Cover Story: Box C/D small nucleolar RNAs (C/D snoRNAs) represent an ancient family of small non-coding RNAs that are classically viewed as housekeeping guides for the 2′-O-methylation of ribosomal RNA in Archaea and Eukaryotes. However, an extensive set of studies now reveals the unexpected diversity of the expression and maturation of C/D snoRNAs, of the regulation of C/D snoRNP biogenesis and of the C/D snoRNP composition, partners and targets. Consequently, the need for an important effort of molecular dissection has emerged, as currently too few mechanisms are in hand to understand this overt variety. Here, we present the data and point out how these advances are necessary for a renewed understanding of C/D snoRNA functions ranging from classical ones (white left part of the figure) to novel ones (red right part of the figure).
