Cover Story: Protein–lipid interactions play crucial roles in membrane protein function but remain challenging to study due to membrane complexity. Molecular dynamics (MD) simulations offer detailed insights, complementing experimental approaches. Our study focuses on YidC, an E. coli membrane insertase critical for protein folding and integration. Using MD simulations, rare lipid fenestration events were captured, where lipids penetrate YidC's vestibule, revealing lipid-mediated regulation of protein insertion. Lipids open YidC’s greasy slide and enhance the flexibility of its C1 domain, vital for substrate recruitment. These findings advance the understanding of membrane protein dynamics and lipid’s regulatory roles. View this paper
