First Report of a Psyllid Vector of ‘Candidatus Phytoplasma pruni’ (Strain 16SrIII-J)

In Graneros, O’Higgins Region, Chile, the mallow psyllid (Russelliana solanicola Tuthill, 1959) from Malva nicaeensis L. was identified as a potential vector of ‘Candidatus Phytoplasma pruni’. Over an 8-month period, 2089 specimens of a species of Psylloidea, including immatures and adults, were captured. We only selected the adults used for transmission trials in Catharanthus roseus (L.) G. Don (periwinkle) plants. By nested PCR, using primer pairs for phytoplasma detection in 16S rRNA and IdpA genes, 7 out of 113 (6.2%) periwinkle plants used in transmission trials were found to be infected by phytoplasmas. Insects that fed on these plants also tested positive for the same phytoplasmas. Periwinkle plants never showed virescence and phyllody, as commonly occurs with phytoplasma 16SrIII-J infection due to the effector SAP54. In this case, using primer pairs for the SAP54 gene, an amplification product was never obtained. Virtual restriction fragment length polymorphism (RFLP) analysis of F2nR2 fragments indicated that the phytoplasma, found in both periwinkle plants and insects used in transmission trials, belongs to the 16SrIII-J ribosomal subgroup. The COI gene of the psyllids samples was amplified and sequenced, showing a similarity ranging from 84.84% to 85.02% with R. solanicola from Solanum tuberosum L. The mitochondrial genome of the psyllid was also sequenced, revealing a 14,835 bp circular DNA molecule with 37 genes. The mallow psyllid transmitted the phytoplasma 16SrIII-J to periwinkle plants. The molecular identification of the insect does not match the morphological one, indicating that the mallow psyllid may constitute a cryptic species within the polyphagous R. solanicola species. This is the first report of a psyllid as a vector of the phytoplasma 16SrIII-J. Full article

Periwinkle (Catharanthus roseus (L.) G. Don) is renowned for its diverse colors and resilience to harsh climates. Still, most commercial cultivars predominantly display flat petals. Using cultivars representing non-wavy, medium-wavy, and extreme-wavy flower forms, we examined morphological differences in both their mature leaves and floral organs. Phenotypes of self-pollinated (S1) and cross-pollinated (F1, F2) populations further underscored their morphological distinctions. Specifically, the extreme-wavy type displayed elliptical leaves, broader than the non-wavy type, with a pronounced acute apex and a notably wrinkled blade surface. The non-wavy type also bore intensely wavy petal margins and exhibited a smaller flower diameter, with a notable absence of a functional pistil, indicating female sterility. The insights gained allowed for early differentiation during the seedling period. This study suggests that the inheritance of these flower forms is regulated by an allele WAVY (Wv), which exhibits incomplete dominance. Concretely, the non-wavy form arises from a recessive homozygous expression (wvwv), the extreme-wavy from a dominant homozygous expression (WvWv), and the medium-wavy from a heterozygous expression (Wvwv). This study provides clarity on morphological descriptions and inheritance patterns of wavy flower forms, facilitating strategic breeding of diverse flower forms in periwinkle. Full article

Tamarisk witches’ broom, yellowing, and little leaf symptoms were observed during 2018–2023 surveys of rural deserts in central regions of Iran with the highest disease incidence up to 72% in Chah Afzal (Yazd province). A verification of the presence and identity of phytoplasmas associated with these symptoms was then performed. Tamarisk tree branch cuttings obtained from symptomatic plants sprouted up to 90.3% but with 15–25 days’ delay compared to the asymptomatic ones and showed internode shortening and witches’ broom, while the branch cuttings from asymptomatic plants had normal growth and sprouted up to 97.8%. Phytoplasma transmission by dodder bridges to periwinkle did not succeed, while nested polymerase chain reaction on the phytoplasma ribosomal gene followed by RFLP and phylogenetic analyses revealed the presence of ‘Candidatus Phytoplasma asteris’, ‘Ca. P. australasiae=australasiaticum’, and ‘Ca. P. trifolii’ (ribosomal subgroups 16SrI-B, 16SrII-D, and 16SrVI-A, respectively) in the samples from symptomatic plants only. Further amplifications were performed on selected phytoplasma-positive samples on tuf and secA genes, and the produced sequences indicated the presence of mixed phytoplasma infection in some of the samples. In particular, in the tuf gene, a mixed infection of ‘Ca. P. australasiae=australasiaticum’ and ‘Ca. P. trifolii’ was detected, while in the secA gene, the presence of ‘Ca. P. asteris’ or ‘Ca. P. tritici’ strains was identified. The first-time detection of diverse phytoplasma strains in symptomatic T. aphylla suggests that this species represent a relevant source of infection for the agricultural crops and for landscape plants especially when temperature allows insect vector transmission, and therefore, it represents a risk in every environment especially in the frame of climatic changes. Phytoplasmas are intracellular pathogenic bacteria that infect a wide range of plant species, including agriculturally important crops and ornamental trees. However, our understanding of the relationship between symptom severity, disease progression, and phytoplasma concentration remains limited due to the inability to inoculate phytoplasmas mechanically into new plant hosts. The present study investigated phytoplasma titer dynamics and symptom development in periwinkle and tomato, both infected with the same potato purple top (PPT) phytoplasma strain using a small seedling grafting approach. Virescence, phyllody, and witches’-broom (WB) symptoms sequentially developed in periwinkle, while in tomato plants, big bud (BB, a form of phyllody), cauliflower-like inflorescence (CLI), and WB appeared in order. Results from quantitative polymerase chain reaction (qPCR) targeting the PPT phytoplasma’s 16S rRNA gene revealed that in both host species, phytoplasma titers differed significantly at different infection stages. Notably, the highest phytoplasma concentration in periwinkles was observed in samples displaying phyllody symptoms, whereas in tomatoes, the titer peaked at the BB stage. Western blot analysis, utilizing an antibody specific to PPT phytoplasma, confirmed substantial phytoplasma presence in samples displaying phyllody and BB symptoms, consistent with the qPCR results. These findings challenge the conventional understanding that phytoplasma infection dynamics result in a higher titer at later stages, such as WB (excessive vegetative growth), rather than in the early stage, such as phyllody (abnormal reproductive growth). Furthermore, the PPT phytoplasma titer was markedly higher in periwinkles than in tomato plants, indicating differing susceptibilities between the hosts. This study reveals distinct host responses to PPT phytoplasma infection, providing valuable insights into phytoplasma titer dynamics and symptom development, with implications for the future management of agricultural disease. The increased use of hexavalent chromium (Cr6+) in various industrial applications has contributed to its elevated levels in the environment, especially the aquatic environment. Thus, there is the potential for accumulation of Cr6+ in the tissues of aquatic organisms and consequent toxic effects. The toxic effects of Cr6+ in aquatic organisms have been widely reported; however, little is known about the patterns of tissue accumulation of Cr6+ and its toxicity in aquatic mollusks. Thus, the present study investigated the effects of Cr6+ exposure on the tissue distribution, proximate composition, and histopathology of an aquatic mollusk, periwinkle (Littorina littorea). The animals were exposed to sublethal concentrations of Cr6+ (0.42, 0.84, and 4.2 mg/L) for 30 days, after which the condition index, tissue accumulation, proximate composition, and histopathological effects were determined. The control animals were maintained in a medium that did not contain Cr6+ (0 mg/L). The condition index did not differ significantly among the groups. The levels of Cr6+ in the tissues differed significantly among the different tissue types while there was no significant effect of the exposure concentration, except in the foot tissue. The proximate parameters (protein, carbohydrates, lipid, crude fiber, and moisture contents) differed significantly among the groups. The protein contents of the exposed animals were significantly lower than those of the control animals and the histological architecture of the major organs was altered in the chromium-exposed animals. The findings from this study indicate a low potential of L. littorea to bioaccumulate Cr6+ in its tissues at the low exposure concentrations tested in this study; as such, its consumption may not pose any serious health risks to humans. However, changes in the proximate composition and histological architecture of the exposed L. littorea show that Cr6+ is potentially toxic to periwinkles. Full article

Fungal elicitation could improve the secondary metabolite contents of in vitro cultures. Herein, we report the effect of Fusarium oxysporum on vinblastine and vincristine alkaloid yields in Catharanthus roseus embryos. The study revealed increased yields of vinblastine and vincristine in Catharanthus tissues. Different concentrations, i.e., 0.05% (T1), 0.15% (T2), 0.25% (T3), and 0.35% (T4), of an F. oxysporum extract were applied to a solid MS medium in addition to a control (T0). Embryogenic calli were formed from the hypocotyl explants of germinating seedlings, and the tissues were exposed to Fusarium extract elicitation. The administration of the F. oxysporum extract improved the growth of the callus biomass, which later differentiated into embryos, and the maximum induction of somatic embryos was noted T2 concentration (102.69/callus mass). A biochemical analysis revealed extra accumulations of sugar, protein, and proline in the fungus-elicitated cultivating tissues. The somatic embryos germinated into plantlets on full-strength MS medium supplemented with 2.24 µM of BA. The germination rate of the embryos and the shoot and root lengths of the embryos were high at low doses of the Fusarium treatment. The yields of vinblastine and vincristine were measured in different treated tissues via high-pressure thin-layer chromatography (HPTLC). The yield of vinblastine was high in mature (45-day old) embryos (1.229 µg g−1 dry weight), which were further enriched (1.267 µg g−1 dry weight) via the F. oxysporum-elicitated treatment, especially at the T2 concentration. Compared to vinblastine, the vincristine content was low, with a maximum of 0.307 µg g−1 dry weight following the addition of the F. oxysporum treatment. The highest and increased yields of vinblastine and vincristine, 7.88 and 15.50%, were noted in F. oxysporum-amended tissues. The maturated and germinating somatic embryos had high levels of SOD activity, and upon the addition of the fungal extracts, the enzyme’s activity was further elevated, indicating that the tissues experienced cellular stress which yielded increased levels of vinblastine and vincristine following the T2/T1 treatments. The improvement in the yields of these alkaloids could augment cancer healthcare treatments, making them easy, accessible, and inexpensive. Full article

Vinca sardoa (Stearn) Pignatti, known as Sardinian periwinkle, is widely diffused in Sardinia (Italy). This species contains indole alkaloids, which are known to have a great variety of biological activities. This study investigated the antileukemic activity against a B lymphoblast cell line (SUP-B15) of V. sardoa alkaloid-rich extracts obtained from plants grown in Italy, in Iglesias (Sardinia) and Rome (Latium). All the extracts showed a good capacity to induce reductions in cell proliferation of up to 50% at the tested concentrations (1–15 µg/mL). Moreover, none of the extracts showed cytotoxicity on normal cells at all the studied concentrations. Full article

The marine and ocean water pollution with different-sized plastic waste poses a real threat to the lives of the next generations. Plastic, including microplastics, is found in all types of water bodies and in the organisms that live in them. However, given the chemical diversity of plastic particles, data on their toxicity are currently incomplete. Moreover, it is clear that different organisms, depending on their habitat and feeding habits, are at different risks from plastic particles. Therefore, we performed a series of experiments on feeding the gastropod scraping mollusk Littorina brevicula with two types of polymeric particles—polymethylmethacrylate (PMMA) and polytetrafluoroethylene (PTFE)—using a special feeding design. In the PMMA-exposed group, changes in gastrointestinal biochemical parameters such as increases in malondialdehyde (MDA) and protein carbonyls (PC) were detected, indicating the initiation of oxidative stress. Similarly, a comet assay showed an almost twofold increase in DNA damage in digestive gland cells compared to the control group. In mollusks fed with PTFE-containing food, no similar changes were recorded. Full article

The use of biobased heterogeneous catalysts made from agricultural waste for producing biodiesel has gained attention for its potential to create a sustainable and low-cost process. The blending of two or more biomass residues to create more viable biobased catalysts is still in its early stages. In this study, a Biobased Composite Heterogeneous Catalyst (CHC) was made by blending the shells of periwinkle (PWS), melon seed-husk (MSH), and locust bean pod-husk (LBP) at a mixing ratio of 67:17:17 using Simplex Lattice Design Mixture, that was then calcined for 4 h at 800 °C. The chemical, structural, and morphological components of the CHC were characterized via XRF, XRD, SEM-EDX, BET, TGA/DSC, and FTIR to assess its catalytic potential. The CHC was employed to synthesize biodiesel from palm kernel oil, and the process optimization was conducted using the Taguchi approach. The XRF analysis showed that the catalyst had 69.049 of Calcium (Ca) and 9.472 of potassium (K) in their elemental and oxide states as 61.592% calcium oxide and 7.919% potassium oxide. This was also supported by the EDX result, that showed an appreciable value of 58.00% of Ca and 2.30% of magnesium, that perhaps provided the active site in the transesterification reaction to synthesize biodiesel. The morphological and physisorption isotherms via SEM and BET showed mesoporous structures in the CHC that were made up of nanoparticles. A high maximum biodiesel yield of 90.207 wt.% was attained under the optimized process conditions. The catalyst could be reused for up to four cycles, and the biodiesel produced met both ASTM D6751 and EN 14214 standards for biodiesel. This study demonstrates that blending PWS, MSH, and LBP waste materials can produce high-quality biodiesel without the need for additional catalysts. Full article

Global consumption of nonrenewable natural aggregate for construction activities is now becoming a significant concern. Reusing agricultural or marine-based wastes could offer a promising alternative to achieve natural aggregate conservation and a pollution-free environment. This study investigated the suitability of using crushed periwinkle shell (CPWS) as a reliable constituent material for sand and stone dust in producing hollow sandcrete blocks. The CPWS was used to partially substitute river sand and stone dust at 5, 10, 15 and 20% in sandcrete block mixes using a constant water–cement ratio (w/c) of 0.35. The weight, density and compressive strength of the hardened hollow sandcrete samples were determined after 28 days of curing along with the water absorption rate. Results showed an increase in the water absorbing rate of the sandcrete blocks as CPWS content increased. Mixes containing 5% and 10% CPWS substitute for sand with 100% stone dust surpassed the minimum targeted strength of 2.5 N/mm2. The compressive strength results suggested that CPWS is most suitable to be deployed as a partial substitute for sand as a constant stone dust material, thus imply that the construction industry can achieve sustainable construction with agro or marine-based wastes in hollow sandcrete production. We studied the reproductive behaviour associated with mate choice by size in three species of White Sea molluscs of the genus LittorinaL. saxatilis, L. obtusata and L. fabalis. Three behavioural patterns were revealed after a comparative analysis of the behaviour of the male snails. The males of L. obtusata, regardless of their size, could mate with all females but preferred to mate with larger ones. The males of L. fabalis chose females of their own size or larger. The males of L. saxatilis mated with females of approximately their own size. In the first case (L. obtusata), there is a free exchange of genes between all the size groups in the population. In the second case (L. fabalis), the gene transfer from small males to females of any size is free but the gene transfer from large males to small females is limited. In the third case (L. saxatilis), the gene flow is limited between groups of individuals of dramatically different size. Thus, in the case of L. saxatilis and L. fabalis, to varying degrees restriction of random mating associated with size assortativity in choosing a mating partner was expressed, which under certain circumstances (divergence of individuals in different econiches, the presence of ecotypes differing in size, lack of stronger natural selection in the other direction) can favour sympatric speciation. The males of all the three periwinkles’ species preferred larger females but sexual selection in favour of larger females was found only in L. obtusata. In the other two species, there were no statistically significant size differences between copulating and non-copulating females. This appears to be due to the occurrence of size-assortative mating in these species, which limits the ability of males to choose larger female partners. Full article

Microbial biofilms have co-evolved with grazing animals, such as gastropods, to develop mutually beneficial relationships. Although microbial biofilms demonstrate resilience and resistance to chemical exposure, pre-existing relationships can be negatively affected by chemical input. In this study, we determined how the grazer, Littorina littorea (common periwinkle sea snail), and a biological surfactant (rhamnolipid) interact on a phototrophic marine biofilm. Biofilms were cultured in 32 twenty-liter buckets at the Queen’s University Marine Laboratory in Portaferry, Northern Ireland on clay tiles that were either exposed to 150 ppm of a rhamnolipid solution or that had no chemical exposure. L. littorea were added into half of the buckets, and biofilms were developed over 14 days. Biofilms exposed to grazing alone demonstrated high tolerance to the disturbance, while those growing on rhamnolipid-exposed substrate demonstrated resistance but experienced slight declines in carbon and stoichiometric ratios. However, when exposed to both, biofilms had significant decreases in stoichiometry and declined in productivity and respiration. This is problematic, as continuing marine pollution increases the likelihood that biofilms will be exposed to combinations of stressors and disturbances. Loss of biofilm productivity within these areas could lead to the loss of an important food source and nutrient cycler within the marine ecosystem. Full article

The potential of using pre-stress NaCl or CaCl2 applications to confer a cross-tolerance to a water deficit was evaluated in periwinkle. The plants initially received five applications of NaCl (0, 30 and 50 mM), or CaCl2 (15 and 25 mM) via irrigation, and then they were cultivated under different water deficit regimes (80, 50 and 20% available water content). The water deficit induced smaller and denser stomata. It promoted a water use efficiency, a proline content and antioxidant enzyme activity. However, it downgraded the aesthetic value (plant stature, flower size and vegetation greenness), magnified the stem bending probability and strongly decreased the floral longevity. It additionally impeded the growth by reductions in the leaf area and photosynthesis. Plants undergoing a water deficit maintained a lower hydration and expressed oxidative damage symptoms, including enhanced chlorophyll and membrane degradation. As the water deficit intensified, these effects were more pronounced. Pre-stress CaCl2 or NaCl applications generally restored most of the water severity-induced effects, with the former being more effective. For CaCl2, the highest concentration (25 mM) was generally optimal, whereas NaCl was the lowest concentration (30 mM). In conclusion, pre-stress CaCl2 or NaCl applications effectively confer a cross-tolerance to the water deficit by promoting the aesthetic value and extending the floral longevity, with the promotive effects being incremental as the water deficit becomes more severe. In this study, the optimal concentration of sodium hydrosulfide (NaSH), salicylic acid (SA), and titanium dioxide nanoparticles (TiO2NPs), and their relative effectiveness on alleviating the adverse effects of water deficit on ornamental quality, were investigated in periwinkle. Plants were cultivated under three water deficit levels (80, 50, and 20% available water content) and received two foliar applications of TiO2NPs (0, 0.5 and 1 mM), NaSH (0.5 and 1 mM), or SA (1 and 2 mM). Water deficit deteriorated ornamental quality, amplified the risk of buckling (lower stem strength) and suppressed longevity. It decreased both light interception (leaf area) and carbon assimilation. Besides impaired hydration status, water-stressed plants underwent oxidative damage as indicated by reduced chlorophyll content, elevated membrane degradation, and lipid peroxidation. Spray treatments improved all traits, besides stem strength and proline content. Additionally, they enhanced carotenoid content and the activities of catalase and peroxidase. Their relative effectiveness (TiO2NPs > NaSH > SA) and optimal concentration (i.e., 0.5 mM (TiO2NPs, NaSH), and 1 mM (SA)) was independent of water deficit level. In conclusion, this study provides practices for improved ornamental quality and longevity independently of water availability, with their positive effect being stronger under irregular or limited water supply. Bois noir, an economically important disease of grapevine yellows that causes significant economic losses in wine production, is associated with ‘Candidatus Phytoplasma solani’ and transmitted to grapevines by cixiids Hyalesthes obsoletus and Reptalus panzeri. Polyphagous planthopper Dictyophara europaea, commonly found in natural habitats, harbors phytoplasmas from distinct groups and is an alternative vector in the open epidemiological cycles of the Flavescence dorée phytoplasma in grapevine in European vineyards. This study addresses the role of D. europaea in the transmission cycle(s) of ‘Ca. P. solani’ among wild habitats, natural reservoir plants, and the vineyard agroecosystem using MLSA and transmission trials with naturally infected adults to grapevine and Catharanthus roseus. The infection rates of D. europaea ranged from 7% to 13% in diverse locations, while reservoir herbaceous plants were infected in the amount of 29%. A total of 13 CaPsol MLSA genotypes were detected in D. europaea (7) and plants (8). Nine of them corresponded to previously identified genotypes. Two new genotypes were found in D. europaea (tuf-b1/S1/V14/Rqg50-sv1 and tuf-b1/S18/V14/Rqg50-sv1) and one in Convolvulus arvensis (tuf-b1/S1/V2-TA/Rqg31-sv1), whereas one was shared by two hosts, Crepis foetida and Daucus carota (tuf-b1/S1/V2-TA/STOL-sv1). Naturally infected D. europaea successfully transmitted the tuf-b1/S1/V2-TA/STOL type to five grapevines and six periwinkles, tuf-b1/S1/V2-TA/Rqg31 to one grapevine, and tuf-b1/S1/V2-TA/Rqg50 to one periwinkle, indicating that D. europaea is an intermediate vector in CaPsol epidemiological cycles.
