The Pre-Catching Sperm Technique Increases the Efficiency of the Intracytoplasmic Sperm Injection Method by Improving Fertilization and Blastocyst Formation Rates

A retrospective cohort study was conducted comparing conventional intracytoplasmic sperm injection (ICSI) with a pre-catching sperm (PCS)-ICSI technique. Cases with at least 0.5 million motile sperm and 5 mature oocytes were included. Conventional ICSI involves simultaneous loading of sperm and oocytes onto the dish, followed by identification, immobilization, and loading of sperm into pipettes for oocyte injection. In the PCS-ICSI technique, suitable sperm were identified and immobilized prior to oocyte loading onto the dish, thus reducing the oocyte exposure time. Variables of interest included rate of fertilized and degenerated oocytes, abnormal fertilization, quality blastocyst formation, and pregnancy outcomes. Statistical analysis utilized Student’s t-test and Fisher’s Exact Test. Our study included 330 PCS-ICSI and 287 conventional ICSI cases. Female age, BMI, AMH, total number of collected oocytes, and rate of abnormal fertilization were similar between groups. The PCS-ICSI group demonstrated an increased rate of oocyte fertilization (84.0% vs. 79.3%, p < 0.001), good quality blastocyst formation (54.9% vs. 48.0%, p < 0.001), and a lower rate of oocyte degeneration (1.4% vs. 3.5%, p < 0.001). Positive pregnancy rate, clinical pregnancy, and live birth rate were similar between groups. The expansion of this technique resulted in increased oocyte fertilization and good quality blastocyst formation, and decreased oocyte degeneration. Further studies will evaluate the effectiveness of this technique in broader patient populations. Full article

Background: Previous studies showed that the fourth SARS-CoV-2 vaccine dose has a protective effect against infection, as well as against severe disease and death. This study aimed to examine whether knowledge of a high-level antibody after the third dose may reduce compliance to the fourth booster dose among healthcare workers (HCWs). Methods: We conducted a prospective cohort study among HCWs vaccinated with the first three doses at Rambam Healthcare Campus, a tertiary hospital in northern Israel. Participants underwent a serological test before the fourth booster vaccine was offered to all of them, with results provided to participants. The population was divided into two groups, namely those with antibodies below 955 AU/mL and those with 955 AU/mL and higher, a cutoff found protective in a previous study. Multiple logistic regression was carried out to compare the compliance to the fourth booster between the two groups, adjusted for demographic and clinical variables. Results: After adjusting for the confounding variables, the compliance was higher in those with antibody levels below 955 AU/mL (OR = 1.41, p = 0.05, 95% CI 1.10–1.96). In addition, male sex and age of 60 years and above were also associated with higher vaccination rates (OR = 2.28, p < 0.001, 95% CI 1.64–3.17), (OR = 1.14, p = 0.043, 95% CI 1.06–1.75), respectively. Conclusions: Knowledge of the antibody status may affect compliance with the booster dose. Considering waning immunity over time, reduced compliance may affect the protection of HCWs who declined the fourth dose. Full article

In this paper, we investigate whether Mandarin short passives contain an implicit external argument (EA; such as an agent or experiencer). Syntactic tests yield inconclusive results in Mandarin. We employ intervention effects as a diagnostic tool and demonstrate that, unlike English, Mandarin short passives do not syntactically project an implicit EA. Our findings are based on spontaneous language data from the CHILDES corpora, encompassing 1182 children aged 2–6 years, and experimental data from 78 Mandarin-speaking children aged 3–6 years. This cross-linguistic structural difference explains the earlier acquisition of short passives in Mandarin compared to English and also raises a learnability question, which we briefly address: how do children know to project an implicit EA or not in their language? ‘Inner mitochondrial membrane peptidase 2 like’ (IMMP2L) is a nuclear-encoded mitochondrial peptidase that has been conserved through evolutionary history, as has its target enzyme, ‘mitochondrial glycerol phosphate dehydrogenase 2′ (GPD2). IMMP2L is known to cleave the mitochondrial transit peptide from GPD2 and another nuclear-encoded mitochondrial respiratory-related protein, cytochrome C1 (CYC1). However, it is not known whether IMMP2L peptidase activates or alters the activity or respiratory-related functions of GPD2 or CYC1. Previous investigations found compelling evidence of behavioural change in the Immp2lKD−/− KO mouse, and in this study, EchoMRI analysis found that the organs of the Immp2lKD−/− KO mouse were smaller and that the KO mouse had significantly less lean mass and overall body weight compared with wildtype littermates (p < 0.05). Moreover, all organs analysed from the Immp2lKD−/− KO had lower relative levels of mitochondrial reactive oxygen species (mitoROS). The kidneys of the Immp2lKD−/− KO mouse displayed the greatest decrease in mitoROS levels that were over 50% less compared with wildtype litter mates. Mitochondrial respiration was also lowest in the kidney of the Immp2lKD−/− KO mouse compared with other tissues when using succinate as the respiratory substrate, whereas respiration was similar to the wildtype when glutamate was used as the substrate. When glycerol-3-phosphate (G3P) was used as the substrate for Gpd2, we observed ~20% and ~7% respective decreases in respiration in female and male Immp2lKD−/− KO mice over time. Together, these findings indicate that the respiratory-related functions of mGpd2 and Cyc1 have been compromised to different degrees in different tissues and genders of the Immp2lKD−/− KO mouse. Structural analyses using AlphaFold2-Multimer further predicted that the interaction between Cyc1 and mitochondrial-encoded cytochrome b (Cyb) in Complex III had been altered, as had the homodimeric structure of the mGpd2 enzyme within the inner mitochondrial membrane of the Immp2lKD−/− KO mouse. mGpd2 functions as an integral component of the glycerol phosphate shuttle (GPS), which positively regulates both mitochondrial respiration and glycolysis. Interestingly, we found that nonmitochondrial respiration (NMR) was also dramatically lowered in the Immp2lKD−/− KO mouse. Primary mouse embryonic fibroblast (MEF) cell lines derived from the Immp2lKD−/− KO mouse displayed a ~27% decrease in total respiration, comprising a ~50% decrease in NMR and a ~12% decrease in total mitochondrial respiration, where the latter was consistent with the cumulative decreases in substrate-specific mediated mitochondrial respiration reported here. This study is the first to report the role of Immp2l in enhancing Gpd2 structure and function, mitochondrial respiration, nonmitochondrial respiration, organ size and homeostasis. Full article

Mitochondrial dysfunction is strongly associated with autism spectrum disorder (ASD) and the Inner mitochondrial membrane protein 2-like (IMMP2L) gene is linked to autism inheritance. However, the biological basis of this linkage is unknown notwithstanding independent reports of oxidative stress in association with both IMMP2L and ASD. To better understand IMMP2L’s association with behaviour, we developed the Immp2lKD knockout (KO) mouse model which is devoid of Immp2l peptidase activity. Immp2lKD −/− KO mice do not display any of the core behavioural symptoms of ASD, albeit homozygous Immp2lKD −/− KO mice do display increased auditory stimulus-driven instrumental behaviour and increased amphetamine-induced locomotion. Due to reports of increased ROS and oxidative stress phenotypes in an earlier truncated Immp2l mouse model resulting from an intragenic deletion within Immp2l, we tested whether high doses of the synthetic mitochondrial targeted antioxidant (MitoQ) could reverse or moderate the behavioural changes in Immp2lKD −/− KO mice. To our surprise, we observed that ROS levels were not increased but significantly lowered in our new Immp2lKD −/− KO mice and that these mice had no oxidative stress-associated phenotypes and were fully fertile with no age-related ataxia or neurodegeneration as ascertained using electron microscopy. Furthermore, the antioxidant MitoQ had no effect on the increased amphetamine-induced locomotion of these mice. Together, these findings indicate that the behavioural changes in Immp2lKD −/− KO mice are associated with an antioxidant-like phenotype with lowered and not increased levels of ROS and no oxidative stress-related phenotypes. This suggested that treatments with antioxidants are unlikely to be effective in treating behaviours directly resulting from the loss of Immp2l/IMMP2L activity, while any behavioural deficits that maybe associated with IMMP2L intragenic deletion-associated truncations have yet to be determined. This work presents refined, updated subregional and regional non-indigenous species (NIS) inventories for the Mediterranean Sea, validated by national and taxonomic experts, with species records observed until December 2020. These datasets will be used as the baselines for the implementation of the Integrated Monitoring and Assessment Programme for the Mediterranean (IMAP) and the Mediterranean Quality Status Report 2023. In total, 1006 non-indigenous species have been found in Mediterranean marine and brackish waters. The highest numbers of NIS were observed in Israel, Türkiye, Lebanon and Italy. Approximately 45 species were categorized as data deficient, either due to lack of consensus on their alien status or the validity of their identification. Polychaeta, Foraminifera and macroalgae were the groups with the highest numbers of controversial species. There was a general increase in the yearly rate of new NIS introductions after the late 1990s, which appears to be slowing down in the last decade, but this may be confounded by reporting lags and differential research efforts. Between 1970 and 2020 there has been a steep increase in the proportion of shared species present throughout all four Mediterranean subregions, which are predominantly transported via shipping and recreational boating. While Lessepsian species are gradually spreading westwards and northwards, there is still a considerable invasion debt accumulating in the eastern and central Mediterranean. Full article

Food and nutrition insecurity continue to risk the lives and wellbeing of millions of people throughout the world today. Further, food and nutrition insecurity are still major challenges in Kenya and have triggered the adoption of a number of modern biotechnologies for agricultural transformation. Consequently, many food technologies have been approved to secure sustainable access to food for millions of people. This study investigated the perceptions and implementation of two technologies for addressing food insecurity in Kenya, namely, genetically modified organisms (GMOs) and the use of antibiotics in livestock production. In particular, the study explored how their implementation can be governed responsibly through approved legislation. Therefore, the knowledge, attitudes and practices, as well as the governance, of GMO technology and antibiotic resistance risks were assessed. In-depth key interviews were conducted for a qualitative survey with triangulation using quantitative data sources. The findings showed that 46% of the population have limited knowledge about GMO technology, with about 79% indicating that foods with GM ingredients were already being consumed in the country despite the government ban. The majority of respondents agree that GMOs can contribute to an increase in the global food supply (65%), make food affordable (57%) and produce more nutritious foods (50%). Further, most agree that GMOs can produce crops more resistant to pests and reduce pesticide use on food crop plants (89). The main concerns reported regarding GMO technology by most respondents included the impact on the environment and human health and the adverse effects on traditional farming practices. About 36% of these respondents indicated that the technology diminishes traditional farming technologies, and 32% reported that it contributes to loss of biodiversity. Notably, 64% reported that GMO technology is a solution to food security and that GM foods are safe. Regarding the use of antimicrobials mainly meant to prevent diseases and access better markets, respondents perceived their use to be associated with a “large level of risk” of antimicrobial resistance (score of 2 on a scale of 1–3) (M = 1.85, SD = 1.06). A total of 56% of the respondents reported that the efforts towards promoting awareness of antibiotic resistance risks and their associated effects on human health are relatively limited. Our findings show that most of the respondents have only observed minimal awareness campaigns. Regarding the governance of the two technologies, 71% and 50% of the respondents reported that scientists and elected officials, respectively, have the greatest roles in the governance of GMOs, with small-scale farmers playing a negligible role. These findings are crucial to the advancement of food innovations that are geared towards achieving food security in Kenya as they highlight the risks associated with the poor governance and implementation of technologies. Therefore, there is a need for a framework for technological risk governance that is sensitive to local values and socio-economic circumstances and that will facilitate the achievement of food security goals. The Supporting and Enhancing NICU Sensory Experiences (SENSE) program promotes consistent, age-appropriate, responsive, and evidence-based positive sensory exposures for preterm infants each day of NICU hospitalization to optimize infant and parent outcomes. The initial development included an integrative review, stakeholder input (NICU parents and healthcare professionals), and feasibility focus groups. To keep the program updated and evidence-based, a review of the recent evidence and engagement with an advisory team will occur every 5 years to inform changes to the SENSE program. Prior to the launch of the 2nd edition of the SENSE program in 2022, information from a new integrative review of 57 articles, clinician feedback, and a survey identifying the barriers and facilitators to the SENSE program’s implementation in a real-world context were combined to inform initial changes. Subsequently, 27 stakeholders (neonatologists, nurse practitioners, clinical nurse specialists, bedside nurses, occupational therapists, physical therapists, speech-language pathologists, and parents) carefully considered the suggested changes, and refinements were made until near consensus was achieved. While the 2nd edition is largely the same as the original SENSE program, the refinements include the following: more inclusive language, clarification on recommended minimum doses, adaptations to allow for variability in how hospitals achieve different levels of light, the addition of visual tracking in the visual domain, and the addition of position changes in the kinesthetic domain. Introduction: Esthesioneuroblastoma (ENB) is a rare malignant neoplasm arising from the olfactory epithelium of the cribriform plate. Although survival is excellent with a reported 5-year overall survival (OS) of 82%, recurrence is frequent and occurs in 40–50% of cases. This study investigates the characteristics of ENB recurrence and the subsequent prognosis of patients with recurrence. Methods: The clinical records of all patients diagnosed as having ENB with subsequent recurrence at a tertiary hospital from 1 January 1960 to 1 January 2020 were retrospectively reviewed. Overall survival (OS) and progression-free survival (PFS) were reported. Results: A total of 64 out of 143 ENB patients had recurrences. In total, 45 out of 64 recurrences met the inclusion criteria and were included in this study. From these, 10 (22%) had a sinonasal recurrence, 14 (31%) had an intracranial recurrence, 15 (33%) had a regional recurrence, and 6 (13%) had a distal recurrence. The average interval from initial treatment to recurrence was 4.74 years. There were no differences in rates of recurrence with respect to age, sex, or types of surgery (endoscopic, transcranial, lateral rhinotomy, and combined). The time to recurrence was shorter for Hyams grades 3 and 4 compared to Hyams grades 1 and 2 (3.75 years vs. 5.70 years, p < 0.05). Patients with recurrence limited to the sinonasal region had a lower overall primary Kadish stage compared to recurrences beyond the sinonasal region (2.60 vs. 3.03, p < 0.05). A total of 9 (20%) out of 45 patients developed secondary recurrence. Following recurrence, the subsequent 5-year OS and PFS were 63 and 56%, respectively. The mean time to secondary recurrence after treatment of the primary recurrence was 32 months, which was significantly shorter than the time to primary recurrence (32 months vs. 57 months, p = 0.048). The mean age of the secondary recurrence group is significantly older than the primary recurrence group (59.78 years vs. 50.31 years, p = 0.02). No statistically significant differences were observed between the secondary recurrence group and the recurrence group in terms of their overall Kadish stages or Hyams grades. Conclusions: Following an ENB recurrence, salvage therapy appears to be an effective therapeutic option with a subsequent 5-year OS of 63%. However, subsequent recurrences are not infrequent and may require additional therapy. Full article

Introduction: The fourth SARS-CoV-2 vaccine dose was found to protect against infection and more importantly against severe disease and death. It was also shown that the risk of symptomatic or severe disease was related to the antibody levels after vaccination or infection, with lower protection against the BA.4 BA.5 Omicron variants. The aim of our study was to assess the impact of the fourth dose on infection and perception of illness seriousness among healthcare workers (HCWs) at a tertiary health care campus in Haifa, Israel, and to investigate the possible protective effect of antibody levels against infection. Methods: We conducted a prospective cohort study among fully vaccinated HCWs and retired employees at Rambam Healthcare Campus (RHCC), a tertiary hospital in northern Israel. Participants underwent serial serological tests at 1, 3, 6, 9, 12 and 18 months following the second BNT162b2 vaccine dose. Only a part of the participants chose to receive the fourth vaccine. A multivariable logistic regression was conducted to test the adjusted association between vaccination, and the risk of infection with SARS-CoV-2. Kaplan–Meier SARS-CoV-2 free “survival” analysis was conducted to compare the waning effect of the first and second, third and fourth vaccines. Receiver Operating Characteristic (ROC) curve was plotted for different values of the sixth serology to identify workers at risk for disease. Results: Disease occurrence was more frequent among females, people age 40-50 years old and those with background chronic lung disease. The fourth vaccine was found to have better protection against infection, compared to the third vaccine; however, it also had a faster waning immunity compared to the third vaccine dose. Antibody titer of 955 AU/mL was found as a cutoff protecting from infection. Conclusions: We found that the fourth vaccine dose had a protective effect, but shorter than the third vaccine dose. Cutoff point of 955 AU/mL was recognized for protection from illness. The decision to vaccinate the population with a booster dose should consider other factors, including the spread of disease at the point, chronic comorbidities and age, especially during shortage of vaccine supply. The COVID-19 pandemic caused an unprecedented disruption of food systems worldwide, with most governments taking severe containment measures to curb the spread. This resulted in unpredicted negative impacts of the agri-food supply chains coupled with food price inflations. Ultimately, this affected the food security and urban livelihoods for most households, who are dependent on urban markets for food supply. This study examined the implications of the pandemic on food prices and commodities supplies to urban markets conducted through structured interviews. A review of the secondary data was also conducted to show the trends of commodity prices over the last 5 years. The high inflation of commodity prices with a decline in sales volumes was reported by most traders (97%) with decreases in supply quantities. Changes in the consumption behaviour in households was reported by consumers (75%), with 65% experiencing reduced food diversity at home. Households adopted varied coping mechanisms, including reduced food portions (52%), reduced food varieties (44%) and skipping meals (32%). Market prices increased by an average margin of 13.8% for grains and pulses with price decline observed for cabbages (−30.8%) and Irish potatoes (−19.4%). The findings may inform policymakers of additional future shock and pandemic control protocols, whose actions would assure food protection of urban livelihoods. Full article

This study assessed humoral response to the third BNT162b2 dose among healthcare workers (HCW). This prospective cohort study of HCW tested for anti-spike antibodies (LIAISON SARS-CoV-2 S1/S2 IgG assay) at 1, 3, 6, 9, and 12 months after receiving the second BNT162b2 vaccine dose (tests 1, 2, 3, 4, and 5, respectively). A third (booster) vaccination dose was introduced before test 4. Linear regression model was used to determine the humoral response following vaccine doses. For each serology test, changes in log-transformed antibody concentrations over time, adjusted for age, sex, underlying diseases, steroid treatment, and smoking were described using the general linear mix model. Serology tests were performed at 3, 6, 9, and 12 months after the second vaccine dose in 1113, 1058, 986, and 939 participants, respectively. The third dose was received by 964 participants before the 9-month tests, 797 of whom participated in the 9- and 12-month serology tests. A significant inverse correlation was noted between time from third dose and antibody concentrations (Spearman correlation −0.395; p < 0.001). Age (p < 0.0001; CI 95% −0.005–−0.004), heart disease (p < 0.0001; CI 95% −0.177–−0.052), immunodeficiency (p < 0.0001; CI 95% 0.251–−0.106), and smoking (p < 0.0001; CI 95% −0.122–−0.040) were significantly associated with decreased antibody concentrations. Female sex (p = 0.03; CI 95% 0.013–0.066) was associated with increased antibody concentrations. The third booster dose had a better effect on immunogenicity, with higher antibody concentrations among tested HCW. Heart disease, smoking, and other known risk factors were associated with decreased antibody concentrations. Full article

The purpose of the study was to examine the mediation roles of student satisfaction and entrepreneurial self-efficacy in the nexus between entrepreneurial education and entrepreneurial self-competencies within a social enterprise context. The study used a cross-sectional survey design, with a sampled population of 185 business students from three universities (Accra Technical University, Cape Coast Technical University and the University of Ghana) in Ghana. A PLS-SEM approach was used to examine the relationships among the independent–dependent constructs in the study. Entrepreneurial education had positive and significant relationships to student satisfaction and entrepreneurial self-efficacy, but it showed an insignificant relationship to entrepreneurial self-competencies. Student satisfaction was also found to relate positively and significantly to entrepreneurial self-efficacy and entrepreneurial self-competencies. Furthermore, both student satisfaction and entrepreneurial self-efficacy were found to fully mediate the nexus between entrepreneurial education and entrepreneurial self-competencies. The study highlights the crucial roles of student satisfaction and self-efficacy in the implementation of entrepreneurial education in higher education institutions. In a discipline that is characterised by paucity, this study provides a unique and original assessment of the important roles of student satisfaction and student self-confidence in building entrepreneurial competencies among students. Objective: The aim of this study was to investigate the long-term oncologic outcome and review the state of the art in the management of olfactory neuroblastomas. Material and Methods: The records of all patients treated for olfactory neuroblastomas in two academic departments between 1975 and 2012 were evaluated retrospectively. Data on epidemiological parameters were collected (age, gender), along with staging (Kadish, Morita), histologic grading (Hyams), time and form of treatment, locoregional control, and disease-specific and overall survival. Patients with other malignant diseases, distant metastases of olfactory neuroblastomas at the time of initial diagnosis, a follow-up time of less than 5 years, or insufficient clinical-pathological data were excluded from further analysis. Results: In total, 53 cases made up our final study sample (26 men, 27 women; male–female ratio 0.96:1). Their mean age was 48.6 years (range: 10–84 years). The mean follow-up time was 137.5 months (4–336 months, SD: 85.0). A total of 5 out of 53 study cases (9.4%) showed metastatic involvement of the neck at the time of initial presentation. Local recurrence was detected in 8/53 (15.1%) and regional recurrence in 7/53 of our study cases (13.2%). Three patients (42.8%) from the group of cases with surgery as the sole form of management (7/53, 13.2%) died due to the disease. The cumulative disease-specific survival and overall survivalfor the whole group of patients were 88.6% and 63.6%, respectively. The cumulative disease-specific survival stratified by Kadish A/B vs. Kadish C/D as well as Hyams I/II vs. Hyams III/IV showed superior results for limited tumors, albeit without significance, and low-grade tumors (highly significant difference). Conclusion: Craniofacial or sometimes solely endoscopically controlled resection can warrant resection of the olfactory neuroblastoma with wide margins. However, locoregional failures and distant metastases can occur after a long period of time. The non-negligible incidence of regional recurrences, partly in unusual localizations, leads us to consider the need to identify the “recurrence-friendly” cases and to perform individualized elective irradiation of the neck in cases with high-risk features. The purpose of the current study was to assess the prevalence of cyst formation at the brain-tumor interface in olfactory neuroblastoma. We used the UCLA patient-based Pathology and Radiology Head and Neck Database (UPP&R HAND) to identify the largest patient cohort reported to date with imaging and pathology data. Eighteen of thirty-one patients (58.1%) had evidence of intracranial extension on MRI, while four (22.0%) demonstrated cyst formation at the brain–tumor interface. The extent of intracranial extension was by far the strongest predictor for intracranial cyst formation, regardless of Hyams tumor grade, using a binary logistics regression model (p = 0.002) and ROC curve analysis (AUC 94.6%). Cyst formation at the brain-tumor interface was an uncommon imaging finding, and tends to occur with a larger component of intracranial tumor extension. Full article
