Dear Colleagues,
Lymphoproliferative neoplasms are rare hematological cancers spanning a broad spectrum of diseases, from indolent to highly aggressive ones. Over the last few years, the treatment landscape of lymphoproliferative diseases (LPDs) has rapidly changed with the advent and incorporation of immunotherapies in the treatment armamentarium, offering remarkable and prolonged remissions in patients who have failed conventional immunochemotherapy options.
Among these newly introduced therapies, T cell engagers (TCEs) and mainly BiTEs, comprise antibodies engineered to bind to a target antigen expressed on a cancer cell and to a trigger molecule on T cells. TCEs act as an immunological synapse by redirecting T cells to recognize cancer cells, inducing their activation and proliferation to kill the target cancer cells. Several TCEs have been approved and licensed in LPD, but challenges and side effects, such as on-target/off-tumor toxicity and risks of cytokine release syndrome, are still a concern. The field is rapidly expanding with over 200 immune cell engagers in clinical trials, testing novel compounds, immune cells beyond T cells and combinatorial regimens even as first-line treatments.
This Special Issue, entitled "Unraveling the Mechanistic Pathways of Immune Cell Engagers in Lymphoproliferative Diseases", aims to collect the most recent advances across the field of immune cell engagers, focusing on mechanisms of action, molecular pathways, avenues of improving safety, reducing side effects, and developing "conditional" immune cell engagers that only activate within the tumor microenvironment.
