Special Issue Information

Dear Colleagues,

Gliomas remain the most aggressive primary brain tumors, yet traditional histopathological approaches, which have long represented the backbone of diagnosis, often fail to explain the striking heterogeneity in prognosis and treatment responses among tumors with similar microscopic features. Today, these limitations are being addressed through rapid advances in molecular insights, which have transformed how we stratify gliomas, predict outcomes, and guide treatment decisions.

A pivotal turning point came with the 2016 WHO Classification of Tumours of the Central Nervous System, which, for the first time, integrated key molecular markers (e.g., IDH mutation, 1p/19q codeletion) into histology, redefining subtypes such as “oligodendroglioma, IDH-mutant and 1p/19q-codeleted” and resolving longstanding diagnostic ambiguities. This framework was further refined in the 2021 WHO update, which incorporated DNA methylation profiling and additional critical markers (e.g., TERT promoter mutation, EGFR amplification) to enable even more precise subtype stratification. These advances have not only clarified subtype-specific biological behaviors but also laid the foundation for precision diagnostics, directly linking molecular features to prognosis and eligibility for targeted therapies. With the rapid evolution of multi-omics technology, new molecular classification systems and biomarkers are continuing to emerge, opening up fresh avenues to improve patient care.

This Special Issue, “Molecular Classification and Precision Diagnostics in Glioma: Recent Advances”, will report the latest advances in this field, covering topics from novel molecular classification schemas and prognostic/therapeutic biomarkers to innovative molecular testing methods. We warmly welcome the submission of original research articles, comprehensive reviews, and technical papers that address new glioma molecular subtypes, the discovery of actionable biomarkers, developments in molecular testing technologies, or strategies to overcome current challenges in translating molecular classification to clinical practice.
