This study aimed to investigate the effects of mental fatigue on physical and cognitive performance (lower-limb power, isometric and handgrip strength, and psychomotor vigilance). Twenty-two physically active young adults (12 males, 10 females; Mage = 20.82 ± 1.47) were randomly assigned to either a Mental Fatigue (MF) or Control group (CON). The MF group showed a statistically significant (p = 0.019) reduction in non-dominant handgrip strength, declining by approximately 2.3 kg (about 5%), while no such change was observed in the CON group or in dominant handgrip strength across groups. Reaction time (RT) was significantly impaired following the mental fatigue protocol: RT increased by 117.82 ms, representing an approximate 46% longer response time in the MF group (p < 0.001), whereas the CON group showed a smaller, non-significant increase of 32.82 ms (~12% longer). No significant differences were found in squat jump performance, indicating that lower-limb explosive power may be less affected by acute mental fatigue. These findings demonstrate that mental fatigue selectively impairs fine motor strength and cognitive processing speed, particularly reaction time, while gross motor power remains resilient. Understanding these effects is critical for optimizing performance in contexts requiring fine motor control and sustained attention under cognitive load. Full article

Huntington’s disease (HD) is a severe and progressive neurodegenerative disease for which therapeutic options have so far been confined to symptomatic treatment. Currently, the diagnosis relies on the signs and symptoms shown by patients; however, by that stage, the psychomotor issues have progressed to a point where reversal of the condition is unattainable. Although numerous clinical trials have been actively investigating therapeutic agents aimed at preventing the onset of disease or slowing down the disease progression, there has been a constant need for reliable biomarkers to assess neurodegeneration, monitor disease progression, and assess the efficacy of treatments accurately. Therefore, to discover the key biomarkers associated with the progression of HD, we employed bioinformatics and machine learning (ML) to create a robust pipeline that integrated differentially expressed gene (DEG) analysis with ML to select potential biomarkers. We performed a meta-analysis to identify DEGs using three Gene Expression Omnibus (GEO) microarray datasets from different platforms related to HD-affected brain tissue, applying both relaxed and strict criteria to identify differentially expressed genes. Subsequently, focusing only on genes identified through the inclusive threshold, we employed 19 diverse ML techniques to explore the common genes that contributed to the top three selected ML algorithms and the shared genes that had an impact on the ML algorithms and were observed in the meta-analysis using the stringent condition were selected. Additionally, a receiver operating characteristic (ROC) analysis was conducted on external datasets to validate the discriminatory power of the identified genes. Based on the results of an inverse variance weighted meta-analysis of the AUCs across both human and mouse cohorts, GABRD and PHACTR1 were identified as the most robust candidates and were selected as key biomarkers for HD. Our comprehensive methodology, which integrates DEG meta-analysis with ML techniques, enabled a systematic prioritization of these biomarkers, providing valuable insights into their biological significance and potential for further validation in clinical research. Background: The COVID-19 pandemic has had a profound impact on pediatric mental health, contributing to a global surge in psychiatric emergencies among children and adolescents. This study aimed to evaluate trends in pediatric emergency department (PED) visits for three key psychiatric conditions—anxiety disorders (ADs), self-injury behaviors (SIBs), and psychomotor agitation (PMA)—before and after the onset of the COVID-19 pandemic. Methods: We conducted a retrospective observational study at a tertiary pediatric hospital in Italy, analyzing all psychiatric presentations to the PED from 1 January 2018 to 31 December 2024. The data were divided into pre-COVID and post-COVID periods and included patient demographics, recurrence of visits, clinical features, hospital admissions, and pharmacological management. Diagnoses were confirmed by chart review. Results: Of 233,867 total PED visits, 1082 were due to primary psychiatric concerns. A marked increase in visits was observed postCOVID: SIB incidence rose from 3.6 to 15.1 per 10,000 visits (p < 0.0001), PMA from 9.4 to 17.8 (p < 0.0001), and AD from 17.7 to 21.6 (p = 0.018). SIB cases showed increased recurrence (from 3.4% to 27.4%, p = 0.004) and greater pharmacological intervention, whereas PMA was associated with a rise in heteroaggression (from 14.3% to 39.8%, p < 0.0001). Pharmacological treatment remained largely consistent, with benzodiazepines and neuroleptics most frequently used. The emerging use of intranasal ketamine was noted in select cases. Conclusions: This study highlights the increasing burden of pediatric psychiatric emergencies in the wake of the COVID-19 pandemic. The findings underscore the urgent need to implement standardized emergency care protocols, strengthen outpatient mental health services, and develop pediatric-specific pharmacological guidelines to improve outcomes in this vulnerable population. Objective: The aim of this study was to investigate the effects of prenatal heavy metal (arsenic, cadmium, manganese) exposure on infant neurodevelopment through a systematic review and meta-analysis, elucidating the toxicological mechanisms and dose–response relationships for consideration in environmental risk assessment. Methods: Following the PRISMA guidelines, systematic searches were conducted in PubMed, Embase, and other databases, ultimately resulting in 17 observational studies involving 6907 participants. Results: Arsenic (As): A 50% increase in arsenic exposure was associated with a reduction of 0.51 points in the Mental Development Index (MDI; 95% CI: −1.43 to 0.4) and 0.15 points in the Psychomotor Development Index (PDI; 95% CI: −0.96 to 0.65). However, these results did not achieve statistical significance (p > 0.05). Cadmium (Cd): Prenatal cadmium exposure significantly decreased the Full-Scale Intelligence Quotient (FSIQ) in children aged 5–9 years. A 50% increase in cadmium exposure resulted in a 0.44-point drop in the FSIQ (95% CI: −0.67 to −0.21, p < 0.05), with stable effects (I2 = 0%). Manganese (Mn): Manganese exposure showed a negative association with the MDI (β = −0.11) and PDI (β = −0.18). However, a high degree of heterogeneity was observed (I2 = 20.89–73.35%), and some studies suggested potential risks even at low exposure levels. Sensitivity analyses indicated that the heterogeneity in the arsenic and manganese results mainly stemmed from individual study differences (e.g., sample characteristics), whereas the cadmium effects were consistent. Conclusions: Prenatal heavy metal exposure (notably cadmium) adversely impacts neurodevelopment, even at low doses. Future research should prioritize critical exposure windows, mixed effects, and sex-specific vulnerabilities. Strengthening environmental monitoring and prenatal guidelines is crucial to mitigate developmental risks. Full article

Background/Objectives: Childhood anemia remains a serious public health issue, negatively affecting cognitive and psychomotor development, with repercussions on school performance and adult productivity. This study aimed to characterize the profile of children aged 6 months to 5 years diagnosed with or at risk of anemia who attended a pediatric hospital in Lisbon, Portugal. Methods: A hospital-based, cross-sectional descriptive study was conducted from September 2023 to September 2024. Descriptive statistics, including frequency distributions and cross-tabulations, summarized participant characteristics and key variables. Results: We observed that 33.3% (74/222) of the children were either anemic or at risk of anemia. Among these, 93.2% (69/74) were confirmed anemic or at risk based on hemoglobin levels. Five children (6.8%) had normal hemoglobin but abnormal red-cell indices, with microcytic (60.0%; 3/5) or normocytic (40.0%; 2/5) patterns. Anemia rates were higher in males (55.1%), children aged 24–59 months, those residing in the Metropolitan Lisbon Area (82.6%), children whose caregivers had only basic or secondary education (58.0%), and those whose mothers were born in foreign countries (48.4%). Microcytic red-cell indices were observed in 63.1% of cases. Serum iron results indicated that 32.0% were pre-anemic and 40.0% anemic. Ferritin levels showed iron-deficiency anemia in 22.2% of tested cases. In addition, 33.3% carried the sickle cell trait, and 35.0% had elevated C-reactive protein, suggesting anemia of inflammation. Conclusions: Anemia is a moderate public health issue, mainly affecting children with less-educated caregivers and migrant mothers. Targeted public health actions, including systematic screening, caregiver education, and multiculturally sensitive interventions, are crucial to address anemia. Full article

Background: This study aimed to evaluate the impact of physical activity levels on selected biochemical markers (glucose, insulin, cholesterol, triglycerides, interleukin-6 [IL-6]), brain-derived neurotrophic factor (BDNF), cognitive functions, and additional macronutrient intake in postmenopausal women. Method: A total of 72 generally healthy women aged 55–73 from western Poland participated in the study. Physical activity levels were assessed using the International Physical Activity Questionnaire (IPAQ), resulting in two distinct groups: 56 women in the lower activity level group and 16 in the higher activity level group. We calculated body mass index (BMI), measured body composition and blood pressure, and conducted cognitive assessments, including the Mini-Mental State Examination (MMSE), motor and psychomotor skills tests, the Clock Drawing Test, and the Geriatric Depression Scale (GDS). Nutritional intake was evaluated using a detailed 3-day food record to analyze macronutrient consumption and total caloric intake. Results: A statistically significant difference in total blood cholesterol levels (p = 0.0277) was observed between the two groups, with the higher physical activity group showing elevated cholesterol levels. Although no other biochemical markers showed statistically significant differences, variations in BDNF, glucose, triglycerides, IL-6, and insulin levels were noted between groups. Moreover, correlations between these markers and cognitive performance, like motor and psychomotor speeds, varied depending on physical activity level. The analyzed dietary pattern of the studied group shows slight deviations from current nutritional recommendations. Conclusions: The findings suggest that physical activity level may influence certain biochemical markers and cognitive functions in postmenopausal women. While these results highlight the potential benefits of physical activity, further research is needed to clarify underlying mechanisms and to validate physical activity as an effective strategy for improving postmenopausal health. Human parechoviruses, officially known as Parechovirus A (PeV-A), are more frequently reported as a significant cause of serious infections in newborns and young infants. We aimed to describe the clinical features and neurological outcomes of PeV-A encephalitis cases identified in Warsaw. Infants with suspected encephalitis were retrospectively identified in three hospitals in the summer of 2022. Cases of confirmed PeV-A infection had their comprehensive demographic, clinical, laboratory, imaging, and outcome data reviewed. The psychomotor development of the children up to the age of 2 years was assessed by using the standardized tools. We identified 18 cases of confirmed encephalitis with a PeV-A infection. Their median age was 16 days. Fourteen cases were included in the analysis, while one patient dropped out after the first visit. Most were boys (9/14), and one patient was born preterm. Three patients had white matter alterations on brain MRI at discharge. No significant neurologic sequelae were observed after acute illness. At the 24-month follow-up, based on the Bayley Scales of Infant and Toddler Development (BSID-IV) and the Brunet–Lézine Scale, the children showed no neurodevelopmental sequelae. Brain MRIs were obtained in all of the participants up to 12 months of age and revealed no significant lesions. Neurodevelopmental complications are not frequent in children after PeV-A encephalitis at 24 months of age. Continued follow-up in larger cohorts is needed to explore the predictors of long-term morbidity. Full article

Background/Objectives: The ability of young adults to control their balance is generally effortless and can occur automatically with minimal cognitive involvement. However, this ability may be compromised when integration conflicts arise due to impairments in vestibular, visual, or somatosensory functions. Hence, psychomotor symptoms linked to emotional states can also influence postural control. The purpose of this study was to understand the effects of anxiety and depression on balance in young adults. Methods: Our study included 50 young adults (21.86 ± 2.63 years), consisting of 13 males and 37 females. Anxiety and depressive symptoms were evaluated using the Hospital Anxiety and Depression Scale (HADS), while balance was assessed through the Modified Clinical Test for the Sensory Interaction on Balance (mCTSIB). Data analysis was conducted using Pearson’s correlation coefficient test and the Kruskal–Wallis test. Results: Pearson’s correlation analysis indicated that young adults exhibited stable postural control. However, a positive correlation (0.259, p < 0.1) was observed between anxiety levels and the sway index. Additionally, positive correlations were found between anxiety and both somatosensory (0.281, p < 0.05) and visual (0.276, p < 0.1) ratios. Conclusions: The results suggest that higher anxiety levels are associated with reduced postural balance, with sensory inputs, particularly visual and somatosensory, playing a key role in this decreased stability. Background: Monochorionic diamniotic (MCDA) twin pregnancies are at risk of complications, particularly selective intrauterine growth restriction. The objective of this study was to evaluate the two-year neurologic outcomes of the eutrophic newborns from monochorionic diamniotic twin pregnancies who were complicated by selective intrauterine growth restriction, compared to newborns from uncomplicated MCDA pregnancies. Our hypothesis was to determine whether selective IUGR in these pregnancies was specifically associated with a risk of delayed psychomotor development at two years old. Methods: We conducted a retrospective–prospective observational cohort study of children from pregnancies and deliveries which were monitored at Hospital Nord of Marseille between 2012 and 2021. The primary outcome measure was the comparison of the Ages and Stages Questionnaire (ASQ) scores at the age of two years between the two groups. The secondary outcome measure was a composite score including the following: neonatal death, grade III or IV intraventricular hemorrhage (IVH) at cerebral MRI or cranial ultrasound, periventricular leucomalacia (PVL) at brain MRI, bronchopulmonary dysplasia (BPD), and necrotizing enterocolitis (NEC) of stages II or III. Results: A total of 57 eutrophic children were included in the group from monochorionic twin pregnancies complicated by selective IUGR and 270 children in the group from MCDA twin pregnancies with no complications. The composite morbidity and mortality criterion, including neonatal death, grade III or IV IVH, the presence of PVL, BPD, and/or stage II or III NEC, was 11% in eutrophic newborns from the MCDA group with IUGR and 5% in the uncomplicated MCDA group, with no statistically significant difference (p = 0.18). The 2-year follow-up allowed for the comparison of a total of 38 eutrophic children from complicated pregnancies and 134 children from uncomplicated pregnancies. The median ASQ score at 24 months was 255 in the complicated pregnancy group and 240 in the uncomplicated pregnancy group, with no statistically significant difference (p = 0.27) after adjustment. Conclusions: Our study did not show a statistically significant difference in the neurodevelopmental follow-up of eutrophic children from monochorionic diamniotic twin pregnancies with selective intrauterine growth restriction compared to newborns from the same pregnancies without complications. Background/Objectives: Action observation therapy (AOT) has gained attention as a rehabilitation method for motor function recovery following nerve injury. Although the total observation time and daily session duration have been studied, the effective observation duration per trial remains unclear. This study examined the effect of different observation durations on manual dexterity, mirror neuron system activity, and subjective psychomotor effort in healthy adults. Methods: Twenty-four healthy right-handed adults participated in this crossover study under four conditions: observing ball rotations with the dominant hand for one, two, or three minutes, or geometric patterns (control) for two minutes. The outcomes included maximum rotations and errors by both hands during a ball rotation task and interpersonal motor resonance (IMR), indicating mirror neuron system activity. These measures were compared before and after intervention. Subjective ratings of concentration, physical fatigue, and mental fatigue were assessed post-intervention. Results: Rotation performance significantly increased for the intervention hand after a 2 min observation and showed a notable effect (p = 0.113, r = 0.48) for the non-intervention hand after a 3 min observation compared to the control. The IMR was significantly greater during the 2 min observation than in the control. Compared to the 1 min observation, the 2 min and 3 min observations resulted in higher mental fatigue, and the 3 min observation showed lower concentration levels. Conclusions: These findings indicate that the observation duration has varying effects on manual dexterity and mirror neuron system activity, with optimal effects observed at specific time intervals while also highlighting the relationship between observational learning and psychomotor effort. The objective of this study was the retrospective analysis of the course, indications for treatment, and effects of therapy for NF1-OPGs. Patients and Methods: We analyzed demographics, clinical and genetic data, imaging and ophthalmological parameters, their impact on therapeutic decisions, and the effectiveness of the therapy in 92 patients. Results: OPGs were unilateral in 55.4% of patients and bilateral in 44.6%. Post-contrast enhancement in MRI was observed in 67.4%. Oncological treatment was required in 16.3% of patients with median age 3.8 years. Factors significant in multivariate analysis contributing to the need of oncological treatment were: amblyopia and proptosis. Factors contributing to amblyopia were: strabismus, proptosis, co-occurrence of epilepsy, bilateral OPGs, and thickness of the optic nerve ≥ 8 mm. The first line of oncological treatment included vincristine + carboplatin or monotherapy with vinblastine. The use of subsequent lines of oncological treatment was necessary in 46.7% patients. Conclusions: The following conclusions, suggest modification of the approach in the management of patients with NF1-OPG, summarize the presented study: (1) perform the first MRI after the age of 1 year, (2) reduce the frequency of follow-up scans in the first year of observation in patients with isolated involvement of intraocular and/or intraorbital segments of the optic nerve, (3) refrain from administering contrast during control MRI examinations of the orbits after OPG diagnosis; (4) in patients with co-occurring psychomotor delay or treated with antiepileptic drugs, do not make decisions about oncological therapy when visual acuity deterioration is observed, without progression in optical coherence tomography (OCT), visual evoked potentials (VEP), and MRI. Objective: Sleep deprivation negatively affects mood and sleepiness across subjective, objective, behavioral, and neuroimaging measures. However, the relationship between abnormal brain dynamics after sleep deprivation and mood or sleepiness impairments, from a temporal perspective, remains underexplored. Electroencephalogram microstate analysis offers a valuable approach for investigating the large-scale dynamics of brain networks. Methods: We implemented a strict sleep deprivation protocol with 71 participants, collecting resting-state electroencephalogram data, subjective sleepiness, objective alertness, and mood assessments after normal sleep and sleep deprivation (SD) conditions. Microstate time parameters were compared between sleep control (SC) and SD conditions. Additionally, we investigated how changes in these parameters correlated with subjective or objective measures of sleepiness and mood changes between SC and SD. Results: SD significantly decreased the mean duration and occurrence of microstate B while increasing those of microstate C. A decrease in microstate B occurrence significantly correlated with a higher Karolinska Sleepiness Scale score, whereas a reduction in microstate B duration indicated an increased response time on the Psychomotor Vigilance Performance. No significant associations were observed between microstate parameters and positive mood decline between SC and SD. Pearson correlation analysis was performed on the positive mood scores in both conditions. The findings demonstrated a significant positive relationship between positive mood scores and the mean duration of microstate B under SD conditions. Conclusions: Using a large SD dataset, this study demonstrated that subjective or objective sleepiness and positive mood were associated with decreased microstate B. These findings suggest that SD disrupts neural dynamics within the visual network. Full article

Objectives: The aim of this study was to determine the performance of cognitive and psychomotor functions in patients after ischemic stroke, taking into account the effectiveness of early and late rehabilitation. Methods: The study included 86 patients with ischemic stroke hospitalized in the Neurological Rehabilitation Unit. The patients were divided into two groups according to the timing of rehabilitation, considering early rehabilitation which started within 30 days of hospital discharge (56 patients), and late rehabilitation which started after 30 days of hospital discharge (30 patients). Cognitive and psychomotor functions were measured in all the study patients using the Integrated System for the Measurement of Psychophysiological Variables called Polypsychograph, including tests assessing memory, attention, eye–hand coordination, and reaction speed. The measurements were repeated after 21 days of post-stroke rehabilitation. Results: Early rehabilitation led to significant improvements in most of the parameters studied, including memory, attention, speed of thinking, and precision of movement. Late rehabilitation was followed by an improvement in the results of the indicators studied to a lesser extent than the early rehabilitation. Improvements in temporal and qualitative parameters were observed in both groups of patients undergoing early and late rehabilitation. Conclusions: In patients after ischemic stroke, early rehabilitation improved cognitive and psychomotor performance to a greater extent than late rehabilitation. Full article

Objectives: Previous studies have assessed the effect of docosahexaenoic acid (DHA, 22:6n-3) and arachidonic acid (ARA, 20:4n-6)-supplemented infant formula on brain development and cognitive function in infants. However, the results have been inconsistent. The aim of this systematic review and meta-analysis was to assess the effect of DHA and ARA supplementation on cognitive function in infants from randomized controlled trials (RCTs). Methods: We systematically searched and identified relevant literature from the PubMed, Web of Science, and Embase databases up to July 2024. Standard methods were applied to assess publication bias, sensitivity analysis, and heterogeneity among the included studies. A total of nine RCTs were included in the study, which comprised 1039 subjects. Results: Meta-analysis showed significantly positive effects of DHA and ARA supplementation on cognitive development in infants (Standardized Mean Difference (SMD): 0.21; 95% CI: 0.03, 0.38). No significant difference was found in Mental Development Index (MDI) score (Weighted Mean Difference (WMD): 0.20; 95% CI: −0.03, 0.43) and Psychomotor Development Index (PDI) score (WMD: 0.12; 95% CI: −0.11, 0.35) in Bayley Scales of Infant and Toddler Development compared with the control group. In subgroup analysis, when DHA/ARA was 0.5–1, PDI had a significant difference (WMD: 0.48; 95% CI: 0.03, 0.93) compared with the control group, with no significant difference between heterogeneity (I2 = 46.4%, p = 0.155). In comparison to the control group, significant differences were observed in MDI when DHA/ARA levels were between 0.5 and 1 (WMD: 0.55; 95% CI: 0.07, 1.02), with no significant difference between heterogeneity (I2 = 51.6%, p = 0.127). Conclusion: When the DHA /ARA was 0.5–1 can significantly improve the cognitive function in infants. Full article

Objectives: To analyse whether breastfeeding (BF) is related to better psychomotor development in the first two years of life. Methods: Prospective longitudinal study of a cohort of children born in Spain (between April 2017 and March 2018) and followed during the first two years of life by their primary care paediatrician in eight visits (LAyDI study—PAPenRed research network). The Haizea-Llevant development chart was used to assess the psychomotor development (DPM), and the subjects were divided according to whether or not they met each milestone. Results: The initial sample was 1946 children (50.1% boys), which varied at each visit from 1946 on the first and second visit to 1076 on the last visit; DPM at 12, 18 and 24 months was compared according to the type of BF at 6 months; at 24 months, significant differences were found in the achievement of milestones in the BF-at-6-months group (“scribbles spontaneously”, p 0.007 and “descends stair”, p 0.002). When comparing the mean duration of BF and exclusive breastfeeding, according to milestones reached or not, statistically significant differences were observed in more milestones at the 24-month visit, including “eats with a spoon” (5.6 months in the group that reached it vs. 2.4 months in the group that did not, p 0.014), and again for the milestone ‘scribbles spontaneously’ (5.6 months vs. 1.8 months, p 0.021), among others. Conclusions: In our study, psychomotor development in the first two years of life does not show major differences in relation to the type of feeding; from this age onwards, the differences may be greater. Many factors influence psychomotor development in the first years of life, with breastfeeding not being an isolated factor. Full article
