Special Issue Information

Dear Colleagues,

Breast and gynecological malignancies represent a substantial burden on women’s health worldwide and are among the most significantly impacted fields by recent advances in molecular biology, and precision medicine.

As regards breast cancer, alongside conventional biomarkers (such as hormone receptors and HER2 status), new predictive markers have recently been introduced, including PIK3CA and AKT1 mutations, PTEN loss, ESR1 mutations, and BRCA1/2 variants. This has gone in parallel with the implementation into clinical practice of CDK4/6 inhibitors, PI3K inhibitors, PARP inhibitors (PARPi), and innovative anti-HER2 therapy.

Biomarker-driven methods are being increasingly often used to classify and treat gynecological cancers, such as ovarian and endometrial cancers. PARPi have revolutionized therapy of HRD-positive ovarian cancer, while immune checkpoint inhibitors have increased the therapeutic options for endometrial cancer. Recent studies are finding novel targets that can be acted on, as ARID1A, MMR deficiency, and HER2 overexpression.

This Special Issue aims to provide a comprehensive and up-to-date overview of targeted therapy for breast and gynecological cancers. We welcome original research articles, reviews, and clinical perspectives that examine novel therapeutic targets, resistance mechanisms, and emerging data from preclinical models. Emphasis will be placed on translational studies, real-world data, and methodologies for integrating targeted therapies into clinical practice.
