Exosomes are membranous vesicles that play a crucial role as intercellular messengers. Cells secrete exosomes, which can be found in a variety of bodily fluids such as amniotic fluid, semen, breast milk, tears, saliva, urine, blood, bile, ascites, and cerebrospinal fluid. Exosomes have a distinct bilipid protein structure and can be as small as 30–150 nm in diameter. They may transport and exchange multiple cellular messenger cargoes across cells and are used as a non-invasive biomarker for various illnesses. Due to their unique features, exosomes are recognized as the most effective biomarkers for cancer and other disease detection. We give a review of the most current applications of exosomes derived from various sources in the prognosis and diagnosis of multiple diseases. This review also briefly examines the significance of exosomes and their applications in biomedical research, including the use of aptamers and antibody–antigen functionalized biosensors. Various factors influence the formation and adjustment of network freight prices, including transportation costs, cargo characteristics, and policies and regulations. The interaction of these factors increases the difficulty of accurately predicting network freight prices through regressions or other machine learning models, especially when the amount and quality of training data are limited. This paper introduces large language models (LLMs) to predict network freight prices using their inherent prior knowledge. Different data sorting methods and serialization strategies are employed to construct the corpora of LLMs, which are then tested on multiple base models. A few-shot sample dataset is constructed to test the performance of models under insufficient information. The Chain of Thought (CoT) is employed to construct a corpus that demonstrates the reasoning process in freight price prediction. Cross entropy loss with LoRA fine-tuning and cosine annealing learning rate adjustment, and Mean Absolute Error (MAE) loss with full fine-tuning and OneCycle learning rate adjustment to train the models, respectively, are used. The experimental results demonstrate that LLMs are better than or competitive with the best comparison model. Tests on a few-shot dataset demonstrate that LLMs outperform most comparison models in performance. This method provides a new reference for predicting network freight prices. Full article

With the rise of carbon capture and storage, liquefied carbon dioxide (LCO2) has emerged as a promising medium for large-scale marine transport. This study evaluates the structural integrity of an IMO Type C cargo tank for a medium-range LCO2 carrier under four conditions: ultimate limit state, accidental limit state, hydrostatic pressure test, and fatigue limit state, based on IGC Code and classification rules. Seventeen load cases were analyzed using finite element methods with multi-step loading to ensure stability. The highest stress occurred at the pump dome–shell junction due to geometric discontinuities, but all stress and buckling criteria were satisfied. The fatigue damage from wave-induced loads was negligible, with low-cycle fatigue from loading/unloading operations governing the fatigue life, which exceeded 31,000 years. The findings confirm the tank’s structural robustness and its suitability for safe, efficient medium-pressure LCO2 transport. Full article

RNA-seq analysis of the highly differentiated human retinal pigment epithelial (RPE) cell-line ARPE-19, cultured on transwells for ≥4 months, yielded 44,909 genes showing 83.35% alignment with the human reference genome. These included mRNA transcripts of RPE-specific genes and those involved in retinopathies. Monolayers were fed photoreceptor outer segments (POS), designed to be synchronously internalised, mimicking homeostatic RPE activity. Cells were subsequently fixed at 4, 6, 24 and 48 h when POS were previously shown to maximally co-localise with Rab5, Rab7, LAMP/lysosomes and LC3b/autophagic compartments. A comprehensive analysis of differentially expressed genes involved in proteolysis revealed a pattern of gene orchestration consistent with POS breakdown in the autophagy-lysosomal pathway. At 4 h, these included elevated upstream signalling events promoting early stages of cargo transport and endosome maturation compared to RPE without POS exposure. This transcriptional landscape altered from 6 h, transitioning to promoting cargo degradation in autolysosomes by 24–48 h. Longitudinal scrutiny of mRNA transcripts revealed nuanced differences even within linked gene networks. POS exposure also initiated transcriptional upregulation in ubiquitin proteasome and chaperone-mediated systems within 4–6 h, providing evidence of cross-talk with other proteolytic processes. These findings show detailed evidence of transcriptome-level responses to cargo trafficking and processing in RPE cells. Full article

As an energy-intensive industry, the aviation sector’s carbon emissions have drawn significant attention. Against the backdrop of the “dual carbon” goals, how to enhance the carbon emission efficiency of airlines has become an urgent issue to be addressed for both industry development and low-carbon targets. This paper constructs an evaluation system for the carbon emission efficiency of airlines and uses the SBM-DDF model under the global production possibility set, combined with the bootstrap-DEA method, to calculate the efficiency values. On this basis, the fuzzy-set qualitative comparative analysis method is employed to analyze the synergistic effects of multiple influencing factors in three dimensions: economic benefits, transportation benefits, and energy consumption on improving carbon emission efficiency. The research findings reveal that, first, a single influencing factor does not constitute a necessary condition for achieving high carbon emission efficiency; second, there are four combinations that enhance carbon emission efficiency: “load volume-driven type”, “scale revenue-driven type”, “high ticket price + technology-driven type”, and “passenger and cargo synergy mixed type”. These discoveries are of great significance for promoting the construction of a carbon emission efficiency system by Chinese airlines and achieving high-quality development in the aviation industry. Waterborne transportation serves as a critical pillar of trunk-line freight systems, offering unparalleled advantages in transport capacity, energy efficiency, and cost-effectiveness. As cargo throughput demands escalate, optimizing lock capacity becomes imperative. This study investigates ship sinkage dynamics through computational fluid dynamics (CFD) simulations for a representative inland cargo vessel navigating the Three Gorges on the Yangtze River. We develop a predictive sinkage model that integrates four key hydrodynamic parameters: ship velocity, draft, water depth, and bank clearance, applicable to both open shallow water and lockage conditions. The model enables determination of maximum safe drafts for lock transit by analyzing upstream/downstream water levels and corresponding chamber depths. Our results demonstrate the technical feasibility of enhancing single-lock cargo capacity while maintaining safety margins. These findings provide (1) a scientifically grounded framework for draft control optimization, and (2) actionable insights for lock operation management. The study establishes a methodological foundation for balancing navigational safety with growing throughput requirements in constrained waterways. Full article

Short-sea shipping, abbreviated SSS, is the transportation of goods by sea over relatively short distances, in contrast to intercontinental ocean and deep-sea shipping. Short-sea shipping (SSS) is important for cargo transportation in some regions of the world with many ports and well-developed liner shipping. The development and improvement of SSS systems is an important scientific and practical task. This article presents theoretical and experimental results of the development and optimization of SSS. A methodology for connecting and evaluating SSS and other transport chains was developed and tested by experimental studies, with the help of which it is possible to assess the efficiency of SSS and other transport chains, e.g., in terms of economy, freight transportation time, and environmental impact. The developed SSS methodology includes sea and land transport corridors, their assessment, and possible ways of optimizing transport chains using a comparative method and can be applied to various transport and logistics chains. The basis for the development and verification of the SSS methodology was the theoretical and experimental results of real short-sea shipping operations. The use of a comparative method based on which transport and logistics chains are assessed allows one to search for the most optimal SSS routes and possible factors that allow optimizing transportation costs and reducing transportation time and environmental impact. PAMAM dendrimers are distinguished by their capacity for functionalization, which enhances the properties of the compounds they transport, rendering them highly versatile nanoparticles with extensive applications in the biomedical domain, including drug, vaccine, and gene delivery. These dendrimers can be internalized into cells through various endocytic mechanisms, such as passive diffusion, clathrin-mediated endocytosis, and caveolae-mediated endocytosis, allowing them to traverse the cytoplasm and reach intracellular targets, such as the mitochondria or nucleus. Despite the significant challenge posed by the cytotoxicity of these nanoparticles, which is contingent upon the dendrimer size, surface charge, and generation, numerous strategies have been documented to modify the dendrimer surface using polyethylene glycol and other chemical groups to temporarily mitigate their cytotoxic effects. The potential of PAMAM dendrimers in cancer therapy and other biomedical applications is substantial, owing to their ability to enhance bioavailability, pharmacokinetics, and pharmacodynamics of active ingredients within the body. This underscores the necessity for further investigation into the optimization of internalization pathways and cytotoxicity of these nanoparticles. This review offers a comprehensive synthesis of the current literature on the diverse cellular internalization pathways of PAMAM dendrimers and their cargo molecules, emphasizing the mechanisms of entry, intracellular trafficking, and factors influencing these processes. Due to the recent significant increase in the scale of both domestic and international cargo transportation, the transport sector is becoming an important factor in the country’s economic development. This implies the need to improve all links in the cargo transportation chain. A key role in it is played by logistics centers, which in their activities must meet both state (CO2 emissions, reduction in road load, increase in transportation safety, etc.) and commercial (cargo transportation in the shortest time and at the lowest cost) requirements. The objective of the paper is freight transportation from China to European countries, reflecting issues of CO2 emissions, reduction in road load, and increase in transportation safety. Transport operations from the manufacturer to the logistics center are especially important in this chain, since the efficiency of transportation largely depends on the decisions made by its employees. They select the appropriate types of transport (air, sea, rail, and road transport) and routes for a specific situation. In methodology, the analyzed problem can be presented as a dynamic multi-criteria decision model. It is assumed that the decision-maker—the manager responsible for planning transportation operations—is interested in achieving three basic goals: financial goal minimizing total delivery costs from factories to the logistics center, environmental goal minimizing the negative impact of supply chain operations on the environment, and high level of customer service goal minimizing delivery times from factories to the logistics center. The proposed methodology allows one to reduce the total carbon dioxide emission by 1.1 percent and the average duration of cargo transportation by 1.47 percent. On the other hand, the total cost of their delivery increases by 1.25 percent. By combining these, it is possible to create optimal transportation options, effectively use vehicles, reduce air pollution, and increase the quality of customer service. All this would significantly contribute to the country’s socio-economic development. It is proposed to solve this complex problem based on a dynamic multi-criteria model. In this paper, the problem of constructing a schedule of transport operations from factories to a logistics center is considered. The analyzed problem can be presented as a dynamic multi-criteria decision model. Linear programming and the AHP method were used to solve it. Full article

The maritime industry remains a significant contributor to global greenhouse gas (GHG) emissions. In this article, a systematic study has been performed on the alternative fuel emissions of large cargo ships under different route scenarios and propulsion systems. For this purpose, a set of key performance indicators (KPIs) are evaluated, including total equivalent CO2 emissions (CO2eq), CO2eq emissions per unit of transport mass and CO2eq emissions per unit of transport mass per distance. The emissions analysis demonstrates that Liquified Natural Gas (LNG) paired with Marine Gas Oil (MGO) emerges as the most viable short-term solution in comparison with the conventional fuel oil propulsion. Synthetic methanol (eMeOH) paired with synthetic diesel (eDiesel) is identified as the most promising long-term fuel combination. When comparing the European Union (EU) emission calculation system (FuelEU) with the International Maritime Organization (IMO) emission metrics, a discrepancy in emissions reduction outcomes has been observed. The IMO approach appears to favor methanol (MeOH) and liquefied natural gas (LNG) over conventional fuel oil. This is attributed to the fact that the IMO metrics do not consider unburned methane emissions (methane slip) and emissions in the production of fuels (Well-to-Tank). Full article

Facing the increasingly severe challenges of global climate change, China has established clear “dual carbon” goals, with the core objective of achieving carbon peak by 2030 or earlier. However, carbon emissions from the road freight industry have remained higher for many years; understanding and estimating the characteristics of truck carbon emissions are critical for developing a low-carbon transportation system. This study takes Heilongjiang Province, a typically cold region, as a case study. By employing the growth curve method, we predicted the time for achieving carbon peak and constructed an improved STIRPAT model to identify key drivers and pathways for emission reduction in the road freight system. The research results show that only by committing to using the economy to reduce carbon emissions and improve energy intensity can the overall carbon emissions of Heilongjiang Province’s cargo transportation system achieve the “dual carbon” goals as soon as possible. If we develop according to the optimistic scenario proposed in this article, by 2030, the total quantity of trucks will reach about 933,720, and the carbon emissions per vehicle will reach about 178.14 t. If we actively increase the proportion of new energy trucks in the overall quantity of trucks, the peak time is expected to be achieved around 2030. The improvement of technological efficiency (e.g., lowering energy intensity) and the advancement of economic development have been identified as effective pathways for carbon emission reduction. Empirical studies indicate that these measures can achieve emission reduction impacts that are approximately 60 times and 10 times greater, respectively, in terms of efficiency, compared to baseline scenarios. Furthermore, energy intensity improvements and structural shifts toward low-carbon vehicles are critical to expediting peak attainment. This study provides a methodological framework for cold-region emission projections and offers actionable insights for policymakers to design tailored emission reduction pathways in the road freight transportation industry. An engineered molecular motor composed of an ATP-dependent kinesin-1 monomer and an ATP-independent diffusing microtubule-associated protein is proposed, and its dynamics are studied theoretically. It is shown that the engineered motor can move directionally on microtubules towards the plus end, bearing great potential for applications in therapeutics or nanorobotics. The engineered motor can have an unloaded velocity similar to the wild-type kinesin-1 dimer, can take a mechanical (either forward or backward) step by hydrolyzing an ATP molecule under any load, and can generate the maximum force that is about half of that generated by the wild-type kinesin-1 dimer. With the global maritime industry accelerating toward carbon neutrality, the adoption of alternative marine fuels has emerged as a pivotal pathway for achieving net-zero emissions. To identify the most promising fuel transition solution for multi-purpose heavy-lift vessels (MPHLVs), which are widely used for transporting large and complex industrial equipment and have specialized structural requirements, this study conducted a comprehensive techno-economic analysis based on a fleet of 12 MPHLVs. An eight-dimensional technical adaptability framework was established, and six types of marine fuel were evaluated. Concurrently, a total cost assessment model was developed using 2024 operational data of the fleet, incorporating the fuel procurement, the carbon allowances under the EU ETS, the FuelEU Maritime compliance costs, and the IMO Net-Zero penalties. The results show that methanol as an alternative fuel is the most compatible decarbonization pathway for this specialized vessel type. A case study of a 38,000 DWT methanol-fueled MPHLV further demonstrates engineering feasibility with minimal impact on cargo capacity, and validates methanol’s potential as a technically viable and strategically transitional fuel for MPHLVs, particularly in the context of stricter international decarbonization regulations. The proposed evaluation framework and engineering application offer practical guidance for fuel selection, ship design, and retrofit planning, supporting the broader goal of accelerating low-carbon development in heavy-lift shipping sector.
