Optimization of the Viability of Microencapsulated Lactobacillus reuteri in Gellan Gum-Based Composites Using a Box–Behnken Design

The growing interest in probiotic bacteria within the food industry is driven by their recognized health benefits for consumers. However, preserving their therapeutic viability and stability during gastrointestinal transit remains a formidable challenge. Hence, this research aimed to enhance the viability of Lactobacillus reuteri through microencapsulation using a binary polysaccharide mixture composed of low acyl gellan gum (LAG), high acyl gellan gum (HAG), and calcium for the microencapsulation of L. reuteri. To achieve this, the Box–Behnken design was applied, targeting the optimization of L. reuteri microencapsulated to withstand simulated gastrointestinal conditions. The microcapsules were crafted using the internal ionic gelation method, and optimization was performed using response surface methodology (RSM) based on the Box–Behnken design. The model demonstrated robust predictive power, with R2 values exceeding 95% and a lack of fit greater than p > 0.05. Under optimized conditions—0.88% (w/v) LAG, 0.43% (w/v) HAG, and 24.44 mM Ca—L. reuteri reached a viability of 97.43% following the encapsulation process. After 4 h of exposure to simulated gastric fluid (SGF) and intestinal fluid (SIF), the encapsulated cells maintained a viable count of 8.02 log CFU/mL. These promising results underscore the potential of biopolymer-based microcapsules, such as those containing LAG and HAG, as an innovative approach for safeguarding probiotics during gastrointestinal passage, paving the way for new probiotic-enriched food products. Background: The aim of this review is to summarize and evaluate the properties of antibacterial polysaccharides for application in dental implantology to identify knowledge gaps and provide new research ideas. Methods: The electronic databases PubMed, Medline, ProQuest, and Google Scholar were used to search for peer-reviewed scientific publications published between 2018 and 2025 that provide insights to answer research questions on the role of antibacterial polysaccharides in combating pathogens in dental implantology without triggering immune reactions and inflammation. Further research questions relate to the efficacy against various dental pathogens and the understanding of the antibacterial mechanism, which may enable the development of functionalized polysaccharides with long-term antibacterial activity. Results: Biomedical implants have revolutionized medicine but also increased the risk of infections. Implant infections are a major problem in implantology and lead to implant failure and replacement. An antibacterial coating could be an excellent strategy to extend the lifespan of implants and improve the quality of the patient’s life. Bacterial resistance to antibiotics poses significant challenges for researchers, forcing them to search for new ways to prevent bacterial infections in implantology. Antibacterial natural polymers have recently received considerable research attention due to their long-term antibacterial activity. Polysaccharides from marine sources, such as chitosan and alginate, or pectin, xanthan, etc., from various plants, appear to be promising biopolymers for such applications in implantology due to their antibacterial activity, biocompatibility, and osteogenic properties. The antibacterial activity of these natural biopolymers depends on their chemical and physical properties. Nanopolysaccharides exhibit higher antibacterial activity than conventional polysaccharides, but their toxicity to human cells must be considered. Their antibacterial activity is based on the disruption of bacterial DNA or RNA synthesis, increased cell wall permeability, membrane disruption, and cytoplasmic leakage. Conclusions: Polysaccharides are a class of natural polymers with a broad spectrum of biological activities. They exhibit antioxidant, immunomodulatory, anticoagulant, anticancer, anti-inflammatory, antibacterial, and antiviral activity. Furthermore, polysaccharides are non-cytotoxic and exhibit good biocompatibility with osteogenic cells. Bactericidal polysaccharides are attractive new antibacterial materials against implant infections and open up new perspectives in implantology. Full article

Biopolymers and bio-based plastics, such as polylactic acid (PLA) and polybutylene succinate (PBS), are recognized as environmentally friendly materials and are widely used, especially in the packaging industry. The purpose of this study was to assess the degradation of PLA- and PBS-based formulations in the forms of granules and films under controlled composting conditions at a laboratory scale. Biodegradation tests of bio-based materials were conducted under controlled aerobic conditions, following the standard EVS-EN ISO 14855-1:2012. Scanning electron microscopy (SEM) was performed using a high-resolution Zeiss Ultra 55 scanning electron microscope to analyze the samples. After the six-month laboratory-scale composting experiment, it was observed that the PLA-based materials degraded by 47.46–98.34%, while the PBS-based materials exhibited a final degradation degree of 34.15–80.36%. Additionally, the PLA-based compounds displayed a variable total organic carbon (TOC) content ranging from 38% to 56%. In contrast, the PBS-based compounds exhibited a more consistent TOC content, with a narrow range from 53% to 54%. These findings demonstrate that bioplastics can contribute to reducing plastic waste through controlled composting, but their degradation efficiency depends on the material composition and environmental conditions. Future efforts should optimize bioplastic formulations and composting systems while developing supportive policies for wider adoption. Full article

Cultured meat is emerging as a sustainable alternative to conventional animal agriculture, with scaffolds playing a central role in supporting cellular attachment, growth, and tissue maturation. This review focuses on the development of gel-based hybrid biomaterials that meet the dual requirements of biocompatibility and food safety. We explore recent advances in the use of naturally derived gel-forming polymers such as gelatin, chitosan, cellulose, alginate, and plant-based proteins as the structural backbone for edible scaffolds. Particular attention is given to the integration of food-grade functional additives into hydrogel-based scaffolds. These include nanocellulose, dietary fibers, modified starches, polyphenols, and enzymatic crosslinkers such as transglutaminase, which enhance mechanical stability, rheological properties, and cell-guidance capabilities. Rather than focusing on fabrication methods or individual case studies, this review emphasizes the material-centric design strategies for building scalable, printable, and digestible gel scaffolds suitable for cultured meat production. By systemically evaluating the role of each component in structural reinforcement and biological interaction, this work provides a comprehensive frame work for designing next-generation edible scaffold systems. Nonetheless, the field continues to face challenges, including structural optimization, regulatory validation, and scale-up, which are critical for future implementation. Ultimately, hybrid gel-based scaffolds are positioned as a foundational technology for advancing the functionality, manufacturability, and consumer readiness of cultured meat products, distinguishing this work from previous reviews. Unlike previous reviews that have focused primarily on fabrication techniques or tissue engineering applications, this review provides a uniquely food-centric perspective by systematically evaluating the compositional design of hybrid hydrogel-based scaffolds with edibility, scalability, and consumer acceptance in mind. Through a comparative analysis of food-safe additives and naturally derived biopolymers, this review establishes a framework that bridges biomaterials science and food engineering to advance the practical realization of cultured meat products. Biopolymer films doped with active substances may become a promising alternative to traditional dressings for skin wounds, as they can deliver drugs while maintaining wound moisture, thus contributing to the healing process. This article describes the preparation of amygdalin-doped biopolymer films for in vitro testing against the bacterial strains typical of chronic wounds: E. coli and S. aureus. Thus, FTIR characterization suggests minimal chemical interaction between amygdalin and the biopolymer matrix components, indicating potential compatibility, while thermogravimetric analysis highlights the thermal behavior of the films as well as the influence of the polymer matrix composition on the amount of bound water and the shift of Tpeak value for the decomposition process of the base polymer. Moreover, the identity of the secondary biopolymer (gelatin or CMC) significantly influences film morphology and antibacterial performance. Full article

Recently, a number of natural biologically active substances have been proven to be attractive alternatives to conventional anticancer medicine or as adjuvants in contemporary combination therapies. Although lignin-based materials were previously accepted as waste materials with limited usefulness, recent studies increasingly report the possibility of their use for novel applications in various industrial branches, including biomedicine. In this regard, the safety, efficiency, advantages and limitations of lignin compounds for in vitro/in vivo applications remain poorly studied and described. This study was carried out to investigate the possibility of using newly synthesized, alkali lignin-based micro-/nano-biopolymer formulations (Lignin@Formulations/L@F) as carriers for substances with antioxidant and/or anticancer effectiveness. Moreover, we tried to assess the opportunity for using an electro-assisted approach for achieving improved intracellular internalization. An investigation was conducted on an in vitro panel of breast cell lines, namely two breast cancer lines with different metastatic potentials and one non-tumorigenic line as a control. The characterization of all tested formulations was performed via DLS (dynamic light scattering) analysis. We developed an improved separation procedure via size/charge unification for all types of Lignin@Formulations. Moreover, in vitro applications were investigated. The results demonstrate that compared to healthy breast cells, both tested cancer lines exhibited slight sensitivity after treatment with different formulations (empty or loaded with antioxidant substances). This effect was also enhanced after applying electric pulses. L@F loaded with Quercetin was also explored only on the highly metastatic cancer cell line as a model for the breast cancer type most aggressive and non-responsive to traditional treatments. All obtained data suggest that the tested formulations have potential as carriers for the electro-assisted delivery of natural antioxidants such as Quercetin. Full article

The textile industry’s reliance on synthetic dyes contributes significantly to pollution, highlighting the need for sustainable alternatives like biopigments. This study investigates the production and application of the biopigment prodigiosin, which was produced by Pseudomonas putida with a yield of 1.85 g/L. Prodigiosin was prepared under acidic, neutral, and alkaline conditions, resulting in varying protonation states that influenced its affinity for cotton and polyester fibers. Three surfactants (anionic, cationic, non-ionic) were tested, with non-ionic Tween 80 yielding a promising color strength (above 4) and fastness results with neutral prodigiosin at 1.3 g/L. Cotton and polyester demonstrated good washing (color difference up to 14 for cotton, 5 for polyester) and light fastness (up to 15 for cotton, 16 for polyester). Cellulose acetate, used in the conventional printing process as a thickener, produced superior color properties compared to commercial thickeners. Neutral prodigiosin achieved higher color strength, and cotton fabrics displayed halochromic properties, distinguishing them from polyester, which showed excellent fastness. Prodigiosin-printed samples also exhibited strong antimicrobial activity against Pseudomonas aeruginosa and retained halochromic properties over 10 pH cycles. These findings suggest prodigiosin as a sustainable dye alternative and pH sensor, with potential applications in biomedical materials, such as antimicrobial and pH-responsive wound dressings. An increasing global population, rising energy demands, and the shift toward a circular bioeconomy are driving the need for more resource-efficient waste management. The increase in the world population—now exceeding 8 billion as of 2024—results in an increased need for alternative proteins, both human and feed grade proteins, as well as for biopolymers and bioenergy. As such, agricultural, forest, and marine waste biomass represent a valuable feedstock for production of food and feed ingredients, biopolymers, and bioenergy. However, the lack of integrated and efficient valorization strategies for these diverse biomass sources remains a major challenge. This literature review aims to give a systematic approach on the recent research status of agricultural, forest, and marine waste biomass valorization, focusing on cascade processing (a sequential combination of processes such as pretreatment, extraction, and conversion methods). Potential products will be identified that create the most economic value over multiple lifetimes, to maximize resource efficiency. It highlights the challenges associated with cascade processing of waste biomass and proposes technological synergies for waste biomass valorization. Moreover, this review will provide a comprehensive understanding of the potential of waste biomass valorization in the context of sustainable and circular bioeconomy. Full article

Marine biomass, particularly from waste streams, by-products, underutilized, invasive, or potential cultivable marine species, offers a sustainable source of high-value biopolymers such as collagen and chitin. These macromolecules have gained significant attention due to their biocompatibility, biodegradability, functional versatility, and broad applicability across health, food, wellness, and environmental fields. This review highlights recent advances in the uses of marine-derived collagen and chitin/chitosan. In alignment with the United Nations Sustainable Development Goals (SDGs), we analyze how these applications contribute to sustainability, particularly in SDGs related to responsible consumption and production, good health and well-being, and life below water. Furthermore, we contextualize the advancement of product development using marine collagen and chitin/chitosan within the European Union’s Blue bioeconomy strategies, highlighting trends in scientific research and technological innovation through bibliometric and patent data. Finally, the review addresses challenges facing the development of robust value chains for these marine biopolymers, including collaboration, regulatory hurdles, supply-chain constraints, policy and financial support, education and training, and the need for integrated marine resource management. The paper concludes with recommendations for fostering innovation and sustainability in the valorization of these marine resources. Full article

Kefiran, the extracellular polysaccharide produced from the Generally Recognized as Safe (GRAS) bacteria in kefir grains, with its well-documented functional and health-promoting properties, constitutes a promising biopolymer with a variety of possible uses. Its compatibility with other biopolymers, such as milk proteins, and its ability to form standalone cryogels allow it to be utilized for the fabrication of films with improved properties for applications in the food and biomedical–pharmaceutical industries. In the present work, the properties of kefiran films were investigated in the presence of milk proteins (sodium caseinate, native and heat-treated whey proteins, and their mixtures), alongside glycerol (as a plasticizer) and cryo-treatment of the film-forming solution prior to drying. A total of 24 kefiran films were fabricated and studied for their physical (thickness, moisture content, water solubility, color parameters and vapor adsorption), mechanical (tensile strength and elongation at break), and optical properties. Milk proteins increased film thickness, solubility and tensile strength and reduced water vapor adsorption. The hygroscopic effect of glycerol was mitigated in the presence of milk proteins and/or the application of cryo-treatment. Glycerol was the most effective at reducing the films’ opacity. Heat treatment of whey proteins proved to be the most effective in increasing film tensile strength, reducing, at the same time, the elongation at break, while sodium caseinates in combination with cryo-treatment resulted in films with high tensile strength and the highest elongation at break. Cryo-treatment, carried out in the present study through freezing followed by gradual thawing of the film-forming solution, proved to be the most effective factor in decreasing film roughness. Based on our results, proper selection of the film-forming solution composition and its treatment prior to drying can result in kefiran–glycerol films with favorable properties for particular applications. Background/Objectives: Dental practitioners widely use dental implants to treat traumatic cases. Titanium implants are currently the most popular choice among dental practitioners and surgeons. The discovery of newer polymeric materials is also influencing the interest of dental professionals in alternative options. A comparative study between existing titanium implants and newer polymeric materials can enhance professionals’ ability to select the most suitable implant for a patient’s treatment. This study aimed to investigate material property advantages of high-performance thermoplastic biopolymers such as PEEK and PEKK, as compared to the time-tested titanium implants, and to find the most suitable and economically fit implant material. Methods: Three distinct implant material properties were assigned—PEEK, PEKK, and commercially pure titanium (CP Ti-55)—to dental implants measuring 5.5 mm by 9 mm, along with two distinct titanium (TI6AL4V) abutments. Twelve three-dimensional (3D) models of bone blocks, representing the mandibular right molar area with Osseo-integrated implants were created. The implant, abutment, and screw were assumed to be linear; elastic, isotropic, and orthotropic properties were attributed to the cancellous and cortical bone. Twelve model sets underwent a three-dimensional finite element analysis to evaluate von Mises stress and total deformation under 250 N vertical and oblique (30 degree) loads on the top surface of each abutment. Results: The study revealed that the time-tested titanium implant outperforms PEEK and PEKK in terms of marginal bone preservation, while PEEK outperforms PEKK. Conclusions: This study will assist dental practitioners in selecting implants from a variety of available materials and will aid researchers in their future research. Full article

The search for sustainable, economically viable, and effective plant protection strategies against pathogenic bacteria, fungi, and viruses is a major challenge in modern agricultural practices. Chitosan (CS) is an abundant cationic natural biopolymer known for its biocompatibility, low toxicity, and antimicrobial properties. Its potential use in agriculture for pathogen control is a promising alternative to traditional chemical fertilisers and pesticides, which raise concerns regarding public health, environmental protection, and pesticide resistance. This study focused on the preparation of chitosan nanoparticles (CS-NPs) through cross-linking with organic molecules, such as tannic acid (TA). Various formulations were explored for the development of stable nanoscale particles having encapsulation capabilities towards low compounds of varying polarity and with potential agricultural applications relevant to plant health and growth. The solution properties of the NPs were assessed using dynamic and electrophoretic light scattering (DLS and ELS); their morphology was observed through atomic force microscopy (AFM), while analytical ultracentrifugation (AUC) measurements provided insights into their molar mass. Their properties proved to be primarily influenced by the concentration of CS, which significantly affected its intrinsic conformation. Additional structural insights were obtained via infrared and UV–Vis spectroscopic measurements, while detailed fluorescence analysis with the use of three different probes, as model cargo molecules, provided information regarding the hydrophobic and hydrophilic microdomains within the particles. Food waste has emerged as a critical worldwide concern, resulting in environmental deterioration and economic detriment. Bio-based natural polymer coatings and films have emerged as a sustainable solution to food preservation challenges, particularly in reducing postharvest losses and extending shelf life. Compared to their synthetic counterparts, these polymers, such as chitosan, starch, cellulose, proteins, and alginate, are derived from renewable sources that are biodegradable, safe, and functional. Within this context, this review examines the various bio-based natural polymer coatings and films as biodegradable, edible alternatives to conventional packaging solutions. It examines the different fabrication methods, like solution casting, electrospinning, and spray coating, and incorporates antimicrobial agents to enhance performance. Emphasis is placed on their mechanical, barrier, and antimicrobial properties, their application in preserving fresh produce, how they promote food safety and environmental sustainability, and accompanying limitations. This review highlights the importance of bio-based natural polymer coatings and films as a promising, eco-friendly solution to enhancing food quality, safety, and shelf life while addressing global sustainability challenges. Over the last few decades, the growing demand for sustainable resources has made biopolymers increasingly popular, as they offer an eco-friendly alternative to conventional synthetic polymers, which are often associated with environmental issues such as the formation of microplastics and toxic substances. Functionalization of biomaterials involves modifying their physical, chemical, or biological properties to improve their performance for specific applications. Cellulose and bacterial cellulose are biopolymers of interest, due to the plethora of hydroxyl groups, their high surface area, and high porosity, which makes them ideal candidates for several applications. However, there are applications, which require precise control of their wetting properties. In this review, we present the most effective fabrication methods for modifying both the morphology and the chemical properties of cellulose and bacterial cellulose, towards the realization of superhydrophobic bacterial cellulose films and surfaces. Such materials can find a wide variety of applications, yet in this review we target and discuss applications deriving from the wettability control, such as antibacterial surfaces, wound healing films, and separation media. The Effect of the Extraction Temperature on the Colligative, Hydrodynamic and Rheological Properties of Psyllium Husk Mucilage Raw Solutions

The aim of the research was to analyse the effect of different extraction temperatures on the colligative, hydrodynamic, and rheological properties of a water-soluble AXs fractions. The research material consisted of raw water extracts of arabinoxylans obtained from the husk at the following temperatures: 40 °C (AX40), 60 °C (AX60), 80 °C (AX80), and 100 °C (AX100). These were characterised in terms of their hydrodynamic, osmotic, and rheological properties, as well as the average molecular mass of the polysaccharide fractions. An increase in extraction temperature resulted in an increase in weight-average molecular mass, from 2190 kDa (AX40) to 3320 kDa (AX100). The values of the osmotic average molecular mass were higher than those obtained from GPC, and decreased with increasing extraction temperature. The dominance of biopolymer–biopolymer interactions was evident in the shape of the autocorrelation function, which did not disappear as the extraction temperature and concentration increased. Furthermore, the values of the second virial coefficient were negative, which is indicative of the tendency of biopolymer chains to aggregate. The rheological properties of the extracts changed from being described by a power-law model (AX40 and AX60) to being described by the full non-linear De Kee model (AX80 and AX100). Full article
