Stutzerimonas, a genus newly separated from the Pseudomonadaceae family in 2022, has attracted considerable attention due to its diverse metabolic capabilities and environmental adaptability. However, the mechanisms underlying its sulfur-oxidizing capacity and survival strategies in extreme environments remain poorly understood. Clarifying potential sulfur-oxidizing microbial groups contributes to a more accurate understanding of energy flow and elemental cycling in hydrothermal ecosystems. In this study, we isolated and identified a sulfur-oxidizing strain, designated 381-2T, from sediments in the Tianxiu hydrothermal field of the northwest Indian Ocean, and proposed it as a new species of Stutzerimonas. Physiological characterizations demonstrated that strain 381-2T could oxidize thiosulfate to tetrathionate and encoded the key sulfur oxidation gene tsdA. Cultivation with sulfide minerals showed that strain 381-2T could influence sulfide mineral weathering through metabolic activities, such as pH regulation, and potentially promote the reprecipitation of metal ions on the microbial surface. Comparative genomic analysis of 322 Stutzerimonas genomes further revealed the widespread presence of the tsdA gene and metal resistance genes, suggesting potential adaptive strategies for survival in hydrothermal environments. This study expands the understanding of Stutzerimonas species and provides insights into their ecological roles in hydrothermal systems. Full article

A series of sulfur dissolving tetrathionate ionic liquids (ILs), featuring imidazolium and pyridinium cationic heads, have been prepared and characterized along with their chloride IL precursors. A novel synthetic approach for the preparation of the proposed tetrathionate ILs has been introduced and successfully tested in the current work, yielding the desired compounds in quantitative yield and high purity, offering a significant advancement over the traditional Volynskii–Smolyaninov reaction. The presented method addresses key challenges of the traditional approach, solving the issues deriving from the influence of the IL cation on the reaction outcome and the unpredictability of the formed polythionate species. The solubility of elemental sulfur in the considered tetrathionate ILs has been investigated at various temperatures, providing good preliminary evidence of the suitability of these ILs as convenient and effective sulfur solubilizing media readily available on the field in “sour” gas extraction plants. Furthermore, the use of ILs instead of traditional organic solvents in operative conditions represents a noteworthy safety improvement due to their lower flammability and volatility. Finally, interesting results were obtained studying binary mixtures of organic solvents and ILs, with cooperative effects or salting-out effects being observed in relation to the type of solvent used. The problem of the foul odor caused by H2S in livestock farms has become a major complaints. In this study, optimal sulfur-oxidizing bacteria (SOB) strains were screened from dairy farm wastewater and the adjacent soil for odor treatment. The strains and physiological functions were determined by identification and genome comparison, and the optimal operating conditions were determined by experiments under different conditions. The identification results showed that the strain that had the highest homology with Halomonas mongoliensis was Halomonas sp. AEB2. The comparative genomic results showed that the average nucleotide identity and DNA–DNA hybridization value were 95.8% and 68.6%, respectively. The optimization results were as follows: sodium succinate-carbon (10 g/L) and ammonium chloride-nitrogen (0.07 g/L). The optimal operating conditions were as follows: seeding rate 4%, temperature 30 °C, stirring speed 90 rpm, and pH 8. The oxidation products of AEB2 were mainly elemental sulfur and tetrathionate, and the metabolic pathway of AEB2 was constructed accordingly. This study suggests a feasible path to reduce H2S emissions from dairy farms, and it provides theoretical support for the restoration of livestock environment and sustainability. Full article

Tetrathionate hydrolase (TTH) is a key enzyme for the oxidation of reduced inorganic sulfur compounds (RISCs) with the S4I pathway, which is distributed in autotrophic or facultative autotrophic sulfur-oxidizing bacteria and archaea. In this study, the enzyme TTHMc from the acidothermophilic archaeon Metallosphaera cuprina Ar-4T, encoded by mcup_1281 and belonging to the pyrroloquinoline quinone (PQQ) family, has been shown to possess tetrathionate hydrolysis activity. The molecular mass of the single subunit of TTHMc was determined to be 57 kDa. TTHMc is proved to be located in the cytoplasm, periplasmic space, and membrane, and the activity of them accounted for 72.3%, 24.0%, and 3.7% of the total activity. Optimal activity was observed at temperatures above 95 °C and pH 6.0, and the kinetic constants Km and Vmax were 0.35 mmol/L and 86.3 μmol/L, respectively. The presence of 0.01 mol/L Mg2+ enhances the activity of TTHMc, while 0.01 mol/L Ca2+ inhibits its activity. The hydrolysis of tetrathionate (TT) by TTHMc results in the production of thiosulfate, pentathionate, and hexathionate. This study represents the first description of TTH in the genus Metallosphaera, providing new theoretical insights into the study of sulfur-oxidizing proteins in acidothermophilic archaea. Perylenediimides (PDIs) are composed of a central perylene ring, on which are grafted two imide groups at the peri positions. Thionated PDIs are characterized by the substitution of one or more oxygen atoms of these imide functions with sulfur atoms. This structural modification alters the electronic properties with a redshift of the optical absorption accompanied by modification of the charge transport characteristics compared to their non-thionated counterparts. These properties make them suitable candidates for applications in optoelectronic devices, such as organic light-emitting diodes and organic photovoltaics. Moreover, the presence of sulfur atom(s) can favor the promotion of reactive oxygen species production for photodynamic and photothermal therapies. These thionated PDIs can be synthesized through the post-functionalization of PDIs by using a sulfurizing reagent. Nevertheless, the main drawbacks remain the difficulties in adjusting the degree of thionation and obtaining tri- and tetrathionated PDIs. Up to now, this thionation reaction has been described almost exclusively using Lawesson’s reagent. In the current study, we present our first investigations into an alternative reagent to enhance selectivity and achieve a greater degree of thionation. The association of phosphorus pentasulfide with hexamethyldisiloxane (Curphey’s reagent) clearly demonstrated higher reactivity compared with Lawesson’s reagent to attain multi-thionated PDIs. Microbial diversity that thrives in the deep subsurface remains largely unknown. In this work, we present the characterization of Citrobacter sp. T1.2D-1, isolated from a 63.6 m-deep core sample extracted from the deep subsurface of the Iberian Pyrite Belt (IPB). A genomic analysis was performed to identify genes that could be ecologically significant in the IPB. We identified all the genes that encoded the formate–hydrogen lyase and hydrogenase-2 complexes, related to hydrogen production, as well as those involved in glycerol fermentation. This is particularly relevant as some of the substrates and byproducts of this process are of industrial interest. Additionally, we conducted a phylogenomic study, which led us to conclude that our isolate was classified within the Citrobacter telavivensis species. Experimentally, we verified the strain’s ability to produce hydrogen from glucose and glycerol and, thus, of performing dark fermentation. Moreover, we assessed the activity of the nitrate and tetrathionate reductase complexes and the isolate’s ability to tolerate high concentrations of heavy metals, especially Zn. These results suggest that C. telavivensis T1.2D-1 can play a role in the carbon, hydrogen, iron, nitrogen, and sulfur cycles that occur in the deep subsurface of the IPB, making it a candidate worthy of further study for possible biotechnological applications. Sulfur oxidation stands as a pivotal process within the Earth’s sulfur cycle, in which Acidithiobacillus species emerge as skillful sulfur-oxidizing bacteria. They are able to efficiently oxidize several reduced inorganic sulfur compounds (RISCs) under extreme conditions for their autotrophic growth. This unique characteristic has made these bacteria a useful tool in bioleaching and biological desulfurization applications. Extensive research has unraveled diverse sulfur metabolism pathways and their corresponding regulatory systems. The metabolic arsenal of the Acidithiobacillus genus includes oxidative enzymes such as: (i) elemental sulfur oxidation enzymes, like sulfur dioxygenase (SDO), sulfur oxygenase reductase (SOR), and heterodisulfide reductase (HDR-like system); (ii) enzymes involved in thiosulfate oxidation pathways, including the sulfur oxidation (Sox) system, tetrathionate hydrolase (TetH), and thiosulfate quinone oxidoreductase (TQO); (iii) sulfide oxidation enzymes, like sulfide:quinone oxidoreductase (SQR); and (iv) sulfite oxidation pathways, such as sulfite oxidase (SOX). This review summarizes the current state of the art of sulfur metabolic processes in Acidithiobacillus species, which are key players of industrial biomining processes. Furthermore, this manuscript highlights the existing challenges and barriers to further exploring the sulfur metabolism of this peculiar extremophilic genus. Full article

The effect of nitrogen and phosphorus availability on the growth of Sulfolobus metallicus was analyzed. This archaeon was subjected to a series of nitrogen and phosphorus limitation conditions to determine their effects on growth. The results indicate that Sulfolobus metallicus showed a relationship between one of the intermediate oxidation products (tetrathionate) and cell concentration during the exponential growth phase in the absence of nitrogen. Furthermore, significant differences were found in the specific growth rates under different scenarios with ammonia and phosphorus limitation, with values of 0.048 h−1 in the ammonia limitation case. The biomass substrate yield obtained was 0.107 gcel·g S−1. Meanwhile, in the absence of phosphorus, the specific growth rate was 0.017 h−1, and the substrate to biomass yield was 0.072 gcel·g S−1. The results indicate that the ability of Sulfolobus metallicus to bio-oxidize H2S depends on the availability of such nutrients (nitrogen and phosphorus), which affect cellular growth and the types of products generated. This, in turn, influences the oxidation process of various sulfur compounds, resulting in changes in the predominant products formed and the final oxidation of sulfate ions. Full article

Extracellular polymeric substances (EPS) produced by microorganisms play a crucial role in various bioprocesses, including bioleaching. The microbial leaching of metal sulfides requires an initial cell attachment, which is facilitated by EPS. These substances are mixtures of polysaccharides, proteins, lipids, and other compounds, and their composition and properties can vary depending on the species, growth conditions, and environmental factors. Despite the significance of iron/sulfur oxidizing species in biomining processes, the knowledge of the interfacial processes between thermoacidophilic archaeal species and mineral surfaces is limited. This study examines the cell surface characteristics and EPS produced by an Acidianus strain. The research was conducted using microscopic techniques, Zeta-potential measurements, spectrophotometric methods, Fourier transform infrared spectroscopy, and fluorescence lectin-binding analysis. The results suggest that non-soluble substrates, such as sulfur or pyrite, induce changes in cell surface structures, including the presence of cell appendages, wider cell envelopes, higher hydrophobicities, and increased EPS production, compared to cells grown with soluble substrates such as tetrathionate or ferrous iron. The EPS mainly consist of proteins and carbohydrates, including glucose, manose, N-acetylgalactosamine, and N-acetylglucosamine residues. This study contributes to a better understanding of the relationship between thermophilic archaea and mineral surfaces in biomining processes. Full article

Acidithiobacillus spp. are prevalent in acid mine drainage, and they have been widely used in biomining for extracting nonferrous metals from ores. The osmotic stress generated by elevated concentrations of inorganic ions is a severe challenge for the growth of Acidithiobacillus spp. in the bioleaching process; however, the adaptation mechanism of these bacteria to high osmotic pressure remains unclear. In this study, bioinformatics analysis indicated that the osmotic stress response two-component system EnvZ-OmpR is widely distributed in Acidithiobacillus spp., while OmpRs from Acidithiobacillus spp. exhibited a far more evolutionary relationship with the well-studied OmpRs in E. coli and Salmonella typhimurium. The growth measurement of an Acidithiobacillus caldus (A. caldus) ompR-knockout strain demonstrated that OmpR is essential in the adaptation of this bacterium to high osmotic stress. The overall impact of OmpR on the various metabolic and regulatory systems of A. caldus was revealed by transcriptome analysis. The OmpR binding sequences of differentially expressed genes (DEGs) were predicted, and the OmpR box motif in A. caldus was analysed. The direct and negative regulation of EnvZ-OmpR on the tetrathionate-metabolic (tetH) cluster in A. caldus was discovered for the first time, and a co-regulation mode mediated by EnvZ-OmpR and RsrS-RsrR for the tetrathionate intermediate thiosulfate-oxidizing (S4I) pathway in this microorganism was proposed. This study reveals that EnvZ-OmpR is an indispensable regulatory system for the ability of A. caldus to cope with high osmotic stress and the significance of EnvZ-OmpR on the regulation of sulfur metabolism in A. caldus adapting to the high-salt environment. Polycyclic aromatic hydrocarbons are versatile building blocks for conjugated materials and can be applied in molecular electronics. Pyrenes are known as the best organic chromophores, and pyrene itself is known as an electron donor. Likewise, quinones are promising electrode materials for lithium-ion batteries. The calculations were performed for pyrene-4,5,9,10-tetrathione, pyrene-4,5,9,10-tetraselenone and pyrene-4,5,9,10-tetratellurone, and the results were compared with those for pyrene-4,5,9,10-tetraone. The results obtained indicate that the sulfur derivative is a suitable candidate for further experimental studies since, although selenium and tellurium compounds present better prospects than 4,5,9,10-tetraoxopyrene, they require the improvement of available synthetic techniques or even the discovery of new ones. Tetrathionates have been found in significantly high concentrations in recycled process waters from massive sulphide ore processing plants. These polythionates react with xanthate added to flotation pulps thus reducing xanthate dosages in solution potentially affecting flotation performance. The current study focused on the effect of the tetrathionate-xanthate reaction on sulphide mineral recoveries. Ore dissolution studies confirmed the generation of tetrathionates by copper-lead-zinc ores. In 20 min, the tetrathionates consumed more than half of the xanthate in solution at pH 7. Rest potential measurements and Fourier transform infrared spectroscopy (FTIR) showed that the degree of collector-mineral interactions of xanthate and both galena and chalcopyrite was greatly reduced in the presence of a 2000 mg/L tetrathionate solution. Microflotation tests showed that chalcopyrite recovery was less sensitive to tetrathionates as indicated by small changes in mineral recoveries. Galena was sensitive to the action of tetrathionates on the mineral surface as the galena recovery significantly declined when floated with xanthate as a collector in both a 500 mg/L tetrathionate solution and a 2000 mg/L tetrathionate solution. These fundamental results lay a sound base on which more discussion into the significance and the effect of tetrathionates on flotation performance of sulphide ores can be developed. Full article

Thiosulfate effluents are generated in the photography and radiography industrial sectors, and in a plant in which thiosulfates are used to recover the gold and silver contained in ores. Similar effluents also containing thiosulfate are those generated from the petrochemical, pharmaceutical and pigment sectors. In the future, the amounts of these effluents may increase, particularly if the cyanides used in the extraction of gold and silver from ores are substituted by thiosulfates, or if the same happens to electronic scrap or in metallic coating processes. This paper reports a study of the oxidation of thiosulfate, with oxygen using copper (II) as a catalyst, at a pH between 4 and 5. The basic idea is to avoid the formation of tetrathionate and polythionate, transforming the thiosulfate into sulfate. The nature of the reaction and a kinetic study of thiosulfate transformation, by reaction with oxygen and Cu2+ at a ppm level, are determined and reported. The best conditions were obtained at 60 °C, pH 5, with an initial concentration of copper of 53 ppm and an oxygen pressure of 1 atm. Under these conditions, the thiosulfate concentration was reduced from 1 g·L−1 to less than 20 ppm in less than three hours. Full article

Sulfur Oxygenase Reductase (Sor) in the Moderately Thermoacidophilic Leaching Bacteria: Studies in Sulfobacillus thermosulfidooxidans and Acidithiobacillus caldus

The sulfur oxygenase reductase (Sor) catalyzes the oxygen dependent disproportionation of elemental sulfur, producing sulfite, thiosulfate and sulfide. Being considered an “archaeal like” enzyme, it is also encoded in the genomes of some acidophilic leaching bacteria such as Acidithiobacillus caldus, Acidithiobacillus thiooxidans, Acidithiobacillus ferrivorans and Sulfobacillus thermosulfidooxidans, among others. We measured Sor activity in crude extracts from Sb. thermosulfidooxidans DSM 9293T. The optimum temperature for its oxygenase activity was achieved at 75 °C, confirming the “thermophilic” nature of this enzyme. Additionally, a search for genes probably involved in sulfur metabolism in the genome sequence of Sb. thermosulfidooxidans DSM 9293T was done. Interestingly, no sox genes were found. Two sor genes, a complete heterodisulfidereductase (hdr) gene cluster, three tetrathionate hydrolase (tth) genes, three sulfide quinonereductase (sqr), as well as the doxD component of a thiosulfate quinonereductase (tqo) were found. Seven At. caldus strains were tested for Sor activity, which was not detected in any of them. We provide evidence that an earlier reported Sor activity from At. caldus S1 and S2 strains most likely was due to the presence of a Sulfobacillus contaminant. Full article
