Dear Colleagues,
The global incidence of thyroid cancer is steadily increasing. While differentiated thyroid cancer (DTC) accounts for the majority of cases and has a generally high survival rate, aggressive DTC variants and other types, such as medullary, anaplastic, or poorly differentiated thyroid cancers, present severe clinical challenges. Identifying patients prone to rapid disease progression, recurrence, tumor dedifferentiation, or therapy resistance remains difficult. Traditional clinico-pathological features and established genetic markers often fall short in predicting individual tumor behavior when used in isolation. This highlights the need for an integrative approach, i.e., combining clinical parameters with comprehensive molecular tools, to optimize risk stratification and ensure timely management for high-risk thyroid cancer patients.
Thyroid cancer pathogenesis is driven by a complex interplay of clinical, genetic, epigenetic, and environmental factors. A holistic understanding of tumor biology paves the way for discovering novel biomarkers and targeted therapies, marking a pivotal step toward precision endocrinology for both well-differentiated and highly aggressive thyroid malignancies. Recent molecular breakthroughs have driven the development of targeted treatments, such as tyrosine kinase inhibitors, which block critical oncogenic pathways in advanced thyroid cancer. Concurrently, immunotherapies are being investigated to stimulate the body's anti-tumor defenses.
This Special Issue will present and disseminate the most recent advances related to the clinical, genetic, and epigenetic characterization of thyroid cancer. We will consider contributions addressing integrative approaches to risk stratification, the discovery of novel biomarkers, and the development of emerging therapeutic strategies for both well-differentiated and highly aggressive thyroid malignancies.
Topics of interest include, but are not limited to, the following:
- Combining clinico-pathological features with genetic and/or epigenetic profiles to predict thyroid cancer recurrence and survival;
- Epigenetic changes and their prognostic or diagnostic value in thyroid cancer;
- Clinical and molecular mechanisms underlying therapy resistance;
- The clinical utility of circulating biomarkers for real-time disease monitoring;
- Preclinical and clinical evaluation of novel genetic and epigenetic therapeutic targets for thyroid malignancies.
