Special Issue Information

Dear Colleagues,

Antibody–drug conjugates (ADCs) are drugs with enormous potential which are now established as a major class of therapeutics for the clinical treatment of cancer. To date, twelve ADCs have been approved by the FDA. ADCs are constructed with four main elements: a monoclonal antibody designed to target a particular cancer-associated antigen, a potent cytotoxic payload or warhead, a “linker” that cleaves off that payload inside cancer cells, and the conjugation technology that attaches the linker and payload to the antibody. ADCs provide a unique opportunity to deliver drugs to tumor cells, thus improving the efficacy of chemotherapy and reducing systemic exposure and toxicity. However, while this all sounds relatively straightforward, the construction has been anything but simple for ADC developers. Emerging linker technologies also raise the exciting possibility of attaching different payloads with different mechanisms of action to the same antibody to improve efficacy and reduction in the development of resistance. The quest for novel payload molecules is driven by several factors, including the need to enhance cytotoxic potency, avoid the development of resistance, and improve physicochemical properties.

In this Special Issue, we aim to publish high-quality manuscripts highlighting novel approaches for payload design and synthesis in the field of ADCs. We welcome manuscript submissions in the form of original research, communication, and perspectives.

Manuscripts should be submitted online at www.mdpi.com by registering and logging in to this website. Once you are registered, click here to go to the submission form. Manuscripts can be submitted until the deadline. All submissions that pass pre-check are peer-reviewed. Accepted papers will be published continuously in the journal (as soon as accepted) and will be listed together on the special issue website. Research articles, review articles as well as short communications are invited. For planned papers, a title and short abstract (about 100 words) can be sent to the Editorial Office for announcement on this website.

Submitted manuscripts should not have been published previously, nor be under consideration for publication elsewhere (except conference proceedings papers). All manuscripts are thoroughly refereed through a single-blind peer-review process. Molecules is an international peer-reviewed open access semimonthly journal published by MDPI.

Submitted papers should be well formatted and use good English. Authors may use MDPI's English editing service prior to publication or during author revisions.
