Ubiquitination Regulates Reorganization of the Membrane System During Cytomegalovirus Infection

Background: During infection with the cytomegalovirus (CMV), the membrane system of the infected cell is remodelled into a megastructure called the assembly compartment (AC). These extensive changes may involve the manipulation of the host cell proteome by targeting a pleiotropic function of the cell such as ubiquitination (Ub). In this study, we investigate whether the Ub system is required for the establishment and maintenance of the AC in murine CMV (MCMV)-infected cells Methods: NIH3T3 cells were infected with wild-type and recombinant MCMVs and the Ub system was inhibited with PYR-41. The expression of viral and host cell proteins was analyzed by Western blot. AC formation was monitored by immunofluorescence with confocal imaging and long-term live imaging as the dislocation of the Golgi and expansion of Rab10-positive tubular membranes (Rab10 TMs). A cell line with inducible expression of hemagglutinin (HA)-Ub was constructed to monitor ubiquitination. siRNA was used to deplete host cell factors. Infectious virion production was monitored using the plaque assay. Results: The Ub system is required for the establishment of the infection, progression of the replication cycle, viral gene expression and production of infectious virions. The Ub system also regulates the establishment and maintenance of the AC, including the expansion of Rab10 TMs. Increased ubiquitination of WASHC1, which is recruited to the machinery that drives the growth of Rab10 TMs, is consistent with Ub-dependent rheostatic control of membrane tubulation and the continued expansion of Rab10 TMs. Conclusions: The Ub system is intensively utilized at all stages of the MCMV replication cycle, including the reorganization of the membrane system into the AC. Disruption of rheostatic control of the membrane tubulation by ubiquitination and expansion of Rab10 TREs within the AC may contribute to the development of a sufficient amount of tubular membranes for virion envelopment. Time-Restricted Eating Improves Glycemic Control in Patients with Type 2 Diabetes: A Meta-Analysis and Systematic Review

Time-restricted eating (TRE), a dietary strategy that aligns food intake with circadian rhythms, has emerged as a promising non-pharmacological approach for improving glycemic control in patients with type 2 diabetes. This systematic review and meta-analysis evaluated the effects of TRE on glycemic outcomes by analyzing eight randomized controlled trials involving 312 participants with type 2 diabetes or impaired fasting glucose. Meta-analyses of six eligible studies demonstrated that TRE significantly reduced fasting glucose (mean difference [MD]: −0.74 mmol/L; 95% CI: −1.13 to −0.36) and glycated hemoglobin (ΔHbA1c) (MD: −0.11%; 95% CI: −0.15 to −0.07) and increased time in range (TIR) for blood glucose (MD: +10.51%; 95% CI: 6.81 to 14.21). Improvements in fasting glucose and HbA1c were modest but consistent, while the increase in TIR showed no between-study heterogeneity, suggesting a robust and reproducible benefit of TRE on glycemic stability. These findings support the clinical feasibility and effectiveness of TRE as a dietary intervention in diabetes management. However, further high-quality trials with standardized protocols and longer follow-up are needed to confirm long-term efficacy and inform guidelines. Full article

Intermittent fasting (IF) and caloric restriction (CR) have garnered attention for their potential to enhance cognitive function, particularly in aging and metabolically compromised populations. This narrative review critically examines whether the cognitive benefits of IF are attributable to its specific fasting patterns or are instead mediated by the production of weight loss, fat loss, and improvements in insulin sensitivity. Evidence from human trials suggests that reductions in body weight, especially visceral adiposity, and improvements in insulin resistance are key factors linked to enhanced cognitive performance. Comparisons between the results of IF and CR trials show comparable cognitive outcomes, supporting the idea that negative energy balance, rather than fasting or altered eating patterns, drive these effects. However, further investigation of specific types of IF patterns, as well as possible cognitive domains to be affected, may identify mechanisms through which IF can yield benefits on neurocognitive health beyond those of loss of body fat and its accompanying inflammatory state. Our research focused on the roost preferences of two bat species in a forest environment. Throughout the Anthropocene, people have heavily altered the landscape. Forested habitation has declined, with remaining forests becoming fragmented and often deprived of old trees that provide shelter opportunities for bats. TReMs (tree-related microhabitats) are essential for forest-dwelling bat species as they provide an opportunity to roost and shelter. Following an infestation of Ips typographus L., the Białowieża Forest is saturated with dead spruce trees. We investigated roost selection in two forest-dwelling species, Barbastella barbastellus and Pipistrellus pygmaeus. To examine similarities or differences in roost selection between species, we radio-tracked 24 barbastelles and 13 soprano pipistrelles over three breeding seasons. We located a total of 48 barbastelle roosts and 15 pipistrelle roosts, together with the characteristics of the surroundings. We found that barbastelles select roosts almost exclusively in dead spruce trees (43/48), while pipistrelles selected roosts predominantly in live (n = 8) and dead (n = 5) broadleaved trees. Our results show that both bat species have clear differences in roost tree preference. In our study area, with an abundance of exfoliating bark, barbastelles showed a preference for roosting under flaking bark despite the availability of crevices within broadleaved trees. Our findings provide useful insight into forestry practices, highlighting the importance of standing dead trees. Full article

Background: The gut microbiota is increasingly recognized as a key modulator in obesity management, influencing host energy balance, lipid metabolism, and inflammatory pathways. With obesity prevalence continuing to rise globally, dietary interventions that promote beneficial microbial shifts are essential for enhancing weight loss outcomes and long-term health. Objective: This study investigated the effects of the multicomponent Weight Loss Maintenance 3 Phases Program (WLM3P), which integrates caloric restriction, a high-protein low-carbohydrate diet, time-restricted eating (10h TRE), dietary supplementation (prebiotics and phytochemicals), and digital app-based support on gut microbiota composition compared to a standard low-carbohydrate diet (LCD) in adults with obesity. The analysis focused exclusively on the 6-month weight loss period corresponding to Phases 1 and 2 of the WLM3P intervention. Methods: In this sub-analysis of a randomized controlled trial (ClinicalTrials.gov Identifier: NCT04192357), 58 adults with obesity (BMI 30.0–39.9 kg/m2) were randomized to the WLM3P (n = 29) or LCD (n = 29) groups. Stool samples were collected at baseline and 6 months for 16S rRNA sequencing. Alpha and beta diversity were assessed, and genus-level differential abundance was determined using EdgeR and LEfSe. Associations between microbial taxa and clinical outcomes were evaluated using regression models. Results: After 6-month, the WLM3P group showed a significant increase in alpha diversity (p = 0.03) and a significant change in beta diversity (p < 0.01), while no significant changes were observed in the LCD group. Differential abundance analysis revealed specific microbial signatures in WLM3P participants, including increased levels of Faecalibacterium. Notably, higher Faecalibacterium abundance was associated with greater reductions in fat mass (kg, %) and visceral adiposity (cm2) in the WLM3P group compared to LCD (p < 0.01). Conclusions: These findings suggest a potential microbiota-mediated mechanism in weight loss, where Faecalibacterium may enhance fat reduction effectiveness in the context of the WLM3P intervention. The high reproductive performance of Nilaparvata lugens (Stål) (Hemiptera: Delphacidae) is one of the key factors contributing to its serious damage. Studies have demonstrated that trehalose hydrolysis, catalyzed by trehalase (TRE), plays an important role in the reproductive regulation of N. lugens. However, it remains unclear how TRE regulates the reproduction of N. lugens. To address this question, the current study was designed to investigate the effects of TRE on the reproductive regulatory network of N. lugens. Specifically, RNA interference (RNAi) was conducted by injecting double-stranded RNA (dsRNA) targeting the TRE genes, and a trehalase inhibitor, validamycin, was used to suppress TRE activity. Subsequently, the relative expression levels of key genes involved in the reproductive regulatory pathway, as well as the triglyceride content in the fat body and ovary, were determined. The results indicated that both dsTREs and validamycin treatment decreased the relative expression of Vitellogenin (Vg) without affecting the expression of its receptor, VgR. However, their impact on the reproductive regulatory network differed: dsTREs injection did not affect the relative expression of JHAMT and Met in the juvenile hormone signaling pathway, but resulted in the upregulation of USP and TOR, and inhibition of InR2 and S6K. In contrast, validamycin injection inhibited the expression of Met, USP, EcR, InR1, TOR, and S6K. Both dsTREs and validamycin inhibited the expression of Fas, but only validamycin decreased the ovarian triglyceride content. These findings suggest that the inhibition of TRE leads to alterations in reproductive regulatory signals, thereby reducing the reproductive capacity of N. lugens. Full article

To address the challenges of weak fault features and strong non-stationarity in early-stage vibration signals, this study proposes a novel fault diagnosis method combining enhanced variational mode decomposition (VMD) with a structurally improved GoogLeNet. Specifically, an improved wild horse optimizer (IWHO) with tent chaotic mapping is employed to automatically optimize critical VMD parameters, including the number of modes K and the penalty factor α, enabling precise decomposition of non-stationary signals to extract weak fault features. The vibration signal is decomposed, and the top five intrinsic mode functions (IMFs) are selected based on the kurtosis criterion. Time–frequency features are then extracted from these IMFs and input into a modified GoogLeNet classifier. The GoogLeNet structure is improved by replacing standard n × n convolution kernels with cascaded 1 × n and n × 1 kernels, and by substituting the ReLU activation function with a parameterized TReLU function to enhance adaptability and convergence. Experimental results on two public rolling bearing datasets demonstrate that the proposed method effectively handles non-stationary signals, achieving 99.17% accuracy across four fault types and maintaining over 95.80% accuracy under noisy conditions. The common oak (Quercus robur L.), though ecologically important and long-lived, has declined in Northern Europe due to historical land use and conifer-dominated forestry. In Latvia, where its distribution is limited, oaks support a rich biodiversity through features like tree-related microhabitats (TreMs) and diverse epiphytic communities. This study compared TreM and epiphyte diversity between planted mature oaks and relict champion oak trees across 16 forest stands. Epiphyte species were recorded using fixed-area frames on tree trunks, and TreMs were categorized following a hierarchical typology. Champion trees hosted significantly more TreMs and a greater variety, including 10 unique TreMs. While overall epiphyte diversity indices did not differ significantly, champion trees supported more specialist and woodland key habitat indicator species. The findings underscore the ecological value of legacy trees, which provide complex habitats essential for specialist taxa and indicators of forest continuity. Conserving such trees is vital for maintaining forest biodiversity and supporting ecosystem resilience in managed landscapes. Full article

Background: Early time-restricted eating combined with energy restriction (eTRE + ER) has been shown to reduce fat mass, diastolic blood pressure (DBP) and fasting glucose more effectively than late TRE with energy restriction (lTRE + ER) or energy restriction (ER) alone. Given the gut microbiome’s sensitivity to circadian rhythms, we examined whether adding TRE, particularly eTRE, to ER alters gut microbiota composition beyond ER alone, and whether such effects persist during follow-up. Methods: We analysed anthropometric, biochemical and gut microbiome data from 76 participants at baseline and after a 3-month intervention (eTRE + ER: n = 33; lTRE + ER: n = 23; ER: n = 20). Follow-up microbiome data 6-months after the end of intervention were available for 43 participants. Gut microbiota composition was assessed via 16S rRNA gene sequencing of stool samples. Results: No significant between-group differences in beta diversity were observed over time. However, changes in alpha diversity differed significantly across groups at the end of the intervention (Shannon: F = 5.72, p < 0.001; Simpson: F = 6.72, p < 0.001; Richness: F = 3.99, p = 0.01) and at follow-up (Richness: F = 3.77, p = 0.02). lTRE + ER led to the greatest reductions in diversity post intervention, while ER was least favourable during follow-up. Although no significant between-group differences were observed at the phylum level either at the end of the intervention or during follow-up, only the eTRE + ER group exhibited a significant decrease in Bacillota and an increase in Bacteroidota during follow-up. At the genus level, differential abundance analysis revealed significant shifts in taxa such as Faecalibacterium, Subdoligranulum, and other genera within the Ruminococcaceae and Oscillospiraceae families. In the eTRE + ER, Faecalibacterium and Subdoligranulum increased, while in other groups decreased. Notably, the changes in Faecalibacterium were negatively correlated with fasting glucose, while the increase in Subdoligranulum was inversely associated with DBP; however, both associations were weak in strength. Conclusions: eTRE + ER may promote beneficial, lasting shifts in the gut microbiome associated with improved metabolic outcomes. These results support further research into personalized TRE strategies for treatment of obesity. Full article

Fruit rot seriously damages pomelo production. Given concerns regarding the safety of chemical agents, biological alternatives are becoming more preferable. Therefore, the experiment aimed to (i) identify the pathogens causing pomelo fruit rot disease and (ii) select Trichoderma spp. strains controlling the determined pathogens in Ben Tre, Vietnam. Three pathogenic fungal strains isolated from diseased pomelo fruits were selected. The three pathogenic fungal strains were randomly injected into 9 healthy pomelo fruits. The strain PCP-B02-A2 led to a completely rotten fruit on day 17 after infection, while strains PCP-B02-B2 and PCP-B03-A1 had infected spots whose lengths were 17.5 and 28.1 mm, became larger, and eventually led to the whole fruit rot. The pathogens were identified by the internal transcribed spacer (ITS) technique as Colletotrichum gloeosporioides PCP-B02-A2, Pseudopestalotiopsis camelliae sinensis PCP-B03-A1, and P. chinensis PCP-B02-B2. Twenty-five Trichoderma spp. strains were isolated. The ITS technique identified four strains, including Trichoderma asperellum TP-B01, T. harzianum TP-B08, T. harzianum TP-B09, and T. asperellum TP-C25. The PCP-B02-A2 strain had antagonism at 66.7–68.7%, while those of PCP-B02-B2 and PCP-B03-A1 were 64.2–71.1% and 55.7–57.4%, respectively. Full article

This study aims to (1) analyze thermal comfort at outdoor sport events held outside of fixed venues or locations; (2) establish a method for evaluating environmental thermal comfort for large-scale, long-term outdoor activities; and (3) provide suggestions for the arrangement of shifts in routes and participants for heat warning and mitigation. Taiwan ReAnalysis Downscaling (TReAD) data, Sky View Factors (SVFs), GSV2SVF tool, and RayMan Pro were applied to analyze and evaluate thermal comfort at the 2021 Torch Relay Round the Island, Taiwan. In this study, modified Physiologically Equivalent Temperature (mPET), Wet Bulb Globe Temperature (WBGT), and Universal Thermal Climate Index (UTCI) were estimated and selected as thermal indicators for the purpose of obtaining a more comprehensive perspective. We also define and present thermal performance with a simple traffic light symbol (green: comfortable/yellow: warm/red: hot) and try to go beyond the concept of heat and visualize it in an easy-to-understand way. Mitochondrial DNA (mtDNA) damage in trabecular meshwork (TM) cells occurs in open-angle glaucoma (OAG). However, current in vitro models for OAG-like changes in TM cells do not explicitly incorporate mtDNA damage. This work validated two methods of mtDNA damage in immortalized TM cells and assessed OAG-associated expression changes. mtDNA was depleted in TM-1 cells via both ethidium bromide (EtBr) treatment and doxycycline (Dox) induction of a mutant (Y147A) version of Uracil DNA Glycosylase 1 (UNG1) in TM-1 cells (TM-1rtTAadv-TRE-UNG1Y147A). Levels of mitochondrial proteins (ATP5F1A, COXII, and COXIV) were measured via western blot. mtDNA levels and mRNA for OAG-associated transcripts (CTGF, FN1, PAI1, and SFRP1) were measured by qPCR. There was a statistically significant decrease in mtDNA levels per cell at all treatment times in both EtBr-treated TM-1 cells and induced TM-1rtTAadv-TRE-UNG1Y147A cells. Protein levels of ATP5F1A were not significantly changed; COXII and COXIV showed significant decreases after both EtBr and Dox induction. Both models resulted in upregulation of CTGF, FN1, and PAI1; additionally, EtBr treatment but not Dox induction resulted in SFRP1 upregulation. In conclusion, two models of mitochondrial depletion were demonstrated in immortalized TM cells; damage was associated with increases in OAG-associated transcripts, supporting a link between mitochondrial damage and glaucoma phenotypes. Primordial Magnetogenesis from Killing Vector Fields

Papapetrou showed that the covariant derivative of a Killing vector field satisfies Maxwell’s equations in vacuum. Papapetrou’s result is extended, in this article, and it is shown that the covariant derivative of a Killing vector field satisfies Maxwell’s equations in non-vacuum backgrounds as well if one allows electromagnetic currents of purely geometric origin. It is then postulated that every Killing vector field gives rise to a physical electromagnetic field and, in a non-vacuum background, a physical electromagnetic current—hereafter called Killing electromagnetic field and Killing electromagnetic current, respectively. It is shown that the Killing electromagnetic field of the flat FLRW (Friedmann–Lemai^tre–Robertson–Walker) universe comprises a Killing magnetic field and a rotational Killing electric field; an upper bound on the Killing magnetic field is derived, and it is found that the upper bound is consistent with the current observational bounds on the cosmic magnetic field. Next, the time-like Killing vector of the Schwarzschild spacetime is shown to give rise to a radial Killing electric field. It is also shown that in the weak field regime—and far from the matter distribution—the back reaction of the radial Killing electric field changes the Schwarzschild metric to the Reissner–Nordström metric, establishing a partial converse of Wald’s result. Drawing upon Rainich’s work on Rainich–Riemann manifolds, the etiological question of how a physical electromagnetic field can arise out of geometry is discussed; it is also argued that detection of the Killing electric field of flat FLRW spacetime may be within the current experimental reach. Finally, this article discusses the relevance of Killing electromagnetic currents and the aforementioned transmutation of Schwarzschild spacetime to Reissner–Nordstrom spacetime, to Misner and Wheeler’s program of realizing “charge without charge”. Debonding, especially in plastic materials, refers to the separation occurring at the interface within a bonded structure composed of two or more polymeric layers. Due to the great heterogeneity of materials and layering configurations, highly specialized expertise is often required to detect the presence and extent of such defects. This study presents a novel approach that leverages transfer learning techniques to improve the detection of debonding defects across different surface types using PICUS, an acoustic diagnostic device developed at Roma Tre University for the assessment of defects in heritage wall paintings. Our method leverages a pre-trained deep learning model, adapting it to new material conditions. We designed a planar test object embedded with controlled subsurface cavities to simulate the presence of defects of adhesion and air among the layers. This was rigorously evaluated using non-destructive testing using PICUS, augmented by artificial intelligence (AI). A convolutional neural network (CNN), initially trained on this mock-up, was then fine-tuned via transfer learning on a second test object with distinct geometry and material characteristics. This strategic adaptation to varying physical and acoustic properties led to a significant improvement in classification precision of defect class, from 88% to 95%, demonstrating the effectiveness of transfer learning for robust cross-domain defect detection in challenging diagnostic applications. Comparison of Rectal and Gastrointestinal Core Temperatures During Heat Tolerance Testing

Background and Objectives: Military capability may be reduced in hot environments with individuals at risk of exertional heat stroke (EHS). Heat tolerance testing (HTT) can be used to indicate readiness to return to duty following EHS. HTT traditionally relies on rectal core temperature (Tre) assessment via a rectal probe. This study investigated the use of gastrointestinal core temperature (Tgi) as an alternative to Tre during HTT. A secondary aim was to compare physiological factors between heat-tolerant and heat-intolerant trials. Materials and Methods: Australian Defence Force personnel undergoing HTT following known or suspected heat stroke volunteered (n = 23 cases participating in 26 trials) along with 14 controls with no known heat illness history. Confusion matrices enabled comparison of HTT outcome based on Tgi and Tre. The validity of Tgi compared to Tre during HTT was assessed using correlation and bias. Comparisons between heat-tolerant and intolerant trials were performed using non-parametric tests. Results: Although Tgi correlated closely with Tre (Spearman’s rank correlation ρ = 0.893; median bias 0.2 °C) there was no consistent pattern in the differences between measures. Importantly, the two measures only agreed on heat tolerance outcome in 80% of trials with Tgi failing to detect heat intolerance identified by Tre in 6 of 8 trials. If Tgi was relied upon for diagnostic outcome, return to duty may occur before full recovery. None of the assessed covariates were related to the difference between Tre and Tgi. In addition, resting heart rate and systolic blood pressure were significantly lower and body surface area to mass ratio significantly higher in heat-tolerant compared to intolerant trials. Conclusions: It is not recommended to rely on Tgi instead of Tre during HTT. Resting heart rate and systolic blood pressure findings point to the importance of aerobic exercise in conveying heat tolerance along with body composition. Full article
