Mechanisms of Resistance to Melanoma Immunotherapy

Dear Colleagues,
The use of immune checkpoint inhibitors targeting programmed cell death protein 1 (PD-1) and/or cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) has dramatically improved the prognosis of patients with metastatic melanoma; however, despite this significant shift in the landscape of melanoma treatment and outcomes, a notable proportion of patients with advanced melanoma do not respond to immune checkpoint inhibitors due to the primary (innate) and secondary (acquired) resistance caused by complex interactions between various intrinsic and extrinsic factors in the tumor microenvironment. Many studies have provided deeper insights into the underlying molecular mechanisms driving resistance and identified critical pathways as well as genetic alterations that contribute to the development of resistance to immune checkpoint inhibitors. The objective of this Special Issue is to cover all aspects of resistance mechanisms to immune checkpoint inhibitors in melanoma while focusing on the recent insights into clinically relevant molecular interactions between the immune system and tumor cells. We discussed the current state of possible methods with which to restore sensitivity to therapy with immune checkpoint inhibitors, the importance of the identification of novel biomarkers for predicting treatment responses, and advances in the development of tailored as well as optimized therapeutic strategies for patients with advanced melanoma.
