Behavioral issues in domestic dogs represent a significant welfare concern affecting both canines and their caregivers, with prevalence rates reported to range from 34 to 86% across the population. Current treatment options, including selective serotonin reuptake inhibitors (SSRIs) like fluoxetine, often present limitations including adverse effects and delayed efficacy. This randomized, placebo-controlled (maltodextrin) study investigated the effects of a novel Lactiplantibacillus plantarum strain (LP815TM) on canine behavioral concerns through gut–brain axis modulation. Home-based dogs (n = 40) received either LP815TM (n = 28) or placebo (n = 12) daily for 4 weeks, with behavioral changes assessed using the comprehensive Canine Behavioral Assessment & Research Questionnaire (C-BARQ) and continuous activity monitoring. After the intervention period, dogs receiving LP815TM showed significant improvements in aggression (p = 0.0047) and anxiety (p = 0.0005) compared to placebo controls. These findings were corroborated by objective activity data, which demonstrated faster post-departure settling, reduced daytime sleep, and improved sleep consistency in the treatment group. Throughout >1120 administered doses, no significant adverse events were reported, contrasting favorably with pharmaceutical alternatives. The concordance between our findings and previous research using different L. plantarum strains suggests a consistent biological mechanism, potentially involving GABA production and vagal nerve stimulation. These results indicate that LP815TM represents a promising, safe alternative for addressing common canine behavioral concerns with potential implications for improving both canine welfare and the human–animal bond. Most research studies have investigated the impact of pet ownership on the mental and physical well-being of elderly populations, supporting the beneficial effect that pets have on their owners. However, few researchers focused on the well-being of both owner and pet. The present study aimed to explore the well-being of pets owned by elderly individuals using an owner assessment tool and the relationship between elderly characteristics and the pet’s health-related quality of life based on the owner’s assessment of their pet’s well-being. Sixty elderly pet owners who made regular visits to veterinary clinics were selected to complete an electronic questionnaire about their pet’s health-related quality of life. The results identified a high agreement percentage on positive indicators related to the pet’s well-being such as “My pet wants to play and My pet responds to my presence” in the happiness domain, “My pet has more good days than bad days” in mental status, “My pet moves normally” in physical status and “My pet keeps him/herself clean” in hygiene which also resulted in a positive relationship with elderly age. Marital status influenced their responses to “My pet responds to my presence and My pet is as active as he/she has been”. The results also support the use of the applied questionnaire to help identify variables that contribute to a pet’s health-related quality of life. The correlation matrix revealed statistically significant positive associations (p < 0.001) among positively phrased items across all domains, as well as among negatively phrased items. These consistent alignments between direct and between reversed items suggest directional coherence and help mitigate potential response bias. Furthermore, the replication of these patterns across multiple domains reinforces the interpretation that the instrument captures a unified construct of pet well-being, In conclusion, based on subjective evaluation of pet-owner relationships, the ownership of pets by elderly individuals could be mutually beneficial to both elderly owners and their pets. Research on human–animal relationships suggests that close bonds with animals can enhance empathy, reduce speciesism, and improve human physical and psychological health. This study investigated whether pet ownership—particularly attachment to a companion animal during childhood—is associated with differences in moral concerns toward all animals in adulthood. It also aimed to explore the potential effects of empathy and speciesism on overall moral concerns toward animals. Using self-report questionnaires among 72 participants recruited online, the analyses revealed a significant effect of animal categories on moral concerns, F(1, 1.98) = 59.37, p < 0.001. Mean moral concern scores were significantly higher for companion animals (M = 6.04, SD = 1.15) than for food animals (M = 4.90, SD = 1.44), unappealing wild animals (M = 4.20, SD = 1.87), and appealing wild animals (M = 5.73, SD = 1.32), p < 0.05. Additionally, childhood pet owners reported greater moral concerns for all animals, F(1, 1.98) = 4.87, η2 = 0.065, p < 0.05. Attachment to a companion animal in childhood was positively correlated with moral concerns for all animal categories. Finally, although attachment and empathy were both positively related to moral concern, only attachment was a significant predictor (p < 0.05). Further research is needed to understand the psychological mechanisms influencing views on animal rights and welfare. Full article

Salmonella is a rapidly spreading and widespread zoonotic infectious disease that poses a serious threat to the safety of both poultry and human lives. Therefore, the timely detection of Salmonella in foods and animals has become an urgent need for food safety. This work describes the construction of an aptamer-based sensor for Salmonella detection, using Fe3O4 magnetic beads and Ag@Au core–shell nanoparticles-embedded 4-mercaptobenzoic acid (4MBA). Leveraging the high affinity between biotin and streptavidin, aptamers were conjugated to Fe3O4 magnetic beads. These beads were then combined with Ag@4MBA@Au nanoparticles functionalized with complementary aptamers through hydrogen bonding and π-π stacking interactions, yielding a SERS-based aptamer sensor with optimized Raman signals from 4MBA. When target bacteria are present, aptamer-conjugated magnetic beads exhibit preferential binding to the bacteria, leading to a decrease in the surface-enhanced Raman scattering (SERS) signal. And it was used for the detection of five different serotypes of Salmonella, respectively, and the results showed that the aptamer sensor exhibited a good linear relationship between the concentration range of 102–108 CFU/mL and LOD is 35.51 CFU/mL. The SERS aptasensor was utilized for the detection of spiked authentic samples with recoveries between 94.0 and 100.4%, which proved the usability of the method and helped to achieve food safety detection. Most studies on relationships between humans and nonhuman animals focus on the benefits of the relationship to humans, the potential detriment or stress to animals, or how humans can better improve husbandry or handling practices in the domestic setting. By comparing existing research in human working relationships and friendships with animal friendships and studies on human–animal interactions, this paper proposes a new framework of an Interspecies Relational Theory that provides approaches for identifying, developing, and maintaining different levels of relationships between humans and nonhuman species. The framework is broken into three stages aligned with existing research in human levels of trust ranging from strategic/calculus-based to working/knowledge/information-based to friendship/affect-based. These levels are also aligned with stages of Maslow’s Hierarchy. The suggested framework can help provide insight into both human and animal perspectives of the human–animal relationship in a variety of veterinary and animal-assisted settings. Full article

Brachycephalic dogs remain highly popular despite the well-documented health problems associated with their conformation. The aim of this study was to investigate differences in the bonding between humans and dogs, behavioral characteristics, awareness of clinical breed-related ailments, and purchasing intentions among Italian brachycephalic dog owners (BDOs) and non-brachycephalic dog owners (NBDOs). A questionnaire, including the DORS and C-BARQ scales, was administered to 320 BDOs and 408 NBDOs. Despite the occurrence of breed-related disorders, 81% of BDOs rated their dog’s health status as good to excellent. Most BDOs were aware of the prevalence of these disorders in the broader brachycephalic dog population. Character and appearance were the main motivations for choosing these breeds. Owners’ satisfaction with specific aspects of ownership, such as veterinary costs and behavior, generally met expectations. Mean scores of perceived emotional closeness (PEC) and dog–owner interaction (DOI) were significantly higher among BDOs compared to NBDOs, while the perceived cost (PC) mean score was higher among NBDOs. The C-BARQ scores for trainability, excitability, and separation-related behaviors were significantly higher in BDOs than in NBDOs. Despite the growing awareness of health issues, the popularity of brachycephalic breeds persists. Scientific evidence may support a revision of brachycephalic breed standards, prioritizing animal welfare while preserving desirable aesthetic traits, promoting more ethical breeding. Full article

Managing relinquished/stray dogs is a significant socio-economic and ethical challenge necessitating structured policies to ensure animal welfare and integration into society. Critical to successful rehoming is careful assessment of a dog’s temperament/behavior and appropriate matching of a dog to an owner. Shelter dogs’ involvement in canine-assisted interventions (CAIs) provides a potential avenue for rehoming through appropriate dog educational programs. CAIs consist of structured interactions between dog and clients targeted at specific groups with physical/psychological/emotional impairments in various settings. However, there is a lack of standardized protocols for assessing shelter dogs’ suitability for CAI, raising ethical concerns regarding selection/training practices. This study explores the ethical dimensions of rehoming shelter dogs for CAI, employing a philosophical framework considering the responsibilities of humans/veterinarians toward companion animals and how societal perceptions of human–animal relationships impact decision-making in this context. An ethical matrix is applied to evaluate the implications of rehoming decisions on canine and human interests. Our findings highlight the need for a balanced, responsible approach that respects the individuality of dogs while promoting their potential roles in human society. Ethical guidelines for the selection and training of shelter dogs for CAI could enhance their welfare, improve adoption rates, and reinforce the value of the human–animal bond. Full article

Dog behaviour that owners perceive as undesirable can compromise dog welfare, impact the owner and human–animal bond, and may result in relinquishment or euthanasia. This longitudinal study explored the type and percentage of owner-perceived undesirable behaviours in 6, 9, 12, 15 and 18-month-old dogs, and examined differences in the prevalence of four commonly reported behaviours across these timepoints. Owners reported their dog’s undesirable behaviours via free-text answers in self-administered surveys. Of the five timepoints studied, the highest percentage of dogs reported by their owners to display one or more undesirable behaviours was in the 12-month survey (42.1%, 513/1219). Barking, jumping up, pulling on the lead and recall issues were the most commonly reported behaviours across all timepoints. Binomial mixed-effects models revealed a significant difference in prevalence of barking, pulling on the lead and recall issues between the timepoints, but no significant difference in the occurrence of jumping up at people. With many dogs in this study being reported to display undesirable behaviour, it is crucial that stakeholders inform dog owners, especially first-time owners, about the behaviours they may experience from young dogs and where to seek appropriate training/behaviour advice to potentially reduce the risk of relinquishment related to behaviour. Circular RNA is a type of noncoding RNA with a special covalent bond structure. As an endogenous RNA in animals and plants, it is formed through RNA splicing. The 5′ and 3′ ends of the exons form circular RNA at the back-splicing sites. Circular RNA plays an important regulatory role in diseases by interacting with the associated miRNAs. Accurate identification of circular RNA can enrich the data on circular RNA and provide new ideas for drug development. At present, mainstream circular RNA recognition algorithms are divided into two categories: those based on RNA sequence position information and those based on RNA sequence biometric information. Herein, we propose a method for the recognition of circular RNA, called circ2LO, which utilizes the LucaOne large model for feature embedding of the splicing sites of RNA sequences as well as their upstream and downstream sequences to prevent semantic information loss caused by the traditional one-hot encoding method. Subsequently, it employs a convolutional layer to extract features and a self-attention mechanism to extract interactive features to accurately capture the core features of the circular RNA at the splicing sites. Finally, it uses a fully connected layer to identify circular RNA. The accuracy of circ2LO on the human dataset reached 95.47%, which is higher than the values shown by existing methods. It also achieved accuracies of 97.04% and 72.04% on the Arabidopsis and mouse datasets, respectively, demonstrating good robustness. Through rigorous validation, the circ2LO model has proven its high-precision identification capability for circular RNAs, marking it as a potentially transformative analytical platform in the circRNA research field. Traditional methods for managing free-roaming cats in Australia primarily depend on legislation and enforcement to achieve compliance. State laws and local regulations mandate confinement, sterilization, registration, and identification and limit the number of cats kept, with penalties for breaches. However, these strategies fail to address underlying issues like financial constraints in low-income areas and the prevalence of semi-owned cats. Containment mandates often result in increased complaints, shelter intake, and euthanasia, without effectively reducing cat-related problems. Research shows that these approaches are expensive, difficult to enforce, and place a disproportionate burden on disadvantaged communities. Moreover, they negatively affect the mental health of shelter staff and animal management officers, who are frequently exposed to euthanasia and ongoing challenges. An alternative “One Welfare” framework, which recognizes the interconnectedness of animal, human, and environmental welfare, has proven more effective. Programs that provide support and resources, particularly for cat sterilization and microchipping, while fostering the human–animal bond improve outcomes for both cats and caregivers. Shifting from punitive measures to collaborative, community-driven strategies is crucial for managing free-roaming cats in a way that benefits animals, people, and the broader community, while protecting wildlife. Methicillin-resistant Staphylococcus epidermidis (MRSE) contamination is commonly found on human skin and medical devices. Herein, we present a sensor utilizing molecularly imprinted polymer nanobodies (nanoMIP) for recognition and electrochemical impedance spectroscopy (EIS) to detect S. epidermidis. Sensor manufacturing involves synthesizing nanoMIP via solid-phase synthesis using whole bacteria as templates. Screen-printed gold electrode (AuSPE)-modified 16-mercaptohexadecanoic acid (MHDA) served to immobilize the nanoMIPs on the sensor surface through an amide bond, with the remaining functional groups blocked by ethanolamine (ETA). Scanning electron microscope (SEM) analysis of the modified AuSPE surface reveals immobilized spherical nanoMIP particles of 114–120 nm diameter, while atomic force microscope (AFM) analysis showed increased roughness and height compared to bare AuSPE. The sensor is selective for S. epidermidis, with a remarkable detection limit of 1 CFU/mL. This research demonstrates that the developed nanoMIP-based sensor effectively detects S. epidermidis. Further research will focus on developing protocols to integrate the nanoMIP-based EIS sensor into medical and industrial applications, ultimately contributing to improved safety for both humans and animals in the future. Metarhizium fungi, essential for ecosystem function and commonly utilised in pest control, often occupy ecological niches contaminated by toxic compounds of both anthropogenic and microbiological origin. The present study reveals the potential of Metarhizium anisopliae for biodegradation of the Fusarium mycotoxin zearalenone (ZEN), a common contaminant of crops that poses a significant threat to human and animal health due to its oestrogenic potential and toxicity. A key aspect of the pathway described is the degradation of ZEN by cleaving the lactone bond, which results in a significant reduction in mycotoxin toxicity, highlighting the fungus’s bioremediation potential. Furthermore, this study provides the first evidence of subsequent degradation of ZEN metabolites through progressive shortening of the aliphatic chain, primarily via alternating oxidation and demethylation, ultimately yielding trihydroxybenzene. Significantly, lactone bond cleavage occurred not only in ZEN itself but also in its reduced forms, the zearalanols, formed through the initial reduction of ZEN to zearalenols. Elevated mRNA levels of cytochrome P450 (CYP450) monooxygenases in M. anisopliae exposed to ZEN indicate their significant involvement in degradation mechanisms. Intriguingly, the inhibition of CYP450 activity resulted in a substantial shift in the quantitative ratio of α- and β-epimers of zearalenols and zearalanols. The observed alteration towards β-form production likely stems from the inhibition of other CYP450-dependent reactions, indirectly influencing ZEN reduction pathways—a particularly noteworthy finding. These insights are crucial for developing strategies to utilise M. anisopliae in the bioremediation of ZEN-contaminated areas. Full article

Human activities significantly contribute to the biodiversity crisis, yet wildlife tourism can promote appreciation for animals and encourage conservation. This study assessed the impact of diving and snorkeling programs at Atlantis Dubai on tourists’ emotional connections with marine life and their conservation attitudes and pro-environmental behavioral intentions. In 2023 and 2024, 346 participants were surveyed before (n = 172) and after (n = 174) their experiences diving and snorkeling with elasmobranchs. Results indicated a notable increase in positive feelings toward sharks and rays, strengthening connections and respect for these creatures. The program positively influenced attitude toward sharks, making participants more inclined to support their protection and foster a deeper connection with the ocean. Although participants’ willingness to recycle improved, overall conservation attitudes and willingness to engage in broader pro-environmental behaviors showed minor change. However, the heightened positive emotions towards sharks resulted in improved perceptions of these animals, stronger conservation attitudes, and a commitment to environmental sustainability. A deeper connection with the ocean reduced fear of sharks. This research suggests that wildlife tourism can facilitate emotional connections with marine life, potentially inspiring conservation action and highlighting the need for targeted strategies to transform these emotional bonds into lasting pro-environmental behaviors. Insight into the Role of the Aryl Hydrocarbon Receptor in Bovine Coronavirus Infection by an Integrated Approach Combining In Vitro and In Silico Methods

It is well known that the host response to different human and animal coronaviruses infection is regulated by the aryl hydrocarbon receptor, a ligand-activated transcription factor. The present study investigates the expression of the aryl hydrocarbon receptor during bovine coronavirus infection, through in vitro and in silico investigations. The in vitro studies demonstrate that the aryl hydrocarbon receptor and as well as its targets, CYP1A1 and CYP1B1, were significantly activated by bovine coronavirus infection in bovine cells (MDBK). During infection, the pretreatment of cells with non-cytotoxic doses of CH223191, a selective inhibitor of the aryl hydrocarbon receptor, resulted in a significant reduction in virus yield and a downregulation in the viral spike protein expression. These findings occurred in the presence of the inhibition of aryl hydrocarbon receptor signaling. Our results reveal that the bovine coronavirus acts on viral replication, upregulating the aryl hydrocarbon receptor and its downstream target proteins, CYP1A1 and CYP1B1. In addition, following the in silico studies, the three-dimensional structural model of the bovine aryl hydrocarbon receptor in complex with the antagonist CH223191 indicates that the molecular mechanism, by which the PASB and TAD domains of the receptor interact with the inhibitor, is mainly driven by an extensive network of hydrophobic interactions, with a series of hydrogen bonds contributing to stabilizing the complex. Interestingly, bioinformatic analyses revealed that the PASB and TAD domains in the human and bovine aryl hydrocarbon receptor present high similarity at the primary sequence and three-dimensional structure levels. Taken together, these findings represent a fundamental step for the development of innovative drugs targeting AhR as a potential object for CoVs therapy. Full article
