Dear Colleagues,
Protein S is a vitamin K-dependent glycoprotein that plays a central role in the regulation of hemostasis through multiple anticoagulant mechanisms, including its function as a cofactor for activated protein C, direct inhibition of Factor IXa, and enhancement of tissue factor pathway inhibitor (TFPI) activity. Beyond its classical role in coagulation, accumulating evidence over the past decades has highlighted its broader biological significance, extending into inflammation, immune regulation, and cancer biology. Protein S exerts anti-inflammatory effects by modulating cytokine production and facilitating the clearance of apoptotic cells through TAM receptor signaling pathways, thereby maintaining vascular and tissue homeostasis.
Dysregulation or deficiency of Protein S has long been associated with thrombotic disorders; however, emerging studies suggest that its imbalance may also contribute to chronic inflammatory states and tumor progression. In the tumor microenvironment, Protein S and its related signaling pathways appear to influence cell survival, immune evasion, and metastasis, underscoring its potential as both a biomarker and therapeutic target.
This Special Issue aims to bring together cutting-edge research and comprehensive reviews that explore the multifaceted roles of Protein S across coagulation, inflammation, and cancer. By integrating insights from basic science and clinical studies, we hope to advance understanding and inspire new translational approaches in this rapidly evolving field.
