Insulin-regulated glucose uptake is a central mechanism in maintaining systemic glucose homeostasis, primarily occurring in skeletal muscle and adipose tissue. This process relies on the insulin-stimulated translocation of the glucose transporter, GLUT4, from specialized intracellular compartments, known as GLUT4 storage vesicles (GSVs), to the plasma membrane. Disruption of this pathway is a hallmark of insulin resistance and a key contributor to the pathogenesis of type 2 diabetes. Recent advances have provided critical insights into both the insulin signalling cascades and the complex biogenesis, as well as the trafficking and fusion dynamics of GSVs. This review synthesizes the current understanding of the molecular mechanisms governing GSV mobilization and membrane fusion, highlighting key regulatory nodes that may become dysfunctional in metabolic disease. By elucidating these pathways, we propose new therapeutic avenues targeting GSV trafficking to improve insulin sensitivity and combat type 2 diabetes. Full article

The apicoplast is a highly specialized organelle in the biosynthesis of essential metabolites in most of the apicomplexan protozoa. This organelle is surrounded by four layers of membranes. However, the molecular mechanisms mediating transmembrane transport are not yet fully understood. In this study, we conducted a phenotypic analysis to investigate the role of a major facilitator superfamily transporter (TgApMFS1) in the survival of the parasite. The results indicated that TgApMFS1 is critical for the survival of Toxoplasma gondii in cell culture conditions. Further analysis indicated that these transporters are crucial for the biogenesis of organelles and the metabolic processes of parasite. Full article

Previous reviews have examined specific facets of Geographic Information Systems (GIS) in transportation planning, such as transit-focused applications and open source geospatial tools. However, this study offers the first systematic, PRISMA-guided longitudinal evaluation of GIS integration in transportation planning, spanning thematic domains, data models, methodologies, and outcomes from 2004 to 2024. This study addresses this gap through a longitudinal analysis of GIS-based transportation research from 2004 to 2024, adhering to PRISMA guidelines. By conducting a mixed-methods analysis of 241 peer-reviewed articles, this study delineates major trends, such as increased emphasis on sustainability, equity, stakeholder involvement, and the incorporation of advanced technologies. Prominent domains include land use–transportation coordination, accessibility, artificial intelligence, real-time monitoring, and policy evaluation. Expanded data sources, such as real-time sensor feeds and 3D models, alongside sophisticated modeling techniques, enable evidence-based, multifaceted decision-making. However, challenges like data limitations, ethical concerns, and the need for specialized expertise persist, particularly in developing regions. Future geospatial innovations should prioritize the responsible adoption of emerging technologies, inclusive capacity building, and environmental justice to foster equitable and efficient transportation systems. This review highlights GIS’s evolution from a supplementary tool to a cornerstone of data-driven, sustainable urban mobility planning, offering insights for researchers, practitioners, and policymakers to advance transportation strategies that align with equity and sustainability goals. The Golgi of goblet cells represents a specialized machine for mucin glycosylation. This process occurs in a specialized form of the secretory pathway, which remains poorly examined. Here, using high-resolution three-dimensional electron microscopy (EM), EM tomography, serial block face scanning EM (SBF-SEM) and immune EM we analyzed the secretory pathway in goblet cells and revealed that COPII-coated buds on the endoplasmic reticulum (ER) are extremely rare. The ERES vesicles with dimensions typical for the COPII-dependent vesicles were not found. The Golgi is formed by a single cisterna organized in a spiral with characteristics of the cycloid surface. This ribbon has a shape of a cup with irregular perforations. The Golgi cup is filled with secretory granules (SGs) containing glycosylated mucins. Their diameter is close to 1 µm. The cup is connected with ER exit sites (ERESs) with temporal bead-like connections, which are observed mostly near the craters observed at the externally located cis surface of the cup. The craters represent conus-like cavities formed by aligned holes of gradually decreasing diameters through the first three Golgi cisternae. These craters are localized directly opposite the ERES. Clusters of the 52 nm vesicles are visible between Golgi cisternae and between SGs. The accumulation of mucin, started in the fourth cisternal layer, induces distensions of the cisternal lumen. The thickness of these distensions gradually increases in size through the next cisternal layers. The spherical distensions are observed at the edges of the Golgi cup, where they fuse with SGs and detach from the cisternae. After the fusion of SGs located just below the apical plasma membrane (APM) with APM, mucus is secreted. The content of this SG becomes less osmiophilic and the excessive surface area of the APM is formed. This membrane is eliminated through the detachment of bubbles filled with another SG and surrounded with a double membrane or by collapse of the empty SG and transformation of the double membrane lacking a visible lumen into multilayered organelles, which move to the cell basis and are secreted into the intercellular space where the processes of dendritic cells are localized. These data are evaluated from the point of view of existing models of intracellular transport. Full article

Firstly, this paper reviews the fundamental theories of solid surface wettability and contact angle hysteresis. Subsequently, it further introduces four typical wettability-engineered surfaces with low hysteresis (superhydrophobic, superamphiphobic, super-slippery, and liquid-like smooth surfaces). Finally, it focuses on the latest research progress in the field of droplet manipulation on open planar surfaces with engineered wettability. To achieve droplet manipulation, the core driving forces primarily stem from natural forces guided by bioinspired gradient surfaces or the regulatory effects of external fields. In terms of bioinspired self-propelled droplet movement, this paper summarizes research inspired by natural organisms such as desert beetles, cacti, self-aligning floating seeds of emergent plants, or water-walking insects, which construct bioinspired special gradient surfaces to induce Laplace pressure differences or wettability gradients on both sides of droplets for droplet manipulation. Moreover, this paper further analyzes the mechanisms, advantages, and limitations of these self-propelled approaches, while summarizing the corresponding driving force sources and their theoretical formulas. For droplet manipulation under external fields, this paper elaborates on various external stimuli including electric fields, thermal fields, optical fields, acoustic fields, and magnetic fields. Among them, electric fields involve actuation mechanisms such as directly applied electrostatic forces and indirectly applied electrocapillary forces; thermal fields influence droplet motion through thermoresponsive wettability gradients and thermocapillary effects; optical fields cover multiple wavelengths including near-infrared, ultraviolet, and visible light; acoustic fields utilize horizontal and vertical acoustic radiation pressure or acoustic wave-induced acoustic streaming for droplet manipulation; the magnetic force acting on droplets may originate from their interior, surface, or external substrates. Based on these different transport principles, this paper comparatively analyzes the unique characteristics of droplet manipulation under the five external fields. Finally, this paper summarizes the current challenges and issues in the research of droplet manipulation on the open planar surfaces and provides an outlook on future development directions in this field. Articulated Tugs and Barges (ATBs) are increasingly recognised for their effectiveness in transporting chemicals, petroleum, bulk goods, and containers, primarily due to their exceptional flexibility and fuel efficiency. Recent projections indicate that the ATB market is on track for significant growth, which is expected to lead to an increase in the annual growth rate from 2025 to 2032. This study aims to analyse the current advancements in ATB technology and provide insights into the ATB fleet and the systems that connect tugboats and barges. Furthermore, it highlights the advantages of this transportation system, especially regarding its role in enhancing energy efficiency within the maritime transport sector. Currently, there is limited information available in the public domain about ATBs compared to other commercial vessels. The analysis reveals that much of the required information for modern ATB design is not accessible outside specialised design companies. The study also focuses on conceptual design aspects, which include the main dimensions, articulated connections, propulsion systems, and machinery, concluding with an evaluation of economic viability. Special emphasis is placed on defining the main dimensions, which is a critical part of the complex design process. In this context, the ratios of length to beam (L/B), beam to draft (B/D), beam to depth (B/T), draft to depth (T/D), and power to the number of tugs cubed (Pw/N3) are established as design control parameters in the conceptual design phase. This aspect underscores the novelty of the present study. Additionally, the economic viability is analysed in terms of both CAPEX (capital expenditures) and OPEX (operational expenditures). While CAPEX does not significantly differ between the methods used in different types of commercial ships, OPEX should account for the unique characteristics of ATB vessels. Full article

Numerical Study of Gas Dynamics and Condensate Removal in Energy-Efficient Recirculation Modes in Train Cabins

Maintaining the required relative humidity values in the vehicle cabin is an important HVAC task, along with considerations related to the temperature, velocity, air pressure and noise. Deviation from the optimal values worsens the psycho-physiological state of the driver and affects the energy efficiency of the train. In this study, a model of liquid film formation on and removal from various cabin surfaces was constructed using the fundamental Navier–Stokes hydrodynamic equations. A special transport model based on the liquid vapor diffusion equation was used to simulate the air environment inside the cabin. The evaporation and condensation of surface films were simulated using the Euler film model, which directly considers liquid–gas and gas–liquid transitions. Numerical results were obtained using the RANS equations and a turbulence model by means of the finite volume method in Ansys CFD. Conjugate fields of temperature, velocity and moisture concentration were constructed for various time intervals, and the dependence values for the film thicknesses on various surfaces relative to time were determined. The verification was conducted in comparison with the experimental data, based on the protocol for measuring the microclimate indicators in workplaces, as applied to the train cabin: the average ranges encompassed temperature changes from 11% to 18%, and relative humidity ranges from 16% to 26%. Comparison with the results of other studies, without considering the phase transition and condensation, shows that, for the warm mode, the average air temperature in the cabin with condensation is 12.5% lower than without condensation, which is related to the process of liquid evaporation from the heated walls. The difference in temperature values for the model with and without condensation ranged from −12.5% to +4.9%. We demonstrate that, with an effective mode of removing condensate film from the window surface, including recirculation modes, the energy consumption of the climate control system improves significantly, but this requires a more accurate consideration of thermodynamic parameters and relative humidity. Thus, considering the moisture condensation model reveals that this variable can significantly affect other parameters of the microclimate in cabins: in particular, the temperature. This means that it should be considered in the numerical modeling, along with the basic heat transfer equations. Full article

Background/Objectives: Magnetic resonance imaging (MRI) is a powerful, non-invasive diagnostic tool capable of capturing detailed anatomical and physiological information. MRI contrast agents enhance image contrast but, especially linear gadolinium-based compounds, have been associated with safety concerns. This has prompted interest in alternative contrast agents. Manganese-based contrast agents offer a promising substitute, owing to manganese’s favorable magnetic properties, natural biological role, and strong T1 relaxivity. This review aims to critically assess the structure, mechanisms, applications, and challenges of manganese-based contrast agents in MRI. Methods: This review synthesizes findings from preclinical and clinical studies involving various types of manganese-based contrast agents, including small-molecule chelates, nanoparticles, theranostic platforms, responsive agents, and controlled-release systems. Special attention is given to pharmacokinetics, biodistribution, and safety evaluations. Results: Mn-based agents demonstrate promising imaging capabilities, with some achieving relaxivity values comparable to gadolinium compounds. Targeted uptake mechanisms, such as hepatocyte-specific transport via organic anion-transporting polypeptides, allow for enhanced tissue contrast. However, concerns remain regarding the in vivo release of free Mn2+ ions, which could lead to toxicity. Preliminary toxicity assessments report low cytotoxicity, but further comprehensive long-term safety studies should be carried out. Conclusions: Manganese-based contrast agents present a potential alternative to gadolinium-based MRI agents pending further validation. Despite promising imaging performance and biocompatibility, further investigation into stability and safety is essential. Additional research is needed to facilitate the clinical translation of these agents. Full article

As sessile organisms, plants adapt to environmental challenges through flexible developmental and physiological programs. Hormones play a central role in this adaptability, integrating environmental signals into coordinated responses that regulate growth and stress tolerance. Comparative studies across photosynthetic lineages reveal that several core hormone functions are remarkably conserved, despite major evolutionary changes in hormone perception, biosynthesis, metabolism, and transport. This conservation suggests that plant hormones have played a pivotal evolutionary role—not only preserving essential biological functions but also enabling increased complexity in plant form and function. A similar dual role is observed in evolutionary endocrinology in animals, where hormones contribute to the emergence and regulation of complex traits. We propose that hormones such as cytokinins, auxins, brassinosteroids, strigolactones, and abscisic acid originated as metabolic derivatives closely tied to core physiological functions essential for survival and reproduction, including reproductive success, nutrient sensing, and dehydration tolerance. Over time, these compounds were progressively integrated into increasingly sophisticated regulatory networks, where they now serve as central coordinators and key targets of evolutionary selection. This model advances our understanding of hormone evolution by providing a structured framework to interpret the persistence, specialization, and integration of plant hormones across evolutionary timescales. The presence of sea ice threatens low ice-class vessels’ navigation safety in the Arctic, and traditional Navigation Risk Assessment Models based on sea ice parameters have been widely used to guide safe passages for ships operating in ice regions. However, these models mainly rely on empirical coefficients, and the accuracy of these models in identifying sea ice navigation risk remains insufficiently validated. Therefore, under the binary classification framework, this study used Automatic Identification System (AIS) data along the Northeast Passage (NEP) as positive samples, manual interpretation non-navigable data as negative samples, a total of 10 machine learning (ML) models were employed to capture the complex relationships between ice conditions and navigation risk for Polar Class (PC) 6 and Open Water (OW) vessels. The results showed that compared to traditional Navigation Risk Assessment Models, most of the 10 ML models exhibited significantly improved classification accuracy, which was especially pronounced when classifying samples of PC6 vessel. This study also revealed that the navigability of the East Siberian Sea (ESS) and the Vilkitsky Strait along the NEP is relatively poor, particularly during the month when sea ice melts and reforms, requiring special attention. The navigation risk output by ML models is strongly determined by sea ice thickness. These findings offer valuable insights for enhancing the safety and efficiency of Arctic maritime transport. Photocuring, including photopolymerization and photocrosslinking, has emerged as a transformative manufacturing paradigm that enables the precise, rapid, and customizable fabrication of advanced battery components. This review first introduces the principles of photocuring and vat photopolymerization and their unique advantages of high process efficiency, non-contact fabrication, ambient-temperature processing, and robust interlayer bonding. It then systematically summarizes photocured battery components, involving electrolytes, membranes, anodes, and cathodes, highlighting their design strategies. This review examines the impact of photocured materials on the battery’s properties, such as its conductivity, lithium-ion transference number, and mechanical strength, while examining how vat-photopolymerization-derived 3D architectures optimize ion transport and electrode–electrolyte integration. Finally, it discusses current challenges and future directions for photocuring-based battery manufacturing, emphasizing the need for specialized energy storage resins and scalable processes to bridge lab-scale innovations with industrial applications. Cold environments challenge the structural and functional integrity of membrane proteins, requiring specialized adaptations to maintain activity under low thermal energy. Here, we investigate the molecular basis of cold tolerance in the peptide transporter PepT1 from the Antarctic icefish (Chionodraco hamatus, ChPepT1) using molecular dynamics simulations, binding free energy calculations (MM/GBSA), and dynamic network analysis. We compare ChPepT1 to its human ortholog (hPepT1), a non-cold-adapted variant, to reveal key features enabling psychrophilic function. Our simulations show that ChPepT1 displays enhanced global flexibility, particularly in domains adjacent to the substrate-binding site and the C-terminal domain (CTD). While hPepT1 loses substrate binding affinity as temperature increases, ChPepT1 maintains stable peptide interactions across a broad thermal range. This thermodynamic buffering results from temperature-sensitive rearrangement of hydrogen bond networks and more dynamic lipid interactions. Importantly, we identify a temperature-responsive segment (TRS, residues 660–670) within the proximal CTD that undergoes an α-helix to coil transition, modulating long-range coupling with transmembrane helices. Dynamic cross-correlation analyses further suggest that ChPepT1, unlike hPepT1, reorganizes its interdomain communication in response to temperature shifts. Our findings suggest that cold tolerance in ChPepT1 arises from a combination of structural flexibility, resilient substrate binding, and temperature-sensitive interdomain dynamics. These results provide new mechanistic insight into thermal adaptation in membrane transporters and offer a framework for engineering proteins with enhanced functionality in extreme environments. This study describes a quite special and interesting atmospheric event characterized by the simultaneous presence of fresh and aged smoke layers. These peculiar conditions occurred on 16 July 2024 at the CNR-IMAA atmospheric observatory (CIAO) in Potenza (Italy), and represent an ideal case for the evaluation of the impact of aging and transport mechanisms on both the optical and microphysical properties of biomass burning aerosol. The fresh smoke was originated by a local wildfire about 2 km from the measurement site and observed about one hour after its ignition. The other smoke layer was due to a wide wildfire occurring in Canada that, according to backward trajectory analysis, traveled for about 5–6 days before reaching the observatory. Synergetic use of lidar, ceilometer, radar, and microwave radiometer measurements revealed that particles from the local wildfire, located at about 3 km a.s.l., acted as condensation nuclei for cloud formation as a result of high humidity concentrations at this altitude range. Optical characterization of the fresh smoke layer based on Raman lidar measurements provided lidar ratio (LR) values of 46 ± 4 sr and 34 ± 3 sr, at 355 and 532 nm, respectively. The particle linear depolarization ratio (PLDR) at 532 nm was 0.067 ± 0.002, while backscatter-related Ångström exponent (AEβ) values were 1.21 ± 0.03, 1.23 ± 0.03, and 1.22 ± 0.04 in the spectral ranges of 355–532 nm, 355–1064 nm and 532–1064 nm, respectively. Microphysical inversion caused by these intensive optical parameters indicates a low contribution of black carbon (BC) and, despite their small size, particles remained outside the ultrafine range. Moreover, a combined use of CIAO remote sensing and in situ instrumentation shows that the particle properties are affected by humidity variations, thus suggesting a marked particle hygroscopic behavior. In contrast, the smoke plume from the Canadian wildfire traveled at altitudes between 6 and 8 km a.s.l., remaining unaffected by local humidity. Absorption in this case was higher, and, as observed in other aged wildfires, the LR at 532 nm was larger than that at 355 nm. Specifically, the LR at 355 nm was 55 ± 2 sr, while at 532 nm it was 82 ± 3 sr. The AEβ values were 1.77 ± 0.13 and 1.41 ± 0.07 at 355–532 nm and 532–1064 nm, respectively and the PLDR at 532 nm was 0.040 ± 0.003. Microphysical analysis suggests the presence of larger, yet much more absorbent particles. This analysis indicates that both optical and microphysical properties of smoke can vary significantly depending on its origin, persistence, and transport in the atmosphere. These factors that must be carefully incorporated into future climate models, especially considering the frequent occurrences of fire events worldwide. Full article

Establishing a high-precision passenger flow prediction model is a critical and complex task for the optimization of urban rail transit systems. With the development of artificial intelligence technology, the data-driven technology has been widely studied in the intelligent transportation system. In this study, a neural network model based on the data-driven technology is established for the prediction of passenger flow in multiple urban rail transit stations to enable smart perception for optimizing urban railway transportation. The integration of network units with different specialities in the proposed model allows the network to capture passenger flow data, temporal correlation, spatial correlation, and spatiotemporal correlation with the dual attention mechanism, further improving the prediction accuracy. Experiments based on the actual passenger flow data of Beijing Metro Line 13 are conducted to compare the prediction performance of the proposed data-driven model with the other baseline models. The experimental results demonstrate that the proposed prediction model achieves lower MAE and RMSE in passenger flow prediction, and its fitted curve more closely aligns with the actual passenger flow data. This demonstrates the model’s practical potential to enhance intelligent transportation system management through more accurate passenger flow forecasting. Full article
