The selectivity of the colorimetric method for Co(II) determination using the nitroso-R-salt (NRS) in samples with complex matrices has been improved. Interferences caused by Cu(II), Fe(II), Fe(III), Mn(II), Al(III) and Ni(II) ions, which were present in the bioleach ate of lithium-ion batteries, have been solved through the sequential addition of masking agents: acetate, fluoride, ethylenediaminetetraacetic acid (EDTA), and strong acids (H2SO4). The absorbance of the NRS-Co(II) complex was typically measured at 525 nm, but it was also studied at 550 nm due to minimal interferences observed at 550 nm. The sequence of the masking agent’s addition showed a significant influence on the interference effect. The optimal sequence was sample, acetate–acetic acid buffer solution with dissolved fluoride, NRS, EDTA and H2SO4. The proposed method demonstrated robust performance at 550 nm, with a relative standard deviation (RSD) around 2%, and good accuracy (RV% around 100%). The limit of detection (LoD) was 0.1 mg L−1 and the limit of quantification (LoQ) was 0.3 mg L−1. The linear range extended up to 15 mg L−1 (R2 = 0.998). Real samples analyzed using the optimized method showed no significant differences when compared to results from atomic absorption spectroscopy, confirming its reliability. Full article

As a core component of industrial robots, the transmission accuracy life (TAL) of rotary vector (RV) reducers constitutes a primary factor determining the high-precision operation of robotic systems. However, current life evaluation methods for RV reducers predominantly rely on conventional bearing strength life calculations, while neglecting its transmission accuracy degradation during operation. To address this limitation, a static analysis model of RV reducers is established, through which a calculation method for transmission accuracy and TAL is presented. Simultaneously, tooth surface and bearing wear models are developed based on Archard’s wear theory. Through coupled analysis of the aforementioned models, the transmission accuracy degradation law of RV reducers is revealed. The results show that during the operation of the RV reducer, the transmission error (TE) maintains relative stability over time, whereas the lost motion (LM) exhibits a continuous increase. Based on this observation, LM is defined as the evaluation metric for TAL, and a novel TAL estimation model is proposed. The feasibility of the developed TAL estimation model is ultimately validated through accelerated transmission accuracy degradation tests on RV reducers. The error between the predicted and experimental results is 11.06%. The proposed TAL estimation model refines the life evaluation methodology for RV reducers, establishing a solid foundation for real-time transmission accuracy compensation in reducer operation. This paper develops a novel modeling framework that integrates time-varying quantile-based spillover effects into a regime-switching realized volatility model. A dynamic spillover factor is constructed by identifying the most influential contributors to Bitcoin’s realized volatility across different quantile levels. This quantile-layered structure enables the model to capture heterogeneous spillover paths under varying market conditions at a macro level while also enhancing the sensitivity of volatility regime identification via its incorporation into a time-varying transition probability (TVTP) Markov-switching mechanism at a micro level. Empirical results based on the cryptocurrency market demonstrate the superior forecasting performance of the proposed TVTP-MS-HAR model relative to standard benchmark models. The model exhibits strong capability in identifying state-dependent spillovers and capturing nonlinear market dynamics. The findings further reveal an asymmetric dual-tail amplification and time-varying interconnectedness in the spillover effects, along with a pronounced asymmetry between market capitalization and systemic importance. Compared to decomposition-based approaches, the X-RV type of models—especially when combined with the proposed quantile-driven factor—offers improved robustness and predictive accuracy in the presence of extreme market behavior. This paper offers a coherent approach that bridges phenomenon identification, source localization, and predictive mechanism construction, contributing to both the academic understanding and practical risk assessment of cryptocurrency markets. Background: Transthoracic echocardiography (TTE) is the primary imaging modality to assess cardiac morphology and function. In athletes, distinguishing physiological adaptations from pathological changes is essential. This study aimed to evaluate long-term cardiac structural and functional changes in professional soccer players. Methods: This retrospective study included 20 healthy male professional soccer players (mean age 21.2 ± 3.4 years) from the German first division, examined annually from 2016 to 2024 (mean follow-up 5.6 ± 2.0 years). TTE parameters associated with the “athlete’s heart” were assessed, including left ventricular end-diastolic diameter (LVEDD), interventricular septal thickness (IVSD), relative wall thickness (RWT), indexed LV mass (LVMi), and left atrial volume index (LAVi), along with 3D-derived LV and RV volumes. Advanced deformation imaging included global longitudinal strain (GLS), right ventricular strain (RVS), and left/right atrial reservoir strain (LASr and RASr, respectively). Baseline and final follow-up values were compared. Results: No significant changes were observed over time in conventional or advanced echocardiographic parameters (e.g., LVEDD: 54.5 ± 3.1 mm vs. 54.6 ± 3.9 mm; p = 0.868; GLS: −18.7% ± 2.2% vs. −18.4% ± 1.9%; p = 0.670). Ventricular volumes and strain values also remained stable throughout follow-up. Conclusions: Over a mean follow-up of more than five years, professional soccer players showed stable cardiac morphology and function without evidence of pathological remodeling. These findings support the concept that long-term high-level training in mixed-discipline sports leads to balanced, physiological cardiac adaptation. Full article

Background: Patients with single right ventricular morphology (SRV) may exhibit impaired function with increased morbidity, mortality, and need for cardiac transplant due to progressive SRV failure after the Fontan procedure. The aim of the study was to longitudinally characterize the cardiac mechanics and trajectory of disease evolution of SRV failure in Fontan patients. Methods: We performed a case-controlled longitudinal study of 52 patients who underwent extracardiac Fontan palliation for SRV between 1994 and 2015 and compared echocardiographic measures of right ventricular (RV) function, RV-systemic vascular coupling and ventricular remodeling between patients who required heart transplants due to SRV failure (study group, n = 26) and those who did not (control group, n = 26). To define the trajectory, measurements were obtained at four matching time points equivalent in duration from Fontan. Results: RV circumferential shortening function declined in both groups over the time period, but was significantly lower (p < 0.01) in the study group farther from the Fontan. RV-systemic vascular coupling, assessed by systolic time interval measures and RV work, was preserved in the control group, but significantly altered (p < 0.001) in the study group. Relative wall thickness decreased, and the minor/major-axis ratio, as an index of ventricular geometry, increased in the study group, but both remained stable in the control group. Conclusions: This study suggests that positive ventricular remodeling with enhanced circumferential systolic function, and preserved RV-vascular coupling, appear to be adaptive and protective mechanisms against RV failure in Fontan with SRV. These indices of cardiac mechanics may serve as clinically relevant quantifiable markers of disease evolution, and early indicators for therapeutic intervention. Full article

The RV-40E reducer is the representative model of the a typical RV reducer, where transmission accuracy and manufacturing cost are two important optimization indicators, while transmission accuracy depends on both the selection of accuracy levels for components, such as bearings, and the design of tolerance values for parts such as cycloidal gears. The selection of accuracy levels, such as bearings, is relatively simple, and studying manufacturing costs requires the use of a large amount of data from manufacturing equipment and processing methods. Therefore, this article mainly studies the design optimization problem of tolerance values closely related to transmission accuracy. The RV-40E reducer is designed based on the experience of engineers. So far, it is unclear whether the tolerance value design of parts such as cycloidal gears meets the optimization requirements of transmission accuracy. This article first explains the research background and the necessity of tolerance value design for RV-40E components. Next, it details the reducer’s structural composition and lists specific part tolerance values. Subsequently, a virtual prototype is established to evaluate transmission errors under different component and part combinations. Finally, through the design of experiment analysis, optimal tolerance values are derived for selected parts as recommended in the final table. The results demonstrate that the optimized tolerance values for RV-40E reducers are basically consistent with the empirical tolerance values designed by engineers, achieving similar results. Background: The RV144 trial in Thailand is the only HIV-1 vaccine efficacy trial to date to demonstrate any efficacy. Genetic signatures suggested that antibodies targeting the variable loop 2 (V2) of the HIV-1 envelope played an important protective role. The ALVAC prime and protein boost follow-up trial in southern Africa (HVTN702) failed to show any efficacy. One hypothesis for this is the greater diversity of subtype C viruses in southern Africa relative to CRF01_AE in Thailand. Methods: Here, we determined whether an ALVAC prime with computationally selected gp120 boost immunogens maximizing coverage of diversity of subtype C viruses in the variable V1 and V2 regions (V1V2) improved the protection of non-human primates (NHPs) from a heterologous subtype C SHIV challenge compared to more traditional regimens. Results: An ALVAC prime with Trivalent subtype C gp120 boosts resulted in statistically significant protection from repeated intrarectal SHIV challenges compared to the control. Evaluation of the immunogenicity of each vaccine regimen at the time of challenge demonstrated that different gp120 combination boosts elicited similar high magnitudes of gp120 and breadth of V1V2-binding antibodies, as well as strong Fc-mediated immune responses. Low-to-no neutralization of the challenge virus was detected. A Cox proportional hazard analysis of five pre-selected immune parameters at the time of challenge identified ADCC against the challenge envelope as a correlate of protection. Systems serology analysis revealed that immune responses elicited by the different vaccine regimens were distinct and identified further correlates of resistance to infection. Conclusions: Computationally designed vaccines with maximized subtype C V1V2 coverage mediated protection of NHPs from a heterologous Tier-2 subtype C SHIV challenge. Full article

Background/Objectives: New anti-tuberculosis compounds are needed to treat patients infected with multi- or extensively drug-resistant Mycobacterium tuberculosis strains. Studies based on spontaneous in vitro mutagenesis can provide insights into the possible modes of action and resistance mechanisms of such new compounds. We evaluated the primary response of M. tuberculosis in vitro to the action of new aroylhydrazones and nitrofuroylamides. Methods: The reference strain H37Rv was cultured on solid media with compounds at increased concentrations relative to MIC. Resistant clones were investigated using whole-genome sequencing and bioinformatics tools to assess the role and potential impact of identified mutations. Results: Some of the mutations are significant (based on in silico analysis), located in essential genes, and therefore of particular interest. Frameshift mutations were observed in (i) Rv2702/ppgK, which is associated with starvation-induced drug tolerance and persistence in mice, and (ii) Rv3696c/glpK, which has been described as a switch on/off mutation associated with drug tolerance. Nonsynonymous substitutions were found in Rv0506/mmpS2, which belongs to the Mmp protein family involved in transport and drug efflux, and in infB, encoding the translation initiation factor IF-2. Conclusions: The primary adaptation of M. tuberculosis to the selective pressure of the tested compounds is complex and multifaceted. It involves multiple unrelated genes and pathways linked to non-specific drug tolerance, efflux systems, or mechanisms counteracting oxidative stress. Full article

This paper examines the improvement in the performance characteristics and the rheological properties of modified bitumen through the addition of the thermoplastic polymer polyvinyl acetate (PVA). PVA is a synthetic polymer derived from the polymerization of the vinyl acetate. The effect of PVA on bitumen and bituminous mixtures was investigated through the conventional (penetration, softening point, force-ductility, elastic recovery, Marshall and Nicholson stripping tests) and Superpave (rotational viscosity (RV), rolling thin film oven (RTFOT), pressure aging vessel (PAV), dynamic shear rheometer (DSR) and beam bending rheometer (BBR)) tests. PVA was added to bitumen at rates of 2%, 4%, 6% and 8% by mass. Based on the bitumen test results, a PVA rate of 6% was selected for the mixture tests. The modification process was carried out at relatively low temperature (150 °C) and mixing time (20 min) based on various trials, considering the short-term aging of the bitumen. With PVA modification, the penetration value of the bitumen decreased while the softening point increased. As a result, the calculated penetration index (PI) increased and the thermal sensitivity of the bitumen decreased. Significant improvements were detected in elastic recovery and force-ductility tests. Additionally, PVA improved the resistance of asphalt to settling and cracking. Similar results were observed in the DSR and BBR tests. Furthermore, the stripping resistance increased and the stability value improved significantly in the mixture tests. Full article

This study aims to design improved inhibitors targeting the thymidylate kinase (TMK) of Mycobacterium tuberculosis (Mtb), the causative agent of infectious disease tuberculosis that is associated with high morbidity and mortality in developing countries. TMK is an essential enzyme for the synthesis of bacterial DNA. We have performed computer-aided molecular design of MtbTMK inhibitors by modification of the reference crystal structures of the lead micromolar inhibitor TKI1 1-(1-((4-(3-Chlorophenoxy)quinolin-2-yl)methyl)piperidin-4-yl)-5-methylpyrimidine-2,4(1H,3H)-dione bound to TMK of Mtb strain H37Rv (PDB entries: 5NRN and 5NR7) using the computational approach MM-PBSA. A QSAR model was prepared for a training set of 31 MtbTMK inhibitors with published inhibitory potencies (IC50exp) and showed a significant correlation between the calculated relative Gibbs free energies of the MtbTMK–TKIx complex formation and the observed potencies. This model was able to explain approximately 95% of the variation in the in vitro inhibition data and validated our molecular model of MtbTMK inhibition for the subsequent design of new TKI analogs. Furthermore, we have confirmed the predictive capacity of this complexation QSAR model by generating a 3D QSAR PH4 pharmacophore-based model. A satisfactory correlation was also obtained for the validation PH4 model of MtbTMK inhibition (R2 = 0.84). We have extended the hydrophobic m-chloro-phenoxyquinolin-2-yl group of TKI1 that can occupy the entry into the thymidine binding cleft of MtbTMK by alternative larger hydrophobic groups. Analysis of residue interactions at the enzyme binding site made it possible to select suitable building blocks to be used in the preparation of a virtual combinatorial library of 28,900 analogs of TKI1. Structural information derived from the complexation model and the PH4 pharmacophore guided the in silico screening of the library of analogs and led to the identification of new potential MtbTMK inhibitors that were predicted to be effective in the low nanomolar concentration range. The QSAR complexation model predicted an inhibitory concentration IC50pre of 9.5 nM for the best new virtual inhibitor candidate TKI 13_1, which represents a significant improvement in estimated inhibitory potency compared to TKI1. Finally, the stability of the MtbTMK–inhibitor complexes and the flexibility of the active conformation of the inhibitors were assessed by molecular dynamics for five top-ranking analogs. This computational study resulted in the discovery of new MtbTMK inhibitors with predicted enhanced inhibitory potencies, which also showed favorable predicted pharmacokinetic profiles. Full article

Increasing evidence suggests that impaired right ventricular (RV) function has a negative effect on placental function, possibly through venous congestion. We report a case series of hepatic and renal venous flow patterns in pregnant women with right ventricular dysfunction after repaired Tetralogy of Fallot (ToF), relative to those observed in normal pregnancy and preeclampsia. Methods: At 20–24 weeks pregnancy, RV function was measured by echocardiography and by cardiovascular magnetic resonance in women with repaired ToF. Combined Doppler-ECG of the hepatic and renal interlobular veins were performed in three women with asymptomatic right ventricular dysfunction. Venous impedance index and pulse transit time were measured and classified as abnormal at >75th and <25th reference percentile, respectively. Results: All three women showed dilated RV and mildly impaired RV function. Both hepatic and intrarenal Doppler flow waves were abnormal and very much resembled the patterns seen in preeclampsia. One of the three women had complications including ventricular tachycardia, intrauterine growth restriction, antenatal bleeding, emergency cesarean section and acute heart failure 2 days postpartum. Conclusions: Pregnant women with mild right ventricular dysfunction after repaired ToF show abnormal venous Doppler flow waves in the liver and kidneys, similar to those observed in preeclampsia. These findings are in line with reported observations on the association between impaired RV function, abnormal return of venous blood, venous congestion and organ dysfunction. The parallel with venous Doppler flow observations in preeclampsia suggest that the venous compartment might play an important role in the etiology of preeclampsia-induced organ dysfunction. Whether this phenomenon directly affects the uteroplacental circulation is to be assessed in future research. Full article

Licochalcone A-Inspired Chalcones: Synthesis and Their Antiproliferative Potential in Prostate Cancer Cells

Prostate cancer remains a significant global health concern, prompting ongoing exploration of novel therapeutic agents. Licochalcone A, a natural product in the chalcone family isolated from licorice root, is characterized by its enone structure and demonstrates antiproliferative activity in the micromolar range across various cell lines, including prostate cancer. Building on our prior success in enhancing curcumin’s antiproliferative potency by replacing the substituted phenol with a 1-alkyl-1H-imizadol-2-yl moiety, we applied a similar approach to design a new class of licochalcone A-inspired chalcones. The synthesis of these target chalcones involved key [3,3]-sigmatropic rearrangement of aryl prenyl ethers and Claisen–Schmidt condensations, yielding three derivative series. These compounds were evaluated for antiproliferative activity in both androgen receptor (AR)-positive and AR-null prostate cancer cell models using WST-1 cell proliferation assay. Systematic evaluation of licochalcone A across four prostate cancer cell lines indicated a modest advantage over enzalutamide, an FDA-approved AR antagonist, in suppressing 22Rv1 cell proliferation. Interestingly, three ester derivatives by replacing the phenol next to the carbonyl with an alkoxide demonstrated similar antiproliferative potency to licochalcone A in both AR-positive and AR-negative prostate cancer cell lines. This suggests that the phenol moiety on licochalcone A may be a promising site for chemical manipulations to enhance anti-prostate cancer activity. Among the synthesized chalcones, nine derivatives showed improved selectivity for AR-positive LNCaP and 22RV1 cells relative to AR-negative PC-3 and DU145 cells, surpassing licochalcone A in selectivity. Additionally, the antiproliferative potency was highly dependent on the R group attached to the imidazole. Most of the derivatives showed antiproliferative potency against androgen receptor-positive LNCaP and 22Rv1 cells, comparable to that of enzalutamide and licochalcone A. These findings suggest that optimization of licochalcone A-inspired chalcones as potential anti-prostate cancer agents warrants further investigation. Full article

Rotavirus (RV) is a significant contributor to diarrhea in both young children and animals, especially in piglets, resulting in considerable economic impacts on the global pig industry. Isoleucine (Ile), a branched-chain amino acid, is crucial for regulating nutrient metabolism and has been found to help mitigate diarrhea. This study aimed to assess the impact of isoleucine supplementation in feed on colonic barrier function, colonic microbiota, and metabolism in RV-infected weanling piglets. A total of thirty-two weaned piglets, aged 21 days, were randomly assigned to two dietary groups (each further divided into two subgroups, with eight replicates in each subgroup), receiving diets with either 0% or 1% isoleucine for a duration of 14 days. One group from each treatment was then challenged with RV, and the experimental period lasted for 19 days. The results showed that dietary Ile significantly increased the secretion of IL-4, IL-10, and sIgA in the colon of RV-infected weanling piglets (p < 0.05). In addition, Ile supplementation notably increased the expression of tight junction proteins, including Claudin-3, Occludin, and ZO-1 (p < 0.01), as well as the mucin protein MUC-1 in the colon of RV-infected weanling piglets (p < 0.05). Gut microbiota analysis revealed that dietary Ile increased the relative abundance of Prevotella and decreased the relative abundance of Rikenellaceae in the colons of RV-infected weanling piglets. Compared with the RV+CON, metabolic pathways in the RV+ILE group were significantly enriched in vitamin digestion and absorption, steroid biosynthesis, purine metabolism, pantothenate and CoA biosynthesis, cutin, suberine, and wax biosynthesis, as well as fatty acid biosynthesis, and unsaturated fatty acid biosynthesis. In conclusion, dietary Ile supplementation can improve immunity, colonic barrier function, colonic microbiota, and colonic metabolism of RV-infected weaned piglets. These findings provide valuable insights into the role of isoleucine in the prevention and control of RV. Full article

Antimycobacterial Activity of Essential Oils from Bulgarian Rosa Species Against Phylogenomically Different Mycobacterium tuberculosis Strains

In this study, we aimed to assess the activity of the essential oils from four Bulgarian oil-bearing roses Rosa damascena Mill., R. alba L., R. centifolia L., and R. gallica L., on the reference strain Mycobacterium tuberculosis H37Rv and clinical M. tuberculosis strains of the Beijing and Latin-American Mediterraneum genotypes. The chemical composition of the essential oils was determined by gas chromatography (GC-FID/MS). Minimal inhibitory concentrations (MIC) were determined using the resazurin method. R. alba oil showed the highest inhibitory activity when tested on all strains of different phylogenetic origins with MIC in the range of 0.16–0.31 mg/mL, while R. gallica oil was the least active (MIC 0.62–1.25 mg/mL). The obtained results show heterogeneity of rose oil action on different mycobacterial strains and we hypothesize that the combined level of geraniol and nerol is a key factor that underlies the antimycobacterial action of the rose oils. Strain Beijing 396 was relatively more susceptible to the rose oils probably due to multiple and likely deleterious mutations in its efflux pump genes. Two clinical MDR strains have likely developed during their previous adaptation to anti-TB drugs certain drug tolerance mechanisms that also permitted them to demonstrate intrinsic tolerance to the essential oils. Further research should investigate a possible synergistic action of the new-generation anti-TB drugs and the most promising rose oil extracts on the large panel of different strains. Full article

Motivated by the comovement of realized volatilities (RVs) of agricultural commodity prices, we study whether multi-task forecasting algorithms improve the accuracy of out-of-sample forecasts of 15 agricultural commodities during the sample period from July 2015 to April 2023. We consider alternative multi-task stacking algorithms and variants of the multivariate Lasso estimator. We find evidence of in-sample predictability but scarce evidence that multi-task forecasting improves out-of-sample forecasts relative to a classic univariate heterogeneous autoregressive (HAR)-RV model. This lack of systematic evidence of out-of-sample forecasting gains is corroborated by extensive robustness checks, including an in-depth study of the quantiles of the distributions of the RVs and subsample periods that account for increases in the total spillovers among the RVs. We also study an extended model that features the RVs of energy commodities and precious metals, but our conclusions remain unaffected. Besides offering important lessons for future research, our results are interesting for financial market participants, who rely on accurate forecasts of RVs when solving portfolio optimization and derivatives pricing problems, and policymakers, who need accurate forecasts of RVs when designing policies to mitigate the potential adverse effects of a rise in the RVs of agricultural commodity prices and the concomitant economic and political uncertainty.
