Ferroptosis in Septic Cardiomyopathy Is Alleviated by Ondansetron: The Critical Role of the HTR3A-ATF3 Axis in Mitochondrial and Oxidative Homeostasis

Background: Emerging evidence has established ferroptosis as a vital factor in the pathogenesis of cardiovascular diseases, especially in septic cardiomyopathy (SCM). Meanwhile, ondansetron (OND), a well-established 5-HT3 receptor antagonist, has gained increasing attention for its pleiotropic effects. However, its potential to modulate ferroptosis in the cardiovascular field remains unexplored. This study aims to fill this gap by exploring the potential of OND as an innovative therapeutic intervention for SCM. Methods: This study utilized both in vitro and in vivo models of septic cardiomyopathy (SCM), which was induced by lipopolysaccharide (LPS) stimulation in neonatal rat cardiomyocytes (NRCMs) and C57BL/6 mice. Through RNA sequencing, as well as molecular and functional assessments—including echocardiography and ferroptosis-related measurements—we revealed the anti-ferroptotic effect of ondansetron (OND). Mechanistically, ATF3 was identified as a pivotal regulator, with its overexpression via AAV9 in vivo and ADV in vitro confirming its role in OND-induced cardioprotection. Results: Ondansetron (OND) showed potent anti-ferroptotic effects in both cellular and murine models of septic cardiomyopathy (SCM). Treatment with OND not only improved cardiac performance but also reduced ferroptotic markers, mitigated lipid peroxidation and iron overload, and bolstered antioxidant defense. Notably, OND administration attenuated oxidative and endoplasmic reticulum (ER) stress while restoring mitochondrial integrity. Mechanistically, the anti-ferroptotic activity of OND was mediated through the HTR3A/ATF3 axis: ATF3 overexpression negated OND’s protective effects, while HTR3A antagonism with VUF10166 recapitulated its benefits. Conversely, HTR3A agonism with PBG attenuated ferroptosis resistance, further implicating this pathway as central to OND’s mechanism. Conclusions: This study demonstrated a novel pharmacological role for ondansetron (OND) in attenuating ferroptosis in septic cardiomyopathy (SCM) via the HTR3A/ATF3 signaling pathway. This finding delineates a novel therapeutic avenue and supports the repurposing of OND beyond its traditional antiemetic use to cardiovascular applications. Full article

Peer-to-peer (P2P) energy trading has emerged as a novel approach to enhancing the coordination and utilization of distributed energy resources (DERs) within modern power distribution networks. This study presents a techno-economic analysis of different DER characteristics, focusing on the integration of photovoltaic (PV) systems and energy storage systems (ESS) within a community-based P2P energy trading framework in Aswan, Egypt, under a time-of-use (ToU) electricity tariff. Eight distinct cases are evaluated to assess the impact of different DER characteristics on P2P energy trading performance and an unbalanced low-voltage (LV) distribution network by varying the PV capacity, ESS capacity, and ESS charging power. To the best of the authors’ knowledge, this is the first study to comprehensively examine the effects of different DER characteristics on P2P energy trading and the associated impacts on an unbalanced distribution network. The findings demonstrate that integrating PV and ESS can substantially reduce operational costs—by 37.19% to 68.22% across the analyzed cases—while enabling more effective energy exchanges among peers and with the distribution system operator (DSO). Moreover, DER integration reduced grid energy imports by 30.09% to 63.21% and improved self-sufficiency, with 30.10% to 63.21% of energy demand covered by community DERs. However, the analysis also reveals that specific DER characteristics—particularly those with low PV capacity (1.5 kWp) and high ESS charging rates (e.g., ESS 13.5 kWh with 2.5 kW inverter)—can significantly increase transformer and line loading, reaching up to 19.90% and 58.91%, respectively, in Case 2. These setups also lead to voltage quality issues, such as increased voltage unbalance factors (VUFs), peaking at 1.261%, and notable phase voltage deviations, with the minimum Vb dropping to 0.972 pu and maximum Vb reaching 1.083 pu. These findings highlight the importance of optimal DER sizing and characteristics to balance economic benefits with technical constraints in P2P energy trading frameworks. Full article

In order to cope with the ecological pressure caused by the uncontrolled expansion of cultivated land in arid areas and ensure regional food security, the implementation of a cultivated land fallowing system has become an effective way to restore the ecology, alleviate the pressure on cultivated land, and increase productivity. In view of this, this paper takes the Tarim River Basin, located in the arid zone of China’s agricultural continent, as the research object. Using a land use transfer matrix and a gravity center migration model, the paper analyzes the spatiotemporal characteristics of cultivated land expansion in the Tarim River Basin from 2000 to 2020. Through remote sensing and the integration of multi-source data, the paper constructs an arable land fallow urgency index (SILF) from multiple dimensions such as human activity intensity, ecological vulnerability, output value, water resources status, and terrain conditions. The research results show that (1) cultivated land in the Tarim River Basin expanded by 15,665.133 km2 in general, which is manifested by spreading around based on existing cultivated land, mainly from the conversion of grassland and unused land; the center of gravity of cultivated land moved 37.833 km to the northeast and 7.257 km to the southwest first. (2) The area of not urgently fallow (NUF) in the watershed showed an overall downward trend, decreasing by 10%, while the area of very urgently fallow (VUF) increased by 16%. VUF is mainly distributed in the marginal areas of cultivated land close to the desert and is gradually expanding into the interior of cultivated land. (3) The overall ecological environment of cultivated land in the watershed is showing a deteriorating trend, and the deterioration is gradually spreading from the edge of the cultivated land to the interior. (4) There are significant differences in the SILF values of different land use types after conversion to cultivated land. The urgency of fallowing cultivated land converted from unused land is the highest, followed by grassland, forest land, water bodies, and construction land. The expanded cultivated land has a higher SILF value than the original cultivated land. The research results can provide insights into regional land resource management, the formulation of cultivated land protection policies, and the ecological restoration of cultivated land. In this study, a hybrid heading control framework for unmanned surface vehicles (USVs) is proposed, combining variable domain fuzzy Proportional–Integral–Derivative (VUF-PID) with an improved algorithmic Beetle Antennae Search–Particle Swarm Optimization–Simulated Annealing (BAS-PSO-SA) optimization to address the multi-objective control challenge. Key innovations include a self-tuning VUF mechanism that improves disturbance rejection by 42%, a weighted adaptive optimization strategy that reduces parameter tuning iterations by 37%, and an asymmetric learning factor that balances global exploration and local refinement. Benchmarks using Rastrigin, Griewank, and Sphere functions show superior convergence and 68% stability improvement. Ocean heading simulations of a 7.02 m unmanned surface vehicle (USV) using the Nomoto model show a 91.7% reduction in stabilization time, a 0.9% reduction in overshoot, and a 30% reduction in optimization iterations. The experimental validation under wind and wave disturbances shows that the heading deviation is less than 0.0392°, meeting the IMO MSC.1/Circ.1580 standard, and an 89.5% improvement in energy efficiency. Although the processing time is 12.7% longer compared to the GRO approach, this framework lays a solid foundation for ship autonomy systems, and future enhancements will focus on MPC-based time delay compensation and Field-Programmable Gate Array (FPGA) acceleration. The integration of Internet of Medical Things (IoMT) technology has revolutionized healthcare, allowing rapid access to medical images and enhancing remote diagnostics in telemedicine. However, this advancement raises serious cybersecurity concerns, particularly regarding unauthorized access and data integrity. This paper presents a novel, user-friendly, visible watermarking scheme for medical images—Visual and User-Friendly Medical Image Watermarking Scheme (VUF-MIWS)—designed to secure medical image ownership while maintaining usability for diagnostic purposes. VUF-MIWS employs a unique combination of inpainting and data hiding techniques to embed hospital logos as visible watermarks, which can be removed seamlessly once image authenticity is verified, restoring the image to its original state. Experimental results demonstrate the scheme’s robust performance, with the watermarking process preserving critical diagnostic information with high fidelity. The method achieved Peak Signal-to-Noise Ratios (PSNR) above 70 dB and Structural Similarity Index Measures (SSIM) of 0.99 for inpainted images, indicating minimal loss of image quality. Additionally, VUF-MIWS effectively restored the ROI region of medical images post-watermark removal, as verified through test cases with restored watermarked regions matching the original images. These findings affirm VUF-MIWS’s suitability for secure telemedicine applications. Full article

In power systems, nonlinear loads cause harmonic distortion, adversely affecting sensitive equipment such as induction motors, power electronics, and variable-speed drives. This paper presents a novel control strategy that integrates with existing hierarchical control systems to mitigate voltage imbalances and harmonic disturbances in AC-islanded microgrids. The proposed method utilizes selective harmonic order filtering through multiple second-order generalized integrators (MSOGI) to extract negative, positive, and harmonic order components. The distributed generation (DG) unit control mechanism is designed to immediately correct voltage imbalances and harmonic disruptions, distributing the compensatory load evenly to rectify real and reactive power imbalances and harmonic disturbances. The microgrid’s control architecture primarily includes droop controllers for real and reactive power of positive sequences, voltage and current regulation inner control loops, an additional loop for correcting imbalances and harmonics, and secondary controllers to maintain voltage magnitude and frequency at nominal levels, ensuring high-quality voltage at the point of common coupling (PCC). The effectiveness of this approach is demonstrated through simulation results on the MATLAB/Simulink platform, proving its ability to effectively mitigate voltage imbalances and harmonic issues with the total harmonic of voltage reduced to approximately THDv = 0.5% and voltage unbalance factor (VUF) within approximately 0.1%. The high penetration of distributed photovoltaics (PV) in distribution networks (DNs) results in voltage violations, imbalances, and flickers, leading to significant disruptions in DN stability. To address this issue, this paper proposes a multi-timescale voltage regulation approach that involves the coordinated control of a step voltage regulator (SVR), switched capacitor (SC), battery energy storage system (BESS), and electric vehicle (EV) across different timescales. During the day-ahead stage, the proposed method utilizes artificial hummingbird algorithm optimization-based least squares support vector machine (AHA-LSSVM) forecasting to predict the PV output, enabling the formulation of a day-ahead schedule for SVR and SC adjustments to maintain the voltage and voltage unbalance factor (VUF) within the limits. In the intra-day stage, a novel floating voltage threshold band (FVTB) control strategy is introduced to refine the day-ahead schedule, enhancing the voltage quality while reducing the erratic operation of SVR and SC under dead band control. For real-time operation, the African vulture optimization algorithm (AVOA) is employed to optimize the BESS output for precise voltage regulation. Additionally, a novel smoothing fluctuation threshold band (SFTB) control strategy and an initiate charging and discharging strategy (ICD) for the BESS are proposed to effectively smooth voltage fluctuations and expand the BESS capacity. To enhance user-side participation and optimize the BESS capacity curtailment, some BESSs are replaced by EVs for voltage regulation. Finally, a simulation conducted on a modified IEEE 33 system validates the efficacy of the proposed voltage regulation strategy. Full article

Currently, the main solution for braking systems for underground electric trackless rubber-tired vehicles (UETRVs) is traditional hydraulic braking systems, which have the disadvantages of hydraulic pressure crawling, the risk of oil leakage and a high maintenance cost. An electro-mechanical-braking (EMB) system, as a type of novel brake-by-wire (BBW) system, can eliminate the above shortcomings and play a significant role in enhancing the intelligence level of the braking system in order to meet the motion control requirements of unmanned UETRVs. Among these requirements, the accurate control of clamping force is a key technology in controlling performance and the practical implementation of EMB systems. In order to achieve an adaptive clamping force control performance of an EMB system, an optimized fuzzy proportional-integral-differential (PID) controller is proposed, where the improved fuzzy algorithm is utilized to adaptively adjust the gain parameters of classic PID. In order to compensate for the deficiency of single-close-loop control and adjusting the brake gap automatically, a cascaded three-closed-loop control architecture with force/position switch technology is established, where a contact point detection method utilizing motor rotor angle displacement is proposed via experiments. The results of the simulation and experiments indicate that the clamping force response of the proposed multi-close-loop Variable Universe Fuzzy–PID (VUF-PID) controller is faster than the multi-closed-loop Fuzzy–PID and cascaded three-close-loop PID controllers. In addition, the chattering of braking force can be suppressed by 17%. This EMB system may rapidly and automatically finish the operation of the overall braking process, including gap elimination, clamping force tracking and gap recovery, which can obviously enhance the precision of the longitudinal motion control of UETRVs. It can thus serve as a BBW actuator of mine autonomous driving electric vehicles, especially in the stage of braking control. Full article

The modulation of biological processes with light-sensitive chemical probes promises precise temporal and spatial control. Yet, the design and synthesis of suitable probes is a challenge for medicinal chemists. This article introduces a photocaging strategy designed to modulate the pharmacology of histamine H3 receptors (H3R) and H4 receptors (H4R). Employing the photoremovable group BODIPY as the caging entity for two agonist scaffolds—immepip and 4-methylhistamine—for H3R and H4R, respectively, we synthesized two BODIPY-caged compounds, 5 (VUF25657) and 6 (VUF25678), demonstrating 10–100-fold reduction in affinity for their respective receptors. Notably, the caged H3R agonist, VUF25657, exhibits approximately a 100-fold reduction in functional activity. The photo-uncaging of VUF25657 at 560 nm resulted in the release of immepip, thereby restoring binding affinity and potency in functional assays. This approach presents a promising method to achieve optical control of H3R receptor pharmacology. Full article

In this study, we propose the use of virtual urban field studies (VUFS) through context-based interface prototypes for evaluating the interaction design of auditory interfaces. Virtual field tests use mixed-reality technologies to combine the fidelity of real-world testing with the affordability and speed of testing in the lab. In this paper, we apply this concept to rapidly test sound designs for autonomous vehicle (AV)–pedestrian interaction with a high degree of realism and fidelity. We also propose the use of psychometrically validated measures of presence in validating the verisimilitude of VUFS. Using mixed qualitative and quantitative methods, we analysed users’ perceptions of presence in our VUFS prototype and the relationship to our prototype’s effectiveness. We also examined the use of higher-order ambisonic spatialised audio and its impact on presence. Our results provide insights into how VUFS can be designed to facilitate presence as well as design guidelines for how this can be leveraged. Full article

Clamping force control is one of the key technologies in the algorithm design and implementation of electro-mechanical braking system, whose control effects directly affect the vehicle braking performance and safety performance. In order to improve the clamping force control performance of electro-mechanical braking (EMB) system, an EMB clamping force control method based on Variable universe adaptive fuzzy PID (VUF-PID) controller is proposed, and stretching factors are added to the fuzzy PID control. According to the operation of the controlled object, the fuzzy theory domain can be adjusted in real time to keep the system in the proper parameter value and improve the adaptive ability of the system. The response characteristics and effectiveness of clamping force under step braking condition, gear switching braking condition and sine braking condition are verified by simulation experiments using MATLAB/Simulink. The results show that the proposed VUF-PID control method has strong tracking characteristics and stability characteristics, and meet the braking requirements under different braking conditions. In this study, a comprehensive computational and structural study was performed to investigate the affinity of our compound library consisting of 757 chalcone-based structures (CHA-1 to CHA-757) for inhibiting the 3CLpro and PLpro enzymes and against twelve selected host-based targets. Our results indicated that CHA-12 (VUF 4819) is the most potent and multi-target inhibitor in our chemical library over all viral and host-based targets. Correspondingly, CHA-384 and its congeners containing ureide moieties were found to be potent and selective 3CLpro inhibitors, and benzotriazole moiety in CHA-37 was found to be a main fragment for inhibiting the 3CLpro and PLpro. Surprisingly, our results indicate that the ureide and sulfonamide moieties are integral fragments for the optimum 3CLpro inhibition while occupying the S1 and S3 subsites, which is fully consistent with recent reports on the site-specific 3CLpro inhibitors. Finding the multi-target inhibitor CHA-12, previously reported as an LTD4 antagonist for the treatment of inflammatory pulmonary diseases, prompted us to suggest it as a concomitant agent for relieving respiratory symptoms and suppressing COVID-19 infection. This paper deals with voltage unbalances and how they can be quantified according to the standards. Firstly, a comparison between the different unbalance voltage factors is conducted in order to remark on their divergences. Secondly, according to the standard that better represents the phenomenon, i.e., EN 50160, a new methodology is proposed to quantify the voltage unbalance factor (VUF). In order to do so, it is recommended to measure the voltage unbalance in three-phase installations by means of a new smart sensor based on a single voltage sensor, which measures the direct-current (DC) voltage at the output of a three-phase diode bridge rectifier, while current methods make use of three voltage sensors (which can measure either phase-to-neutral voltages or phase-to-phase voltages). Furthermore, both simulation and experimental results have been carried out to validate the proposed methodology. Finally, a new voltage unbalance factor (and the corresponding methodology to obtain it from the measured DC voltage) is proposed. Full article

The discovery of the human histamine H4 receptor (H4R) has contributed to our understanding of the role of histamine in numerous physiological and pathological conditions, including tumor development and progression. The lymph nodes of patients with malignant lymphomas have shown to contain high levels of histamine, however, less is known regarding the expression and function of the H4R in T-cell lymphoma (TCL). In this work we demonstrate the expression of H4R isoforms (mRNA and protein) in three human aggressive TCL (OCI-Ly12, Karpas 299, and HuT78). Histamine and specific H4R agonists (VUF8430 and JNJ28610244) significantly reduced cell viability in a dose-dependent manner (p < 0.05). The combined treatment with the H4R antagonist (JNJ7777120, 10 µM) reversed the effects of the H4R ligands. Importantly, we screened a drug repurposing library of 433 FDA-approved compounds (1 μM) in combination with histamine (10 μM) in Hut78 cells. Histamine produced a favorable antitumor effect with 18 of these compounds, including the histone deacetylase inhibitor panobinostat. Apoptosis, proliferation, and oxidative stress studies confirmed the antitumoral effects of the combination. We conclude that the H4R is expressed in TCL, and it is involved in histamine-mediated responses. Full article

A3 adenosine receptor (A3AR) agonists have emerged as potent relievers of neuropathic pain by a T cell-mediated production of IL-10. The H4 histamine receptor (H4R), also implicated in pain modulation, is expressed on T cells playing a preeminent role in its activation and release of IL-10. To improve the therapeutic opportunities, this study aimed to verify the hypothesis of a possible cross-talk between A3AR and H4R in the resolution of neuropathic pain. In the mouse model of Chronic Constriction Injury (CCI), the acute intraperitoneal co-administration of the A3AR agonist IB-MECA (0.5 mg/kg) and the H4R agonist VUF 8430 (10 mg/kg), were additive in counteracting mechano-allodynia increasing IL-10 plasma levels. In H4R−/− mice, IB-MECA activity was reduced, lower pain relief and lower modulation of plasma IL-1β, TNF-α, IL-6 and IL-10 were shown. The complete anti-allodynia effect of IB-MECA in H4R−/− mice was restored after intravenous administration of CD4+ T cells obtained from naïve wild type mice. In conclusion, a role of the histaminergic system in the mechanism of A3AR-mediated neuropathic pain relief was suggested highlighting the driving force evoked by CD4+ T cells throughout IL-10 up-regulation.
