Background/Objectives: The detection of ovarian cancer remains challenging due to the lack of reliable serum biomarkers that reflect malignant transformation rather than mere tumor presence. We developed a novel biotest using an immortalized human fallopian tube epithelial cell line (TY), which exhibits anchorage-independent growth (AIG) in response to cancer-associated serum factors. Methods: Sera from ovarian and breast cancer patients, non-cancer controls, and ID8 ovarian cancer-bearing mice were tested for AIG-promoting activity in TY cells. Results: TY cells (passage 96) effectively distinguished cancer sera from controls (68.50 ± 2.12 vs. 17.50 ± 3.54 colonies, p < 0.01) and correlated with serum CA125 levels (r = 0.73, p = 0.03) in ovarian cancer patients. Receiver operating characteristic (ROC) analysis showed high diagnostic accuracy (AUC = 0.85, cutoff: 23.75 colonies). The AIG-promoting activity was mediated by HGF/c-MET and IGF/IGF-1R signaling, as inhibition of these pathways reduced phosphorylation and AIG. In an ID8 mouse ovarian cancer model, TY-AIG colonies strongly correlated with tumor burden (r = 0.95, p < 0.01). Conclusions: Our findings demonstrate that the TY cell-based AIG assay is a sensitive and specific biotest for detecting ovarian cancer and potentially other malignancies, leveraging the fundamental hallmark of malignant transformation. This discussion explores the (dis)engagement of Jamaican RastafarI from the party political process, using RastafarI elder Mortimo Planno’s notion of “politics without a party” as a strand shaping and tying together the multiple threads of the exploration. The discussion examines how RastafarI has engaged with partisan politics/political parties from Independence (1962) until today. It highlights the differing ways of approaching politics among Rastas, including the minority, who have entered representative politics in a bid to [as yet unsuccessfully] change the tribal and compromised state of Jamaican politics. The decentralized nature of the RastafarI movement allows for diverse expressions of RastafarI political thought and action, but can present challenges for unified political mobilization on a large scale. Nonetheless, with or without direct partisan involvement, RastafarI has adapted and re-presented itself in response to changes in the local and global context, thus becoming a potent political force. So, despite this general lack of engagement with “statical” matters, RastafarI is and continues to be a significant political movement on several fronts, through movements, music, and symbols rather than traditional electoral routes. Increased Oxygen Treatment in the Fermentation Process Improves the Taste and Liquor Color Qualities of Black Tea

Black tea is widely consumed worldwide, and its characteristic taste and color result from fermentation, where polyphenols are enzymatically oxidized to generate major pigments, including theaflavins (TFs), thearubigins (TRs), and theabrownins (TBs). This study investigated the effects of increased oxygen treatment during fermentation on the flavor attributes and chemical properties of Congou black tea. Fresh tea leaves (variety “Fuyun 6”) were subjected to four oxygen treatments: 0 h (CK), 1 h (TY-1h), 2 h (TY-2h), and 3 h (TY-3h), with oxygen supplied at 8.0 L/min. Sensory evaluation revealed that oxygen-treated samples exhibited tighter and deeper-colored leaves, a redder liquor, fuller taste, and a sweeter fragrance compared with CK. Chromatic analysis showed significant increases in redness (a*) and luminance (L*), alongside reduced yellowness (b*), indicating enhanced liquor color. Chemical analyses demonstrated elevated levels of TFs, TRs, and TBs in oxygen treatments, with TRs showing the most pronounced increase. Non-targeted metabolomics identified 2318 non-volatile and 761 volatile metabolites, highlighting upregulated flavonoids, phenolic acids, and lipids, and downregulated catechins and tannins, which collectively contributed to improved taste and aroma. Optimal results were achieved with 2–3 h of oxygen treatment, balancing pigment formation and sensory quality. These findings can provide a scientific basis for optimizing oxygen conditions in black tea fermentation to improve product quality. Full article

Cholesterol accumulation plays a significant role in the pathogenesis of metabolic-dysfunction-associated steatotic liver disease (MASLD), yet changes in cholesterol homeostasis in MASLD remain insufficiently investigated. This study aimed to examine alterations in cholesterol synthesis and absorption by measuring plasma levels of endogenous cholesterol precursors (as markers of synthesis) and phytosterols (as indicators of absorption). A total of 124 MASLD patients and 43 healthy individuals were included. Our results showed higher plasma concentrations of lathosterol in the MASLD group (p = 0.006), in parallel with comparable concentrations of desmosterol (p = 0.472) and all analyzed phytosterols in both groups. Correlation analysis showed that both lathosterol and desmosterol were positively associated with non-invasive hepatic steatosis indices: FLI, HSI, and TyG index (p < 0.01, p < 0.01, and p < 0.05, respectively). Multivariate linear regression further confirmed that these synthesis markers remained significant predictors of FLI (p = 0.010), HSI (p = 0.013), and TyG index (p = 0.002), even after adjusting for other relevant variables. These findings indicate that MASLD is associated with a shift in cholesterol homeostasis towards enhanced endogenous cholesterol synthesis. Full article

Objectives: This study sought to assess the impact of diabetes and hypertension on wound healing and recovery in orthopedic patients, with an emphasis on laboratory correlations. Materials and Methods: This study included 67 orthopedic patients, divided into a geriatric group (n = 49, ≥65 years) and a control group (n = 18). Clinical and laboratory assessments were performed at admission and discharge. Data were analyzed statistically. Results: Geriatric patients showed a higher triglyceride glucose-body mass index (TyG-BMI), glucose, cholesterol, C-reactive protein (CRP), interleukin-6 (IL-6), and leukocytes and lower hemoglobin and platelets (PLTs), with poorer healing and well-being. Elevated CRP, IL-6, and urea and decreased protein and hemoglobin persisted in this group. Diabetes improved outcomes in older adults, while hypertension worsened them in younger patients. Favorable outcomes correlated with higher triglycerides, fibrinogen, hemoglobin, and red blood cells (RBCs), while they did not correlate with elevated CRP, IL-6, leptin, urea, creatinine, and white blood cells (WBCs). Conclusions: Key predictors of healing and well-being included CRP, hemoglobin, RBC, and hematocrit in older patients and hypertension, CRP, hemoglobin, and leptin in younger individuals. Age-specific metabolic and inflammatory profiles influence recovery trajectories and may be used to predict problems in both recovery and patients’ well-being. Further research is required to better understand the correlations between these factors. Full article

In this study, we examined the microclimates at eight entrances to a karst system distributed between an elevation of 812 and 906 m in Southern Spain. The karst system, characterised by subvertical open tectonic joints that form narrow shafts, developed on the slope of a mountainous area with a Mediterranean climate and strong chimney effect, resulting in an intense airflow throughout the year. The airflows modify the entrance temperatures, creating a distinctive pattern in each opening that changes with the seasons. The objective of this work is to characterise the outflows and find simple temperature-based parameters that provide information about the karst interior. The entrances were monitored for five years (2017–2022) with temperature–humidity dataloggers at different depths. Other data collected include discrete wind measurements and outside weather data. The most significant parameters identified were the characteristic temperature (Ty), recorded at the end of the outflow season, and the rate of cooling/warming, which ranges between 0.1 and 0.9 °C/month. These parameters allowed the entrances to be grouped based on the efficiency of heat exchange between the outside air and the cave walls, which depends on the rock-boundary geometry. This research demonstrates that simple temperature studies with data recorded at selected positions will allow us to understand geometric aspects of inaccessible karst systems. Dynamic high-airflow cave systems could become a natural source of evidence for climate change and its effects on the underground world. Full article

Background: Metabolic syndrome (MS) is a major public health concern linked to an elevated risk of type 2 diabetes and cardiovascular disease. Simple, reliable screening tools are needed for early identification, especially in working populations. Objective: To compare the diagnostic accuracy of body mass index (BMI), waist-to-height ratio (WtHR), triglyceride–glucose index (TyG), and waist–triglyceride index (WTI) for detecting MS based on NCEP ATP III and IDF criteria in a large cohort of Spanish workers. Methods: This cross-sectional study analyzed data from 386,924 Spanish workers. MS was diagnosed using NCEP ATP III and IDF definitions. The four indexes were evaluated by sex using a receiver operating characteristic (ROC) curve analysis. Area under the curve (AUC), optimal cut-off points, and Youden’s index were calculated. Results: TyG and WTI had the highest AUC values in men (0.911 and 0.901, respectively) for NCEP ATP III-defined MS, while WtHR and WTI achieved the best performance in women (0.955 and 0.953, respectively). WtHR outperformed BMI in all subgroups. Optimal cut-off values were identified according to sex and the definition of MS: TyG (8.95 men, 8.51 women), WtHR (0.54 men, 0.51 women), and WTI (170.6 men, 96.5 women), supporting their practical implementation in occupational health programs. All indexes showed significant discriminatory capacity (p < 0.001). Conclusions: TyG, WtHR, and WTI are more effective than BMI in detecting MS among Spanish workers, with sex-specific patterns. Their ease of use and diagnostic strength support their adoption in occupational health programs for early cardiometabolic risk detection. Full article

Background/Objectives: The consumption of refined carbohydrates has increased globally. It is associated with inflammation and oxidative stress, both recognized as risk factors for cardiovascular disease. This study investigated the effects of a refined carbohydrate-rich diet on the vascular reactivity of rat aorta. Methods: We acclimatized adult male Wistar rats for two weeks and then randomly assigned them to two experimental groups: a control (CT) group and a high-carbohydrate diet (HCD) group. The CT group received standard laboratory chow for 15 days, while the HCD group received a diet composed of 45% sweetened condensed milk, 10% refined sugar, and 45% standard chow. After the dietary exposure period, we evaluated the vascular reactivity of aortic rings, gene expression related to inflammation, superoxide dismutase activity, and biochemical parameters, including cholesterol, triglycerides, fasting glucose, and glucose and insulin tolerance. Results: The results demonstrate a reduction in vascular reactivity caused by endothelial alterations, including increased NO production, which was observed as higher vasoconstriction in the presence of L-NAME and aminoguanidine and upregulation of iNOS gene expression. In addition, increased production of free radicals, such as O2-, was observed, as well as immune markers like MCP-1 and CD86 in the HCD group. Additionally, the HCD group showed an increase in the TyG index, suggesting early metabolic impairment. GTT and ITT results revealed higher glycemic levels, indicating early signs of insulin resistance. Conclusions: These findings indicate that short-term consumption of a refined carbohydrate-rich diet may trigger oxidative stress and endothelial dysfunction, thereby increasing the risk of cardiovascular complications. The introduction of virus resistance genes into traditional tomato varieties offers a strategy to preserve genetic diversity and enhance commercial viability. However, the homozygous presence of these genes has been associated with negative effects on yield and fruit quality. This two-year study evaluated the impact of introducing the Tm-2a, Sw-5 and Ty-1 genes, which are associated with resistance to ToMV, TSWV and TYLCV, respectively, on the agronomic yield, fruit characteristics and metabolic profile of Muchamiel-type cultivars. Four hybrids were obtained by crossing two breeding lines carrying the resistance genes in homozygosis (UMH1139 and UMH1200) with two traditional susceptible varieties (MC1 and MC2). Hybrids matched or exceeded the agronomic performance of their parents. Fruit morphology of the hybrids was similar to traditional parents. The presence of Ty-1 correlated with reduced organic acid concentration, though hybrids exhibited higher levels than the homozygous line, UMH1200. No negative effects on soluble sugars or secondary metabolites were observed. Genotypes carrying resistance genes, breeding lines and hybrids exhibited higher flavonoid contents, suggesting a potential role in virus response. Hybrids maintained or improved the bioactive profile of traditional varieties. These findings support the development of Muchamiel-type hybrids that combine the presence of virus resistance genes in heterozygosity with the desirable traits of traditional tomatoes. Full article

Tomato (Solanum lycopersicum) is an essential vegetable crop cultivated worldwide, but its production is highly vulnerable to tomato yellow leaf curl disease (TYLCD), which is transmitted by whiteflies (Bemisia tabaci). Management strategies typically focus on controlling either the virus or its vector. This study evaluates the effectiveness of multi-disease and insect-resistant tomato lines, developed by the World Vegetable Center (WorldVeg), which integrate Ty-1/Ty-3 genes for virus resistance and WF2-10 and WF3-09 genes for whitefly resistance. Virus accumulation, whitefly settling behavior, and adult mortality were assessed among multi-resistant lines, a Ty-resistant line, a whitefly-resistant line, and a susceptible check using preference bioassays, controlled inoculation experiments, and acylsugar quantification. Multi-resistant lines exhibited significantly higher acylsugar concentrations, reduced whitefly preference for settling, and increased whitefly adult mortality. Additionally, these lines displayed less severe disease symptoms and lower virus accumulation over time than Ty-resistant, whitefly-resistant, and susceptible controls. These findings highlight the superior efficacy of combined virus and vector resistance in mitigating tomato yellow leaf curl Thailand virus (TYLCTHV) transmission. This research underscores the importance of integrated genetic resistance as a key element of sustainable integrated pest management strategies, offering an environmentally friendly solution for safeguarding global tomato production. Background: Childhood obesity is a significant global health concern. Butyrylcholinesterase (BChE) has been shown to play a role in lipid metabolism. This study aimed to assess BChE activity, obesity-related and lipid-related indices, and dyslipidemia in obese and non-obese children, and to investigate the associations of these parameters with obesity among Thai children. Methods: The study included 661 Thai children, consisting of 338 with obesity and 323 with a normal weight. Anthropometric measurements, metabolic parameters, obesity- and lipid-related indices, and BChE activity were evaluated. Results: The obese group exhibited significantly higher BChE activity and obesity-related and lipid-related indices compared to the non-obese group (p < 0.01). Additionally, metabolic parameters—including glucose levels, triglyceride-glucose (TyG) index, and TyG-related indices—as well as the lipid profile, which included triglycerides (TG), non-high-density lipoprotein cholesterol (non-HDL-C), and very-low-density lipoprotein cholesterol (VLDL-C), were all significantly elevated in the obese group (p < 0.01). Obesity was associated with dyslipidemia (p < 0.01). Moreover, BChE activity showed a positive correlation with obesity-related and lipid-related indices, along with several metabolic parameters (p < 0.002). The upper stratum of BChE activity (OR = 5.356), the non-HDL-C/HDL-C ratio (OR = 2.185), and the TG/HDL-C ratio (OR = 1.703) were found to be effective in evaluating and predicting the risk of obesity, even after adjusting for potential covariates (p < 0.01). Conclusions: These findings indicate a significant relationship between obesity and increased BChE activity, lipid-related indices, and dyslipidemia in Thai children. Therefore, changes in BChE activity may be considered a factor associated with obesity, enhancing its potential as a marker for obesity assessment. Full article

Remotely Sensing Phytoplankton Size Structure in the Mediterranean Sea: Insights from In Situ Data and Temperature-Corrected Abundance-Based Models

Since the mid-1980s, the Mediterranean Sea’s surface and deeper layers have warmed at unprecedented rates, with recent projections identifying it as one of the regions most impacted by rising global temperatures. Metrics that characterize phytoplankton abundance, phenology and size structure are widely utilized as ecological indicators that enable a quantitative assessment of the status of marine ecosystems in response to environmental change. Here, using an extensive, updated in situ pigment dataset collated from numerous past research campaigns across the Mediterranean Sea, we re-parameterized an abundance-based phytoplankton size class model that infers Chl-a concentration in three phytoplankton size classes: pico- (<2 μm), nano- (2–20 μm) and micro-phytoplankton (>20 μm). Following recent advancements made within this category of size class models, we also incorporated information of sea surface temperature (SST) into the model parameterization. By tying model parameters to SST, the performance of the re-parameterized model was improved based on comparisons with concurrent, independent in situ measurements. Similarly, the application of the model to remotely sensed ocean color observations revealed strong agreement between satellite-derived estimates of phytoplankton size structure and in situ observations, with a performance comparable to the current regional operational datasets on size structure. The proposed conceptual regional model, parameterized with the most extended in situ pigment dataset available to date for the area, serves as a suitable foundation for long-term (1997–present) analyses on phytoplankton size structure and ecological indicators (i.e., phenology), ultimately linking higher trophic level responses to a changing Mediterranean Sea. The Beidou Global System (BDS-3) innovatively achieves autonomous navigation using inter-satellite links (ISL) across the entire constellation, but it still faces challenges such as the limitations of the prior constraint orbital accuracy and the overall constellation rotation. The gradual availability of satellite laser ranging (SLR) data, with advantages of high precision and no ambiguous parameters, can provide new ideas for solving the current problem. This work firstly deduces the mathematical model for orbit determination by combining inter-satellite links and the introduced satellite laser ranging observations, then designs orbit determination experiments with different prior orbit constraints and different observation data, and finally evaluates the impacts of the prior orbits and the introduction of SLR observations from two dimensions: orbit accuracy and constellation rotation. The experimental results using one month of measured data show the following: (1) There is good consistency among different days, and the accuracy of the prior orbits affects the performance of the orbit determination and the consistency. Compared with broadcast ephemerides, using precise ephemerides as prior constraints significantly improves the consistency, and the orbit accuracy can be increased by about 75%. (2) The type of observation data affects the performance of the orbit determination. Introducing SLR observations can improve the orbit accuracy by approximately 13% to 26%. (3) Regardless of whether broadcast ephemerides or precise ephemerides are used as prior constraints, the constellation translation and rotation still exist after introducing SLR observations. Among the translation parameters, TX is the largest, followed by TY, and TZ is the smallest; all three rotation parameters (RX, RY, and RZ) show relatively large values, which may be related to the limited number of available satellite laser ranging stations during this period. (4) After considering the constellation translation and rotation, the orbit accuracy under different prior constraints remains at the same level. The statistical root mean square error (RMSE) indicates that the orbit accuracy of inclined geosynchronous orbit (IGSO) satellites in three directions is better than 20 cm, while the accuracy of medium earth orbit (MEO) satellites in along-track, cross-track, and radial directions is better than 10 cm, 8 cm, and 5 cm, respectively. Triglyceride–Glucose-Based Anthropometric Indices for Predicting Incident Cardiovascular Disease: Relative Fat Mass (RFM) as a Robust Indicator

Background/Objectives: The triglyceride–glucose (TyG) index is a recognized marker for cardiovascular disease (CVD) risk linked to insulin resistance. Combining TyG with anthropometric indicators (AIs) may improve risk prediction, but the comparative value of different AIs, including novel ones like Relative Fat Mass (RFM), is unclear. This study aimed to identify which combination of TyG and AIs has the strongest association with incident CVD in a middle-aged and elderly Chinese cohort. Methods: In this prospective study, we evaluated the association between the cumulative average of TyG combined with eight AIs (TyG-AIs) and incident CVD, heart disease, and stroke. Using data from 5192 participants in the China Health and Retirement Longitudinal Study (CHARLS), we used multivariable logistic regression to compare the predictive value of these composite indices. Results: During follow-up, 1382 (26.6%) participants developed CVD. After full adjustment, the TyG index alone was only significantly associated with stroke. In contrast, most TyG-AIs showed stronger associations with all outcomes. Notably, the index combining TyG with Relative Fat Mass (TyG-RFM) exhibited the most robust associations with total CVD (OR = 2.236), heart disease (OR = 1.679), and stroke (OR = 3.288) when comparing the highest to lowest quartiles. Conclusions: Cumulative average TyG-AIs, particularly TyG-RFM, demonstrated more robust associations with incident CVD than the TyG index alone. The TyG-RFM index shows promise as a valuable tool to improve cardiovascular risk stratification, especially for identifying at-risk non-obese individuals. Full article

Background/Objective: Shift work has been increasingly associated with adverse metabolic outcomes, including insulin resistance (IR), a key contributor to cardiometabolic diseases. However, few large-scale studies have explored the association between shift work and validated IR indices across sociodemographic and lifestyle variables, stratified by sex. To investigate the relationship between shift work and four surrogate markers of insulin resistance—TyG índex, TyG-BMI, METS-IR (Metabolic score for insulin resistance), and SPISE-IR (Single-Point insulin Sensitivity estimator)—in a large Spanish working population and to assess how sociodemographic and behavioral factors modify these associations. Methods: This cross-sectional study included 53,053 employed adults (31,753 men and 21,300 women) across various regions and labor sectors in Spain. Participants were classified as shift or non-shift workers based on their work schedules. Data were collected during routine occupational health assessments and included anthropometric, clinical, biochemical, and behavioral variables. IR indices were calculated using standard formulas. Statistical analyses included t-tests, chi-square tests, and multivariable logistic regression, with stratification by sex. Results: Shift workers exhibited significantly higher levels of TyG, TyG-BMI, and METS-IR, and lower SPISE values compared to non-shift workers (all p < 0.001). These differences persisted after stratification by sex, age, education, and lifestyle factors. Logistic regression analyses confirmed that shift work independently predicted high IR risk across all indices, with adjusted ORs ranging from 1.49 to 1.89. Physical inactivity, low adherence to the Mediterranean diet, and alcohol consumption were the strongest modifiable predictors. Men exhibited a higher burden of IR than women across all indices. Conclusions: Shift work is independently associated with elevated insulin resistance in both men and women. These findings emphasize the importance of workplace health interventions targeting physical activity, diet, and substance use, particularly in populations engaged in non-standard work schedules.
