Roads impact medium- and large-sized mammal populations through both collisions and barrier effects. This study examined how landscape characteristics influence roadkill occurrences along the Dom Pedro I highway (SP-065), located in the Cantareira-Mantiqueira Corridor, São Paulo State, Brazil. The SP-065 crosses important remnants of the Brazilian Atlantic Forest, a global hotspot for biodiversity. Roadkill records were obtained from the Environmental Company of the State, and land use data were extracted from the MapBiomas platform. We analyzed seven landscape variables (in percentage): native forest, pasture, agriculture, forestry, urban areas, mosaic of uses, and water bodies, considering multiple spatial scales. Mammal species were grouped functionally by home range size and tolerance to anthropogenic environments. In total, 1418 roadkills of 24 species were recorded, including eight threatened species. Capybaras (Hydrochoerus hydrochaeris) were the most frequently killed species. Generalized linear models showed that, for Group G1 (small home range, common in human-modified areas), roadkills were positively associated with native forest and pasture, and negatively with mosaic landscapes. For Group G3 (large home range, tolerant to anthropogenic areas), agriculture had a positive effect, especially within a 3000 m radius. For Group G5 (capybara), roadkills increased with pasture and agriculture, while mosaic uses had a negative effect. Since pasture and agriculture were frequently linked to higher roadkill rates, environmental impact assessments should consider these land-use types when planning mitigation actions. Ultimately, responsibility for roadkill extends beyond highway managers to rural landowners and local governments, as land-use patterns around roads strongly influence mammal movement and mortality. In Brazil, the presence of capybaras (Hydrochoerus hydrochaeris) in urban environments has raised concerns due to ecological, sanitary and coexistence challenges. This study investigates the growing presence of capybaras on Santa Catarina Island, Brazil, exploring their integration into urban areas. We interviewed 1505 residents of the Island to understand capybara distribution, public perceptions and associated risks. Beliefs about management strategies were also assessed. The findings reveal that capybaras are present in all neighborhoods of Florianópolis, the sole city in the Island. Participants expressed positive views of capybaras, describing them as “beautiful” and “cute” and showing concerns for their well-being in urban environments. Despite these positive perceptions, participants demonstrated limited knowledge about capybara biology (88%) and potential problems, such as disease transmission (37%), traffic accidents (10%), pet conflicts (56%) and garden damage (42%). Most respondents agreed with the statements: “Capybaras are part of nature and should be left undisturbed, even in urban areas” (70%) and “It is important for people to see capybaras in the city, as long as their population is not causing problems” (82%). After risk awareness, more participants rated these issues as highly relevant: disease transmission (97%), destruction of urban gardens (77%), pet incidents (92%) and traffic accidents (98%). This led to greater acceptance of population control, though elimination remained unpopular. Sociodemographic factors influenced opinions. Addressing knowledge gaps can foster ethical, supported management strategies. This case places Florianópolis within a broader South American phenomenon of urban capybara conflicts, highlighting the global challenge of managing human–wildlife interactions in cities. Tick-borne piroplasmids are apicomplexan protozoa that infect a wide range of vertebrate hosts, with significant implications for animal and human health. This study investigated the occurrence and genetic diversity of piroplasmids in wild mammals, unconventional pets, and associated ticks in Goiás state, midwestern Brazil. Between April 2023 and January 2024, 105 blood samples, 22 tissue samples, and 300 ticks were collected from 21 mammalian species housed in wildlife screening centers, zoos, and veterinary clinics. Molecular screening targeting the 18S rRNA gene of piroplasmids detected a 25.7% (27/105) overall positivity, with gray brockets (Subulo gouazoubira) and South American tapirs (Tapirus terrestris) showing the highest infection rates. Three tick samples tested positive, including two Amblyomma sculptum nymphs and a male of Amblyomma dubitatum collected from a tapir and capybara (Hydrochoerus hydrochaeris). Cytauxzoon brasiliensis was reported, for the first time, in cougars (Puma concolor) from Goiás state, midwestern Brazil, indicating the role of this feline as a host of this parasite. Babesia goianiaensis was confirmed in a capybara, and Theileria terrestris in tapirs. Phylogenetic analyses clustered gray brockets-associated Theileria sequences with Theileria sp. previously detected in Neotropical deer from Brazil and Theileria cervi. While the phylogenetic analysis of amino acid sequences of the cytochrome c oxidase subunit III separated Theileria genotypes detected in S. gouazoubira from T. cervi, hsp70-based phylogenetic inferences clustered the genotypes detected in Tapirus terrestris with Theileria terrestris, suggesting host-specific evolutionary lineages. These findings contribute to the understanding of Piroplasmida diversity and circulation in South American wild mammals, emphasizing the need for enhanced molecular surveillance to elucidate transmission dynamics, assess potential health risks, and contribute to the establishment of wildlife conservation and One Health strategies. Along with exposure to parasites and other biological disease vectors, animal faeces can also contain heavy metals and metalloids. We quantified metals, metalloids, and non-metals in the faeces of capybara (Hydrochoerus hydrochaeris) that live in parks in the city of Campo Grande (Brazil). Quantification of metalloids was obtained after acid digestion using an inductively coupled plasma optical emission spectrometer. Higher mean concentrations in mg/kg of aluminium (Al) (140.322), arsenic (As) (0.010), cadmium (Cd) (1.042), chromium (Cr) (26.866), cobalt (Co) (1.946), copper (Cu) (50.764), lead (Pb) (8.762), manganese (Mn) (291.469), molybdenum (Mo) (3.634), nickel (Ni) (5.475), and zinc (Zn) (100.027) were quantified in samples of faeces of capybara that live on the banks of a lagoon that receives input from streams that cross the city. According to the risk assessment, potential risks to the health of children and adults may occur due to the presence of Al, As, Cd, Co, Cu, and Mn through involuntary oral ingestion of faeces, via inhalation and dermal contact. The hazard index (HI) due to oral ingestion was greater than 1 for children and adults. Therefore, we believe that faeces of H. hydrochaeris can be considered as a bioindicator of environmental pollution in urban parks. Full article

This study explored the role of the hydrological regime as a trigger factor for wildlife roadkill along a 22 km road crossing the Taim Wetland, a Ramsar site of international importance in South Brazil. The north–south crossing of BR-471, a federal highway, causes fatalities to numerous animals due to collisions with vehicles. An 11-year long-term time series study encompassing monthly roadkill incidents, water level, and rainfall was analyzed by considering three different periods related to a Wildlife Protection System (WPS): (i) 3 initial years before the WPS implementation (BWPS); (ii) 4 intermediate years after the WPS implementation (AWPS), which includes fences, fauna tunnels, cattle guard stocks, bumps, and speed control; (iii) 4 final years during partial destruction of the WPS (PDWPS). A pseudo-2D full hydrodynamic cell model was employed to fill missing water level data. The model had a good to very good performance (NSE: 0.73 to 0.87; R2: 0.79 to 0.90). The relationship between roadkill incidents and the WPS periods (BWPS, AWPS, and PDWPS) was modeled using Generalized Additive Models for Location, Scale, and Shape (GAMLSS), considering rainfall and water level as predictor variables. The analysis revealed a higher incidence of wildlife roadkill in BWPS compared to AWPS and PDWPS, suggesting the effectiveness of the implemented measures. Critical season and interplay between water levels, rainfall, and the roadkill were assessed. Mammals was the most common roadkill class identified (~92%), followed by reptiles (13%) and birds (2%), with no change in these percentual in the BWPS, AWPS, and PDWPS. Among mammals, capybara (Hydrochoerus hydrochaeris) and coypu (Myocastor coypu) were the most frequent victims (~93% of mammals). Winter, followed by autumn, recorded the highest number of roadkill incidents (>60%), and this pattern remained consistent during the three periods. While rainfall did not emerge as a determining variable for roadkill, water levels above certain thresholds (>3.3 m) drastically diminished the effectiveness of the WPS, mainly due to fauna tunnel submersion. These findings offer valuable insights for enhancing wildlife conservation strategies in this protected area by incorporating hydrological information providing a baseline for designing WPS in similar environments. Piroplasmids (order Piroplasmida) are a diverse group of tick-borne protozoa that may cause disease in animals and occasionally in humans. Novel Piroplasmida clades and species have been found in wild animals from Brazil based on the phylogenetic assessment of near-complete 18S rRNA, mitochondrial and heat-shock protein genes. For instance, a putative novel Babesia species has been detected in capybaras and Amblyomma ticks in three Brazilian states. The present work aimed to describe, using phylogenetic assessments based on distinct molecular markers, this novel Babesia species in capybaras and associated ticks (Amblyomma sculptum and Amblyomma dubitatum) sampled in Goiânia city, Goiás state, midwestern Brazil. While the phylogenetic analysis based on both near-complete 18S rRNA and hsp-70 genes positioned the sequences obtained from capybara blood samples into a new clade sister to the Babesia sensu stricto clade, the phylogenetic inference based on the COX-3 amino acid positioned the obtained sequences from capybara blood samples and A. sculptum ticks also into a clade sister to the Theileria sensu stricto clade, highlighting the inappropriateness of this marker inferring evolutionary relationships among piroplasmids. Pairwise distance analysis demonstrated that the divergence rates between the 18S rRNA sequences detected in capybaras and other Piroplasmida already described were very high and ranged from 9.4 to 12.9%. Genotype analysis based on the near-full 18S rRNA sequences of the Piroplasmida detected in capybaras and associated ticks demonstrated the occurrence of high genotype diversity at an intra-species level. In conclusion, phylogenetic analyses based on distinct molecular markers supported the description of Babesia goianiaensis nov. sp. in capybaras and associated Amblyomma ticks. Additionally, a novel phylogenetic clade, apart from the previously described ones, was described in the present study and contributed to untangling the complex evolutionary history of the Piroplasmida. Brazilian spotted fever (BSF) is the most important tick-borne diseases affecting humans in Brazil. Cases of BSF have recently been reported in the Goiás state, midwestern Brazil. All cases have been confirmed by reference laboratories by seroconversion to Rickettsia rickettsii antigens. Because serological cross-reactions among different rickettsial species that belong to the spotted fever group (SFG) are common, the agent responsible for BSF cases in Goiás remains unknown. From March 2020 to April 2022, ticks and plasma were collected from dogs, horses and capybaras (Hydrochoerus hydrochaeris), and from the vegetation in an area where BSF cases have been reported and two areas under epidemiological surveillance in Goiás. Horses were infested by Amblyomma sculptum, Dermacentor nitens and Rhipicephalus microplus; dogs by Rhipicephalus sanguineus sensu lato (s.l.), Amblyomma ovale and A. sculptum, and capybaras by A. sculptum and Amblyomma dubitatum. Adults of A. sculptum, A. dubitatum, Amblyomma rotundatum and immature stages of A. sculptum and A. dubitatum, and Amblyomma spp. were collected from the vegetation. DNA of Rickettsia that did not belong to the SFG was detected in A. dubitatum, which was identified by DNA sequencing as Rickettsia bellii. Seroreactivity to SFG and Rickettsia bellii antigens was detected in 25.4% (42/165) of dogs, 22.7% (10/44) of horses and 41.2% (7/17) of capybaras, with higher titers for R. bellii in dogs and capybaras. The seropositivity of animals to SFG Rickettsia spp. antigens demonstrates the circulation of SFG rickettsiae in the region. Further research is needed to fully determine the agent responsible for rickettsiosis cases in this area. Full article

It is important to examine the animal welfare implications of all aspects of zoo operations, including out-of-hours public events. Research to date has indicated variable responses across species and event types. The current research aimed to understand and quantify the impact of a Christmas lights event. Four species: Rothschild giraffe (Giraffa camelopardalis rothschildi; n = 2) in one exhibit and capybara (Hydrochoerus hydrochaeris; n = 4), lowland tapir (Tapirus terrestris; n = 3) and vicuña (Lama vicugna; n = 5) in a mixed species exhibit were observed. Data were collected from 16:00–20:00 between 28 October 2021 and 11 January 2022. The event ran from mid-November to the end of December 2021. Five-minute behavioural observations were undertaken once per hour using instantaneous scan sampling with a one-minute inter-scan interval. A further six days of 12 h observations were conducted to enable a more detailed investigation post-event. Data collected were compared on non-event and event days using Mann–Whitney U tests (event vs. non-event) and Kruskal–Wallis tests (pre-event, event, post-event periods). Kruskal–Wallis tests and one-way ANOVAs were undertaken to compare behaviours during three time periods (12:00–16:00, 16:00–20:00, 20:00–00:00) over 12 h. Mixed behavioural responses were seen across the study species. Capybara spent more time in their house from 16:00–20:00 on event nights compared to non-event nights (p < 0.001) and tapir only engaged in vigilant behaviour from 16:00–20:00 when the event was held, (p = 0.044). There were no differences in frequency of behaviour between pre-event, event, and post-event observation periods, with the exception of capybara, who spent more time OOS in the pre-event period than during (p < 0.001) or after the event (p < 0.001). The results of the project, undertaken as part of an evidence-based management programme, highlighted that the event did not have any overtly negative impacts on the individuals studied. Except for the giraffe, all individuals had free access to inside and outside environments, and it is believed this choice enabled animals to be active in managing their response to the event. It is recommended that future work observe animals over 24 h to understand whether events lead to behavioural changes the day after events or if animals reverted to normal activity once the event ended. Capybaras (Hydrochoerus hydrochaeris) are the largest rodents on Earth. While capybaras are hosts for various tick species, there is limited information regarding the tick-borne pathogens they can carry. We investigated the presence of piroplasmids and Ehrlichia spp. in capybaras and their associated ticks in two peri-urban areas in Goiás state, central-western Brazil. Blood samples collected from 23 capybaras were used to investigate the presence of piroplasmids and Ehrlichia spp. in stained-blood smears and by PCR. Ticks collected from the capybaras were identified morphologically and also tested using PCR for the same pathogens. A total of 955 ticks were collected, including 822 (86.1%) Amblyomma sculptum, 132 (13.8%) Amblyomma dubitatum, and one (0.1%) unidentified larva of Amblyomma sp. Neither the capybaras nor ticks were positive for Ehrlichia spp. However, a stained-blood smear examination revealed the presence of ring-stage and pyriform-shaped merozoites in the erythrocytes of one (4.4%) capybara. In the same way, 47.8% (11/23) and 19.9% (36/181) of blood samples and ticks, respectively, were positive for piroplasmids in the PCR. We successfully sequenced a partial 18S rRNA gene fragment of four samples (two capybaras, one A. sculptum, and one A. dubitatum), and the phylogenetic reconstruction disclosed that the organism reported in the present study clusters within the genus Babesia. Further research is required for a formal delineation of this species (designated as Babesia sp. strain Capybara) and to investigate the hypothesis of A. dubitatum and A. sculptum ticks being vectors. This study aims to compare the morphological features and chemical composition of dental enamel of extinct and extant terrestrial vertebrates of Crocodylia: Purussaurus sp. (extinct) and Melanosuchus niger (extant), and Rodentia: Neoepiblema sp. (extinct) and Hydrochoerus hydrochaeris (extant). To obtain structural and chemical data, superficial and internal enamel were analyzed by Scanning Electron Microscopy (SEM) and Energy Dispersive Spectroscopy (SEM-EDS). Organic, mineral, and water content were obtained using polarizing microscopy and microradiography on ground sections of four teeth, resulting in a higher organic volume than previously expected (up to 49%). It is observed that both modern and fossil tooth enamel exhibit the same major constituents: 36.7% Ca, 17.2% P, and 41% O, characteristic of hydroxyapatite. Additionally, 27 other elements were measured from superficial enamel by inductively coupled mass spectrometry (ICP-MS). Zinc was the most abundant microelement detected, followed by Pb, Fe, Mg, and Al. Morphological features observed include enamel rods in the rodent teeth, while incremental lines and semiprismatic enamel were observed in the alligator species. The fossil enamel was in an excellent state for microscopic analyses. Results show that all major dental enamel’s physical, chemical, and morphological features are present both in extant and extinct fossil tooth enamel (>8.5 Ma) in both taxa. Full article

This is the second of two literature reviews that focuses on the female reproductive anatomy and reproductive technologies used in selected neo-tropical hystricomorphic rodents. The rodents chosen included the agouti (Dasyprocta leporina), the capybara (Hydrochoerus hydrochaeris), and the paca (Cuniculus paca). Over seventy references were used, spanning the years 1919–2021. Knowledge of the reproductive tract is important in understanding any unique features, which may affect the use of reproductive technologies. Some unique characteristics common to these species included the presence of a vaginal closure membrane and a lobulated placenta with a vascular sub-placenta. The capybara had hyperpigmentation of the vagina that was unique to each individual, while the agouti and paca had accessory corpora lutea, in addition to the main one. Some reproductive technologies have been used, with attempts at estrous induction and synchronization taking place within the past five years. Even though most work has been done over the past twenty years, there is still a dearth of information. Full article

This review, which is the first of two, focuses on the male reproductive anatomy and reproductive technologies used in Neo-tropical hystricomorphic rodents with the potential for domestication, which are the agouti (Dasyprocta leporina), the capybara (Hydrochoerus hydrochaeris) and the paca (Cuniculus paca). We consider over seventy references spanning from 1965 to 2020, with the majority of work being done in the past twenty years. Knowledge of the reproductive tract and reproductive technologies is critical to the conservation and preservation of these species. Although all three animals had similarities in their anatomy, such as no overt scrotums and testes located intra-abdominally in the inguinal region, some had unique features—for example, the agouti and the paca had penile spines, and two lateral penile cartilages. High spermatogenic efficiency was noted in the agouti and the paca, making them good candidates for increasing their reproductive performance in conservation programs. A review of the literature has shown that there is increasing work taking place on the reproductive technologies used in these animals; however, a lot of work is still lacking, as, to the author’s knowledge, standard protocols and artificial insemination procedures are yet to be established. Capybaras (Hydrochoerus hydrochaeris) are affected by a wide range of protozoan and metazoan-derived parasitic diseases. Among parasites of free-ranging capybaras are soil-, water-, food- and gastropod-borne parasitosis, today considered as opportunistic infections in semiaquatic ecosystems. The overlapping of the capybara’s natural ecological habitats with human and domestic animal activities has unfortunately increased in recent decades, thereby enhancing possible cross- or spillover events of zoonotic parasites. Due to this, three synanthropic wild capybara populations in the Orinoco Basin were studied for the occurrence of gastrointestinal parasite infections. A total of forty-six fecal samples were collected from free-ranging capybaras in close proximity to livestock farms. Macroscopical analyses, standard copromicroscopical techniques, coproELISA, PCR, and phylogenetic analysis revealed thirteen parasite taxa. In detail, the study indicates stages of five protozoans, four nematodes, one cestode, and three trematodes. Two zoonotic parasites were identified (i.e., Plagorchis muris, and Neobalantidium coli). The trematode P. muris represents the first report within South America. In addition, this report expands the geographical distribution range of echinocoelosis (Echinocoleus hydrochoeri). Overall, parasitological findings include two new host records (i.e., P. muris, and Entamoeba). The present findings collectively constitute baseline data for future monitoring of wildlife-derived anthropozoonotic parasites and call for future research on the health and the ecological impact of this largest semiaquatic rodent closely linked to humans, domestic and wild animals. The capybara, Hydrochoerus hydrochaeris, is the largest extant rodent of the world. To better understand the correlation between size and body mass, and biochemical parameters of capybaras from areas with different degrees of anthropization (i.e., different food supplies), we sampled free-ranging capybaras from areas of natural landscapes (NLs) and human-modified landscapes (HMLs) in Brazil. Analyses of biometrical and biochemical parameters of capybaras showed that animals from HMLs were heavier (higher body mass) than those from NL, a condition possibly related to fat deposit rather than body length, as indicated by Body Condition Index (BCI) analyses. Biochemical parameters indicated higher serum levels of albumin, creatine kinase, cholesterol, fructosamine and total protein among capybaras from HMLs than from NLs; however, when all adult capybaras were analyzed together only cholesterol and triglycerides were positively correlated with body mass. We propose that the biochemical profile differences between HMLs and NLs are related to the obesity condition of capybaras among HMLs. Considering that heavier animals might live longer and reproduce more often, our results could have important implications in the population dynamics of capybaras among HMLs, where this rodent species is frequently represented by overgrowth populations that generate several levels of conflicts with human beings. Trichuriasis is the clinical disease of animals infected with the parasite of the genus Trichuris. This review attempts to present information on Trichuris spp. infestation in neo-tropical rodents that are utilized for meat consumption by humans. Neo-tropical rodents utilized for meat production can be divided into two categories: those that have been domesticated, which include the guinea pig (Cavia porcellus), and those that are on the verge of domestication, such as the capybara (Hydrochoerus hydrochaeris), lappe (Cuniculus paca/Agouti paca), and agouti (Dasyprocta leporina). This document reviews the literature on the species of Trichuris that affects the rodents mentioned above, as well as the clinical signs observed. The literature obtained spans over sixty years, from 1951 to 2020. Trichuris spp. was found in these neo-tropical rodents mentioned. However, there is a dearth of information on the species of Trichuris that parasitize these animals. The capybara was the only rodent where some molecular techniques were used to identify a new species named T. cutillasae. In most cases, Trichuris spp. was found in combination with other endoparasites, and was found at a low prevalence in the lappe and guinea pig. The presence of Trichuris spp. ranged from 4.62–53.85% in the agouti, 4.21–10.00% in the lappe, 50% in the capybaras, and 1–31% in guinea pigs. Further work must be done towards molecular identification of various Trichuris spp. present in these rodents, as well as the clinical effect of infection on the performance of agouti, lappe, capybara, and guinea pigs.
