Dear Colleagues,

Psychoactive alkaloids such as caffeine and nicotine have shaped human cognition and behavior for centuries, offering powerful yet imperfect tools for modulating alertness, mood, and performance. Their rapid efficacy, accessibility, and affordability have made them deeply integrated into modern life. Despite their benefits, challenges, including tolerance, desensitization, and addiction, underscore the need for safer, more sustainable neuroactive alternatives. This Special Issue explores the molecular and neuropharmacological underpinnings of classical alkaloids. It examines emerging next-generation bioactive compounds—from botanicals to synthetic analogs—aimed at achieving enhanced efficacy with improved safety profiles. By bridging molecular insights with translational advances, this issue aims to illuminate innovative strategies for neuropharmacological interventions, performance enhancement, and preventive health.
