Dear Colleagues,

Marine natural products have extraordinary chemical diversity, biochemical specificity and other molecular properties that make them suitable for consideration as lead structures for drug discovery. In this regard, one of the main problems is the typical low bioavailability, which can complicate both the structural characterization and possible future biological and pharmacological developments. For these reasons, an important step in the study and investigation of bioactive marine metabolites is the development of chemical synthesis and synthetic strategies aimed at the preparation of pure compounds and analogs both for structural confirmation and/or for large-scale preparation. Marine natural products could be a pivotal starting point to pave the way to new classes of biologically active compounds with pharmacological potential.

This Special Issue is the third version on these subjects (the first and second editions are available at https://www.mdpi.com/journal/marinedrugs/special_issues/Synthesis_Inspired and https://www.mdpi.com/journal/marinedrugs/special_issues/Synthesis_Immunomodulatory) and aims to collect original research articles regarding synthetic strategies for secondary marine metabolites and/or analogs that can favor applications and/or can solve structural challenges common in the field of natural substances.

A great effort to discover new therapeutic ingredients is often initiated through the discovery of the existence of novel marine natural products. Since substances produced by the marine environment might be structurally more complex and unique than terrestrial natural products, there have been cases of misassignments of their structures despite the availability of modern spectroscopic and computational chemistry techniques. When it comes to refutation to erroneously or tentatively proposed structures empirical preparations through organic chemical synthesis has the greatest contribution along with close and sophiscated inspection of spectroscopic data. Herein, we analyzed the total synthetic studies that have decisively achieved in revelation of errors, ambiguities, or incompleteness of the isolated structures of marine natural products covering the period from 2018 to 2021.
