters such as age and EDSS score may independently influence QoL, regardless of the DMT used. Full article

Recent sampling efforts in northern Algeria and the investigation of some collection material has revealed the presence of taxa with conchological and genital morphological similarities to the Candidula-like group. A detailed investigation of the anatomy of the genitalia combined with the analysis of two mitochondrial and two nuclear markers of representatives of the known genera as well as two Algerian taxa has been conducted. The result confirms the affiliation of the latter in the Candidula-like complex. Comparative anatomical analysis led to the description of a new genus, Kherattolactea gen. nov., endemic to northern Algeria with Xerophila keratae, Kobelt, 1892 as the type species. The anatomical distinction corroborates the result of the molecular analysis that reveals apomorphic nucleotide substitution. Moreover, the result of the combined analysis provides the first confirmed record of the genus Orexana Chueca, Gómez-Moliner, Madeira & Pfenninger, 2018, in north Africa, represented by the species Orexana lemoinei Kobelt, 1882 nov. comb. A new Xerogyra species, Xerogyra halgassi nov. sp., from the Apennine Mountain has been described based on the combination of morphoanatomical and molecular traits. For the first time, the morphoanatomical description of the known Xerogyra species has been presented. Full article
