TasA gene, encoding a functional amyloid protein critical for biofilm formation and antimicrobial activity, was cloned from the endophytic strain Bacillus amyloliquefaciens BS-3, isolated from rubber tree roots. This study identified the shortest functional TasA variant (483 bp, 160 aa) reported to date, featuring unique amino acid substitutions in conserved domains. Bioinformatics analysis predicted a signal peptide (1–27 aa) and transmembrane domain (7–29 aa), which were truncated to optimize heterologous expression. Two prokaryotic vectors (pET28a and pCZN1) were constructed, with pCZN1-TasA expressed solubly in Escherichia coli Arctic Express at 15 °C, while pET28a-TasA formed inclusion bodies at 37 °C. Purified recombinant TasA exhibited potent antifungal activity, achieving 98.6% ± 1.09 inhibition against Colletotrichum acutatum, 64.77% ± 1.34 against Alternaria heveae. Notably, TasA completely suppressed spore germination in C. acutatum and Oidium heveae Steinmannat 60 μg/mL. Structural analysis via AlphaFold3 revealed that truncation enhanced protein stability. These findings highlight BS-3-derived TasA as a promising biocontrol agent, providing molecular insights for developing protein-based biopesticides against rubber tree pathogens. Full article

In this paper, we propose an optoelectronic feed-forward millimeter-wave generator based on the Mach–Zehnder interferometer (MZI) structure. The phase noise of the local oscillation (LO) input is extracted by loop design and used for phase noise suppression of the output, thereby optimizing the phase noise performance of the generator output. The scheme achieves separation of the phase noise by using an MZI structure and a mixing-frequency oscillator to realize the differential and integration process of the phase noise from the LO input source, respectively. Then, it is combined with a feed-forward operation to skillfully realize phase noise rejection of the resulting high-frequency output. The proposed scheme has been demonstrated to facilitate millimeter-wave generation at 40 GHz and 50 GHz. The measured phase noise is as low as −120 dBc/Hz at a 10 kHz offset, and the experimental setup achieves phase noise suppression of up to 36 dB at this frequency offset. Through systematic theoretical analysis and experimental verification, the excellent capabilities of the proposed scheme in high-frequency signal generation and phase noise suppression are fully demonstrated, which provides a new technological path for high-performance millimeter-wave generation, avoiding the deterioration of the phase noise introduced using high-frequency optoelectronic devices other than photodetectors (PDs) to process the signals. Urban expansion has a profound impact on both society and the environment. In this study, VOSviewer 1.6.16 and CiteSpace 6.3.R1 were used to conduct a bibliometric analysis of 2987 articles published during the period of 1992–2022 from the Web of Science database in order to identify the research hotspots and trends of urban expansion and its driving factors. The number of articles significantly increased during the period of 1992–2022. The spatiotemporal characteristics and driving forces of urban expansion, urban growth models and simulations, and the impacts of urban expansion were the main research topics. The rate of urban expansion showed regional differences. Socioeconomic factors, political and institutional factors, natural factors, path effects, and proximity effects were the main driving factors. Urban expansion promoted economic growth, occupied cultivated land, and affected ecological environments. Big data and deep learning techniques were recently applied due to advancements in information techniques. With the increasing awareness of environmental protection, the number of studies on environmental impacts and spatial planning regulations has increased. Some political and institutional factors, such as subsidies, taxation, spatial planning, new development strategies, regulation policies, and economic industries, had controversial or unknown impacts. Further research on these factors and their mechanisms is needed. A limitation of this study is that articles which were not indexed, were not included in bibliometric analysis. Further studies can review these articles and conduct comparative research to capture the diversity. Full article

Owing to high electrical conductivity, layered structure, and abundant surface functional groups, transition metal carbides/nitrides (MXenes) have received enormous interest in the field of gas sensors at room temperature. In this work, we synthesize a heterostructured nanocomposite with indium oxide (In2O3) decorated on titanium carbide (Ti3C2Tx) nanosheets by electrostatic self-assembly and develop it for high-selectivity NO2 gas sensing at room temperature. Self-assembly formation of multiple heterojunctions in the In2O3/Ti3C2Tx composite provide abundant NO2 gas adsorption sites and high electron transfer activity, which is conducive to improving the gas-sensing response of the In2O3/Ti3C2Tx gas sensor. Assisted by rich adsorption sites and hetero interface, the as-fabricated In2O3/Ti3C2Tx gas sensor exhibits the highest response to NO2 among various interference gases. Meanwhile, a detection limit of 0.3 ppm, and response/recovery time (197.62/93.84 s) is displayed at room temperature. Finally, a NO2 sensing mechanism of In2O3/Ti3C2Tx gas sensor is constructed based on PN heterojunction enhancement and molecular adsorption. This work not only expands the gas-sensing application of MXenes, but also demonstrates an avenue for the rational design and construction of NO2-sensing materials. Full article

Under the influence of global climate change, water conservancy projects located in the high-latitude cold regions of the world are facing severe challenges. This study addresses the contradiction between water supply stability and ecological flow during the dry season in cold regions. Taking Linhai Reservoir as the core, it integrates the HEC-RAS hydrodynamic model with multi-source data such as basin topography, hydro-meteorological data, and water conservancy project parameters to construct a multi-scenario water supply scheduling model during the dry season. The aim is to provide scientific recommendations for different reservoir operation strategies in response to varying frequencies of upstream inflow, based on simulations conducted after the reservoir’s completion. Taking into account winter runoff reduction characteristics and engineering parameters, we simulated the relationships between water level and flow, ecological flow requirements, and urban water shortages. The results indicate that in both flood and normal years, dynamic coordination of storage and discharge can achieve a daily water supply of 120,000 cubic meters, with 100% compliance for the ecological flow rate. For mild and moderate drought years, additional water diversion becomes necessary to achieve 93.5% and 89% supply reliability, respectively. During severe and extreme droughts, significantly reduced reservoir inflows lower ecological compliance rates, necessitating emergency measures, such as utilizing dead storage capacity and exploring alternative water sources. The study proposes operational strategies tailored to different drought intensities: initiating storage adjustments in September for mild droughts and implementing peak-shifting measures by mid-October for extreme droughts. These approaches enhance storage efficiency and mitigate ice blockage risks. This research supports the water supply security and river ecological health of urban and rural areas in Mudanjiang City and Hailin City and provides a certain scientific reference basis for the multi-objective coordinated operation of reservoirs in the same type of high-latitude cold regions. The inclusion of adsorption thermodynamic analysis and performance benchmarking with existing adsorbents reinforces both the theoretical significance and practical applicability of this study. The modified rubber-derived carbon demonstrated a remarkably high DBT adsorption capacity of 254.45 mg/g. These results establish it as a promising alternative to conventional materials such as commercial activated carbon, zeolites, and even metal–organic framework materials. In addition to confirming the superior performance of the adsorbent, the findings provide a deeper understanding of the DBT adsorption mechanism and offer a solid scientific basis for large-scale fuel desulfurization applications. This research highlights the potential of transforming end-of-life tire waste into value-added functional materials and contributes to the advancement of sustainable and efficient desulfurization technologies. Future work should focus on optimizing surface functionalization and regeneration strategies to further improve long-term adsorption stability and practical deployment. Full article

Background/Objectives: Visual impairment (VI) represents a significant health challenge among older adults, particularly due to their limited understanding of health information. This study aimed to investigate and compare the health information-seeking behavior (HISB) of older adults with VI across different stages of eye care. Methods: A cross-sectional comparative study was conducted in 248 older adults with VI in various stages of eye care, including the pre-visit stage (n = 84), treatment stage (n = 83), and follow-up stage (n = 81) at the Shenzhen Eye Hospital from July to October 2024. Participants completed an HISB questionnaire encompassing four dimensions: attitude, needs, sources, and barriers. Results: The overall mean score of HISB varied significantly among the different eye care stages. The treatment stage had the highest scores (3.70 ± 0.32), particularly in attitudes toward health information and information needs. Poor self-reported overall health facilitated HISB in each stage. In the pre-visit stage, higher income was associated with increased HISB, while a lack of internet access reduced it. In the treatment stage, higher education was associated with higher HISB, whereas moderate-to-severe VI and certain income levels were associated with negative effects. In the follow-up stage, rural residence and spousal or child caregiving emerged as key facilitators of HISB. Conclusions: The treatment stage is a critical period for HISB engagement in older adults with VI. Healthcare providers should consider stage-specific factors of HISB to optimize health information delivery. Full article

Background/Objectives: Dominant leg use in badminton may contribute to lower limb asymmetry, potentially affecting performance and injury risk. This study investigated the effects of a 12-week integrated neuromuscular training (NMT) program on sports performance. Methods: Twenty-four well-trained male badminton players (age: 13.5 ± 1.15 years) were randomly assigned to groups based on maturation status (pre-peak height velocity [pre-PHV] and post-peak height velocity [post-PHV]; n = 12 each). All participants completed two NMT sessions weekly. Pre- and post-training assessments included a 20 m sprint, countermovement jump (CMJ), agility t-text, hexagon test, and Y-balance test. Results: Both groups improved significantly across most tests. The post-PHV group (ES: 0.70–1.35) showed greater improvements in sprinting, CMJ, and agility, while the pre-PHV group (ES: 0.39–1.23) improved more in balance and asymmetry. Conclusions: These results underscore the need for age- and maturity-specific training strategies to optimize performance and address asymmetries in youth athletes. Full article

The high-dimensional integral–differential nature of the neutron transport equation and the complexity of nuclear reactors result in high computational costs. A set of reduced-order modeling frameworks based on Proper Orthogonal Decomposition (POD) is developed to improve the computational efficiency for neutron diffusion calculations while maintaining accuracy, especially for small samples. For modal coefficient calculations, three methods—Galerkin, radial basis function (RBF), and Deep Neural Network (DNN)—are introduced and analyzed for molten salt reactors. The results show that all three reduced-order models achieve sufficient accuracy, with neutron flux L2 errors below 1% and delayed neutron precursor (DNP) L2 errors below 2.4%, while the acceleration ratios exceed 800. Among these, the POD–Galerkin model demonstrates superior performance, achieving average L2 errors of less than 0.00658% for neutron flux and 1.01% for DNP concentration, with an acceleration ratio of approximately 1800 and excellent extrapolation ability. The POD–Galerkin reduced-order model significantly enhances the computational efficiency for solving neutron multi-group diffusion equations and DNP conservation equations in molten salt reactors while preserving the solution accuracy, making it ideal for a liquid fuel molten salt reactor in the case of small samples. Environmental DNA (eDNA) metabarcoding was utilized to compare the fish species composition, diversity, and their relationships with environmental factors in four medium-sized drinking water reservoirs (Tongjiqiao, Andi, Shafan, and Jinlan) within the Qiantang River Basin during both wet and dry seasons. A total of 44 fish species belonging to 6 orders, 15 families, and 40 genera were detected, with Cyprinidae being identified as the dominant family (68.2%). Silver carp (Hypophthalmichthys molitrix) and bighead carp (Hypophthalmichthys nobilis) were identified as the most abundant species, representing 81.94% and 99.98% of the relative eDNA abundance, respectively. The fish communities were characterized by river-resident species (59.1%), pelagic species (43.2%), and omnivorous (43.2%) and carnivorous (43.2%) feeding habits. Significant influences of seasonal and reservoir-specific variations on physicochemical parameters, species composition, and ecological traits were observed. However, no significant differences in community diversity (Chao1, Pielou_e, Shannon, and Simpson indices) or distribution patterns were detected between wet and dry seasons. In contrast, Jinlan Reservoir was found to exhibit distinct diversity and distribution patterns compared to the other three reservoirs, which was consistent with the relative eDNA abundance of H. molitrix and H. nobilis. Through a Spearman correlation analysis, the relative abundance of H. molitrix was revealed to be negatively correlated with community diversity, while H. nobilis was shown to have positive correlations (except with Chao1). This suggests that the excessive stocking of H. molitrix may lead to reduced diversity, whereas moderate stocking of H. nobilis could promote diversity restoration. Among physicochemical factors, Chao1 richness was found to be negatively correlated with conductivity, pH, phosphate–phosphorus (PO4-P), and the total nitrogen to total phosphorus ratio (TN/TP), while Shannon and Simpson diversity indices were negatively correlated with nitrate nitrogen (NO3-N) concentrations. A redundancy analysis (RDA) identified NO3-N and permanganate index (CODMn) as the primary physicochemical drivers of fish community structure, indicating that while physicochemical differences were found to influence species composition and diversity, their effects were considered relatively limited. These findings suggest that the overwhelming dominance of H. molitrix and H. nobilis in the reservoirs may reduce the influence of seasonal variations and cross-reservoir physicochemical disparities on fish community dynamics. Ticks and tick-borne pathogens pose a significant threat to human and animal health, yet the diversity and prevalence of tick-borne microorganisms in karst regions remains inadequately explored. In October 2023, a total of 274 Rhipicephalus microplus ticks were collected from livestock in Guizhou Province, which boasts the largest karst area in China. Pathogen identification was subsequently performed using PCR amplification, Sanger sequencing, and phylogenetic analysis. High microbial diversity was noted, with five bacterial species from the order Rickettsiales detected, including those from the genera Rickettsia (family Rickettsiaceae), Anaplasma, and Ehrlichia (family Anaplasmataceae). The overall prevalence of infection with at least one pathogen was remarkably high at 94.5%. The highest positive rate was observed for Candidatus Rickettsia jingxinensis at 90.9%. A novel Ehrlichia species, provisionally designated as Candidatus Ehrlichia carsus, was identified with a positive rate of 16.8%. In addition, Anaplasma marginale, Ehrlchia minasensis and Ehrlichia canis were detected in 15.3%, 4.7% and 1.5%, respectively. The co-infections involving two or three rickettsial species were observed in 34.3% ticks. These findings highlight the high diversity and prevalence of tick-borne rickettsial agents in the karst area, underscoring the need for enhanced surveillance and effective tick control to mitigate disease risks to both humans and livestock. Background and Objectives: Sarcopenia is a notable comorbidity of rheumatoid arthritis (RA), affecting about one third of patients. However, the characteristic of muscle function and its association with RA disease remains unknown. Materials and Methods: This cross-sectional study collected clinical data from a real-world Chinese RA cohort. Sarcopenia was defined as both myopenia and low muscle function (LMF). Myopenia was defined as appendicular skeletal muscle mass index (ASMI) < 7.0 kg/m2 in men and <5.7 kg/m2 in women. LMF was defined as low muscle strength (LMS, hand grip < 28 kg in men and <18 kg in women) or low physical performance (LPP, 6 m gait speed < 1.0 m/s). Results: Among 1125 RA patients recruited in this study, 928 RA patients were eligible for analysis. The prevalence of sarcopenia, myopenia, LMF, LMS, and LPP in all RA patients was 36.5%, 46.1%, 69.0%, 57.8%, and 37.1%, respectively. According to their trends in age and disease activity, there were 111 (11.9%) patients in the young (age < 50 years) and remission (CDAI ≤ 2.8) subgroup, 199 (21.4%) patients in the young and active (CDAI > 2.8) subgroup, 198 (21.3%) patients in the old (age ≥ 50 years) and remission subgroup, and 420 (45.2%) patients in the old and active subgroup. Compared with the two remission subgroups, respectively, the young and active subgroup had significantly lower grip strength, higher prevalence of sarcopenia, LMF, and LMS, and worse activity function. After adjustment for potential confounders, multivariate multinominal logistic regression analysis showed that the young and active subgroup was positively associated with sarcopenia (OR = 3.193, 95%CI: 1.477–6.899), LMF (OR = 2.390, 95%CI: 1.207–4.731), and LMS (OR = 3.520, 95%CI: 1.743–7.110). Conclusions: Worse muscle strength, rather than reduced physical performance, is more common in patients with active RA at a young age. It underscores the critical need for early identification and intervention of muscle dysfunction to improve their quality of life. Full article

Interferon-γ (IFN-γ), a member of the Type II IFN family, is a crucial cytokine in the immune system and serves as an important indicator of immune response. Intracellular cytokine staining (ICS) is a technique used to analyze the production of cytokines within individual cells, and it has a wide range of applications in the fields of immunological monitoring, vaccine trials, and the study of infectious diseases. This study aimed to prepare monoclonal antibodies against duck IFN-γ protein and to establish an ICS protocol for detecting the duck IFN-γ protein. The duIFN-γ-His or duIFN-γ-Fc gene was cloned into the pEE12.4 expression vector and expressed as a recombinant protein of size 20.2 KDa or 54.9 KDa in 293F cells. The purified recombinant proteins were inoculated into BALB/c mice to generate splenic lymphocytes capable of secreting anti-duIFN-γ antibodies, and hybridoma cells were obtained after fusion with SP2/0 cells. A new hybridoma cell line named 24H4, which stably secreted IgG3 κ subtype antibody against duck IFN-γ, was established. This monoclonal antibody (mAb) was identified by Western blot to recognize duck IFN-γ antibodies, and the indirect ELISA results showed that its ability to recognize IFN-γ protein reached 0.001 μg/mL. The established ICS method was used to stain PBMCs after Concanavalin A (ConA) stimulation, and duck IFN-γ protein was successfully detected by flow cytometry, indicating that the ICS method was successful. In this study, we provide a crucial tool for subsequent research on duck cellular immune responses by using the monoclonal antibody 24H4. Full article

Using Generative Artificial Intelligence (GAI) in education has opened new avenues for innovation, yet its role as an interactive tool with learners remains underexplored. Research in this domain faces challenges from pedagogical complexities and the variability of AI tools. To address these gaps, this study developed an online project-based learning (PBL) platform incorporating a GAI plug-in and conducted a year-long experiment to analyze its impact. Three sets of experimental analyses were performed to examine learners’ methods, cognitive processes, and learning effectiveness. The findings reveal that GAI significantly influenced students’ learning approaches, cognitive engagement, and learning effectiveness. Additionally, the study demonstrates that PBL offers an effective framework for investigating the educational implications of GAI, providing new insights for future research in this evolving field.
