VD9136 Positively Modulates the Pathogenicity of Verticillium dahliae to Cotton

Histidine triad (HIT) family proteins contain a conserved histidine triad motif and play key roles in fungal metabolism and pathogenicity. This study focused on VD9136, a member of the HIT family in Verticillium dahliae, aiming to elucidate its biological function and mechanism underlying its role in cotton pathogenesis. A systematic investigation of the VD9136 gene in V. dahliae was conducted using bioinformatics analysis, gene knockout, genetic complementation, and pathogenicity assays. The results showed that VD9136 protein consists of 136 amino acids and is a stable, neutral, and weakly hydrophilic protein that lacks transmembrane domains and signal peptides; it is localized to the extracellular space via a non-classical secretion pathway. Its secondary structure is predominantly composed of α-helices and random coils. Phylogenetic analysis revealed that VD9136 is closely related to VliHIT, a homologous protein from V. longisporum, the pathogen responsible for Verticillium wilt in rapeseed. The promoter region of VD9136 contains multiple cis-acting elements, including light-responsive, hormone-responsive, and stress-responsive elements, indicating that its transcription may be regulated by multiple signaling pathways. VD9136 was significantly upregulated during the early stage of cotton infection (6–24 h post-inoculation). Pathogenicity assays demonstrated that V. dahliae knockout mutants lacking VD9136 exhibited a significant reduction in virulence, as evidenced by a lower disease index, decreased fungal biomass within plant tissues, and attenuated vascular browning in cotton plants. The pathogenic phenotype was successfully restored in genetic complementation strains. This study identified VD9136 as a key regulatory factor in the pathogenic process of V. dahliae, and its loss of function reduces the pathogenicity of V. dahliae. The findings provide a theoretical basis for elucidating the pathogenic mechanism of cotton Verticillium wilt and for developing corresponding prevention and control strategies. Full article

This work presents the Theory of Spacetime Impedance (TSI), a phenomenological framework in which the vacuum is modeled as a distributed reactive medium with an effective RLC structure. At the classical level, the vacuum is characterized by permeability, μ0, permittivity, ε0, and impedance, Z0, so that the speed of light follows from the vacuum’s constitutive reactive properties. The TSI introduces a reactive–dissipative term, RH, as an effective mechanism associated with irreversibility, decoherence, and entropy production, providing a physical basis for the arrow of time. At the quantum level, TSI incorporates a quantum RLC triad associated with the electron, defined by quantum inductance, LK, quantum capacitance, CK, and von Klitzing resistance, RK. When normalized by the Compton wavelength, these quantities admit a direct comparison with μ0 and ε0, identifying the fine-structure constant as an impedance scaling factor between classical and quantum regimes. Within this unified reactive picture, inductive, capacitive, and resistive responses are respectively associated with gravitation, electromagnetism, and thermodynamic irreversibility, offering a complementary bridge across quantum, relativistic, and macroscopic domains. Oncometabolites and hypoxia-regulated exosomes orchestrate hypoxia-inducible factor (HIF)–driven macrophage reprogramming across chronic cardiometabolic and oncologic conditions. In type 2 diabetes (T2D) and obesity, regional hypoxia in expanding white adipose tissue (WAT) reconfigures macrophage immunometabolism and chemokine signaling, recruits C-C chemokine receptor 2 (CCR2+) monocytes, and skews adipose-tissue macrophages toward M1-like programs that sustain low-grade inflammation and blunt the physiological M1-to-M2 transition during wound repair. In atherosclerotic plaques, lipid-core hypoxia stabilizes HIF-1α, amplifies nuclear factor kappa-light-chain-enhancer of activated B cells/reactive oxygen species (NF-κB/ROS) signaling, increases matrix metalloproteinase-2/-9 (MMP-2/-9) release, and reduces ATP-binding cassette transporter A1 (ABCA1)-mediated cholesterol efflux, weakening the fibrous cap. In tumors, poorly perfused niches accumulate lactate and succinate, which act as paracrine cues. Lactate activates PKA/cAMP pathways and promotes immunosuppressive tumor-associated macrophages (TAMs), whereas succinate signals through succinate receptor 1 (SUCNR1) to reinforce HIF-1α–dependent transcription and M2-like programming. In parallel, hypoxia-regulated exosomes deliver microRNAs such as miR-301a-3p, which suppress phosphatase and tensin homolog (PTEN) and activate PI3Kγ, thereby augmenting immunosuppression and programmed death-ligand 1 (PD-L1) expression. Clinically, this hypoxia–oncometabolite–exosome triad links oxygen debt with macrophage state, plaque destabilization, impaired wound repair, and tumor immune escape. Translational entry points include selective HIF-2α inhibition, phosphoinositide 3-kinase gamma (PI3Kγ) blockade, SUCNR1 targeting, and exosome-based miRNA modulation, while a biomarker panel comprising HIF-1α, vascular endothelial growth factor A (VEGF-A), and MMP-9 offers a pragmatic readout of hypoxia burden, macrophage programming, and therapeutic response. We conducted a focused narrative review (PubMed, Scopus, Web of Science; English; 2003–2025), prioritizing mechanistic and translational studies on hypoxia–HIF, lactate/succinate, and hypoxia-regulated exosomes across T2D, atherosclerosis, and cancer. The role of metapragmatics in maintaining interactional coherence and achieving intersubjectivity has been variously underscored in the last three decades. In particular, raising metapragmatic awareness has become increasingly salient in research on intercultural communication and foreign/second language teaching. However, the topic has not been hitherto discussed in connection with heritage languages, and this is a gap that the present paper aims to fill. Based on interviews with Greek Melburnians who belong (in triads or dyads) to the same family but to different generations, a typology of metapragmatic awareness indicators encountered in the data is presented. Quantitative examination of one type of indicators—those oriented towards the addressee—indicates a decrease in their use across three generations. Similarly, examination of the variants of second-person pronouns and/or verb endings (the T/V distinction) brought to the fore alternations in the T and V forms, indicative of linguistic insecurity, as well as an increasing preference for the informal variants across three generations. The qualitative analysis of extracts from the interviews shed further light on the insecurity regarding the T/V distinction. Overall, the results point to changes in the communicative style of Greek Melburnians, namely away from positive politeness features (typical of the Greek society) towards English interactional norms, and the fostering of a hybrid communicative style—in alignment with their hybrid identities. It is suggested that politeness issues be integrated into the teaching of Greek as a heritage language. Full article

Green technologies are actively being used to produce nanosized zinc oxide, which is in demand for water purification processes to remove pollutants. Despite the success of the green synthesis of ZnO nanoparticles, no scientific approach exists for selecting plant extracts to produce nanoparticles with the desired properties. This study shows that the antioxidant activity of the plant extracts used is a key parameter influencing the properties of the resulting ZnO nanoparticles. This conclusion is based on the results of nanoparticle synthesis with the use of various plant extracts. The antioxidant activity of the extracts increases in the following order: plum–gooseberry–black currant–strawberry–sea buckthorn. The synthesized ZnO nanoparticles were characterized by UV–visible spectroscopy, transmission electron microscopy, and energy-dispersive X-ray spectroscopy. The catalytic properties of ZnO nanoparticles were tested under the degradation of a synthetic methylene blue dye after exposure to UV light. We found that with an increase in the AOA of plant extracts, the size of the nanoparticles decreases, while their photocatalytic activity increases. The smallest (d = 13 nm), most uniform in size (polydispersity index 0.1), and most catalytically active ZnO nanoparticles with a small band gap (2.85 eV) were obtained using the sea buckthorn extract with the highest AOA, pH 10 of the reaction mixture and 0.1 M Zn(CH3COO)2∙2H2O as a precursor salt. ZnO nanoparticles synthesized in the sea buckthorn extract demonstrated the highest dye photodegradation efficiency (96.4%) compared with other nanoparticles. The established patterns demonstrate the “antioxidant activity–size–catalytic activity” triad can be considered as a practical guide for obtaining ZnO nanoparticles of a given size and with given properties for environmental remediation applications. Background: Genetic studies have found that a germline BRCA1 gene mutation is the origin of highly increased cancer risk. Clinical studies have suggested that increased cancer risk in type-2 diabetes may be attributed to unhealthy lifestyle factors and bad habits. Purpose: Patients with either BRCA1 gene mutation or type-2 diabetes similarly exhibit increased cancer risk, insulin resistance, and fertility disorders. It was suggested that these three alterations derive from a common genomic failure, and its recognition may shed light on the unsolved secret of cancer. Results: (1) Germline mutations on ESR1, BRCA1, and CYP19A genes encoding estrogen receptor alpha (ERα), genome safeguarding BRCA1 protein, and CYP19 aromatase enzyme cause genomic instability. BRCA1 and ESR1 gene mutations specifically cause breast cancer, while error in the CYP19A gene leads to cancers in the endometrium, ovaries, and thyroid. (2) ERα, BRCA1, and CYP19 aromatase proteins are transcription factors creating the crucial DNA stabilizer circuit driven by estrogen regulation. Liganded ERα drives a second regulatory circuit to also control cell proliferation, in partnership with various growth factors. In a third regulatory circuit, liganded ERα drives cellular glucose supply in close interplay with insulin, IGF-1, and glucose transporters. (3) Impaired expression or activation of each transcription factor of the triad leads to defective estrogen signaling and endangers regular cell proliferation, insulin sensitivity, and fertility. (4) Impaired estrogen signaling caused by either genetic or lifestyle factors alarms the hypothalamus, which issues neural and hormonal commands throughout the body to restore estrogen signaling. (5) When the compensatory actions cannot restore estrogen signaling, the breakdown of genomic regulation leads to cancer initiation. (6) Lifestyle factors that upregulate estrogen signaling decrease cancer risk, while downregulating estrogen signaling increases it. Conclusions: Increased cancer risk, insulin resistance, and infertility all originate from defective estrogen signaling. Full article

Façade systems in hot, high-insolation climates are required to simultaneously mitigate cooling loads, ensure high-quality daylight, and, where feasible, harvest on-site electricity demands that are often in tension. This study assesses and compares two efficient façade strategies for a fully glazed office prototype in Hail, Saudi Arabia: semi-transparent photovoltaic glazing (STPV10–30%VLT) and parametrically tuned split louvers (18 depth–spacing–tilt configurations). Using a unified parametric workflow (Rhino/Grasshopper), Radiance/honeybee for daylight metrics, ASHRAE-55 heat-balance metrics for thermal comfort, and EnergyPlus for end-use and PV yield, to evaluate annual and solstice performance across cardinal orientations. Optimized split louvers maintained UDI300–1000lx and effectively suppress glare, but incur substantial lighting-energy penalties. In contrast, STPV with 10–20% VLT broadly meets daylight targets while strongly reducing cooling and lighting demand, delivering whole-façade energy savings of up to 50–94% depending on orientation, but could be net-neutral to slightly adverse north 3% when daylight penalties dominate. Thermal comfort responses mirrored these trends: summer PMV was near 0 to +0.5 for both systems, with winter under-heating evident when solar gains are strongly suppressed. Overall, in hot-arid, highly glazed offices, STPV of 10–20%VLT provides the most balanced triad of daylight quality, cooling reduction, and net energy benefit, while optimized louvers excel where glare control is paramount but require careful daylight-control integration. A significant body of research has documented the aggressive and antisocial tendencies of individuals with dark triad personality traits. Although the prevalence of dark personalities in online environments is often criticized, there is a need to explore effective strategies to mitigate or stop such behaviors. This study aims to shed light on the intriguing phenomenon of “Giants on the Internet, cowards in real life” by examining the longitudinal relationship between dark triad traits, bullying victimization, and cyberbullying. Study 1 revealed that adolescents tend to display heightened tendencies towards cyberbullying after experiencing real-life victimization. Study 2, on the other hand, showed a reduction in cyberbullying behaviors among those with dark triad traits following experiences of bullying. These findings highlight the paradoxical mechanisms underlying the relationship between bullying victimization, dark triad traits, and cyberbullying. Consequently, this study introduces the new label, “From Online Aggression to Offline Silence,” to describe this dynamic. Full article

Given the continued existence, personal success, and organizational damage of ‘bad’ leadership (e.g., with Bankman-Fried, Lay, Welch, Stonecipher), how do we—in academia—do more good than harm in studying and informing others about it? We address that question to improve leadership research more generally. We do so through a dialectic inquiry approach where we join current conversations about teaching and research related to the private benefits and the public costs produced by ‘bad’ leadership. As part of the dialectic, we offer a new model of the phenomenon—where ‘bad’ behaviors can be used to win a contest for leadership where the leader can then act badly or not. Furthermore, we inquire as to the efficacy of how the literature diagnoses ‘bad’ leadership (at either stage), suggests treatments, and attempts to impact practice. We ground our inquiry in two foundational definitions of ‘bad’-ness—one based on psychology’s dark triad, and one based on economics’ self-interest-seeking with guile agent. We critique a relevant, representative set of recent examples to make our points about the significant issues involved in the three elements, across two stages and two definitions. This antithesis-like critique provides the grounds for a set of synthesizing recommendations to improve leadership studies, in light of practical (and often unnecessarily self-imposed) constraints. The Dark Triad (Machiavellianism, narcissism, and psychopathy) has been traditionally associated with externalizing behaviors and a lack of empathy. However, emerging evidence suggests that these traits also relate to internal emotional vulnerabilities (such as depression, alexithymia, and anhedonia). This study aimed to examine the association between Dark Triad traits and emotional variabilities (alexithymia and anhedonia) in the general population, with a particular focus on sex differences. A total of 492 French-speaking adults completed a battery of validated self-report measures online, including the SD3fr, BDI-II, TAS-20, and PAS. Comparative and multivariate analyses were conducted separately for males and females. High SD3 scores were associated with greater depression, alexithymia (especially difficulty in identifying feelings), and anhedonia in the overall sample. These associations remained significant after adjusting for depression severity. Sex differences emerged: in females, high Dark Triad traits were strongly linked to emotional dysfunction, while no such associations were observed in males. These findings support the presence of sex-specific emotional correlates of the Dark Triad, particularly in females, where Machiavellianism and psychopathy were linked to emotional distress. Clinical implications are discussed in light of hormonal, sociocultural, and emotional regulation differences. Full article

In informal economic contexts, migrant entrepreneurs have been extolled as highly resilient and adaptable. This study critically investigates the adverse psychological foundations inherent in such enterprises, focusing on how dark triad personality traits emerge in the leadership orientations of marginal migrant entrepreneurs. Following a qualitative ethnographic approach, this research engaged 10–15 migrant employees through participant observation, field notes, and semi-structured interviews in an informal economic context. Thematic analysis revealed five dominant patterns: narcissistic leadership with entitlement and emotional disrespect; Machiavellian behavior of manipulation and deception; psychopathic detachment in emotional callousness; absence of light triad actions such as empathy, humility, and selflessness; and moral disengagement through rationalizations such as “everyone does it” or system blame. Migrant business owners prefer to rationalize their exploitative acts as being necessary for economic survival, thus legitimizing immoral conduct and suppressing moral self-regulation. The findings indicate that marginality not only drives entrepreneurial innovation, but also has the potential to create exploitative inclinations that are institutionally and morally unchecked. Solving this issue requires not only mere psychological awareness, but also systematic reforms that foster ethical robustness and emotional sensitivity. This study ultimately asserts the need to reframe migrant entrepreneurship discourse, including both ethical and psychological accountability. Full article

Flourishing refers to a state of optimal well-being and life satisfaction, while the Light Triad traits—Kantianism, Humanism, and Faith in Humanity—represent a prosocial and benevolent approach to interpersonal relationships. This study explores the relationship between these traits and Flourishing among adults in the Honduras Central District. A quantitative, non-experimental, cross-sectional methodology with a relational scope was used in a sample of 715 Honduran adults. Results indicate that Kantianism, Humanism, and Faith in Humanity significantly and positively explain the participants’ levels of Flourishing. Men and women reported similar levels of Flourishing, Faith in Humanity, and Kantianism. However, women scored significantly higher in Humanism. This study highlights the role of the Light Triad traits in predicting Flourishing, emphasizing the need for educational and policy initiatives that foster traits like Kantianism, Humanism, and Faith in Humanity for personal and societal development. Background and Objectives: The primary purpose of the article was to examine the relationship between dark and light personality traits in university students enrolled in the Faculty of Sport Sciences. Materials and Methods: Data were collected from 518 students (208 female, 310 male) with an average age of 20.89 (±2.25). The Short Dark Triad and the Light Triad Scales were used. Harman’s single-factor analysis minimized measurement error, and various statistical methods assessed the effect of gender and age on personality traits. Results: Results indicated a positive correlation among dark personality traits, while light personality traits exhibited more complex relationships. Gender and age were found to significantly influence certain personality traits. Conclusions: This study contributes to the sports literature by exploring the role of demographic variables in personality formation. Full article

In this paper, we present a new class of extended oscillators in light of chaos theory. It is based on dynamical complex systems built on the concept of self-describing with a stopping criterion process. We offer an effective studying approach with a specific focus on learning, provoking students’ thinking through the triad of enigmatics–creativity–acmeology. Dynamic processes are the basis of mathematical modeling; thus, we can reach the goal of the above-mentioned triad by the proposed differential systems. The results we derive strongly confirm the presence of symmetry in the outcomes of the proposed models. We suggest a stochastic approach to structuring the proposed dynamical systems by modeling the coefficients that drive them by some discrete probability distribution that exhibits symmetry or asymmetry. We propose specific tools for researching the behavior of these systems. Full article

Mesenchymal stem cells (MSCs) have shown great potential for the treatment of liver injuries, and the therapeutic efficacy greatly depends on their homing to the site of injury. In the present study, we detected significant upregulation of hepatocyte growth factor (HGF) in the serum and liver in mice with acute or chronic liver injury. In vitro study revealed that upregulation of miR-9-5p or miR-221-3p promoted the migration of human MSCs (hMSCs) toward HGF. Moreover, overexpression of miR-9-5p or miR-221-3p promoted hMSC homing to the injured liver and resulted in significantly higher engraftment upon peripheral infusion. hMSCs reduced hepatic necrosis and inflammatory infiltration but showed little effect on extracellular matrix (ECM) deposition. By contrast, hMSCs overexpressing miR-9-5p or miR-221-3p resulted in not only less centrilobular necrosis and venous congestion but also a significant reduction of ECM deposition, leading to obvious improvement of hepatocyte morphology and alleviation of fibrosis around central vein and portal triads. Further studies showed that hMSCs inhibited the activation of hepatic stellate cells (HSCs) but could not decrease the expression of TIMP-1 upon acute injury and the expression of MCP-1 and TIMP-1 upon chronic injury, while hMSCs overexpressing miR-9-5p or miR-221-3p led to further inactivation of HSCs and downregulation of all three fibrogenic and proinflammatory factors TGF-β, MCP-1, and TIMP-1 upon both acute and chronic injuries. Overexpression of miR-9-5p or miR-221-3p significantly downregulated the expression of α-SMA and Col-1α1 in activated human hepatic stellate cell line LX-2, suggesting that miR-9-5p and miR-221-3p may partially contribute to the alleviation of liver injury by preventing HSC activation and collagen expression, shedding light on improving the therapeutic efficacy of hMSCs via microRNA modification.
