Dear Colleagues,
The tumor microenvironment (TME) plays a pivotal role in tumor progression, metastasis, immune evasion, and therapeutic resistance. Despite the recent success of immunotherapies and molecularly targeted agents, the TME remains a formidable barrier to conventional cancer therapies, largely due to its complex and heterogeneous nature. To overcome the physical and biological hurdles posed by the TME, innovative bioengineering strategies to target and reprogram the TME are urgently needed. As the field moves towards precision medicine, the integration of intelligent materials, advanced nanotechnology, and data-driven design offers immense opportunities to transform cancer treatment paradigms. This Special Issue aims to present and disseminate the most recent advances related to TME engineering and targeted therapeutic delivery. The Issue will focus on three interconnected, cutting-edge directions: (1) advanced TME-responsive materials for enhanced targeted drug delivery; (2) engineering platforms for the remodeling of the TME for enhanced anti-cancer therapy; and (3) the application of artificial intelligence (AI) and omics data in guiding the design of TME engineering and targeted delivery systems. We welcome contributions on a variety of bioengineering platforms, including inorganic and organic nanoparticles, biomaterials, oncolytic viruses, extracellular vesicles, hydrogels, and scaffolds, which target, respond to, or reprogram multiple TME signatures, including physicochemical, metabolic, and immune aspects. In this Special Issue, original research articles and reviews are welcome. Topics of interest for publication include, but are not limited to, the following:
- Intelligent TME-responsive materials (e.g., pH, enzyme, or hypoxia-responsive) for improved tumor targeting and spatiotemporally controlled drug release.
- Biomimetic delivery systems and engineered extracellular vesicles for homologous targeting and immune evasion.
- Genetically engineered oncolytic viruses armed with immunomodulatory genes for synergistic tumor therapy.
- Nanomedicine and biomaterials for immune microenvironment modulation and immunotherapy potentiation.
- Nanoplatforms targeting tumor metabolic reprogramming (e.g., lactate modulation) to re-educate the immune microenvironment.
- Vascular normalization and stroma degradation strategies for enhanced drug penetration in solid tumors.
- Machine learning-assisted rational design and inverse screening of nano-carriers for precision delivery.
- Integration of single-cell omics and bioinformatics for identifying novel TME targets and personalized therapeutic strategies.
