Dear Colleagues,
The tumor microenvironment contains various inter-reacting components, including infiltrating immune cells, stromal and endothelial cells, soluble factors, and extracellular matrix proteins. Profiling the immune components of this complex network is now mandatory to develop novel immunotherapeutic targets and new predictive biomarkers. Single-cell technologies, including single-cell RNA transcriptomics, multiparameter flow cytometry, and computational techniques have revolutionized cancer immunology. The advancement of these technics in the field of innate immunity now allows for the deep immune-profiling of different tumor-infiltrating innate immune cell types, capturing their heterogeneity and dynamic interactions with other components of the tumor microenvironment. There is growing evidence that different innate immune cell types might determine the outcome of cancer immunosurveillance and patient survival. Innate immune cells such as macrophages and cytotoxic natural killer (NK) cells and unconventional gamma delta (gd) T-cells might contribute to tumor elimination through adaptive immune activation or direct tumor cell killing. Furthermore, research into innate immune checkpoint inhibitors and the generation of "universal" donor cells for cell-based adoptive therapies represent only a few examples of many therapeutic strategies investigating the application of innate immunity in cancer immunotherapy.
Therefore, the main goal of this Special Issue is to provide a forum to discuss the progress of innate immune complexity and delve into innate mechanisms and therapeutic strategies targeting innate immune regulators.
